1.Different methods for the diagnosis of endometrial histological comparative study

Objective To evaluate the feasibility of endometrial sampler Pipelle for endometrial histologic diagnosis.Methods Using prospective and self-control methods,200 patients with endometrial biopsy firstly used Pipelle endometrial sampler for endometrial tissue,then followed by diagnostic curettage,the same pathologist evaluated the specimen quality and made the histologic diagnosis.Results Totally 200patients completed the observation,the specimen satisfaction of Pipelle was 93.0% (in this 200 cases,186cases were satisfactory),its pathological accuracy was 85.0% (in this 200 cases,170 cases' pathological results are highly consistent with diagnostic curettage).There was no significant difference between two kinds of endometrial sampling (P>0.05).There was no pain for patients during the Pipelle using process.Conclusion Pipelle could obtain satisfactory samples used for histological diagnosis in normal endometrium,simple hyperplasia,complex hyperplasia,atypical hyperplasia and endometrial cancer disease,because its pathological accuracy is so close to the diagnostic curettage,which may be used as a routine screening tool of endometrial diseases.
2.Prevalence of HIV infection and sexual behaviors with both men and women among currently married men who have sex with men

Objective To find out the status of HIV infection,sexual behaviors with both men and women as well as condom use among currently married men who have sex with men (MSM) so as to document for HIV intervention targeting this sub-population,in Beijing,Chengdu,Harbin and Zhengzhou cities.Methods “Snowballing” sampling method was used to recruit subjects in four cities.Participants were investigated by anonymous questionnaire containing demographics,sexual orientation,both homosexual and heterosexual behaviors and condom use during the past 6 months.Blood samples from participants were used to test HIV antibodies.Results Of the 858 participants for four cities,the average age was 38.3 (SD=9.1) years.36.7% of them had completed the junior high school or under.The percentage of participants who identified themselves as homosexual,bisexual,and heterosexual or undecided were 40.9%,54.3% and 4.8%,respectively.The overall HIV prevalence was 8.0%,and among four cities the HIV prevalence in Chengdu was the highest ( 13.3% ).Results from the Multiple logistic regression analysis showed that never used condoms when having anal sex with men during the past 6 months among participants who did not have a city residential card,identifying himself as having homosexual orientation,living in Harbin,having first anal sex experience before 18 years of age the range of OR value was 1.5-2.1.In addition,the faetors associated with never using condoms in vaginal sex with women during the past 6 months were as follows:being over 46 years old,not having a city residential card,living in Beijing,identifying himself as heterosexual orientated or with unknown sexual orientation,never using condoms in anal sex with men during the past 6 months,the range of OR value was 1.7-5.9.Conclusion The prevalence of HIV infection seemed to be quite high among those currently married MSM.High rates of unprotected homosexual and heterosexual behaviors might accelerate the spreading of HIV from currently married MSM to their wives.
3.Mortierella isabellina lipid-soluble extracts improves lipid-induced renal injury in rats.

OBJECTIVETo investigate the effect of the Mortierella isabellina lipid-soluble extracts on improving hyperlipidemia and renal damage in lipid-induced renal injury rats.
METHODSThirty two male Wistar rats were divided into four groups: normal group (N), model group (M), the group treated with Mortierella isabellina lipid-soluble extracts (T) and Simvastatin (S). N group was fed a standard diet and the other groups were fed a high-fat diet. T and S groups were orally administered by Mortierella isabellina lipid-soluble extracts and Simvastatin respectively for 12 weeks. N and M groups were orally administered by the normal saline. Twenty four-hour urinary protein excretion was detected. The levels of serum ALB, TP, BUN, Scr, TG, TC, LDL and TG, TC, LDL in the kidney were measured at week 12. The histological change, ultrastructural change and lipid deposits in kidney were evaluated.
RESULTSAt the 12th week, compared with M group, twenty four hour urinary protein excretion and the levels of TC, LDL in serum and renal tissue in T and S group decreased (P < 0.01 or P < 0.05), but the levels of ALB and TP in T and S group increased markedly (P < 0.01). There was no significant difference of the levels of BUN, Scr, and TG among the four groups. The protein casts in renal tubules, inflammatory cell infiltration in tubulointerstitium as well as mild mesangial cell proliferation and expanded mesangial matrix were obseverd in M group. Electron micrograph demonstrated massive fusion of foot process of podocytes accompanied with rich lipid droplets in proximal tubular epithelial cells in M group.
CONCLUSIONThe lipid-soluble extracts of Mortierella isabellina has beneficial effect on improving hypercholesterolemia and kidney damage in lipid-induced renal injury rat.

4.Study on effects of Tripterygium wilfordii polycoride in resisting macrophage inflammation and regulating inflammation via TLR4/NF-κB.

To investigate the effect of Tripterygium wilfordii polycoride (TWP) on LPS-induced macrophage inflammatory response, particularly the inhibitory effect on inflammatory factors TNF-α and IL-1β and the regulatory effect on inflammation via TLR4/NF-κB. The MTT method was adopted to test the effects of tested drugs, TWP, dexamethasone (DXM) and azathioprine (AZA) on cell growth to define the appropriate concentration. LPS was used to induce the inflammatory reaction in mouse RAW264. 7 cell lines. The Elisa kit was adopted to test the release level of TNF-α and IL-1β. The Western blotting was applied to test the protein expressions of TNF-α and IL-1β. The RT-PCR was adopted to test the expressions of TLR4 and NF-κB. According to the results, TWP could inhibit the release of macrophage inflammatory factors TNF-α and IL-1β in a dose dependent manner. All of TWP groups showed a weaker efficacy than that of the DXM group. But the TWP high dose group revealed a better effect on TNF-α and equal effect on IL-1β compared with the AZA group. TWP show an equal or better effect in down-regulating TLR4 and NF-κB p65 expressions in a dose dependent manner than DXM and AZA. In conclusion, TWP could inhibit TLR4 and NF-κB p65, which may be related to the down-regulation of TLR4 and NF-κB p65 receptor expressions.

5.Clinical observation of femtosecond laser capsulorhexis with toric intraocular lens implantation

Objective To evaluate the clinical effect of femtosecond laser assisted cataract surgery with Toric IOL implantation on correcting astigmatism and IOL rotating stability.Methods A prospective,nonrandomized,controlled study was conducted in 68 patients (82 eyes) diagnosed as age-related cataract with more than 1.0 diopter (D) regular astigmatism,including 41 eyes undergoing Toric IOL implantation with femtosecond laser assisted cataract surgery,and the other eyes receiving Toric IOL with phacoemulsification.And observation was performed for the uncorrected visual acuity (UCVA) before operation and 6 months after operation,preoperative corneal astigmatism,anticipated and postoperative residual astigmatism 6 months after operation.Then,comparison was performed for the degree of Toric lens axis rotation,as well as the horizontal and vertical movement of the IOL and the degree of vertical deflection in the capsule at 1,3,and 6 months after operation.Results In the observation group,preoperative UCVA (logMAR),preoperative corneal astigmatism,postoperative BCVA and the residual astigmatism at 6 months after operation was 1.03 ± 0.32,(2.09 ± 0.73) D,0.13 ± 0.11 and (0.33 ± 0.27) D,respectively,while the corresponding data in the control group was 0.92 ± 0.40,(1.69 ± 0.64) D,0.13 ± 0.09,(0.26 ± 0.22) D;the postoperative visual acuity in both groups was significantly improved,and residual astigmatism was significantly decreased at 6 months after operation (all P < 0.001).There was no significant difference in Toric lens axis after femtosecond laser assisted surgery and phacoemulsification at 1 month,3 months,6 months after operation (all P > 0.05),but the differences in the IOL movement in horizontal and vertical direction were statistically significant at the different time-points (all P < 0.05),and there was significant differences in the degree of vertical deflection (all P < 0.05).Conclusion Both femtosecond laser assisted cataract surgery and phacoemulsification combined with Toric IOL can achieve a better stability and predictability.
6.Thermal stability of neuraminidase in influenza vaccine

Abstract:Objective To analyze the stabilities of neuraminidase(NA)in influenza vaccine at different temperatures and
provide a reference for further complete understanding of overall shelf life of vaccines. Methods Monovalent bulks of
influenza H1N1,H3N2 and B vaccines were stored at 4(low temperature),25(room temperature)and 37 ℃(changed
temperature)for 0. 5,2,7,24 and 48 h separately,using that at 100 ℃(extreme temperature)for 1 h as control,and
determined for NA activity by enzyme⁃linked lectin method. Results The NA activities of influenza H1N1 vaccines stored at
25 and 37 ℃ decreased significantly with the increasing of time. No significant decreases were observed in H3N2 and B
vaccines even after storage at two non⁃storage temperatures for 48 h. However,all the NA activities of three vaccines
decreased at 100 ℃. Conclusion Both H3N2 and B vaccines showed high stability at abnormal storage temperatures not
more than 37 ℃,while H1N1 vaccine was relatively sensitive to the temperature for storage.
7.The experimental study of creating a new rat scarring model by inserting absorbable gelatin sponge into rats' excisional wounds.

OBJECTIVETo explore the possibility of creating a rat new scar model by inserting gelatin sponge into rat excisional wounds.
METHODSTwo full-thickness wounds were created in each of total 49 SD rats. In the Experimental group (n = 19), a regular incisional wound (1 cm) was created on the left side, and an excisional wound of 1.0 cm x 0.2 cm was created on the right side with a gelatin sponge inserted. In control 1 group (n = 15), an excisional wound with sponge insertion was created on both sides of rats. In control 2 group (n = 15), two excisional wounds were created on both sides, and only one side wounds were inserted with a sponge. Animals were sacrificed at various time points for different examinations.
RESULTSThe wound/scar width increased 4 - 11 times in inserted wounds than in regular incisional wounds (P < 0.01), with an obvious delay of epithelialization. No difference in wound/scar width was found in both sides of wounds of control 1 group at various locations. In contrast to the linear scar of sponge-inserted wounds, contracted and irregular scar was found in non-inserted wounds of control 2 group.
CONCLUSIONSGelatin sponge insertion can create a thick linear scar in rat wounds, and thus provides a new model for scar research.

8.Study on dermal absorption of Imidacloprid in vitro.

OBJECTIVEThe dermal absorption of Imidacloprid was studied to understand the effects of concentrations and skin reservoir on pesticide risk assessment in in vitro absorption studies.
METHODSBy using Franz diffusion cell and the transdermal barrier of viable Wistar rat abdomen skin or frozen ones, the imidacloprid content in the receptor fluid and skin was determined by LC/MS/MS method, and the absorption effects were compared between two concentrations of Imidacloprid solutions and two types of skin, respectively.
RESULTSAll percentages reported are % of applied dose. In vitro studies using viable skin, the Imidacloprid content in the receptor fluid of high and low concentration was 6.8%, 6.6% respectively; and 10.7%, 1.3% in skin, thus total absorption was 17.5% and 7.9%. And in vitro studies using both viable and frozen skin under the same concentration circumstances, the Imidacloprid content in the receptor fluid of viable and frozen skin was 6.6% and 0.7% respectively, in skin was 1.3% and 10.7%, and total absorption was 7.9% and 11.4%.
CONCLUSIONComparison of these in vitro results showed that either concentrations or skin reservoir had an effect on the dermal absorption. During 6h exposure, the high concentration in viable skin had the maximum dermal absorption value, which was the worst-case exposure estimate, also the best single estimate for pesticide risk assessment.

9.Expression of Hippo signaling pathway core element MST1 in acute leukemia patients and its significance.

This study was aimed to investigate the expression of Hippo signaling pathway core element MST1 gene in acute leukemia (AL) patients, and explore its relation with AL pathogenesis and prognosis. 50 newly diagnosed patients with AL, 33 normal people, 23 patients with AL in complete remission, 12 refractory or relapsed patients were tested for the expression of MST1 gene by real-time quantitative PCR, Western blot was used to further validate the level of MST1 protein expression. And combined with clinical data, prognostic factors of the patients were analyzed. The results indicated that compared with the normal people, the expression level of MST1 in newly diagnosed patients with AL significantly decreased (P < 0.05), but significantly increased in AL patients with complete remission (CR), the difference of expression was statistically significant before CR and after CR (P < 0.05). Compared with refractory or relapsed patients, the expression level of MST1 gene in newly diagnosed patients was not significantly different (P > 0.05). Besides, the expression level of MST1 between the patients with CR and the normal people was not significantly different (P > 0.05). It is concluded that the low expression of MST1 may be related with the pathogenesis and prognosis of AL.

10.Bacillus cereus growth model established in cooked rice

Objective Establishment of growth model of Bacillus cereus in cooked rice.Methods To study the effects of temperature (10,15,20,25,30,34,37 and 43℃) on the growth of Bacillus cereus in rice.And then the SGompertz and SLogistic models were selected as the primary growth models to fit the growth curve of Bacillus cereus in cooked rice at variable storage temperatures.Using the fitness (R2),accuracy factor (Af) and deviation factor (Bf) as evaluation index,quadratic polynomial model and square root model were selected further to fit and to establish the secondary growth models of Bacillus cereus in cooked rice.Results The SGompertz model could be better fitting the growth of Bacillus cereus at different temperatures,and therefore was chosen as the primary growth model of Bacillus cereus in rice.For the developed square root model,Af was 1.12 and 1.24,Bf was 0.99 and 1.03,R2 values were 0.9537 and 0.8503;respectively.For the developed quadratic polynomial model,Af was 1.24 and 1.11,Bf was 0.92 and 0.92,R2 values were 0.9550 and 0.9462;respectively.Conclusion The quadratic polynomial model can well predict the growth of Bacillus cereus in cooked rice,which proves to be reliable.
