1.Effects of lack of mitochondria topoisomerase on migration ability of gastric cancer cells

Objective To investigate the effect of mitochondrial topoisomerase(TOP1MT) on migration of gastric cancer cells.Methods Through siRNA of TOP1MT,the expression of TOP1MT in MKN45 and MKN28 gastric cancer (GC) cells reduced.The effect on GC cell migration was proved by using scratch test and Transwell experiments.Results Compared with the control group,TOP1MT knockout significantly increased the migration of GC cells of MKN28 and MKN45 (t=48.69,P<0.05;t=23.42,P<0.05).Immunohistochemical staining further proved that lack of TOP1MT was associated with lymph node metastasis in gastric cancer (F=14.97,P<0.05).Conclusion Low expression of TOP1MT can significantly enhance the migration of GC cells.
2.Expression and bioactivity of staphylococcal enterotoxin O

Objective To clone the mature peptide gene of seo, construct two kinds of prokaryotic fusion expression vector, obtain recombinant staphylococcal enterotoxin O(SEO) and evaluate its biological activity. Methods The seo gene was amplified from ZNZ2-3 strain of Staphylococcus xylosus by designed primers and it was cloned into fusion vector pGEX-6P-1 and pET28a. The soluble recombinant protein(GST-SEO) was expressed in E. coli BL21 host cells, the purified GST-SEO was obtained by a single step of affini-ty chromatography using Glutathinione Sepharose 4B colume. Another fusion protein(P28-SEO) with his-tag was expressed in E. coli BL21 ( DE3), the purified P28-SEO protein was harvested by nickel chelate affinity chromatography method. The antigenicity of recombinant SEO was tested by Western blot. The stimulating effects of recombinant SEO on mouse lymphocytes was tested by MTT. The activity of Caspase 3 and the ap-optosis of DNA ladder were tested. Results The nucleotide sequence of the cloned seo gene was the same as that of reported in GenBank. The soluble recombinant protein GST-SEO was expressed at 25% expression level, and P28-SEO was 22%. The Western blot by GST-SEO-positive sera demonstrated that the recombi-nant SEO had good immunogenicity. The MTY assay of the enterotoxin activity showed that low dose of re-combinant SEO stimulated the proliferation of mice spleen lymphocytes, higher doses will lead to apoptosis. Conclusion Two of the recombinant SEO still have superantigen activity, and can lead to apoptosis of mice splenocytes.
3.Evaluation value of the MRI parameter and immunohistochemistry in the neoadj uvant chemotherapy for HER-2 positive breast cancer

Objective To explore the evaluation value of MRI parameters and immunohistochemistry in the neoadjuvant chemotherapy (NAC)for the HER-2 positive breast cancer.Methods 7 6 patients with locally advanced HER-2 positive breast cancer were analyzed retrospectively.According to the postoperative pathology,patients were divided into pathological complete response (PCR)group and non-PCR (Non-PCR)group.First,univariate and multivariate Logistic regression analysis were used to analyze the predictors of PCR,and then we assessed the evaluation value of the assessment model which was built on multivariate logistic regression.Results Univariate regression analysis showed that menstrual status,ER expression,K-i 67 expression,ADC value,tumor diameter, ΔADC%(NAC2-NAC0),ΔD%(NAC0-NAC2) had predictive value for PCR;the parameters ΔADC% and ΔD%,the expression of ER and K-i 67 were independent predictors of PCR based on multivariate Logistic regression analysis.The AUC of the assessment model model was 0.873.Conclusion Based on the heterogeneity of breast cancer,combine MRI with immunohistochemistry parameters could improve the prediction in the middle of NAC,and provide imaging basis for the adj ustment of follow-up chemotherapy regimen.
4.Comparsion of the clinical application between Da Vinci robot and open operation in thyroid benign tumor with diameter greater than 5 cm

Objective:To evaluate the feasibility and safety of robotic thyroidectomy for treatment of thyroid benign tumor with diameter larger than 5cm.Methods:The clinical data of 36 patients (in the robot group) who received Da Vinci robot thyroid benign tumor surgery in the thyroid breast Surgery Department of the 960th Hospital of the PLA (Former Jinan Military General Hospital of PLA) from Jan. 2014 to Jun. 2019 were retrospectively analyzed, and compared with 48 patients (in the open group) who received open surgery in the same period.The operative time, intraoperative blood loss, length of hospital stay, postoperative drainage volume, postoperative parathyroid hormone, cosmetic effect, incidence of surgical complications and postoperative recurrence rate of the two groups were observed. Statistical software SPSS16.0 was used for statistical analysis of the data.Results:Both groups successfully completed the operation. Compared with the open group, the operation time ( t=11.232, P<0.001) and postoperative drainage volume ( t=2.892, P=0.003) of the robot group were more than those of the open group, and the aesthetic effect was better ( t=3.291, P=0.024). Intraoperative blood loss ( t=1.575 , P=0.118), hospital stay ( t=-1.784, P=0.091), postoperative PTH ( t=1.892, P=0.086), incidence of surgical complications ( χ2=3.291, P=0.610), visual analogue score of postoperative pain ( t=-2.431, P=0.081) and postoperative recurrence rate were not significantly different ( P>0.05). No recurrence occurred during follow-up. Conclusion:For patients with a selective benign thyroid tumor with a diameter of 5-8 cm, the bilateral axillo-breast and transaxillary approach for thyroid surgery by Da Vinci robot are safe, effective and have better cosmetic results.
5.The evaluation of the tumor diameter combined ADC values for the therapeutic efficacy of neoadj uvant chemotherapy in each subtype of breast cancer

Objective To study the values of change of diameter (ΔD%)on DCE-MRI and ΔD% combined with ADC for the therapeutic effect evaluation in the neoadjuvant chemotherapy(NAC)of each subtype of breast cancer.Methods 121 women with invasive breast cancer were enrolled in this study,including 56 cases of Luminal A type,42 cases of Luminal B type,13 cases of Her-2 type and 10 cases of triple negative type.Results Imaging evaluation results obtained by single applying DCE-MRI was analyzed with the pathological results after surgery for the consistency,and the Kappa values of Luminal A,Luminal B,Her-2 and triple negative type were 0.594,0.617,0.401,0.690,0.800,respectively.When combining ΔD% with ΔADC%,the Kappa values were 0.844,0.792,0.854, 0.847 and 0.800,respectively.The sensitivity of combined parameters in evaluating pCR after NAC was significantly increased in Luminal type cancers,however,with a slightly increasing in Her-2 type,and constant in triple negative type.Conclusion ΔD% combining with ΔADC% is better than single parameter in evaluating pCR for Luminal and Her-2 types cancer,but not for triple negative type.
6. Clinical Significance of Matrix Metalloproteinase-7 in Inflammatory Bowel Disease

Background: Matrix metalloproteinase-7 (MMP-7) is a proteolytic enzyme involved in wound healing, tissue remodeling by regulating a variety of extracellular matrix and non-matrix substrates, and is an important mediator of tissue damage during inflammation in inflammatory bowel disease (IBD). Aims: To investigate the clinical significance of MMP-7 in IBD. Methods: A total of 38 cases of colon biopsy samples from patients with IBD from January 2019 to January 2021 at the Second Affiliated Hospital of Zhengzhou University were collected, including 20 ulcerative colitis (UC) and 18 Crohn's disease (CD). The paracancerous tissues of 5 patients with colon cancer were served as the control group. The expression of MMP-7 in colon tissue was detected by immunohistochemical staining, and its correlation with C-reactive protein (CRP) and white blood cell (WBC) was analyzed. Results: Compared with the control group, the expression of MMP-7 was significantly up-regulated in both active UC group and severe CD group (P<0.05). The expression of MMP-7 was gradually up-regulated with the increase of severity. The expression of MMP-7 in severe UC and CD group was significantly increased than that in remission UC and CD group (P<0.05). No significant difference in the expression of MMP-7 was found between patients with different gender, age, and whether treated with anti-TNF-α agents (P>0.05). The expression of MMP-7 in colon tissue of patients with active IBD was positively correlated with CRP and WBC (P<0.001). Conclusions: The expression of MMP-7 is correlated with the activity and severity of IBD, but not correlated with gender, age, and whether treated with anti-TNF-α agents. To some extent, MMP-7 can indicate the severity of intestinal inflammation, and may be an important indicator of inflammatory activity in IBD.
7.Study on characteristic chromatogram of Yao medicine Kadsura longipedunculata and its anwulignan content and anti-inflammatory activity

OBJECTIVE To establish characteristic chromatogram of Yao medicine Kadsura longipedunculata and the method for the content determination of its main component anwulignan, and evaluate the anti-inflammatory activity of anwulignan. METHODS HPLC method was performed with acetonitrile-0.5% phosphoric acid solution as the mobile phase for gradient elution. The characteristic chromatogram of K. longipedunculata was established and similarity was evaluated by Similarity Evaluation System for Chromatographic Fingerprint of TCM (2012 edition). The content of anwulignan in K. longipedunculata was determined. Lipopolysaccharide induced RAW264.7 macrophages were selected as inflammatory cell model to investigate the effects of anwulignan on the levels of tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) and IL-6. RESULTS The similarities of characteristic chromatogram for 10 batches of K. longipedunculata ranged 0.901-0.994, and 9 common peaks were determined; 3 components were identified, such as changnan schisantherin E, kadsulactone A, anwulignan. The contents of anwulignan were (0.72±0.05)-(1.21±0.03) mg/g(n=3). Anwulignan of 0.125-0.5 μg/mL greatly decreased the levels of TNF-α, IL-1β and IL-6 in the supernatant of inflammatory model cells (P<0.05 or P<0.01). CONCLUSIONS HPLC characteristic chromatogram of K. longipedunculata and the method for the content determination of anwulignan are all established, and anwulignan may be the active ingredient of anti-inflammatory effect in K. longipedunculata.
8. Current status and progress of 2019 novel coronavirus pneumonia

Recently, the 2019 novel coronavirus (2019-nCoV) pneumonia outbroke in Wuhan and rapidly spread to all over China and even the world. Because of the strong infectivity and various clinical symptoms, it has brought certain difficulties to the epidemic prevention and control. Currently there is no specific drug for 2019-nCoV. Previous drugs used to treat other coronaviruses may be effective, but further clinical trials remain needed. We reviewed literature on the epidemiology, etiology, clinical manifestations, imaging manifestations, laboratory examination, diagnosis, complications, treatment and outcome of 2019-nCoV pneumonia.
9. Effectiveness of chloral hydrate sedation in pediatric patients who receive cone beam computed tomography examinations

Objective:
To discuss the success rate and image quality in pediatric patients who used chloral hydrate before their cone beam computed tomography exam.
Methods:
1752 patients aged 1 to 6 were selected for this retrospective study. They were divided into sedated group (219 cases) and non-sedated group (1 533 cases). The success rate and image quality were compared between two groups.
Results:
The sedated group had a higher success rate to non-sedated group: 99.5%(218/219) vs. 90.4% (1 386/1 533). The motion artifact in sedated group was lower than non- sedated group with I degree: 4.8% (15/314) vs. 20.1%(327/1 630) and II degree: 0.3%(1/314) vs. 12.2%(199/1 630).
Conclusion
Giving chloral hydrate to pediatric patients before their CBCT exam would improve both success rate and image quality, and reduce unnecessary radiation expose.
