1.Comparison of curative effect between r-tPA application and urokinase in the interventional therapy for acute and severe pulmonary embolism

Objective To study the different curative effect between r-tPA application and urokinase in the interventional therapy for acute and severe pulmonary embolism.Methods Analyzed the clinical date of 22 cases acute and severe pulmonary embolism patients,and classified them into two groups according to the application of r-tPA and urokinase to compare the changes of their blood pressure,heart rate,arterial partial pressure of oxygen,hemachrome and pulmonary arterial pressure before and after therapy,meanwhile,record the thrombolysis time that the two groups took.Results The thrombolysis effect time adopting urokinase was remarkably longer than that thrombolysis adopting r-tPA(P<0.05); The blood pressure,heart rate,arterial partial pressure of oxygen of the two groups gained remarkable improvement after therapy(P<0.05).The pulmonary arterial pressure of the urokinase group dropped obviously after operation(P<0.05),while the r-tPA group dropped not so significantly after operation.The hemachrome of the two groups did not dropped significantly after operation.Conclusion The r-tPA and urokinase are both effective to interventional therapy for acute and severe pulmonary embolism,while r-tPA can clearly shorten the time in thrombolysis and could reduce any bleeding risk.

4.Identification of Bletillae Rhizoma and its adulterants by SNPs in ITS2.

To establish a molecular identification method for Bletillae Rhizoma, this paper extracted genome DNA from Bletillae Rhizoma and its adulterants. The sequences of rDNA ITS2 were sequenced after amplifying. Then multiple alignments of ITS2 were constructed phylogenetic tree with Neighbor Joining by MEGA 5. 1 and found out SNPs loci. The result showed that rDNA ITS2 region could identify Bletillae Rhizoma and its adulterants. There existed the SNPs loci, which could identify Bletilla striata and B. ochracea. Furthermore, we designed specific primers against the SNPs loci of B. striata and B. ochracea, then screened primers and optimized the PCR amplification conditions. Finally, the DNA of B. striata and B. ochracea were specifically amplified by BJ59-412F, BJ59-412R and HHBJ-225R. The length of amplification products were respectively about 350 bp and 520 bp that were effectively identified of B. striata and B. ochracea. While, the adulterants of Bletillae Rhizoma were no-reaction occurring. To sum up, the amplification conditions of the primers can identify B. striata, B. ochracea and their adulterants successfully at the same time. This method was easy, time-saving, and reliable, which can be used as a rapid method for molecular identification of Bletillae Rhizoma.

5.Rapid molecular identification of pseudostellariae radix.

To establish a convenient and rapid method for identification of Pseudostellariae Radix by molecular identification, the rDNA-ITS sequences of Pseudostella riaheterophylla and its adulterants had been aligned to find out specific fragment. The specific primers against the fragment were designed and the PCR amplification conditions were optimized. The fluorescence reaction of the PCR products colored by 100 x SYBR Green I was observed under UV. The concentration of reaction buffer included 5.5 μL DNA Taq polymerase premix, 10 pmol Tzs-2F and 10 pmol Tzs-2R, 20-80 ng template DNA, and plus double sterile distilled water to 25 μL. The PCR thermal profile was as follows: predenaturation at 95 degrees C for 1 min, followed by 30 cycles of denaturation at 95 degrees C for 5 seconds, primer annealing and extension at 56 degrees C for 15 seconds, then it was extension at 72 degrees C for 30 seconds. The fluorescence reaction of Pseudostellariae Radix showed green fluorescence, while adulterants had not. Extraction, amplification DNA and all steps of molecular identification could be completed successfully in 40 minutes. The approach could amplify DNA template of Pseudostellariae Radix specificity, and its product with 1 μL 100 x SYBR Green I could engender green fluorescence under UV. The method was simple and accurate, so it could be used for identification of Chinese traditional medicine.

7.Effects of course diary in preventing lumen catheter infection in hemodialysis patients

Objective To explore the effects of course diary in preventing lumen catheter infection in patients who received hemodialysis (HD).Methods Totally 105 patients who received HD in Civil Aviation General Hospital between January 2013 and December 2016 were selected by convenience sampling and divided into an observation group (53 patients) and a control group (52 patients) according to the random number table. Patients in the control group received conventional nursing, while patients in the observation group, on this basis, were supervised by nurses to implement the course diary including content related to infection prevention, and the nurses undertook to keep records. 12 weeks later, the blood flow volume, urea clearance index (KT/V), incidence of insufficient blood flow and infection rate were compared between the patients in the two groups.Results The patients in the control group showed a higher blood flow volume than the patients in the observation group (P<0.05), and there was no statistically significant difference in the urea clearance index (P>0.05) before the intervention, but the patients in the observation group showed a higher blood flow volume and urea clearance index than the patients in the control group (P<0.05) after the intervention. There was no statistically significant difference in the incidence of insufficient blood flow (P>0.05) before the intervention, while the patients in the observation showed a lower incidence of insufficient blood flow than the patients in the control group (P<0.05) after the intervention. And the patients in the observation group showed a lower infection rate than the patients in the control group (P<0.05) after the intervention.Conclusions Course diary can help to improve the blood flow volume and urea clearance index, reduce the incidence of pathogens, and lower the infection rate in patients receiving HD with lumen catheter.
8.Effect of progressive muscle relaxation combined with health education on the psychological state of the maintained hemodialysis patients

Objective To evaluate the effect of progressive muscle relaxation combined with health education on the psychological state of the maintained hemodialysis patients and provide basis for the psychological intervention for them. Methods Forty-two MHD patients were randomly assigned to the intervention group and control group. Patients in the control group received the conventional nursing, and patients in the intervention group received the conventional therapy combined with nursing intervention for two times. Results After the first intervention, the various psychological testing indicators of intervention group were obviously lower than those of control group and there was statistically significant difference between two groups(P < 0. 05 ). The differences of index of physiological chemistry between two groups had no statistical significance (P > 0. 05 ). After the second intervention, the various psychological testing indicators of intervention group were further lower than those of control group and there was statistical significant difference ( P < 0. 01 or P < 0. 05 ). The Hb and Ca2 + of the intervention group were remarkably higher than control group,while PO43+ and K+ of intervention group were significantly lower than control group. Both differences had statistical significance(P < 0. 05 ). Conclusions Progressive muscle relaxation combined with health education can greatly improve the psychological and physiological status of MHD patients.
9.Expression of Attractin in male reproductive tract of human and mice and its correlation with male reproduction.

The expression of Attractin mRNA and protein in testis and semen of human and male mice was investigated. Human testis and semen samples were all collected from Reproductive Center of Renmin Hospital, Wuhan University in December, 2012. Testis samples were collected from 7 cases of obstructive azoospermias when they were subjected to diagnosed testis biopsy, and 30 normal human semen samples were obtained from those cases of semen analysis. Adult mice testis tissues were obtained from 10 2-month-old male BALB/c mice, and 60 male mice at different ages were classified into 10 groups (day 1, 5, 10, 15, 21, 28, 35, 42, 56, and 120 respectively, n=6 each). The expression of Attractin mRNA and protein in testis was detected by RT-PCR and Western blotting respectively. Human semen samples were centrifuged into sperm plasma (SP) and sperm extract (SE), and mice sperm samples were collected from the epididymis of 10 adult male BALB/c mice. Western blotting was used to determine the Attractin protein expression level. Attractin mRNA and protein were expressed in the testis of both patients with obstructive azoospermias and adult Bcl/B mice. Quantitative RT-PCR revealed that no Attractin mRNA was detectable in day 1 male BALB/c mice group. The Attractin mRNA and protein levels were low on the day 10, and increased with age until day 56. On the day 120, the expression levels of Attractin were decreased. As for human semen samples, Attractin protein was expressed in both SP and SE, but didn't exist in samples from the epididymis of male BALB/c mice. It was suggested that Attractin acted as a novel active substance and was involved in male reproduction in both human and BALB/c mice, but it exerted a different expression profile in different mammal species.
10.Effect of zuojin pill and fanzuojin pill on the growth metabolism of enterobacteria by microcalorimetry.

OBJECTIVETo study the effects of Zuojin Pill and Fanzuojin Pill (ZJP and FZJP) on the growth metabolism of enterobacteria from the biothermodynamic viewpoint, and to objectively validate the difference between the two drugs for investigating a new way in congener Chinese drugs research.
METHODSThe growth metabolism power-time curves of enterobacteria affected by ZJP and FZJP in different concentrations were obtained by microcalorimetry. And the activities of the two drugs were assessed depending some related characteristic parameters, including growth rate constant (k), maximum output power (P(max)), peak time (T(p)) , maximum heat-output (Q1), and bacteria inhibition rate (I).
RESULTSWith the drugs' concentration increasing, T(p) was prolonged, k value reduced in enterobacteria, i.e. , the two drugs both inhibited the growth of enterobacteria in different extents. Comparison of inhibiting rates showed that the bacteriostasis activity of ZJP was greater than that of FZJP.
CONCLUSIONSThere was objective difference in activities of inhibiting enterobacteria between ZJP and FZJP. Microcalorimetry could express the features at real-time, on-line and sensitively. It was able to determine the activity of drugs accurately and quickly in appraising congener Chinese drugs, like ZJP and FZJP, by microcalorimetry.
