1.Proliferation of antigen specific T cells in vitro and analysis of phenotype and function

Objective To explore a new peptide-based approach independent of HLA to generate antigen-specific CD+ CD8+T cells. Methods Peripheral blood mononuclear cells(PBMC) were stimula- ted for 6 h with IE-1 peptide pool. Then the activated IFN-γsecreting ceils were tested by immunomagnetic selection. And the selected cells were cultured with radio-inactivated PBMC in medium with 100 IU/ml IL-2 for 4 weeks. Results The generated T cell lines consisted of IE-1 specific CD4+ T (6.88%) and CD8+ T cells 92.99%, which demonstrated antigen-specific killing and cytokine secretion. Conclusion T ceils can be proliferated with this new procedure, and maintain its phenotype and antigen-specific function.
2.Immunogenecity of cytomegalovirus-infected fibroblasts

AIM: To analyze the capability of cytomegaIovirus (CMV)-infected human embryonic lung fibroblasts (HELFs) to induce immune response. METHODS: HELFs were infected with cytomegalovirus and stained with antibody against HLA-A2 molecular, the expression of HLA-A2 was detected by FCM. The infected HELFs were incubated with individual pp65 peptide NLVPMVATV. While the uninfected and unloaded infected HELFs served as control respectively. After PBMC was added to the differently treated HELFs and incubated, the immune response was measured with IFN-γ release as readout. RESULTS: The expression of HLA-A molecular on infected fibroblasts diminished markedly compared with that on the uninfected. The peptides expressed on the infected HELFs together with those pulsed externally induced a stronger response than the infected HELFs alone. CONCLUSION: Although CMV can downregulate the expression of MHC Ⅰ on the infected cells, it can not decrease the capacity of cells to present peptides loaded externally, and therefore still induce immune response to some extent.
3.Analysis of Tetrandrine Anti-hepatic Fibrosis by Assessing the Liver Enzyme Spectrum

Hepatic fibrosis of rat was induced by CCl_4 and treated with tetrandrine(Tet). Results showed that Tet had obvious effect of anti-hepatic fibrosis which can be borne out by determining the activities of liver enzyme spectrum consisting of ALT,AKP,ChE,MAO and r-GT in serum. The meaurement can be observed dynamically, is noninvasive and more rapid,accurate and convenient than invasive hepatic biopsy. It provides valuable biochemical indexes for diagnosis,curative effect and prognostic judgement of hepatic fibrosis.
4.Evaluation on measurement uncertainty of detecting procedure of amino acids and carnitines by using non-derivatized tandem mass spectrometry method

Objective To evaluate the measurement uncertainty of the detecting procedure of amino acids and carnitines by the Waters ACQUITY TQD tandem mass spectrometer and PerkinElmer NeoBaseTM non‐derivatized MSMS kit ,and to discuss the meaning of evaluation .Methods According to the method provided by the Medical Laboratories‐Evaluation and Expression of Measurement Uncertainty ,the internal quality control was detected by using two different batch codes of kit for 20 d ,once before and after the routine sample detection on each working day ,the detector were randomly changed ,and then the measurement uncer‐tainty introduced by measurement reproducibility was calculated;the amino acid external assessment quality control data in 20 times of neonatal hereditary metabolic disease tandem mass spectrometry screening provided by the National Center for Clinical Laborato‐ry in 2014 and 2015 ,and the same and 16 times of carnitines external assessment quality control data were performed the statistics , and then the measurement uncertainty introduced by bias was calculated .Next the relative standard uncertainty and the relative ex‐panded uncertainty according to the measurement uncertainty introduced by bias and measurement reproducibility were calculated . The allowed total error indicators of amino acid and carnitines external quality assessment in the neonatal hereditary metabolic dis‐ease tandem mass spectrometry screening by the National Center for Clinical Laboratory were used as the target expanded uncer‐tainty .Results The relative expanded uncertainties of citrulline ,methionine ,phenylalanine ,propionyl carnitine ,octanoyl carnitine , dodecanoyl carnitine ,palmitoyl carnitine and octadecanoyl carnitine were 19 .1% -26 .1% (k=2) ,which were smaller than the tar‐get uncertainty .The relative expanded uncertainties of leucine ,tyrosine ,valine ,free carnitine were 31 .0% -43 .3% (k=2) ,which were greater than the target uncertainty .The uncertainty of isovaleryl carnitine needed to be estimated separately .Conclusion As‐sessing the measurement uncertainty of the detecting procedure of amino acids and carnitines by the non‐derivatized tandem mass spectrometer method can not only provide an opportunity for continuously improving the detection quality ,but also can help the ex‐perimental technique staffs to interpret the test data correctly and the clinician to use the detection reports correctly .
5.Influence of interferon-α therapy on CD8 T memory subsets in patients with chronic hepatitis B

Objective To investigate the influence of interferon-a therapy on CD8 T memory subsets in patients with chronic hepatitis B(CHB) and correlation between the effect of IFN-α and CD8 T memory subsets. Methods Blood samples from 57 patients with CHB were collected before treatment (0 week), at 12 weeks and 24 weeks of treatment with pegylated IFN-α. Assays were performed on freshly isolated peripheral blood mononuclear cells ( PBMCs). For phenotype analysis, All data were acquired on a flow cytometer instrument and prepared for analysis. Results A significantly higher frequency of CD8+ TEM and lower frequency of CD8+ TCM in inactive HBsAg carriers than that in CHB patients prior to treatment was observed (P <0.05). The proportion of CD8 + TCM was higher in group nonresponders than in group respond-ers, and the proportion of CD8 + TEM was lower in group nonresponders than in group responders (P < 0.05 ). The average dosage of IFN-α applied to patients with response was significantly higher than nonresponders. Conclusion The dominance of circulating effector memory T cells may be associated with elimination of viral infection, and possibly benefit for response to therapy with IFN-α.
6.Progress in the application of ketamine in the perioperative period

Ketamine is a racemic mixture composed of an equal amount of L-ketamine and D-ketamine. It has a long history of use as an anesthetic, with various effects such as relieving anxiety, reducing suicidal ideation, and treating chronic pain. However, the widespread use of ketamine is limited by adverse reactions such as dizziness, nausea, elevated blood pressure, and potential abuse. Esketamine has been a hot drug on the domestic market in recent years. As a right-handed isomer of ketamine, Esketamine has the aforementioned effects while reducing the incidence of adverse reactions and improving patient tolerance. It has broad application prospects in multiple clinical environments in the fields of psychotherapy and anesthesia. Therefore, this article provides a review of the application and pharmacological characteristics of ketamine during the perioperative period, briefly describing the pharmacological effects and possible mechanisms of ketamine, and introduces the clinical application progress of ketamine in sedation, analgesia, and anti anxiety and depression during the perioperative period. This article also explores the application prospects of ketamine in the perioperative period, in order to provide reference and guidance for the promotion and application of ketamine, and to provide new research directions.
