: The recovery of spinal cord injury(SCI) is a difficult problem in neuroscience research field, one of the reasons is that the ability of injured spinal cord regeneration is limited. Recent years many experiments prove that neurotrophin-3 is very important in SCI regeneration recovery, the gene transplantation is effective. This article makes a summary of SCI treatment research advancement.
2.Expression of CXC chemokine receptor 4 in clear cell renal cell carcinoma

Objective To investigate the expression of CXC chemokine receptor 4 (CXCR4) in clear cell renal cell carcinoma (ccRCC) and its relationship with the clinical pathological parameters of ccRCC.Methods The expression of CXCR4 was detected by immuno-histochemistry method in 63 cases of ccRCCs,20 cases of para-carcinoma tissues and 20 cases of normal renal tissues.The correlation between expression level of CXCR4 and clinical pathological parameters of ccRCC patients were analyzed,and the clinical significance of its expression in ccRCC was evaluated.Results The positive expression rate of CXCR4(49.2%) in ccRCCs was significantly higher than that in para-carcinoma tissues (15%) and normal renal tissue (10%),and the difference was statistically significant (P < 0.05).The expression level of CXCR4 and the clinical stage and pathological grade of ccRCC were correlated (P < 0.05),and was associated with lymph node transfer (P < 0.05).The CXCR4 negative group overall survival rate [55.2% (16/29)] and the average survival time(46 months) was significantly better than the positive group [38.5% (10/26),32 months;P < 0.05].Conclusions The expression level of CXCR4 in ccRCC is correlated with the clinicopathological parameters and prognosis.CXCR4 is expected to be an important marker for diagnosis and prognosis evaluation of renal cell carcinoma.
3.Expression of Chemokine CXCL12 and Its Receptor CXCR4 in Endometrial Carcinoma Tissues

Objective To observe the translations of chemokine CXCL12 and its receptor CXCR4 in endometrial carcinoma tissues,and to evaluate their significance in clinical pathologic features of endometrial carcinoma.Methods From Jan.2012 to Dec.2014,52 tissue specimens of patients with endometrial carcinoma were admitted to the hospital,aged 55.6 ± 19.2 years,and 26 cases of normal endometrium as control,aged 52.3±16.5 years old.Expressions of CXCL12 and CXCR4 in the tissue specimens were detected by immunohistochemistry,and to analyze the relationship between the expressions and clini-cal pathological features,and FIGO stage,cell differentiation,lymph node metastasis and pathological type in endometrial carcinoma.Results In endometrial carcinoma,the positivity rates of CXCR4 and CXCL12 were 76.92% and 69.23% re-spectively.The positive expression rates of which were 34.62% and 30.77% in normal endometrium tissues,statistically significant (χ2 =11.826,P <0.01).The expression of CXCR4 was positively correlated with the differentiation of endome-trial carcinoma cells (P <0.05).In endometrial carcinoma,with the exception of CXCR4 expression and cell differentiation was positively related (r=0.386,P <0.05),CXCL12 and CXCR4 expression and clinical stage,pathological type and lymph node metastasis no correlation was found (P >0.05).Conclusion The results showed that the expression of CXCL12 and CXCR4 in endometrial carcinoma tissues was significantly increased,which may play an important role in the development of endometrial cancer.
4.Clinical observation of curative effect of high intensity focused ultrasound treatment in liver metastasis of colorectal cancer

Objective To investigate the clinical efficacy and safety of the high-intensity focused ultrasound therapy in patients with colorectal liver metastases. Methods 28colorectal lesions in 16 patients with liver metastases after HIFU treatment lines observed patient vital signs,the main organ,imaging studies (CT or MRI,etc. ) ,complications and prognosis of cancer cases. Results All patients with stable vital signs after treatment, liver, kidney function was normal,6-month follow up, six more than 80% reduced lesion size,lesion size decreased in 18 of 30% to 50% ,4tumor volume did not change significantly; 16 patients, 1 patient was lost. 15 cases were followed uPfor 6 to 24 months,5 patients were transferred because of lesions disseminated in death within 6 months after surgery, calculated with the Kaplan-Meier median survival time was 13 months; 1 patient skin burning streak marks, healed after 1 week after treatment. 1 patient treatment areas of the chest skin, mild subcutaneous edema. Conclusion The treatment of patients with liver metastasis of colon cancer lesions was a non-invasive, safe and effective treatment.
5.Effects of Salvia injection on synaptic plasticity of neonatal rats with hypoxic-ischemic brain damage

Objective To observe the effects of Salvia injection on the brain pathology,expression of synaptophysin and the synaptic ultrastructure at different time points in neonatal rats after hypoxic-ischemic brain damage (HIBD).Methods One hundred and fifteen healthy newborn SD rats of 7-day-old were randomly divided into normal group,sham group,Salvia high dose group,Salvia low dose group,and the HIBD model group.The HIBD model was prepared by ligation of the left carotid artery combined with hypoxic environment.The rats(in normal group,sham and HIBD model groups) were injected with sterile saline[9.0 mL/(kg · d)],while the rats in high and low dose groups were injected with Salvia injection [9.0 mL/(kg · d) and 4.5 mL/(kg · d) respectively].The intraperitoneal injec tion lasted for 7 and 14 days.The rats' brains were collected at one day,7 days and 14 days after the modeling respec tively.The specimens of brain tissue were observed through hematoxylin-eosin (HE),the expression of synaptophysin (SYP) was determined by using immunohistochemistry method,and the synaptic ultrastructure in the frontal cortex was observed through transmission electron microscope.Results Finally,106 newborn rats were included in statistics analysial.(1)General observation:after HIBD modeling,the neonatal rats were mostly in the left-lateral position,with difficulties in turning over,balance abnormalities,limb shaking and other abnormal behaviors,and the delay of eyes open with the left upper eyelid ptosis.(2) Pathology:the disordered and deep stained nerve cells,the degenerated and necrotic neurons were observed in the brain tissues of the HIBD model group.(3) Expression of synaptophysin:mean density of SYP in the HIBD model group was significantly lower than the normal group on day 1,day 7,and day 14 (P <0.05);the SYP expression in the Salvia intervention groups increased compared with the HIBD model group(P < 0.05).(4) Synaptic ultrastructure:the incomplete structure of the frontal cortex neurons,the swelling organelle and the synaptic structure damage were observed in the HIBD rats.Compared with the HIBD model group,the neuronic and synaptic ultrastructure were improved by the intervention treatment of high and low dose Salvia injection.Conclusions The mechanism of Salvia injection in treatment of neonatal rats with HIBD may be associated with the improvement in neuronal ultrastructure and synaptic reorganization.
6.Significance of combined detection of procalcitonin,C-reactive protein and lipopolysaccharide for early diagnosis of bacterial infection in newborns

Objective To study the significance of combined detection of procalcitonin ( PCT ) , C-reactive protein( CRP) and lipopolysaccharide( LPS) for early diagnosis of bacterial infection in newborns. Methods Clinical data of ninety-eight newborns from neonatal ward of our hospital were retrospectively studied. Fifty cases with bacterial infectious diseases were selected as infection group,in the same period,48 cases with non-bacterial infectious diseases were selected as control group. In the 24 hours after admission before use of antibiotics,all of cases were picked blood used for testing CRP,PCT,LPS and blood culture, and the results were contrasted and analyzed. Results The levels of serum PCT,CRP and LPS in infection group were respectively significantly higher than those in control group,and the differences were statistically significant(P<0. 05,respectively). In gram-positive bacterium group,the positive rate of combined detection of serum PCT and CRP was obviously higher than that of single detection of PCT or CRP ( 91. 3% vs. 60. 9%,P<0. 05;91. 3% vs. 56. 5%,P<0. 05 respectively) . In gram-negative bacteria group,positive rate of combined detection of serum PCT and LPS was obviously higher than that of single detection of PCT or LPS respectively(88. 9% vs. 59. 3%,P<0. 05;88. 9% vs. 66. 7%,P<0. 05 respectively). Conclusion Joint detection can improve the diagnostic efficiency, and reduce missed diagnosis. And we can identify disease which predominantly infected by positive bacteria or negative bacteria through joint detection,which can contribute to the choice of clinical antibiotic drugs.
7.Preparation Process of Xuangui Zhitong Dispersible Tablets

OBJECTIVE:To prepare Xuangui zhitong dispersible tablets and optimize its formulation technology. METHODS:Using disintegration time as index,single factor test was conducted for filler,disintegrating agent,the types and amount of adhe-sives and compression pressure. The amount of mixed disintegrating agent,avicel and gum arabic were optimized by orthogonal test. The tablet quality by optimized formulation was detected,and disintegration time,the content and dissolution rate of tetrahy-dropalmatine were determined;the similarity of in vitro dissolution rate of dispersible tablets and dropping pills were evaluated by similarity factor test. RESULTS:The optimized formulation was composed of 25% MCC as fillers,9% PVPP and 9% L-HPC as mixed disintegrants,85% ethanol solution as adhesives,micro-silica gel 2%,compression pressure of 3.0 kg/cm2. The average dis-integration time was 1.22 min,and the content of tetrahydropalmatine was 1.097 mg/g. The accumulative dissolution rate was more than 80% at 10 min and more than 90% at 15 min. The similarity factor f2 of dissolution curve was 62,using dropping pills as ref-erence preparation. CONCLUSIONS:Xuangui zhitong dispersible tablet had a rapid disintegration and the behavior of dissolution is similar to Xuangui zhitong dropping pills.
8.Effects of microtubule disassembly on hypoxic injury of cultured cardiomyocytes

Objective To investigate whether microtubule disassembly plays an important role in the pathogenesis of the opening of mitochondria permeability transition pore (MPTP) in hypoxic cardiomyocytes and the decrease of its activity, resulting in its hypoxic injury. Methods Neonatal rat cardiomyocytes in primary culture were randomized as normoxia group (A), hypoxic group (B), normoxia treated with microtubule destabilizing agent (Colchicine) group (C), hypoxia treated with microtubule stabilizing agent (Taxol) group (D). At 0.5, 1, 3, 6, 12 h after treatment, polymeric tubulin was detected by immunofluorescence and Western blotting, mitochondria permeability transition pore (MPTP) open by coloading with calcein AM and cobalt chloride, and the activity of cells by measuring the mitochondrial-dependent reduction of MTT to formazan. Results Early microtubule disassembly, MPTP open and activity decrease of cardiomyocytes in both groups B and C were observed at 0.5 h after treatment. These phenomena all became more and more significant with the prolongation of treatment. However, microtubule disassembly, MPTP open and activity decrease of cardiomyocytes of group D were significantly lower than those of group B. Conclusion Microtubule disassembly happened at 0.5 h after hypoxic treatment. Microtubule stabling agent Taxol and destabilizing agent Colchicine can regulate microtubule integrity efficiently. The microtubule damage plays an important role in the hypoxic injury of cardiomyocytes.
9.Construction and identification of Rattus norvegicus microtubule-associated protein 4 gene recombinant adenovirus

Objective To construct recombinant adenovirus vector containing Rattus norvegicus microtubule-associated protein 4 gene,and transfect it into the rat cardiac myocytes cultured in vitro.Methods A pair of primers were designed,and full length MAP4 DNA was cloned from rat total mRNA by PCR.The PCR product was double-digested with restriction endonucleases NheⅠ and NocⅠ,and inserted orientationally into pShuttle2.The plasmid of pShuttle2-MAP4 was double-digested with restriction endonucleases NheⅠ and NocⅠ,and inserted BD Adeno-X~(TM) Virul DNA,named pAd2-MAP4.The non-recombinant adenovirus was screened out with PacⅠ, pAd2-MAP4 was linerized with SwaⅠ,and the recombinant virus genome was transfected into HEK293 cell line for packaging and amplification of Ad-MAP4 virus.The recombinant adenovirus was transfected into rat cardiac myocytes and MAP4 was identified by immunohistochemistry.Results The recombinant adenovirus-MAP4 was constructed successfully and the titer was about 2.3?10~(8) pfu/ml.The expression of MAP4 was enhanced at 48 h after the transfection.Conclusion We have successfully constructed a recombinant adenovirus Ad-MAP4 that has enforced the expression of MAP4 in vivo.
10.Case-control study on effects of external fixation combined with limited internal fixation for the treatment of Pilon fractures of Rüedi-Allgower type III.

OBJECTIVETo analyze the effects of three surgical operations in the treatment of Pilon fracture of Rüedi-Allgower type III, and put forward the best therapeutic method.
METHODSThe clinical data of 33 patients with Pilon fracture who received surgical operations (plaster immobilization group, 10 cases; distal tibia anatomical plate group, 11 cases; external fixation with limited internal fixation group, 12 cases) from October 2009 to January 2012 were analyzed. There were 5 males and 5 females, ranging in age from 24 to 61 years in the plaster immobilization group. There were 7 males and 4 females, ranging in age from 21 to 64 years in the distal tibia anatomical plate group. There were 7 males and 5 females, ranging in age from 23 to 67 years in the external fixation with limited internal fixation group. The Ankle X-ray of Pilon fracture after operation, ankle score, early and late complications were collected. Bourne system was used to evaluate ankle joint function.
RESULTSAfter 8 months to 3 years follow-up, it was found that three kinds of treatment had significant differences in the outcomes and complications (P < 0.05): the external fixation with limited internal fixation group got the best results. The number of anatomic reduction cases in the external fixation with limited internal fixation group (7 cases) and the distal tibia anatomical plate group (8 cases) was more than the plaster immobilization group (2 cases). According to the ankle score, 8 patients got an excellent result, 3 good and 1 poor in the limited internal fixation group ,which was better than those of distal tibia anatomical plate group (5 excellent, 4 good and 2 poor) and the plaster immobilization group (3 excellent, 4 good and 3 poor). The number of early and late complications in the external fixation with limited internal fixation group was more than those in the plaster immobilization group and the distal tibia anatomical plate group (P< 0.05).
CONCLUSIONTreatment of external fixation with limited internal fixation in the treatment of Pilon fracture of Rüedi-Allgower type III is effective and safe.
