AIM: To study the effect of cigarette smoke medium(CSM) on the gene expression and activity of gelatinases from alveolar macrophages(AMs) in the rat,and then to explore their role in the pathogenesis of chronic obstructive pulmonary disease(COPD).METHODS: AMs were obtained from BALF of the rats that had smoked for 12 weeks.CSM was produced following the method of Wirtz and colleagues,and the cultured AMs were respectively stimulated for 24 h by 0%,1%,3%,5%,10%, 15% CSM.The mRNA levels of MMP-9 and MMP-2 were detected by semi-quantitative RT-PCR,and the enzyme activity was measured by Zymography.RESULTS: When the concentration of CSM was below 5%,the expression and activity of MMP-9 and MMP-2 signficantly increased with the concentration of CSM in a dose-depended manner(P
2.Exploration into rules of combined Chinese and Western medical treatment on immune infertility.

In order to explore the rules of combined Chinese and Western medical treatment on immune infertility, the study was carried out by searching relative primary documents from databases and 26 articles (dealing with 5865 cases) were screened out. Excel was used to perform the frequency analysis on the Western drugs and 27 Chinese recipes emerging in the documents separately. It was discovered that the combined use of Chinese and Western medicines has its superiority. Low dose glucocorticoids together with vitamine is the main Western treatment used, and dexamethasone is the most frequently used preparation of glucocorticoids. Among the 72 Chinese drugs presented in the 27 Chinese recipes, 13 appeared for more than 1800 times, they were Angelica sinensis, Salvia miltiorrhiza, Radix Paeoniae Rubra, Radix Astragali, Poria, Carthamus tinctorius, Phellodendron amurense, Scutellaria baicalensis, Anemarrhena asphodeloides, Rehmannia glutinosa, Cuscuta chinensis, Radix Paeoniae Alba and Radix Glycyrrhiza.

3.Comparison of therapeutic effects of peripheral facial paralysis in acute stage by different interventions.

OBJECTIVETo compare the therapeutic effects of peripheral facial paralysis in acute stage by different interventions and explore the better treatments of peripheral facial paralysis.
METHODSOne hundred and thirty one cases of Bell's facial paralysis were randomly divided into three groups. In acupuncture group (44 cases), Dicang (ST 4), Jiache (ST 6), Hegu (LI 4), Yangbai (GB 14) and Taiyang (EX-HN 5), etc. were applied; in electroacupuncture group (45 cases), the selection of acupoints and needling method were same as those in acupuncture group, and the electroacupuncture therapy was applied on Dicang (ST 4), Xiaguan (ST 7), Yangbai (GB 14) and Taiyang (EX-HN 5) in acute stage; in medication and acupuncture group (42 cases), Prednisone and Acyclovir were taken by oral administration, Vitamin B1 and Vitamin B12, were applied by intramuscular injection in acute stage, and acupuncture was applied by the way which was same as that in acupuncture group during quiescent and recovery stages. The curative effects were evaluated by House-Brackmann Grading Scale, and the failed rates were observed by follow-up after one and three months.
RESULTSThe cured and markedly effective rates were 79.6% (35/44), 93.4% (42/45) and 78.6% (33/42) respectively in acupuncture group, electroacupuncture group and medication and acupuncture group, and the result in electroacupuncture group was superior to those in acupuncture group and medication and acupuncture group (P < 0.05). The cured rates above tympanichord were 54.2% (13/24), 85.2% (23/27) and 48.0% (12/25) in acupuncture group, electroacupuncture group and medication and acupuncture group, and the result in electroacupuncture group was superior to those in acupuncture group and medication and acupuncture group (P < 0.01). There was no significant differences of cured rates below tympanichord among three groups (P > 0.05); and the failed rate in electroacupuncture group was much lower than those in acupuncture group and medication and acupuncture group by follow-up after one and three months (all P < 0.01).
CONCLUSIONThe peripheral facial paralysis is effectively treated by electroacupuncture in acute stage, and it suggests that electroacupuncture should be applied early during the acupuncture treatment of peripheral facial paralysis.

Abstract:
The demand for reliable toxicological data of chemicals runs through every link of occupational health work. The
prevention of occupational diseases involves high requirements for the standardization of chemical toxicity assessment in
occupational health institutions. Good laboratory practice (GLP) emphasizes the integrity of the test process to trace and
supervise the whole process of the test, which is conducive to the standardization of chemical toxicity identification. Therefore,
the standardized construction of GLP laboratories is an important starting point for occupational health institutions to carry out
chemical toxicity identification. In the construction and management process of GLP laboratories for chemical toxicity
identification, occupational health institutions need to build a sound organization and operation system, carry out systematic
training and assessment of personnel, establish standard operating norms and emphasize their importance, strengthen the
management of facility environment and laboratory, pay attention to quality control and process supervision, and constantly
improve their own ability level. To actively adapt to social development and market demand, to provide strong support for
occupational health work.
5.Protoplasts isolation, purification and plant regeneration of Pinellia cordata.

The main factors which affected the isolation, purification and cultivation of Pinellia cordata protoplasts from leaves were studied. The results indicated that the optimum enzyme solution for P. cordata leaves was 13% CPW + 1.0% Cellulose +0.1% Pectolase, at pH 6.0, temperature (25-28 degrees C ) for 4 h. The sucrose density gradient centrifugation was adopted to purificate the protoplasts collected, when 25% sucrose was used as mediator, centrifugating at 500 rpm for 10 min. When the protoplasts were shallow liquid and liquid-solid double layer cultured on the medium of MS + 0.5 mg x L(-1) 6-BA + 0.25 mg x L(-1) NAA + 13% mannitol at the density of 2.5 x 104 protoplasts/mL, or fed and nursed cultured at the density of 100-500 protoplasts/mL, cell division could be observed for 3 days; granular calli appeared for 30 days. Calli was proliferated on the medium of MS + 0.5 mg x L(-1) 6-BA + 0.25 mg x L(-1) NAA solidified by 0.55% agar, and differentiated and regenerated after 5-6 months. Plant generation of P. cordata is successfully established.

6.The Procaryotic Expression, Purification and Activity Analysis of VIP-sTNFRII

A prokaryotic expression plasmid containing VIP (vasoactive intestinal peptide) and sTNFRII(soluble tumor necrosis factor receptor II ) genes was constructed. The sTNFRII was cloned by PCR by using special primers which contained VIP gene ORF and a linker in its forward primer. The amplified fragment was inserted into the expression vector pET32a between BamHI and Hind III restriction sites. Transformed E.coli DH5 by pET32a-VIP- sTNFRIIexpressed the fusion protein. After being identified, the protein was purified by ion exchange chromatography and by hydrophobic interaction chromatography. The reconstructed protein showed high bio-activity and could be applied for further use.
7.Detection of quorum-sensing pathway and construction of luxS gene allelic exchange plasmid of Streptococcus mutans.

OBJECTIVETo detect the AI-2 quorum-sensing pathway and construct the luxS g-ene allelic exchange plasmid of Streptococcus mutans.
METHODSTo detect AI-2 pathway in Streptococcus mutans, the Vibrio harveyi BB170 was used as reporter strain. The PCR fragments of the upstream and downstream regions of luxS and the Erythromycin resistance gene were amplified with the primers respectively, and these fragments were ligated into pUC19 vector with double endonuclease reaction sequentially, the ligated DNAs were transformed into Escherichia coli DH5alpha, then the reconstructed plasmids were isolated and identified by restricted endonuclease digestions.
RESULTSStreptococcus mutans Ingbritt C could induce luminescence of BB170, suggesting the presence of AI-2 quorum sensing pathway in Streptococcus mutans, and such stimulatory activity was maximal at the mid-log growth phase. The recombinant plasmid pUCluxKO was digested by PstI-BamHI, and the digest product were 1000 bp and 5000 bp. When the pUCluxKO was digested by BamHI-KpnI, the digest product were 1500 bp and 4500 bp. While it was digested by KpnI-EcoRI, the digest product were 1000 bp and 5000 bp. All PCR product was in a single belt respectively.
CONCLUSIONSThe recombinant plasmid was cloned effectively and can be used in the construction of S.mutans luxS mutant.

8.Investigation of iodine nutritional status and thyroid function among pregnant and lactating women from rural areas of Jinlin province in 2009

Objective To investigate the iodine nutritional level and thyroid function of pregnant and lactating women in rural areas of Jilin province. Methods The investigation sites were selected from rural areas of three towns (Baoshan, Mingcheng and Yantongshan of Panshi county, Jilin province) in 2009. The pregnant and lactating women were selected as subjects in these three towns. The blood samples were collected and the thyroid function (including serum TT3, TT4, FT3, FT4) were measured with chemiluminescence, and serum thyroglobulin antibodies(TgAb), thyromicrosome antibody(TMAb), and thyroglobulin(Tg) were measured with radioimmunoassay (RIA). The urine samples were collected three times within one month and were measured for iodine concentration by As-Ce catalytic spectrophotometry method. Results In the pregnant women, serum TT3 was higher than that of healthy pregnant women, accounted for 14.3%(8/56), while serum TT4, TT3, FT4 were lower than those of healthy pregnant women, accounted for 3.6%(2/56),5.4% (3/56), and 1.8%(1/56), respectively. In the lactating women,serum TT3 was higher than that of healthy lactating women, accounted for 3.6%(2/56), while serum TT4, FT4 were lower than those of healthy lactating women, accounted for 1.8%(1/56), respectively. Five per cent to 20% of the pregnant and lactating women had higher TgAb and TMAb. Conclusions Existing salt iodine level is appropriate for pregnant women and lactating women, but there was a tendency towards hypothyroid in some women. Routine monitoring of urinary iodine and thyroid function should be carried out among pregnant and lactating women.
9.Prokaryotic expression and purification of the capsid protein of porcine getah virus and preparation of its polyclonal antibody.

Based on a pair of specific primers, a 804-bp fragment was amplified from the plasmid pT-Cap containing Cap gene of Porcine Getah Virus(PGETV) and cloned into the prokaryotic expression vector pCold I which carried the His tag, this recombinant plasmid was then determined by enzyme digestion, PCR and DNA sequencing. This recombinant plasmid pCold-Cap was transformed into E. coli Rosetta 2, and PGETV Cap fusion protein was expressed through IPTG induction. The results showed that the Cap gene obtained efficient and soluble expression in Rosetta 2 induced by 0. Immol/L IPTG under 15"C for 24h, the expression quantity was 40. 2%. The product had a molecular mass about 32. 3kD as expected. The target protein was separated in gel slices and used to immunize Balb/c mice. The polyclonal antibody with high titer against Cap protein specifically analyzed by Western blot was obtained. The successful preparation of the polyclonal antibody laid the foundation for the further study on the detection and identification of PGETV.
