Objective:TCM Gegen extract(puerarin)and glucose tolerance factor(GTF) can regulate glucose metabolism and lipid metabolism. The purpose of research is to observe effect of the comoles compound of puerarin and GTF at molecular level on diet-induced obesity in rats. Methods:Male SD obese rats, induced by high fat feed, were randomly divided into fi ve groups:obese model group, positive medicine control group, the groups with small, middle and large of puerarin and GTF,and normal control group. The rats were given medicine for four weeks, body weight of every week, and lipid were measured. Results:After being fed the comoles compound of puerarin and GTF at molecular level for four weeks, the weight growth of rats was slowed down, the level of the following quota:TG,TC,serum insulin and serum leptin declined, the level of HDL-C increased.Compared with obese model group, there was a difference(P

Objective To study the mechanism that glial cell line-derived neurotrophic factor (GDNF)promotes human glio-ma cells proliferation. Methods We divided glioma samples into two groups, including low-grade glioma group and high-grade glio-ma group, while cerebral contusion patients were treated as the control group,12 cases in each group.C6 glioma cell lines were di-vided into three groups, such as GDNF group,BSA(bovine serum albumin)group and control group.CCK-8 (cell counting kit-8) was used to detect the cell proliferation, while Western blot was used to detect the expression of AKT,p-AKT,β-catenin and p-β-catenin in each group. Results Comparing with the control group, the expression levels of AKT,p-AKT,β-catenin and p-β-catenin in glioma group had a significantly increased (P <0.05).Meanwhile,the high-grade gliomas group also had a significant increase in those more than low-grade gliomas group (P <0.05).CCK-8 test showed that the cell proliferation in GDNF group was significant-ly higher than the control group (P <0.05),and the expression levels of p-AKT,β-catenin and p-β-catenin proteins all had a signifi-cant increase (P <0.05).However,the expression level of AKT had no obvious difference. Conclusion GDNF might promote the proliferation of glioma cells by up-regulating the expression of p-AKT,β-catenin and p-β-catenin.

Objective To observe the expression pattern of caspase-3 and HCLS1-associated protein X-1 (HAX-1) at different time after cerebral contusion in rat, and explore the new method for estimating the injury interval. Methods The cerebral contusion model was established using adult SD male rats. Then the rats were randomly allocated into 8 groups: 2 h, 6 h, 12 h, 1 d, 3 d, and 7 d after cerebral con-tusion, sham-operation and normal control. Expression of caspase-3 and HAX-1 protein after cerebral contusion in rat was detected by Western blotting. Laser scanning confocal microscope was used to ob-serve the number of HAX-1 positive cells and TUNEL-stained cells after cerebral contusion. Results The expression of caspase-3 increased parallelly with the time after cerebral contusion and reached the peak value on 3 d. The expression of caspase-3 decreased gradually and still maintained a high level expression on 7 d (P<0.05). The expression of HAX-1 positive cell went up after injury, and reached the peak value at 6 h (P<0.05), then turned down gradually after 12 h and went out of detection after 3 d. The number of TUNEL-stained cells increased obviously at 2 h and reached the peak value on 3 d. The number of TUNEL-stained apoptotic cells decreased gradually and still maintained a high level expres-sion on 7 d (P<0.05). Conclusion The expression of caspase-3 and HAX-1 after cerebral contusion has time sequential regularity, which may provide new evidence for forensic diagnosis of cerebral contusion interval.

Objective:In this article, pueraria extract combined with glucose tolerance factor(GTF) , extracted from yeast, at molecular level. We studied the mechanism ofnew compatibility ofTCM on target organ in treating obesity and analyzed the TCM's theory ofcompatibility with mutual reinforcement and searched a new breakthrough on grouping prescriptions ofTCM.Methods:We study the pharmacodynamics ofpueraria extract(puerarin)combining with GTF by being compared with puerarin, GTF and control medicine respectively by determining the gene expression ofleptin receptor(OB-R).Results:At the fourth week, compared with the obesity model group, the expression ofOB-R in the puerarin combining with GTF group increased significantly(P

BACKGROUND:Currently, different methods of model establishment have been derived from different injury modes of spinal cord injury.Traditional physical injury modeling methods have their own advantages and disadvantages, and there is a lack of more effective and stable animal models of spinal cord injury. OBJECTIVE:To establish a reproducible,controllable,trauma-free,low-mortality,more stable, widely applicable, and short-term postoperative care rat model of spinal cord injury. METHODS:Forty Sprague-Dawley rats with similar body mass and ages were randomly divided into a control group and an improved group, with 20 rats in each group.Animal models of spinal cord injury in the control group were constructed using a clip model method, while the improved group used a modified ligation method based on the compression method to make the spinal cord injury models using suture ligation based on fenestration.Postoperative comparisons were made between the two groups, assessing urination behavior, hematuria, pyuria(infection rate),mortality, scoliosis rate and Basso-Beattie-Bresnahan locomotor rating scale scores at 1,3,5,and 7 days after modeling. RESULTS AND CONCLUSION:Compared with the conventional modeling method, the modified ligation method based on the compression method resulted in faster recovery of urination behavior, lower hematuria rate, lower infection rate, lower mortality rate, lower scoliosis rate, and more concentrated and stable Basso-Beattie-Bresnahan scores(all below 2 points within 1 week). This proves that the modified ligation method based on compression is more suitable for the establishment of spinal cord injury models in rats.
