3.Hepatic bile acids and the expression of FXR,SHP,and Cyp7α1 in normal rats during pregnancy and lactation

Objective To investigate the accumulation of liver bile acids and bile acid regulated nuclear receptor farnesoid X (FXR) and its target gene cholesterol 7α hydroxylase (Cyp7α1) and small different dimer partner (SHP) in normal rats during pregnancy and lactation .Methods SD rats were maintained in the SPF‐grade facilities for time‐controlled mate .Livers were collect‐ed at Gestation Day (GD) 10 ,14 ,and 19 and Postnatal Day (PND) 1 ,7 ,14 and 21 d .Hepatic bile acids were quantified with a total bile acid assay kit ,and mRNA expression of FXR ,Cyp7α1 and SHP was determined by RT‐PCR .Western blot was used for Cyp7α1 protein quantification .Results In livers of normal rats ,bile acids was not elevated during pregnancy but was increased during lacta‐tion .The bile acid synthesis rate limiting enzyme Cyp7α1 increased during lactation but were normal during pregnancy .SHP was 6 fold higher at the late pregnancy ,and slightly higher during lactation .FXR was increased at postpartum .Conclusion Under normal circumstances ,hepatic bile acid synthesis during pregnancy was controlled by SHP feedback ,while during lactation ,hepatic bile acids increased ,along with increased Cyp7α1 and FXR .
4.The impact of aerobic combined strength and balance exercise on cognitive function in patients with cognitive impairment no dementia

Objective To evaluate the effects of aerobic combined strength and balance exercise on cognitive function and satisfaction in patients with cognitive impairment no dementia. Methods Four hundred and twenty patients with cognitive impairment no dementia were divided into control group (200 cases) and experimental group (220 cases) by random digits table method. The control group received general health education and rehabilitation training. The experimental group received aerobic combined strength and balance exercise. The patients were assessed with Montreal Cognitive Assessment (MoCA) to evaluate their cognition before training, as well as after training. And the patients′satisfaction with the questionnaire was evaluated. Results The total scores of MoCA, scores of name, attention, language, memory and directionafter training were (23.47 ±2.38), (2.77 ±0.42), (5.09 ±0.86), (2.50 ±0.65), (3.42 ±0.68), (5.03 ± 0.10) points in the experimental group, and (21.20±2.55), (2.31±0.76), (4.71±1.10), (2.35±0.70), (2.23±0.81), (4.48±0.96) points in the control group, and there were significant differences, P<0.05. There were no significant differences in the space and abstract thinking between experimental group and control group:(3.61±0.91) points vs. (3.45±1.09) points, (1.83±0.76 ) points vs. (1.72±0.52) points, P>0.05. The scores of satisfaction in the experimental group was significantly higher than that in the control group:(49.33 ±1.57) points vs. ( 48.20 ±2.14) points, P<0.01. Conclusion The application of aerobic combined strength and balance exercise could improve the cognitive function and patients′satisfaction.
5.Relationship between homocysteine metabolism abnormality and pre-clampsia

Objective To study the serum levels of homocysteine(Hcy) and the polymorphism of C677T mutation of methylenetetrahydrofolate reductase (MTHFR) gene in pregnant women to explore their role in the pathogenesis of pre-clampsia. Methods Fifty pregnant women with preclampsia (study group) including 14 mild pre-eclampsia and 36 severe pre-eclampsia and 40 healthy ones (control group) were selected. Hey concentration was determined by fluorescence polarization immunoassay. The mutations of the 677C→T and 1298A→C of MTHFR were investigated by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results The mean level of plasma Hey in the study group was higher than that of control 1(12. 00±4. 59) μmol/L vs (7. 85:1:1.51) μmol/L, P<0.05], and that of the severe pre-eclampsia group was higher E(13. 30±2. 06) μmol/L, P<0. 01]. However, no significant difference was found between the control and the mild pre-clampsia group[(8. 63±3. 94) μmol/L, P>0.05]. There was significant difference in the genotype frequencies of MTHFR 677CT, TT genotype(C/T:42.0% ;T/T:14. 0%)and the total frequency of mutant T (T: 35.0%) allele between the study and control groups (P < 0. 05). Conclusions Hyperhomocysteinemia plays an important role in the pathogenesis of pre-clampsia. The polymorphism of MTHFR C677T might be associated with the development of pre-clampsia as it affects the metabolism of Hey.

8.Meta-analysis on the randomized controlled trials of breviscapine injection plus conventional therapy for unstable angina pectoris

Objective To systematically evaluate the clinical effectiveness and safety of Breviscapine injection combined with conventional therapy in the treatment of unstable angina pectoris (UAP).Methods The literature were systematicly retrieved about the randomized controlled trials (RCTs) of Breviscapine injection for UAP. The Cochrane Risk of Bias Assessment Table was used to evaluate the methodological quality of the RCTs and then the data were extracted and meta-analysed by RevMan 5.3 software.Results A total of 15 RCTs with 1202 participants were included. In the meta-analysis, the combination of Breviscapine injection and conventional therapy in the treatment of UAP can achieve better angina pectoris curative effect (Z=7.74,P<0.01), theRR(95% CI) was 1.23 (1.17,1.30), and the electrocardiogram curative effect (Z=6.26,P<0.01), the RR(95% CI) was 1.32(1.21, 1.44). Furthermore, Breviscapine injection can improve hemorheologic parameters. And there was no major or serious adverse drug event.Conclusions The Breviscapine injection combined with conventional therapy shows good therapeutic effect on UAP.
9.Levonorgestrel-releasing intrauterine system for treatment of dysmenorrhea associated with adenomyosis

Objective To evaluate the efficacy and safety of a levonorgestrel-releasing intrauterine system(LNG-IUS)for the treatment of dysmenorrhea associated with adenomyosis.Methods We recruited 48 women with moderate or severe dysmenorrhea associated with adenomyosis.All women were inserted of LNG-IUS into their uterine cavity from days 5-7 of their periods and maintained for 12 months.We compared the visual analogue scale(VAS)scores and verbal rating scale(VRS)scores of their dysmenorrhea and dyspareunia at baseline and 12 monthes follow-up.Results Forty-four women completed the study. There were significant differences between mean VAS and VRS scores changes of dysmenorrhea and dyspareunia at baseline and 12 monthes follow-up,those of dysmenorrhea dropping from 75?13 to 11?11 and 2.3?0.4 to 0.4?0.3,those of dyspareunia dropping from 54?19 to 4?4 and from 1.6?0.8 to 0.2?0.2 respectively.Overall 29 women(66%)were very satisfied or satisfied with the one-year treatment. Conclusion Insertion of LNG-IUS alleviates moderate or severe dysmenorrhea associated with adenomyosis remarkably.
10.Role of renal cortex endothelin and NO in the development of proteinuria

AIM: To test the hypothesis that locally produced renal endothelin(ET) and nitric oxide(NO) play more important role in initiating proteinuria than circulating ET and NO. METHODS: (1) Rat nephrotic syndrome model was made by single injection of adriamycin, and the urinary protein excretion per 24 hours for every two days was measured. (2) Rat plasma and renal cortex were collected respectively 4 days, 8 days, 32 days and 56 days after the injection, and renal cortex homogenate was further prepared; ET of both plasma and renal cortex homogenate were determined by radioimmunoassay; NO of both plasma and renal homogenate were measured by the method of Griess. (3) The correlation between urinary protein and ET or NO concentration was analyzed respectively. RESULTS: (1) The urinary protein excretion of normal rat was 4-7 mg/d, that in nephrotic rat at the 4 time points was 5, 10, 241 and 201 mg/d, there was a significant increase of that at day 8, peaked at day 32, and less decrease at day 56. (2) The plasma ET in nephrotic rats of the 4 time points was 134, 150, 538 and 445 ?g/L, respectively, which of day 8, day 32 and day 56 shown significant higher than that of normal control (126-129 ?g/L). (3) The ET in renal cortex homogenate of nephrotic rat of the 4 time points was 364 6, 652 3, 1 526 8, 1 393 6 pg/g, all of that was much more higher than that of normal rat [(235 9-246 1) pg/g,P
