1.Interleukin-4 decreases expression of epithelial sodium channel ?-subunit in A549 cells

Objective To observe the effects of IL-4 on the expression of epithelial sodium channel(ENaC) ?-subunit.Methods A549 cells were cultured with IL-4 at the concentration of 0,0.1,1 and 10 ng/ml for 12 h or with 1 ng/ml IL-4 for 3,6,12,24 and 48 h.Then ?-ENaC protein and mRNA expressions were detected by Western blotting and RT-PCR respectively.Results IL-4 decreased the expression of ?-ENaC in A549 cells in a time-and dose-dependent manner,via gene transformation.When A549 cells were cultured with IL-4 at the concentration of 1 ng/ml for 3,6,12,24 and 48 h,the expression of ?-ENaC protein and mRNA were decreased in a time-dependent manner.Statistical analysis showed culture for 6 h had a significant difference compared with other time points(P

Objective To study the juxtaglomerular cell tumor. Methods The clinical characteristics,diagnosis,surgical teratment and the prognosis of 5 patients were retrospectively studied with review of the literature. Results All the patients had hypokalemia.Peripheral serum renin levels were recorded in 4 patients and 3 of them had high levels of peripheral serum renin activity and hyperaldosteronism while these were normal in the other 1.1 patient had been preoperatively diagnosed as a right adrenal tumor with a concurrent right kidney tumor while the post-operative histopathological studied revealed normal adrenal tissue and a juxtaglomerular cell tumor of the right kidney.All patients underwent simple tumor resection.The peripheral serum renin activity,hyperaldosteronism,hypokalemia,and hypertension became normal after the tumor resection.At a mean follow-up of 58 months (range 15 to 120 months ) no tumor recurrence and hypertension has been documented. Conclusions Juxtaglomerular cell tumor is a begin tumor of kidney,being a rare, curable cause of high levels of peripheral serum renin activity,hyperaldosteronism, hypokalemia and hypertension.Differential diagnosis includes primary hyperaldosteronism and renal artery stenosis.The prognosis is good with tumor resection.
3.Captopril suppresses lectin-like oxidized LDL receptor-1 protein in rats after acute lung injury

Objective To study the expression of lectin-like oxidized LDL receptor-1 (LOX-1) protein in acute lung injury (ALI) and the effect of captopril on this expression in order to investigate its role in the process and preliminary intervention.Methods Sprague-Dawley rats were randomly divided into 3 groups,normal control group,lipopolysaccharide (LPS) groups,and a LPS+captopril group,each group 10 rats.ALI model was induced by intratracheal injection of LPS (5 mg/kg),then those of LPS+captopril group were give an intraperitoneal injection of captopril (1.25 mg/kg).The animals of normal control group received an intratracheal injection of normal saline.In 6 h after LPS injection,the level of p(O2),wet/dry ratio (W/D),concentration of total protein in bronchoalveolar lavage fluid (BALF) and lung tissue histopathological changes were examined.The expression of LOX-1 protein in the lung tissue was measured by Western blot analysis.HE staining was used to examine the pathological changes of the lung.Results Histological examination showed that extensive lung inflammation were seen in the LPS group,which manifested by accumulation of significant numbers of neutrophils.The level of p(O2) in LPS group [(6.86?0.75) kPa] was decreased compared with that in sham group [(12.14?0.60) kPa,P
4.Experimented gene therapy for prostate cancer with HSV-TK/GCV system

Purpose: To investigate the effect of HSV-TK/GCV system on human prostate cancer. Methods: MTTassay, flow cytometry( FCM), optical and electron microscopy were used to determine the sensitivity of GCV on prostate cancer cell( PC-3m) after being tranfected with HSV-TK gene. Results: It was found that GCV had significant cytotoxic activity on HSV-TK gene-transduced prostate cancer cells, but little effect on the non-transduced cells. Conclusions: The experiment indicates that HSV-TK/GCV system has significant antitumor activities on the TK gene-transduced prostate cancer cells in vitro.
5.Preliminary study on peroxisome proliferator activated receptor gamma of lymphocytes in patients with asthma

Objective To explore the peroxisome proliferator activated receptor gamma (PPARγ) expression characteristic of lymphocytes in patients with asthma .Methods Collected blood samples from healthy subjects (health control group) and asthmatic patients(asthmatic group) before treatment ,2 and 4 days after treatment .Expression levels of PPARγtested with Q-PCR .Analyzed eosinophil percentage of induced sputum ,IL-5 concentration in sputum supernatant measured with ELISA kits .Results Compared with healthy control group ,the eosinophil percentage and IL-5 concentration were higher in asthmatic group before treatment ;meanwhile the expression level of PPARγwas at the lowest .After treatment ,PPARγgradually increased accompanied with eosino-phil percentage and IL-5 concentration gradually decreased .Conclusion Asthmatic patients had a lowest PPARγ expression level . Their recovery perhaps attributed to the up-regulation of PPARγin lymphocytes .The anti-inflammatory effect of PPARγachieved might be via inhibiting the function of Th2 cells .
6.Update on perivascular epithelioid cell tumors of the urinary system

The perivascular epithelioid cell (PEC) is a cell type constantly present in a group of tumors called PEComas(perivascular epithelioid cell tumors). PEC expresses myogenic and melanocytic markers,such as HMB45. PEComa is a widely accepted entity now. PEComas are related to the genetic alterations of tuberous sclerosis complex (TSC), an autosomal dominant genetic disease due to losses of TSC1 or TSC2 genes. PEComas are rare in the urinary system and there are some open questions about PEComas regarding its histogenesis, the definition of epithelioid angiomyolipoma and the identification of the histological criteria of malignancy. This review provides an update on PEComas of the urinary system.

Objective To study the in vitro effects of sonodynamic management on human ovarian cancer cell line HRA. Methods Hematoporphyrin was selected as a sonosensitizer and ultrasound with certain intensity was used to activate hematoporphyrin.Then MTT assay and clony-forming efficiency assay were applied to determine the growth inhibitory effects of ovarian cancer cells. Electron microscope was applied to detect the morphological changes of the cells. Cell apoptosis was analyzed using flow cytometry. 8.Treatment on osteopenia of maintenance hemodialysis patients with salmon calcitonin

Objective To study the effect of long-term salmon calcitonin on bone mineral density (BMD), bone metabolism biochemical indicators and subjective score of bone pain in maintenance hemedialysis (MHD) patients with osteopenia. Methods Thirty-four MHD patients diagnosed as osteopenia by dual-energy X-ray absorptiometry (DXEA) were enrolled in this study. All the patients were treated with hypodermic injection of salmon calcitonin (50 U, thrice a week) for 12 months. The detecting parameters were as follows: BMD with DEXA in lumbar spine (L1-L4), femoral neck, troch, inter, and Ward's triangle before and after the study;serum bone metabolism biochemical indicators before and 6 and 12 months after the study;subjective scores of bone pain before and 1, 6, and 12 months after the study. Results Thirty-two patients were followed-up successfully. As compared to BMD parameters before study, the total T-score (-1.98± 2.20 vs 1.26±1.88, P=0.009) and total Z-score (-0.90±2.15 vs 0.08±2.05, P=0.002) of lumbar spine, the total T-score (-1.72±1.53 vs 1.06±1.58, P=0.016) and totle Z-score (-0.66±0.80 vs 0.08±1.08, P=0.029) of hip, the T-score of L3 (-2.02±2.51 vs 1.24±2.02, P=0.033), the Z-score of L2 (-0.44±1.82 vs 0.06±1.63, P=0.016), the Z-score of femoral troch (-0.65±1.11 vs 0.48±1.12, P=0.034) and the Z-score of inter (-0.58±0.94 vs 0.02±1.12, P=0.006) were increased significantly after study. But there were no significant differences in other examined regions and serum biochemical parameters. The subjective scores of bone pain were decreased rapidly for 41.7% after 1 month (P<0.01) and 76.6% after 6 months (P<0.01). The subjective score of bone pain after 12 months was similar to 6 months. The side effects of salmon calcitonin included nausea and vomitting in 5 cases (14.71%, 5/34), dizziness, blushing and flustered in 1 case respectively (3.13%,1/32). Conclusions Long-term hypodermic injection of salmon calcitonin can improve BMD and bone pain for MHD patients with osteopenia but has no significant effect on serum bone metabolism biochemical indicators. Salmon calcitonin is safe for MHD patients with seldom side effects, such as nausea and vomitting.
9.Effects of continuous quality improvement on dietary and nutrition management in patients with chronic kidney disease

Objective To evaluate the effects of continuous quality improvement (CQI) management on nutritional status,renal function progression,and compliance of low protein diet in patients with chronic kidney disease (CKD).Methods Totally 115 CKD patients who were regularly followed up in CKD clinic services were recruited in this study.Plan,Do,Check,and Act (PDCA) method was adopted to manage the dietary of these patients for 12 months.The clinical indicators and diet compliance before and after receiving CQI management were compared.Results After receiving the CQI management,the nutritional status of patients was well maintained;meanwhile,the average hand strength and the hemoglobin,serum albumin,total cholesterol,and triglyceride levels showed no significant changes (all P > 0.05).Subjective feelings of patients were improved.The modified Subjective Global Assessment of Nutrition (mSGA) score was decreased from 7.0 (7.0,8.0) to 7.0 (7.0,7.0) (P =0.000).The estimated glomerular filtration rate (eGFR) calculated by formula of modified MDRD was decreased from (40.74 ± 14.49) to (37.94 ± 16.86) ml/(min · 1.73 m2) (P =0.000),and the average descended speed was (2.81 ±7.42) ml/(min · 1.73 m2) per year.The creatinine clearance rate had no statistical difference between pre-and post management (P =0.910),and the average descended speed was (0.19 ± 17.01) ml/min per year.The daily protein intake (DPI) and protein equivalent of nitrogen appearance rate (PNA) were both significandy descended:DPI/kg decreased from (0.79 ± 0.27) to (0.64 ± 0.15) g/ (24 h · kg) (P =0.000),and PNA/kg dropped from (1.02 ± 0.32) to (0.82 ± 0.24) g/ (24 h · kg) (P =0.000).The scores of awareness and compliance of patients on low protein diet were significantly increased after CQI management (P =0.000).Conculsion Applying CQI on dietary and nutrition management in CKD patients can maintain the good nutritional status and improve the compliance of low protein diet.
10.The study of mouse lymphoma cell EL4 transfected with mB7-2 gene

Objective:To assess the potential of mouse immune-costimulating signal B7-2 in inducing immune effect.Methods:(1)The specific mB7-2cDNA fragment from LPS-stimulated mouse splenocytes was obtained by reverse transcription and polymerase chain reaction.The obtained fragment was inserted to pLXSN plasmid.Then the plasmid pLXSN-mB7-2 was packaged into PA317 cells;(2)The EL4/mB7-2 was obtained by infecting the EL4 by the concentrated virus particles produced by PA317/mB7-2;(3)In vitro,the secretion of IL-2 of EL4/mB7-2 stimulated-lymphocytes was detected by using mixed lymphocytes culture.Results:pLXSN-mB7-2 and transgenetic EL4/mB7-2 cells were obained successfully.IL-2 production in 24h in the supernatant stimulated by EL4/mB7-2 was much higher than wild EL4 cells.Conclusion:EL4/mB7-2 can activated T cell to produce IL-2.This research lay foundation for the research of the function of immune-costimulating signal in the tumor immunity and treatment,the mechanism of autoimmune disease and organ transplantation.
