1.Inhibitory role of epigallocatechin-3-gallate in proliferation of human na-sopharyngeal carcinoma cells by targeting P53/miR-34a

AIM:To study the effect of epigallocatechin-3-gallate (EGCG) on the proliferation of human naso-pharyngeal carcinoma ( NPC) cells, and to explore its mechanism by targeting miR-34a.METHODS: Nasopharyngeal carcinoma CNE-2Z cells were treated with various concentrations of EGCG .The ability of cell proliferation was detected by CCK-8 assay, 5-ethynyl-2-deoxyuridine (EdU) incorporation assay and colony-forming assay.The cell cycle distributions were analyzed by flow cytometry .The protein levels of P53 and Notch1 were detected by Western blot .The expression of miR-34a and Notch1 mRNA was measured by real-time PCR.RESULTS:EGCG effectively inhibited the proliferation and colony formation of CNE-2Z cells in a dose-dependent manner , which was related to its induction of cell cycle arrest at G 0/G1 phase.The expression of P53 and miR-34a in CNE-2Z cells was significantly increased after treated with EGCG , while the expression of Notch1 at mRNA and protein levels was markedly suppressed .CONCLUSION:EGCG induces cell cycle arrest and suppresses cell proliferation by regulating the P 53/miR-34a/Notch1 pathway in NPC cells.

The etiology of deep vein thrombosis (DVT) is still not elucidated nowadays. Based on the accordance between DVT incidence and the anemophilous pollen concentration in the air, we proposed the hypothesis that allergic reaction induced by anemophilous pollen may cause "idiopathic" DVT, and proinflammatory factors may play an important role in the thrombosis process.
3.A preliminary study on the MATP gene knockout in a mouse melanoma cell line using CRISPR-Cas9 system

Objective To knockout the MATP gene of mouse melanoma cell line B16F10 using CRISPR/Cas9 system,and to lay foundation for the functional study of MATP gene.Methods Specific primers of MATP were designed according to the report in http://crispr.mit.edu/ website.The primers were linked to pCAS9/gRNA1 vector.Then the positive vector was transfected into mouse melanoma B16F10 cells,and monoclonal cell lines were obtained by the infinite dilution method.After the genomes of different monoclonal cell lines were extracted and sequenced,the cell lines with MATP gene cleavage were screened,and the expression of MATP in these cell lines was verified by Western-blot analysis.Results Three MATP gene knockout cell lines were successfully obtained.The western-blot results showed that the cell lines did not express MATP protein.Conclusions The knockout of MATP gene in B16F10 cell line can be successfully achieved using the pCAS9/gRNA1 vector.
4.Effect of inferior vestibular and part of mandibular branch of trigeminal neurotomy on sound-induced masseter reflex potentials.

OBJECTIVE:
To evaluate the influence of inferior vestibular and part of mandibular branch of trigeminal neurotomy on sound-induced masseter reflex potentials.
METHOD:
Twenty guinea pigs were randomly divided into four groups, including 5 normal control guinea pigs, 5 received unilateral inferior vestibular neurotomy, 5 received unilateral inferior alveolar nerve neurotomy, and 5 received auriculotemporal nerve neurotomy. Click sound-induced masseter reflex potentials were recorded in four groups, respectively.
RESULT:
The thresholds of negative peak (NP) of click sound-induced masseter reflex potentials in normal control group were (90.00 +/- 8.16) dBnHL. The response rates of the NP of the masseter reflex potentials using 100, 90, 80 and 70 dBnHL monaural acoustic stimulation with unilateral recording were 100%, 70%, 40% and 0, respectively. The mean latencies of the NP were (6.55 +/- 0.25) ms, (6.61 +/- 0.16) ms and (6.70 +/- 0.13) ms, when elicited by 100,90 and 80 dBnHL acoustic stimulation respectively. There was no significant difference between the stimulus intensity and the mean latency of the NP (P > 0.05). Negative peak of click sound-induced masseter reflex potentials was not observed in the inferior vestibular neurotomy group. The NP was preserved in the inferior alveolar nerve and auriculotemporal nerve neurotomy groups. There were no significant difference of the mean thresholds and latencies of NP between normal control group and inferior alveolar nerve and auriculotemporal nerve neurotomy group (P > 0.05).
CONCLUSION
Click sound-induced masseter reflex potentials originates from vestibular afferents. Afferent of inferior alveolar nerve and auriculotemporal nerve can not influence the vestibular evoked masseter reflex potentials.

5.DNA microarray screening analysis in children with profound hearing impairment in Hubei province.

OBJECTIVE:
To investigate characteristics of molecular etiology of children with profound sensorineural hearing loss in Hubei province, and to provide reference for deafness treatment and genetic counseling.
METHOD:
Three hundred and six children with profound sensorineural hearing loss in Hubei province were enrolled, their genomic DNA were extracted from peripheral blood and a deafness gene test chip was used to screen nine hot spot mutation in the GJB2, GJB3, SLC26A4, and mitochondria 12SrRNA gene. All patients with SLC26A4 gene mutation were given temporal bone CT scan.
RESULT:
One hundred and thirty-two (43.14%) out of 306 children were found carrying at least one pathogenic gene mutation. The mutation rates of GJB2, SLC26A4 and mitochondria DNA 12SrRNA gene were 29.41% (90/306), 13.72% (42/306) and 0.65% (2/306), respectively. None out of 306 children was detected GJB3 gene mutation. Thirty-six patients carrying SLC26A4 gene mutation were detected enlarged vestibular aqueduct by CT scan.
CONCLUSION
Mutations of GJB2 and SLC26A4 gene are two major pathogenic gene for genetic hearing loss in children. 235delC mutation is the main mutation type, followed by IVS7-2A> G mutation type. The screening of SLC26A4 gene common mutations contribute to the diagnosis of enlarged vestibular aqueduct syndrome.

6.Application of laparoscopic ultrasonography assistance in minimally invasive surgery for patients with uterine fibroids

Objective To investigate the effect of laparoscopic ultrasonography assistance in minimally invasive surgery for uterine leiomyoma patients and provide scientific basis for reducing the recurrence rate of uterine leiomyoma. Methods 156 cases of uterine leiomyoma from January 2011 to June 2014 were divided into control group and observation group according to the digital table method, 78 cases in each. The control group were treated with conventional laparoscopic surgery, while the observation group with laparoscopic ultrasonography assistance, then compare the postoperative residue, recurrence in 12 months at different time points and the number of fibroids diameter, analyze the relationship between number of uterine muscle tumor and residual recurrence. Results There were no significant differences in operation time, blood loss, length of hospital stay and anal exhaust time between the two groups. The proportion of patients in the observation group and the recurrence rate within 12 months were significantly lower than those in the control group, the number of uterine leiomyomas in the observation group was significantly lower than that in the control group at 3 months, 6 months, 9 months and 12 months after operation. The patients in the two groups were followed up for 3 months and 6 months the maximum diameter of uterine leiomyoma was not statistically significant, the observation group 9 months and 12 months after the maximum diameter of uterine fibroids was significantly lower than the control group, and the difference was statistically significant; with the patient's uterine muscle the number of residual tumor and the recurrence rate were significantly increased. When the number of uterine leiomyomas was 10 or more, the residual rate was 100.0% and the recurrence rate was 80.0%. Conclusion Laparoscopic ultrasonography assistance in minimally invasive surgery for patients with uterine fibroids can effectively reduce the postoperative residual rate and recurrence rate, worthy of clinical promotion.
7.Diagnosis and treatment for the corticomedullary mixed tumor of adrenal gland.

OBJECTIVETo study and summarize the diagnosis and treatment for the corticomedullary mixed tumor of adrenal gland.
METHODSThe clinical data of 25 cases of adrenal corticomedullary mixed tumor from January 2000 to April 2008 were analyzed retrospectively, which including 9 males and 16 females. The ages were from 25 to 60 years old, and the average age was 39 years old. Thirteen cases had paroxysmal hypertension and 11 cases had central obesity, as well as 8 cases with hypokalemia. There were different degree abnormalities in plasma endocrine hormones in laboratory examination. Every case underwent b-ultrasound and CT normal plus extensive scan to make the diagnosis.
RESULTSAdrenalectomy was performed in the 25 cases, which contain 9 cases of open operations and 16 cases of endoscopic adrenalectomies. All of the cases had blood pressure fluctuation during dissection of the adrenal tumors, with the highest blood pressure reached to 230/140 mm Hg (1 mm Hg = 0.133 kPa). Postoperative histopathological study revealed that the pathological changes was corticomedullary mixed tumor of adrenal gland, which was supported by immunohistochemical study.
CONCLUSIONSIn cases with complex phenomenon that can't explain with single cortical or medullary changes, it must beware of the mixed pathological changes in adrenal gland.

8.Parapelvic cyst of kidney.

OBJECTIVETo study the clinical features, treatment and diagnosis of parapelvic cyst.
METHODSTwenty-three patients of parapelvic cyst of the kidney were reviewed retrospectively. Fourteen cases (61%) complained of lumbar pain or discomfort, and 4 patients (17%) accompany hematuria and hypertension.
RESULTSIn 15 patients receiving surgery, 2 were treated by nephrectomy, one by radical nephrectomy for misdiagnosis. Postoperative diagnosis confirmed a cyst. Eight patients were treated conservatively for cyst being small and without clinical symptoms. Nineteen cases were followed up for 0.5 - 12.0 years.
CONCLUSIONSUltrasonography and CT scan are the main diagnostic methods. Enhanced CT is extremely helpful in differential diagnosis of hydronephrosis. Surgical management is suitable for big cysts, lumbar pain, hematuria, hypertension and other complications.
