Journal of Chinese Physician 2015;17(4):516-518
Objective To investigate the relationships between serum inflammatory mediators and myocardial injury in rats with sepsis. Methods Thirty male Sprague Dawley (SD) rats were randomly divided into normal control group and sepsis (0,24,48,72 h) groups. The rats subjected to sepsis were induced with cecum ligation perforation (CLP).Tumor necrosis factor alpha (TNF-α),interleukin-6 (IL-6),cardiac troponin Ⅰ (cTnI) and B-type natriuretic peptide precursor (NT-proBNP) in the venous blood were measured at 0 h,24 h,48 h,and 72 h after CLP.The lactic acid (Lac) of the arterial blood, cardiac index (CI),acute physiology and chronic health evaluation (APACHE) Ⅱ score were also determined simultaneously. Results The levels of TNF-α,IL-6,cTnI,NT-proBNP,and Lac in sepsis (24 h,48 h,and 72 h) groups were significantly elevated compared to normal control and sepsis (0 h) group. In addition, the number of sepsis (24 h,48 h,and 72 h) groups progressively increased (P <0.05).Conclusions It existed a close relationship between serum inflammatory mediators and myocardial injury, and made myocardium damage after CLP.

International Journal of Cerebrovascular Diseases 2013;21(12):933-937
Stroke is a cerebrovascular disease type with high morbidity and mortality,of which ischemic stroke accounts for about 80%.Now,it is believed that the inflammatory mechanisms play an important role in the progress of pathological physiology of ischemic stroke.Peripheral T lymphocytes infiltrate to the damaged area within 24 hours after cerebral ischemia and are involved in the advance of inflammatory process of brain tissue. As a subtype of T lymphocytes, regulatory T cells are mainly located in the ischemic penumbra; however,at present, its role in ischemic brain injury remains controversial. The research on the mechanisms of regulatory T cells in ischemic stroke may contribute to further understanding of the pathogenesis of ischemic stroke and find new therapeutic targets.

Objective To explore the mechanism of synthesis metabolism intensified by human growth hormone(GH) on the basis of total parenteral nutrition(TPN) for postoperative gastrointestinal carcinoma patients. Methods Totally 94 cases of postoperative gastrointestinal carcinoma patients were randomly divided into TPN group and TPN+GH group. The levels of TNF-?,IL-1,IL-6,CRP were detected on postoperative day 1,3 and 7.Results The levels of TNF-?,IL-1,IL-6,CRP of TPN+GH group were significantly lower than those of TPN group(P

Objective To investigate the role of cyclophilin A (CypA) and CD147 in the process of skin aging. Methods Twenty cases of tissue samples from junior group(<15 years old), middle age group(30-40 years old)or old age group (>65 years old) were collected from photophobic and exposal parts of skin, respectively. Immunohistochemistry (IHC) and in situ hybridization (ISH) were carried out to semi-quantitatively detect the expression level of CyPA and CD147. Results IHC demonstrated that both CyPA and CD147 were expressed in both photophobic and exposal parts of normal human skin in all 3 groups. The expression levels of both CyPA and CD147 were increased with increase in age. There were significant differences in both CyPA and CD147 expression among 3 groups (P<0.05). CyPA and CD147 were also positively correlated in all 3 groups. Similar results were achieved by ISH. Conclusion The interaction between CD147 and CyPA might play an important role in the process of skin aging.

There exist a number of mercury-resistant bacterial in environment, Mer operon is involved in the resistant mechanism, MerRTPA of Mer operon encodes the proteins related to the regulation, transport and reduction of mercury ion, respectively. The toxic mercury ion is transported by MerTP from medium to cytoplasmic mercuric reductase, MerA, and deoxidized to non-toxic and volatile element mercury, Hg(0). Bacterial mercury-resistant system originated from ancient times, and evolved into the Mer operon with diversity by gene integration and insertion. Mercury-resistant bacteria highly specifically absorb mercury ion, and can be used in recovering the mercury-polluted environment as well as the genetic selective marker.

In gene manipulation, different selectable markers with various linkers are necessary. In order to get selectable markers directly, we constructed from pBlueScript SK(-) a series of particular plasmids, pSKsymKm, pSKsymBle, pSKsymEry, pSKsymHyg and pSKsymGm, each contains Kanamycin, Bleomycin, Erythromycin, Hygromycin or Gentamycin resistance cassette. By restriction enzyme digestion and gel extraction, any of five antibiotic resistance genes with specific ends can be conveniently obtained.

Objective To investigate the effects of diabetic mellitus on the opening of mycardial mitochondrial permeability transition pore (MPTP) in rats, and to explore the role of the opening of MPTP in the development of diabetic cardiomyopathy. Methods Sixteen male Wistar rats were randomly divided into normal control group (NC) and diabetic mellitus group (DM) (8 each).Diabetic mellitus model was reproduced by a single intraperitoneal injection of streptozotocin (STZ).After 12 weeks, the production rate of mitochondrial reactive oxygen species (ROS) and the change in mitochondrial membrane potential (MMP) in myocardial mitochondria were assayed by fluorospectrophotometer.Cardiomyocyte apoptosis was evaluated by terminal-deoxynucleotidyl transferase mediated deoxy-UTP nick end labeling (TUNEL),and the glutathione (GSH) level in serum was measured by spectrophotometer. Results Compared with NC group [13.01?3.31U/(s?mg)],the production rate of mitochondrial ROS was significantly increased in DM group [28.79?6.11U/(s?mg),P

Objective To investigate the effect of telmisartan on mitochondrial membrane potential and cardiomyocyte apoptosis of rats with type 1 diabetes. Methods Type 1 diabetic rat model was reproduced by intraperitoneal injection of streptozotocin to adult male Wistar rats.Sixteen diabetic rats were randomly divided into two groups (8 each):DM group and T group (rats were treated with telmisartan).Another 8 normal rats served as control (Con group). After 12 weeks, the levels of reactive oxygen species (ROS) were detected by fluorescent probe DCFH-DA,and the changes in mitochondrial membrane potential (?m) was detected by fluorescent probe JC-1.Cardiomyocyte apoptosis was evaluated by TUNEL.Results The levels of blood glucose were higher and body weight was lower in both DM and T group than in Con group at 7th day and 12th week after streptozotocin injection (P

Objective To investigate the effects of telmisartan on oxidative stress in type 1 diabetic rats. Methods Type 1 diabetic rat model was established by intraperitoneal injection of streptozotocin (STZ) in adult male Wistar rats.Sixteen diabetic rats were randomly divided into two groups (8 each):diabetic mellitus group (DM) and telmisartan group (T).Eight normal rats were employed as the control group (Con group). After 12 weeks, body weight (BW) and heart weight (HW) were measured to calculate HW/BW.Glutathione (GSH),malondialdehyde (MDA),the activity of superoxide dismutase (SOD) were evaluated by spectrophotometer.Ultrastructure of myocardial cells was observed by transmission electron microscope. Results There was no significant difference in blood glucose levels between DM group and T group at the 7th day and 12th week after STZ injection, but the blood glucose levels in the both groups were significantly higher than that in Con group (P0.05).The MDA level was higher (P
