1.Expression and clinical significance of circulating miR-103a-2 in serum from patients with nonalcoholic fatty liver disease

Objective To investigate the significance of miR-103 as noninvasive biomarker for diagnosis of nonalcoholic fatty liver disease (NAFLD).Methods The serum samples were collected from healthy subjects and NALFD patients to detect biochemical parameters.NucleoZOL method was used to isolate serum miRNA.SYBR Green qPCR was used for relatively quantitative analysis of miR103a-2.The correlations between miR-103a-2 and biochemical parameters were analyzed by Spearman method.Results Compared with healthy control group,the levels of total cholesterol (TC),triacylglyceride (TG),alanine aminotransferase (ALT) and glutamyl transpeptidase (γ-GGT) were all significantly increased in NAFLD group (P < 0.01).Compared with the NAFLD group accompanying ortholiposis,the levels of TC,TG and low density lipoprotein-cholesterol (LDL-C) were all significantly increased in the NAFLD group accompanying dyslipidemia (P <0.01).Compared with the healthy control group,the level of serum miR-103a-2 increased in both NAFLD with ortholiposis group (P < 0.05) and NAFLD with dyslipidemia group (P < 0.01).The level of serum miR-103a-2 was positively correlated with serum TC (r =0.495,P =0.001).Conclusion Serum miR-103a-2 may be a more sensitive parameter for noninvasive screening of NAFLD than the parameters of blood lipid.
2.Rebuilding laboratory work flow in regional laboratory information management system

Objective To explore how to optimize laboratory work flow in regional laboratory information management system (LIS) .Methods Upgraded both software and hardware ,reduced manual operation ,and to avoid the mistake and improve the effi‐ciency .Results Compared to the original process ,the optimized one reduces the time line by 57% .Furthermore ,time cost of other individual processes ,such as patient information input ,sample collection and sample classification were also shortened significantly . Conclusion By work flow rebuilding ,we not only improved the efficiency of laboratory work by saving time cost ,but also reduced the error rate ,which finally satisfied our patients .
3.Effects Origin-Preserving Decoction on the Proliferation of Marrow Hematopoietic Cells in Aplastic Anemia (AA)

Objective:To investigate the effects of Origin-Preserving Decoction on the proliferation of marrow hematopoietic cells in aplastic anemia mice. Methods:Based on experimental AA models induced by Benzene and 60Co radiation in mice, the experiments including semi-solid colony culture (CFU-GM、CFU-E、BFU-E) and liquid culture (MTT Assay) were undertaken to investigate the effects of Origin-Preserving Decoction on the proliferation of marrow hematopoietic cells in AA mice. Results:Origin-Preserving Decoction (3g/kg) (ig.) could improve the outputs of CFU-GM, CFU-E and BFU-E in AA mice damaged by benzene and 60Co radiation: MTT assay showed that Origin-Preserving Decoction could stimulate the hematopoeisis of bone marrow of AA mice radiated by 60Co. Conclusions: Origin-Preserving Decoction has a good future for AA therapy.
4.Rapid diagnosis of hereditary neuropathy with liability to pressure palsies by multiplex ligation-dependent probe amplification

Objective To introduce the application of multiplex ligation-dependent probe amplification (MLPA) assays in the diagnosis of patients with hereditary neuropathy with liability to pressure palsies (HNPP).Methods Copy numbers of the exons in peripheral myelin prolein 22 (PMP22) gene,tektin 3 (TEKT3) gene and cytochrome c oxidase assembly protein 10 (COX10) gene were analyzed by MLPA in 8 patients diagnosed with HNPP clinically and 5 normal controls.Results Among the 8 patients,7 patients were identified to have deletion mutations according to their reduced peak area of PMP22 gene,TEKT3 gene and COX10 gene compared with that of normal controls.One patient with normal peak area of PMP22 gene,TEKT3 gene and COX10 gene showed no deletion of these genes.Conclusions MLPA assays can detect the copy numbers of genes in HNPP region through semi-quantitative analysis in a rapid,accurate way,which may be utilized widely in the genetic diagnosis among HNPP patients.
5. The relationship between core members' individual social capital and performance among HIV/AIDS related community-based organization

Objective:
To investigate the relationship between core members' social capital and performance among HIV/AIDS-related community-based organizations (CBO).
Methods:
From July to December in 2015, a total of 327 core members from 212 HIV/AIDS-related CBO in 8 provinces were recruited based on the prevalence of HIV/AIDS (e.g., Yunnan, Hunan, and Sichuan are in high epidemic level; Anhui, Hubei, Shandong, and Jilin are in middle epidemic level; and Gansu is in low epidemic level) by multistage stratified cluster sampling and convenient sampling method. A questionnaire was administered in this study, including general demographic information, core members' social capital, individual performance and organizational performance. Multivariate logistic regression model was used to analyze the relationship between core members' social capital and performance among CBO.
Results:

Objective To investigate the expression levels of adiponectin and visfatin in the gingival crevicular fluid (GCF) and serum of patients with obesity and chronic periodontitis.Methods The study included 80 patients.Among them,20 were healthy controls (H group),20 had periodontitis (CP group),20 had obesity (OB group),and 20 had both obesity and periodontitis (OB+CP group).The patients' baseline information and clinical parameters were recorded,and the GCF and serum were collected.The concentration of adiponectin and visfatin in the GCF and serum were then assayed using ELISA.Use the SPSS 17.0 software package for statistical analysis.Results The concentration of adiponectin in the serum of the CP group,the OB group,and the OB+CP group were significantly lower than that of the H group (P < 0.05).The concentration of adiponectin in the serum of the OB+CP group was lower than that of both the CP and OB groups (P < 0.05).The concentration of adiponectin in the GCF of the CP group,the OB group,and the H group were higher than that of the OB+CP group (P < 0.05).The concentration of adiponectin in the GCF of the CP group,the OB group,and the H group were lower than that of the OB+CP group (P < 0.05).Conclusion Obesity may aggravate the destruction of periodontal tissue by inhibiting the expression of adiponectin and promoting the expression of visfatin.
7.Attributes and influencing factors of care needs for maternal after perinatal loss based on the Kano model

Objective:To investigate the attributes and influencing factors of care needs for maternal after perinatal loss, and to provide a reference for promoting maternal physical and mental health and improving the quality of care.Methods:From February to June of 2023, a cross-sectional study was used, 222 maternal after perinatal loss were selected by the convenience sampling method and completed a battery of questionnaires, including a general information questionnaire, the Distress Thermometer (DT), the Perceived Social Support Scale (PSSS), the Attitudes Toward Seeking Professional Psychological Help Scale-Short Form (ATSPPH-SF) and Kano Model-based Questionnaire on Care Needs for Maternal after Perinatal Loss.Results:A total of 207 maternal after perinatal loss were included in the complete data, (31.12±4.55) years old, (68.64 ± 10.70) of PSSS, (16.10 ± 3.43) of ATSPPH-SF. The care needs for maternal after perinatal loss included 4 must-be quality, 8 one-dimensional quality and 13 attractive quality. Multiple linear regression analysis showed that education level ( t=2.28), the PSSS score ( t=2.15) and the ATSPPH-SF score ( t=3.94) were the main influencing factors of care needs for maternal after perinatal loss (all P<0.05). Conclusions:Health care professionals should gradually improve the nursing service system according to the priority division of care needs attributes of maternal after perinatal loss, and develop personalized care according to different influencing factors.
8.Adiponectin gene polymorphism and postpartum type 2 diabetes in pregnant women with gestational diabetes mellitus

Objective:To investigate the relationship between adiponectin (ADIPOQ) gene polymorphism and postpartum type 2 diabetes mellitus (T2DM) in pregnant women with gestational diabetes mellitus (GDM).Methods:A retrospective study was conducted on 236 GDM postpartum women admitted to the Affiliated Hospital of Jining Medical College from June 2020 to June 2021 as observation subjects. They were divided into a T2DM group and a non T2DM group based on the occurrence of T2DM after delivery. The clinical data of the two groups were compared. The double deoxygenation end termination method was used to detect the single nucleotide polymorphism (SNP) of the ADIPOQ gene, and the four loci rs17366568, rs822395, rs1501299, and rs2241766 were classified. The relationship between ADIPOQ genotype polymorphism and postpartum T2DM was analyzed using a logistic regression model.Results:The G allele carrying the rs2241766 locus in ADIPOQ gene was negatively correlated with the occurrence of T2DM ( OR=0.71, 0.68, P<0.05). Compared with T2DM patients with TT genotype, the GT+ GG genotype at the rs2241766 locus had a lower risk of occurrence for gestational age ≥2 and HbA 1c>85%. Similarly, T2DM patients with pre pregnancy body mass index (BMI)>25 kg/m 2 were more likely to be carriers of the rs2241766 TT genotype ( P=0.026). The (GT+ TT) genotype carrying the T allele at the rs1501299 locus was a protective factor for gestational age and HbA 1c in T2DM patients. Conclusions:The rs2241766 and rs1501299 polymorphisms of the ADIPOQ gene are associated with susceptibility to postpartum T2DM in GDM women. Individuals with rs2241766 and rs1501299 mutant genotypes belong to the high-risk population for T2DM.
9.Changes in tuberculosis incidence in BRICS countries from 1990 to 2019 and incidence trend prediction

Objective:To understand the trend in the disease burden of tuberculosis in the BRICS countries (Brazil, Russia, India, China, and South Africa) from 1990 to 2019, forecast the incidence of tuberculosis in the BRICS countries from 2020 to 2030.Methods:This study used data from the Global Burden of Disease Study 2019 to describe the incidence trends from 1990 to 2019 and epidemiological characteristics of tuberculosis in the BRICS countries. The Bayesian-age- period-cohort model was used to predict the incidence of tuberculosis in the BRICS countries from 2020 to 2030.Results:Although the age-standardized incidence rates (ASIR) of tuberculosis were on the decline in all BRICS countries, the magnitude of the decline varied, with the most significant decline in China and the least in South Africa. In 2019, there were gender and age differences in the incidence of tuberculosis in all BRICS countries. Except for South Africa, the ASIR in males were higher than that in females and except for South Africa and Russia, the incidence rates of tuberculosis showed an increasing trend with age in all BRICS countries. From 2020 to 2030, the incidence rates of tuberculosis in the BRICS countries were projected to be declining, except it was likely to increase among women in Russia. However, the expected decrease of tuberculosis incidence in the BRICS countries by 2030 would be less than 80% as compared to that of 2015.Conclusions:Positive results have been achieved in the prevention and treatment of tuberculosis in BRICS countries. But the situation is still serious, with demographic differences in the incidence of tuberculosis in different countries.
10.Construction of a medical service experience system based on staff role play

Objective:To build a medical service experience system based on role play of hospital staff, and explore its contribution to upgrading medical service, elevating medical service quality, and improving patients′ satisfaction.Methods:An indicator system for medical service experience was developed via literature review. 20 newly recruited hospital employees in 2019 were sampled randomly to form a service experience team, who were arranged to experience the full medical service process of outpatients and inpatients based on role play. The questionnaire for service quality based on patient perception and expectation was used to survey the expectation and perception values of those role players on their medical service experiences, and summarize the defects found on such six dimensions as hospital reliability, responsiveness, tangibility, assurance, empathy and cost-effectiveness. After due corrections of these defects, further experiences and patient satisfaction survey were made to assess the effects of such actions taken. Paired t test was used respectively in statistical analysis of service experience value and actual perception value, as well as the perception values before and after the actions taken, and χ2 test was used for a statistical analysis of patient satisfaction. Results:Role players experienced respectively at the emergency, outpatient and inpatient departments of the hospital for a week in August 2019. Statistics of their experiences indicated≥0.50 points difference between the mean expectation and perception values on doctor-patient communication, patient help responsiveness, outpatient process, examination report delivery duration, service attitude, patient trust on medical workers, medical environment and patient privacy protection.Following a 3-month reform, role players found improvements in such aspects as hospital reliability, responsiveness, tangibility, assurance, empathy and cost-effectiveness; patient satisfaction improved sizably in December 2019 over January of the same year( P<0.05). Conclusions:The construction and application of the medical service experience system based on role play of hospital staff prove highly useful in upgrading medical service actions, improving medical service quality, and improving patient satisfaction.
