Objective To examine expression of PTEN gene in ovarian cancer cisplatin-sensitive cell line OV2008 cells and cisplatin-resistant cell line C13K cells,and evaluate the effect of wild-type PTEN gene on reversing cisplatin-resistance of C13K cells and underlying mechanisms.Methods The expression of PTEN mRNA and protein in OV2008 and C13K cells were detected by semi-quantitative RT-PCR and western blot.Recombinant eukaryotic expression plasmid containing human wild-type PTEN gene was transfected into C13K cells by lipofectamine 2000.The expression of PTEN mRNA was monitored by RT- PCR and the expression of PTEN,protein kinase B(AKT),phospho-AKT(p-AKT)protein were analyzed by western blot in PTEN transfected and untransfected C13K cells.Proliferation and chemosensitivity of cells to cisplatin were measured by methyl thiazolyl tetrazolium(MTT),and cell apoptosis was detected by flow cytometry after treatment with cisplatin.Results(1)The expression of PTEN mRNA and protein(1.02 ?0.05,1.02?0.07)in OV2008 cells were significantly higher than those in C13K cells,which were 0.45 ?0.03 and 0.55?0.03 respectively(P

Nowadays,oil-pollution of seawater in the world has severely threatened the security of sea entironment.Bioremediation offers one available option for an oil spill response.The aspects as follows are introduced some evolvement of microbial ecology,including new method of survey of microbial diversity without cultivation,new isolated method and the properties of main hydrocarbon degradated strain.But we have little or no understanding of the vast majority of marine bacteria that remain uncultured,and more efforts should be made to improve current methods for isolating oil-degrading or oil-emulsifying bacteria,not only for assessing the fate and effects of the spilled oil,but also for isolating novel bacteria that would be useful for the petroleum industry.
5.Stem cell factor promotes the proliferation and osteogenic differentiation of stem cells from human exfoliated deciduous teeth.

OBJECTIVETo investigate the effect of stem cell factor on the proliferation and osteogenic differentiation of human deciduous dental pulp stem cells.
METHODHuman dental pulp tissues were harvested from extracted deciduous teeth and digested by collagenase and dispase. The stem cells from human exfoliated deciduous teeth (SHED) obtained were cultured in the presence of 3 or 10 µmol/L stem cell factor, and the proliferation of the cells was assessed by MTT assay. The influence of stem cell factor on alkaline phosphatase (ALP) activity was evaluated using ALP kit. Bone sialoprotein and osteocalcin mRNA expression in the treated cells were examined by real-time PCR.
RESULTMTT assay indicated that both 3 and 10 µmol/L stem cell factor promoted the proliferation of SHED. Stem cell factor enhanced ALP activity in the SHED, and the effect was more obvious at 10 µmol/L. Treatment of the cells with stem cell factor up-regulated the mRNA expressions of bone sialoprotein and osteocalcin.
CONCLUSIONStem cell factor can promote the proliferation and osteogenic differentiation SHED, suggesting the effect of stem cell factor in promoting tooth regeneration.

6.Study on two strains of mouse developing dampness-heat models infected by Dengue virus.

OBJECTIVETo establish two strains of mouse developing dampness-heat syndrome models infected by Dengue virus and to compare the difference in infection, so as to choose a suitable mouse strain for modeling.
METHODSAccording to the modeling methods of the seasonal febrile disease of the dampness-heat syndrome in Chinese medicine, BALB/C and C57BL/6 mice were respectively treated with complex factors as the high glucose and high fat forage + high-temperature chamber + Dengue virus. At the same time a normal control group, the virus infection group (modeled by Dengue virus infection), and the dampness-heat group (modeling by pure dampness heat circumstance) were set up. Changes of the body temperature, platelet counts, virus in the separate serum, pathological changes of the liver, and serological indicators were observed to compare the modeling difference.
RESULTSAfter modeling low-grade fever appeared in mice in the high-temperature chamber. Compared with the normal control group, the platelet count decreased in mice of the BALB/C model group. AST increased in both BALB/C and C57BL/6 mice and the virus infection group. TC and TG increased in BALB/C model group and the dampness-heat group, with statistical significance (P<0.05). Various degrees of pathological changes were shown in the liver tissue of each group, with the most severe one in the BALB/C model group. The serum virus titers were detected with Real-time PCR after modeling. The virus load was 2.9 x 10(4) - 5.5 x 10(4) copies/mL. No significant difference was found among these groups.
CONCLUSIONSThe mouse model of dampness-heat syndrome infected by Dengue virus was primarily established. When compared the infection between BALB/C mice and C57BL/6 mice, BALB/C mice were more suitable for modeling.

Hemolytic diseases of newborn (HDN) can cause miscarriage, premature birth, fetal edema, fetal intrauterine anemia and even fetal death in early pregnancy. Neonatus with HDN can have jaundice, anemia, hepatosplenomegaly, edema and nuclear jaundice sequelae. This article reviewed the diagnosis and treatment of two patients with HDN caused by anti-E and anti-Ec combined with anti-Fyb, and reviewed the relevant literature on the epidemilogy, the diagnosis and treatment of HDN in order to improve the understanding of the disease.
9.Cooperation of reconstruction of breast and anterior axillary fold with island latissimus dorsi musculocutaneous flap

Objective To study nursing cooperation of second-stage reconstruction of breast and anterior axillary fold with island latissimus dorsi musculocutaneous flap after Radical Mastectomy. Methods Thirteen patients with severe defect of thoracic wall underwent second-stage reconstruction of breast and anterior axillary fold with island latissimus dorsi musculocutaneous flap and breast implants 1 ~ 4 years after the Radical Mastectomy for breast cancer. The nursing cooperation including preoperative preparation of staffs, surgical instruments, and articles, and methods of intraoperative nursing cooperation were retrospectively analyzed.Results Procedure success was gained in all 13 cases, without operation beingg delayed because of surgicalinstruments or poor cooperation. Flaps survived intraoperatively and postoperatively. Primary healing of incision without any infection was observed for all patients. Satisfactory mammary contour was achieved for all patients uneventfully with absence of any complications. Conclusions Careful preoperative preparation of articles and psychological nursing to gain best cooperation of the patients, and skilled intraoperative nursing cooperation are essential for procedure success.
10.Effects of interferon-γ on biological characteristics and immunomodulatory property of human umbilical cord-derived mesenchymal stem cells.

The aim of this study was to investigate the effects of interferon (IFN)-γ on biological characteristics and immunomodulatory property of human umbilical cord-derived mesenchymal stem cells (hUC-MSC). hUC-MSC were treated with IFN-γ 10 ng/ml (IFN-γ group) or without IFN-γ (control group). The phenotype of hUC-MSC was detected by flow cytometry. The proliferation status was detected by CCK-8 method, and its differentiation ability was assessed by oil red O and von Kossa staining. The production of PGE-2 was measured by ELISA, and the mRNA expression levels of COX-2, IDO-1 and IDO-2 in hUC-MSC were detected by real-time quantitative PCR. Furthermore, the proliferation of human peripheral blood mononuclear cells (hPBMNC) was evaluated after co-culture with hUC-MSC, IFN-γ pretreatment or not. The results showed that after IFN-γ stimulation, the expression of SSEA-4 on hUC-MSC decreased significantly [(8.15 ± 2.94) vs (16.42 ± 8.5), P < 0.05], and the expression of CD54 increased [(96.64 ± 3.29) vs (84.12 ± 10.73), P = 0.051]. The immunomodulatory property of hUC-MSC on the proliferation of hPBMNC was enhanced (P < 0.05). All the above mentioned effects were IFN-γ concentration-dependent. When hUC-MSC were stimulated by IFN-γ for 24 h, the production of PGE-2 secreted by hUC-MSC decreased significantly (P < 0.01). The mRNA expression level of COX-2 also decreased though the difference did not reach to statistically significant level. Compared with control group, IDO-1 expression level in IFN-γ group increased significantly (P < 0.01), and the mRNA expression level of IDO-2 remained unchanged. It is concluded that IFN-γ can influence the phenotype of hUC-MSC and enhance the immunomodulatory property of hUC-MSC.
