Objective To observe the change of serum fibrinogen(FIB) ,procalcitonin(PCT) and hypersensitve C reactive protein (hs-CRP) in children with infectious pneumonia and its clinical significance .Methods One hundred and four cases of children with bacterial infectious pneumonia (bacterial group) ,109 cases with non-bacterial infectious pneumonia(non-bacterial group) and 110 healthy children under going physical examination (control group) were included into this study ,the serum FIB ,PCT and hs-CRP level were observed in each group .Results The FIB ,PCT and hs-CRP levels in the bacterial group were higher than those in the non-bacterial group and control group (P<0 .05);the FIB ,PCT and hs-CRP had no statistical difference among the virus group , mycoplasma group and chlamydia group had no atoctistical(P>0 .05) .The positive rate of combined detection of FIB ,PCT and hs-CRP in the non-bacterial group and bacterial group was significantly higher than that of single index detection (P<0 .05) .The FIB level in the bacterial group was positively correlated with PCT and hs-CRP(r=0 .445 ,0 .396 ,P<0 .05) .PCT and hs-CRP showed a positive correlation (r=0 .487 ,P=0 .021) .Conclusion The combined detection of FIB ,PCT and hs-CRP can effectively identify bacterial infection and non-bacterial infection and which has an important clinical significance for the diagnosis and treatment of pathogenic infection of pneumonia .

Objective To prepare rabbit anti-human RBBP10 polyclonal antibody and then character them.Methods New Zealand rabbit was immunized with RBBP10 protein,Polyclonal antiserum was analyzed by ELISA and Western blot.Results The titer of the antiserum obtained in this experiment determined by ELISA was up to 1∶16000 and the sensitivity was at least 10?g/?l.Western blot results show the antibody has high specificity.Conclusions Preparation the Rabbit anti-human RBBP10 Polyclonal antibody in this method is successfully,The expression of the RBBP10 protein in tumor can be detected in immunohistochemistry method,it is useful.

Objective To study the effects of cinobufacin(cino) combined with fluorouracil(5-FU) on inhibiting proliferation and inducing apoptosis of human gastric carcinoma cells in vitro. Methods The experiment was divided into control group,cinobufacin group,5-FU group and cino+5-FU group. Cell morphological variation,cell inhibitory rate, cell cycle and ratio of apoptotic cell of human gastric carcinoma cell line BGC-823 were studied by cell culture, inverse microscopy, fluoroscopy, MTT assay and flow cytometry on different concentrations of cino and 5-FU. AIM:To investigate the protective effects of sodium ferulate(SF)on apoptosis in cultured hippocampal neurons induced by sodium nitroprusside(SNP),and the effect of SF on expression of bcl-2 and bax.METHODS:The primary cultured hippocampal neurons were exposed to 50 ?mol SNP,a nitric oxide-donor,for 24 h after pretreatment with different concentrations of SF(10-160 ?mol/mL)for 6 h.Then neuronal viability was tested by MTT assay.Fluorescent staining with Hoechst 33258 and agarose gel electrophoresis was used to analyze apoptosis.The expressions of bcl-2,bax mRNA and protein were tested by RT-PCR and Western blotting.RESULTS:Pretreatment with SF(10-160 ?mol/L)for 6 h increased the survival rate of neurons.SF prevented the neuronal nuclei from shrinkage,condensation and cleavage and blocked neuronal nuclear DNA fragmentation induced by SNP.SF also increased the expressions of bcl-2 mRNA and Bcl-2 protein and decreased the expressions of bax mRNA and Bax protein.CONCLUSION:SF prevents the cultured hippocampal neurons against SNP neurotoxicity.The mechanism of protection is related to the increase in Bcl-2 level and the decrease in Bax level.As a result,the ratio of Bcl-2/Bax is changed.

Objective To establish a simple ,stable acute liver injury model induced by CCl4 to observe effects of hepatocyte transplantation .Methods CCl4 plant oil with different concentration of 20% and 50% was used in mice by intraperitoneal injection , of which the dose was 2 mL/kg ,and then materials were taken at different time points respectively .Mice survival rate ,alanine amin-otransferase (ALT) ,aspartate aminotransferase (AST) and the pathological changes of the liver were detected .Results Mice sur-vival rate in 20% CCl4 intraperitoneal injection was significantly higher than that of 50% .ALT and AST in experiment group were significantly higher than that of control group ,but there was no significant difference between two experimental groups .Pathologi-cal examination showed that mice liver cells showed typical cytoplasmic ,ballooning ,scattered punctate ,piecemeal necrosis and in-flammatory cell infiltration in 20% CCl4 intraperitoneal injection ;while in 50% CCl4 ,there was obvious fibrosis ,in addition to the mentioned heavier lesions .Conclusion 20% -50% CCl4 intraperitoneal injection in 2 mL/kg dose can induce different degrees of relatively stable liver injury ,and its concentration determines the degree of liver injury .Acute liver injury induced by 20% -50%CCl4 was an ideal model for hepatocyte transplantation experiment .

Objective To study the expression of Stat5,E-cadherin and Vimentin in thyroid carcinoma and the correlation among them. Method The expression of Stat5,E-cadherin and Vimentin in 149 cases of thyroid specimens(including 23 cases of nodular goiter and 126 cases of thyroid carcinoma)was detected by immunohisto-chemistry SP method and the correlation analysis was conducted with the main clinical pathological parameters. Results The positive rate of Stat5 and Vimentin in thyroid carcinoma was 85.71% and 77.78% respectively, which was significantly higher than that in nodular goiter(26.09%and 8.70%)(P<0.05)and the positive rate of E-cadherin in thyroid carcinoma was 25.40%,lower than that in nodular goiter(86.96%)(P<0.05). Three kinds of protein expression were not significantly correlated with gender and age (P > 0.05),but obviously with lymph node metastasis and pathological type(P<0.05). Stat5 was negatively correlated with the expression of E-cadherin (r=-0.335,P=0.000)but positively with the expression of Vimentin(r=0.218,P<0.05). Conclusion The high expression of Stat5 and EMT in thyroid carcinoma tissues indicate that Stat5 may be involved in the EMT process of thyroid carcinoma ,thus it can promote the invasion and metastasis of thyroid carcinoma.
