Objective:To construct the recombinant plasmid expressing STAT6 shRNA and observe the effects of RNAi suppressed STAT6 gene expression. Methods:shRNA and pIRES2-EGFP-STAT6 plasmid were cotransfected into COS7 cell,to investigate the suppression effect of shRNA on STAT6 under fluorescent microscope . This effect was confirmed by Western blot at protein expression level. Results: STAT6 protein exhibitited high expression in COS7 cells after transfecting pIRES2-EGFP-STAT6 plasmid into COS7 cells alone ; compare with the negative group,the level of the STAT6 protein in the two shRNAs group were 23.1% and 9.6%. Conclusion:STAT6 shRNA were successfully constructed.This technique deserves further study as a potential therapentic tool for asthma .

Objective To investigate the expression of matrix metalloproteinase-8 (MMP-8) in human breast cancers and its relationship with the clinicopathological parameters,evaluate the relevance of MMP-8 levels to metastases in breast cancer.Methods Human specimens were obtained from 50 patients with invasive ductal carcinomas,and 20 normal breast glands were enrolled.Intact clinical parameters were aviliable for all patients.The protein expression of MMP-8 was determined by immunohistocbemistry technology SP.Results The rate of MMP-8 positive breast cancer was significantly higher than that in normal breast glands [68 ％ (34/50) vs 15 ％ (3/20),P ＜ 0.05].The expression of MMP-8 showed a negative correlation with breast cancer staging.The expression of MMP-8 in stage Ⅰ was higher than that in stage Ⅱ and Ⅲ/Ⅳ[94 ％ (15/16),57 ％ (13/23),55 ％ (6/11),P ＜ 0.05].Lymph node negative patients had a higher MMP-8 expression than patients with lymph node metastases [84 ％ (21/25) vs 50 ％ (13/25),P ＜ 0.05].Conclusion MMP-8 expression is associated with human breast cancer metastases and might be involved in inhibiting metastases.

Myelodysplastic syndrome(MDS) is a malignant clonal disease of hematopoietic system,characterized by dysplasia and ineffective hematopoiesis.Therefore,the morphology of bone marrow and peripheral blood cells are crucial in the diagnosis of MDS.The morphological features of marrow cells,the significance of morphological diagnosis,the progress in the study of morphological diagnosis and some major issues on morphological diagnosis of MDS are discussed in this article.

Objective To study the quantitative analysis of myocardial reperfusion after percutaneous coronary intervention (PCI) using myocardial contrast echocardiography (MCE) in patients with coronary diseases. Methods The contrast agent of SonoVue via vein route using contrast pulse sequencing (CPS)MCE were performed in 15 patients with coronary diseases before and after the PCI. The intensity of amplitude (A) ,slope(β), A ×β, the contrast arrival times (AT) and the arrival peak times (TTP) were calculated with the autotracking contrast quantification software. Results Clear images of the left ventricle were well displayed in all the patients. There were satisfied images in the normal perfusion area of the myocardium,but not in the abnormal. AT,TTP and A ×β were significantly different between the abnormal and the normal perfusion area ( P ＜0. 05). The myocardial reperfusion significantly increased in the group that the coronary stenosis was greater than 70% after PCI( P ＜0. 05). There was no change in the group that the coronary stenosis was less than 70% ( P ＞ 0.05). Conclusions PCI can efficiently improve the myocardial microcirculation. MCE can accurately, rapidly, and efficiently reveal the reperfusion status of myocardium after PCI.

BACKGROUND: Thrombotic diseases always accompany with blood rheological abnormality. Whether allicin has effects on hemorheological indexes and physiological characteristics of erythrocyte needs to further study.OBJECTIVE: To investigate the effect of allicin on hemorheological indexes and physiological characteristics of erythrocyte in rats with acute blood hyperviscosity.DESIGN: Purely randomly controlled animal study.SETTING: College of Food Science and Nutritional Engineering, China Agricultural University.MATERIALS: A total of 50 Wistar rats of half gender, weighing (200±20) g and aged from 3 to 4 month were provided by Beijing Haidian Tongli Experimental Animal Cultivation Center. Allicin solution was provided by Shanghai Hefeng Pharrnaceutical Co., Ltd. (cencentration: 15 g/L; batch number: 20051001). The main equipments were detailed as follows: NXE-1 cone-plate viscometer (Chengdu Device Factory), DXC-300 nuclear pore membrane erythrocyte deformation radiometer (Device Factory Affiliated to Shanghai Medical University), 650-60 fluorescence polarization (Hitachi Company, Japan) and trace high speed centrifugation (Huaxing Aviation Aircraft Company, Aerospace Faulty).METHODS: The experiment was carried out in the Laboratory of College of Food Science and Nutritional Engineering,China Agricultural University from February to April 2006. All rats were randomly divided into 5 groups, including control group, model group, low-dosage allicin group, moderate-dosage allicin group and high-dosage allicin group, with 10 in each group. Except control group, rats in other 4 groups were perfused with 5 mL tap-water every day fcr 7 successive days. On the seventh day, rats were subcutaneously injected with 0.1% 0.8 mL/kg adnephrin; 2 hours later, rats were dripped in iced water for 5 minutes; 4 hours later, the rats were injected again to establish models of acute blood hyperviscosity. Two days later, all rats in the control group and model group were intraperitoneally injected with saline hematocrit and erythrocyte aggregation index were measured with viscosity determination method; erythrocyte filtration index was measured with nuclear pore membrane flow rate method; erythrocyte membrane fluidity (erythrocyte fluorescence polarization) was measured with fluorescence polarization technique; erythrocyte osmotic fragility test (concentration of NaCl solution from the beginning hemolysis to complete hemolysis) was performed to compare various indexes among different groups.MATN OUTCOME MEASURES: ① Effect of allicin on erythrocyte aggregation (whole blood viscosity,plasma viscosity,hematocrit and erythrocyte aggregation index);② effect of allicin on erythrocyte deformability index (filtration index);③ effect of allicin on erythrocyte osmotic fragility (concentration of NaCl solution from the beginning hemolysis to complete hemolysis);④ effect of allicin on erythrocyte membrane fluidity (erythrocyte membrane fluorescence polarization).RESULTS: Among 50 rats, 8 rats did not have enough blood collection or died; therefore, other 42 rats were involved in the final analysis. ① Effect of allicin on erythrocyte aggregation: As compared with that in the model group, whole blood viscosity was decreased 14.43%, 9.71% and 4.15% in high-dosage, moderate-dosage and low-dosage groups,respectively (P ＜ 0.05-0.01); meanwhile, whole blood viscosity of low shear rate was decreased 15.89%, 14.27% and 6.86%, respectively (P ＜ 0.01), and plasma viscosity was decreased 11.33% (P ＜ 0.05), 9.78% (P ＜ 0.05) and 3.49% (P＞ 0.05), respectively. And the effects mentioned above were shown a dose-effect relationship. Allicin in a high dosage could obviously decrease hematocrit (P ＜ 0.05) and the decreasing degree was 6.47%.② Effect of allicin on erythrocyte deformability index: Filtration index was higher in the model group than that in the control group (0.228±0.025, 0.175±0.043, P ＜ 0.05); however, filtration index was lower in the high-dosage allicin group (0.176±0.034) than that in the model group (P ＜ 0.05). ③ Effect of allicin on erythrocyte osmotic fragility: Concentration of NaCl was higher in the model group than that in the control group at the beginning of hemolysis and at the phase of complete hemolysis. Moreover, allicin in various dosages could improve the increase of erythrocyte osmotic fragility, but there were no significant differences among different dosage groups. ④ Effect of allicin on erythrocyte membrane fluidity: Erythrocyte membrane fluorescence polarization was increased in the model group, and this suggested that membrane fluidity was decreased. Allicin could improve the decrease of erythrocyte membrane fluidity, but there were no significant differences among various dosage groups.CONCLUSION: Allicin can raise erythrocyte deformability, decrease blood viscosity, improve blood fluidity, prevent and cure blood stasis syndrome.

Objective To investigate the influence of different scout image on the radiation dose of head and chest spiral scanning in CT using automatic tube current modulation(ATCM).Methods CT scanning was performed on the head-neck and chest phantom with ATCM.Five different scout images through the five various positions that include anteroposterior(AP),posteroanterior(PA),lateral,AP,lateral,PA and lateral.The phantom was scanned three times for each position.Then each scout image was scanned by spiral technique once more.ROI in orbital center and C5 upper edge level were selected for Head and neck phantom,ROI in the apical and tracheal bifurcation level were selected for chest phantom.The contrast to noise ratio (CNR) was measured and recorded.The organ dose of eye lens and mammary glands were measured by thermoluminescent dosimeters (TLD) in every scanning(the average of 3 measurements),The cumulative value of scout image and spiral scanning were calculated.The volume CT dose index (CTDIvol) of each scan was recorded,and the cumulative value of CTDIvol was calculated.Results In the five scout images of the head-neck phantom mode,the maximum value of accumulated radiation dose of eye lens and CTDIvol appear on PA scout image(18.354 and 26.43 mGy respectively),while the minimum value appear on the lateral scout image(11.847 and 18.20 mGy respectively).In the chest phantom mode,the maximum value of the accumulated radiation dose of the mammary gland and CTDIvol emerge from the AP scout image (6.873 and 9.42 mGy respectively),while the minimum value of the mammary accumulated radiation dose appear on lateral scout image(4.592 mGy),the minimum value of CTDIvol appear on AP plus lateral scout images(3.94 mGy).The accumulated radiation dose of eye lens and CTDIvol value in PA scout image mode were 54.9％(6.507/11.847) and 45.2％(8.23/18.20) higher than those of the lateral scout image mode.The accumulated radiation dose of mammary gland and CTDIvol value in AP scout image mode were 42.5％ (2.051/4.822) and 136.7％ (5.44/3.98) higher than that of PA plus lateral scout images mode.In the head-neck phantom mode,the CNR value of orbital center and C5 upper edge level were 102.55 to 115.89,161.01 to 204.52 respectively.In the chest phantom mode,the CNR value of the apical and tracheal bifurcation level were 82.74 to 164.00,83.12 to 121.49 respectively.Conclusion The choice of the scout image had significant effect on the radiation dose and the sensitive organ dose in CT.

Medicine is related to people's wellbeing,which is an eternal topic that all mankind concern.Studying on the process from the origin to the development of medicine can figure out the development rules of medicine,gain experience from it,and provide guidance for better development of medicine in the future.Medical anthropology is a new discipline which focuses on the study of disease and culture.From this special perspective,studying on the process from the origin to the development of medicine can help us to have a profound and comprehensive understanding of medicine.This paper reviewed the relevant concepts of medical anthropology and the research status and research characteristics of the origin and development of Chinese and Western Medicine,providing a reference for the colleagues.

OBJECTIVE To investigate the relationship between state trait anxiety and behavioral status in patients with sudden deafness. METHODS From April 2015 to June 2016, 140 cases of hospitalized patients with sudden hearing loss were selected as the observation group, and 140 healthy volunteers in our hospital for medical examination were selected as the control group. Average data, state trait anxiety, quality of life, and behavior survey were investigated in the two groups and correlation analysis was conducted. RESULTS The state trait anxiety score of the observation group was 51.20±4.19 points, and the control group was 34.40±3.49 points, and the state trait anxiety score of the observation group was significantly higher than that of the control group(t=12.949, P<0.05). The scores of physical, compulsive, interpersonal sensitivity, hostility and depression in the observation group were significantly higher than those in the control group(P<0.05). The problem solving and help scores in the observation group were significantly lower than that of the control group, while the fantasy and self accusation scores was significantly higher than that of the control group(P<0.05). In the observation group, Pearson correlation analysis shows anxiety scores were significantly correlated to the dimensions scores of quality of life, fantasy and self accusation scores(P<0.05), and were significantly negative correlated to the problem solving and help scores (P<0.05). CONCLUSION The patients with sudden deafness is accompanied by state trait anxiety and fantasy, self accusation and other bad behavior, it can seriously affect the patient's quality of life and form vicious circle, so the nursing intervention should be strengthened in clinical practice.

β-Catenin is an adhesion molecule that mediates intercellular adhesion.It can be activated by the classical Wnt signaling pathway and promote cell survival.During cerebral ischemia,the stability of β-catenin decreases and the phosphorylated β-catenin level increases.Up-regulation of β-catenin can inhibit the schemia-induced neuronal apoptosis,promote neurogenesis,maintain the stability of blood-brain-barrier,and protect against cerebral injury.

OBJECTIVE:To evaluate the therapeutic efficacy of oxygen-driven aerosol inhalation of Ventolin plus Atrovent and Budesonide suspension for acute episode of intermediate or severe degree of bronchial asthma. METHODS:A total of 48 patients with bronchial asthma were divided into two groups randomly:26 cases in the treatment group received normal saline (1 mL) +Ventolin (2 mL) + Atrovent (2 mL) and Budesonide suspension(4 mL) q6 h in addition to the routinetherapy by high-flow oxygen driven in addition to the routine therapy; 22 cases in the control group received Ventolin inhaler(200 ?g) q 6 h. The improvement of clinical symptoms in the two groups after treatment for 3 days,the forced expiratory volume in one second (FEV1),forced vital capacity (FVC),peak expiratory flow (PEF) at 3 h and 5 days,respectively,the blood sugar level,arterial blood gas index as well as duration of systemic use of hormone and the hospital days were compared between two groups. RESULTS:After treatment for 3 days,the effective rate in the treatment group(24/26) was significantly higher than in the control group (14/22),showing significant differences between two groups. At 3 h,there were significant differences between the treatment group and the control group in FEV1 and PEF but not in FVC; at 5 days,there were significant differences between the treatment group and the control group in FEV1,FVC and PEF. On day 5,there were significant differences between two groups in PaO2,PaCO2 and blood sugar level as well as in the duration of systemic use of hormone and the hospital days. CONCLUSION:Aerosol inhalation of Ventolin plus Atrovent and Budesonide suspension shows remarkable therapeutic efficacy for acute episode of intermediate or severe degree of bronchial asthma.
