Journal of Applied Clinical Pediatrics 2006;0(22):-
Objective To explore the effect of preoperative chemotherapy for children with malignant extragonadial germinoma. Methods Twenty patients with malignant extrogonadal germinoma had been treated with preoperative chemotherapy from Mar.2003 to Mar.2007.Protocol 1 of chemotherapy was to improve PEB:(cisplatin plus etoposide plus bleomycin A5);Protocol 2 was VAC(vincristine plus actinomycin D plus cyclophosphamide) the average time span of the chemotherapy was 16 weeks.Upon completion of chemotherapy, tumor resection was performed. The content of alpha fetoprotein(AFP) was determined before operation and compared with the content before chemotherapy.Re-gular follow-up procedure was taken after operation. Results All 20 patients received chemotherapy and underwent operation. The removal rate of resection operation was 100%,the process of operation was smooth. The volume of tumors in 14 patients decreased over 50% compared with those before chemotherapy. The response rate was 70%.The content of AFP decreased significantly in 15 patients(75%).Fifteen patients had been living without tumor for up to 2 years, for 5 cases the survival time went beyond 5 years. Conclusions Malignant extrogonadal ger-minoma in children grow infiltratively, bring detrimental effects to neighboring organs, and bring obstacles to resection operation.Chemotherapy for children with malignant extragonadal germinoma before operation can reduce volume of tumor and bleeding during operation, raise the resectability rate and lower the content of AFP.It will enhance the effectiveness of operation and improve prognosis. Therefore,it can be used as a conventional clinical method.

Recent Advances in Ophthalmology 2017;37(8):714-718
Objective To establish the model of autoimmune dry eye and analyze the level of cytokines and the pathological changes of autoimmune dry eye rabbits treated with human umbilical cord mesenchymal stem cells (MSCs).Methods Twentyfour female New Zealand rabbits were randomly divided into three groups:Normal control group, dry eye group and MSCs treatment group,8 cases in each group. After treated with MSCs for six weeks, the pathological changes were observed, and the expression of Th1 and Th17 cell differentiation related cytokines mRNA in lacrimal gland tissue was detected by real-time quantitative PCR.The percentage of regulatory T cells (CD4 +Foxp3 + cells) in the lacrimal gland and spleen tissues of the dry eye model group and MSCs treatment group was measured by flow cytometry. Results After treatment with MSCs for 6 weeks,HE staining from each rabbit lacrimal gland showed that, the normal control group showed no or only a few lymphocytes; The lacrimal gland cells were atrophied in the dry eye group, the distribution of lymphocyte scattered around the gland and small blood vessels;Compared with dry eye group, the lacrimal gland lymphocyte infiltration reduced in MSCs treatment group, and the cell morphology were better.Compared with dry eye group, lymphocyte infiltration and aggregation in the conjunctival conjunctiva in MSCs treatment group were significantly reduced, the epithelial structure was intact, and the degeneration and atrophy cells were rare. The expression of inflammatory factors including interferon-γ(IFN-γ),Interleukin-17 (IL-17) and transcription factor T-bet mRNA were decreased in MSCs treatment group, there were significant difference compared with dry eye group (all P < 0.05);Thl7 cell associated cytokines IL-17 expression levels were decreased, but no significant difference was found between two groups (P > 0.05);The expression of transcription factor RORrt mRNA in dry eye group was significantly decreased, the difference was statistically significant (P < 0.05).The expression of inflammatory factors TNF-α in the lacrimal gland tissue of MSCs treatment group was significantly lower than that in the dry eye model group, while the expression of the anti-inflammatory factor TGF-β was significantly higher than that in the dry eye model group, and the differences were statistically significant (all P<0.05).The regulatory T cells (CD4 + Foxp3 + cells) in the lacrimal gland of dry eye model group accounted for 10% of lymphocytes was, while the treatment group accounted for 27.8%,MSCs group was significantly higher than dry eye group, the difference was statistically significant (P < 0.05).Conclusion MSCs can reduce the histopathological changes of the immune dry eye,and they may have immunoregulatory effects on autoimmune dry eye.The mechanisms of immunomodulatory effects may be related to the balance of inflammatory cytokines and anti-inflammatory cytokines.

Tianjin Medical Journal 2014;(10):969-971,1059
Objective To investigate the effect of LY294002 on proliferation of cultured human glomerular mesan-gial cells, and to study on the mechanism underlying this proliferation. Methods Different concentrations of LY294002 (0.02,0.2,2,20,100 mg/L) were administrated to the cultured human glomerular mesangial cells. The effects of mesangial cell proliferation were measured using CCK-8 colorimetric assay. Under appropriate concentrations, proliferation of cultured mesangial cells were measured using CCK-8 at 0.5, 1, 24, 48 hours of drug administration time. Results LY294002(0.02 mg/L)didn’t obviously inhibited the growth of mesangial cells(P>0.05), the inhibition rate was 5.05%. The effect of LY294002 on the cells decreased with rising concentration (0.2 to 20 mg/L) in a dose-dependent manner (P<0.05). Mesan-gial cells would stop growing with the increasing concentration. LY294002 didn’t obviously inhibited the growth of mesan-gial cells at 2 mg/L during 0 to 1 h(P>0.05), but inhibited proliferation gradually from 1 to 48 h (P<0.05). Conclu-sion Within the certain range of concentration and time, the LY294002 inhibits the proliferation of human glomerular me-sangial cells, and PI3K/Akt/mTOR signaling pathway may regulate the proliferation of human mesangial cell.

To analyze the composition regularity of prescriptions containing Scutellaria baicalensis in Drug Standard of Ministry of Public Health of the Peoples Republic of China--Chinese Patent Medicines and Preparations on the basis of the traditional Chinese medicine inheritance support system (TCMISS), in order to provide reference for new drug R&D. the platform's software V2.0 was applied to establish a database of prescriptions containing S. baicalensis. The software's statistical statement module, association rules and improved mutual information method and other data mining technologies were adopted to analyze commonly used drugs, combination rules and core combination of S. baicalensis prescriptions. Having analyzed 477 prescriptions containing S. baicalensis, the researchers summarized 45 most commonly used drug combinations, whose ingredients mostly had functions of heat-clearing and damp-drying, purging fire for removing toxin and hemostasis. Drugs adopted in core combinations were relatively concentrated and selected according to definite composition methods. There were 23 diseases that S. baicalensis were most frequently applied in the treatment. Having compared three highly frequent diseases--cold, cough and dizziness, the researchers concluded that S. baicalensis could show different therapeutic effects through different combination ratios. Therefore, TCMISS (V2.0) is an important tool in analyzing the composition regularity of traditional Chinese medicines. The longitudinal and parallel comparison method is an effective method for studying the clinical composition regularity of S. baicalensis, while providing reference for new drug R&D.
