1.A rat model of repeated seizures induced by pentylenetetrazol at different maturational stages and the role of NF-?B in the pathogenesis of epilepsy

AIM: To observe histopathologic changes and NF-?B expression in hippocampus in neonatal and matural rats after repeated seizures, and to explore the role of NF-?B in the pathogenesis of epilepsy in premature brain of rats. METHODS: Neonatal rats and mature rats were divided into 2 experimental groups at 10 days and 60 days after birth (P10 and P60). Convulsions were induced by repeated injection of pentylenetetrazol (PTZ) intraperitoneally for first 5 days. The animals in control group were injected with NS at the same volume in the same conditions. The neurons in CA1, CA3, dentate granule (DG), as well as in hilar were counted by thionin staining, in order to observe the profile of the necrosis and apoptosis. NF-?B expression was examined by immunohistochemistry assay. Timm's method of silver sulfide staining was adopted to observe the mossy fiber sprouting. RESULTS: (1) In immature rats (10 days old), neurons in CA1, CA3 and hilar demonstrated no differences from controls, whereas adult rats (P60) had a significant decrease in number of neurons in CA1 and CA3 (8.22?1.88, 5.62?1.68 vs 6.31?1.50, 3.62?1.40). In adult rats, neurons in dentate granule showed no differences with controls, whereas immature rats with daily seizures had a significant increase (23.25?3.06 vs 16.25?1.58). (2) There was prominent sprouting in the CA3 stratum pyramidal layer in all experimental rats after 5 daily seizures, regardless of the age. 2.The value of digital subtraction angiography in diagnosis and planning endovascular treatment of children with ischemic cerebrovascular diseases

Objective To evaluate the value of digital subtraction angiography(DSA) in diagnosis and planning endovascular treatment of cerebrovascular diseases. Methods Thirty-one children suspected with ischemic cerebrovascular diseases were treated by urokinase and dextran after they underwent DSA.Results Twenty-four cases (77%) had cerebrovascular abnormalities. Two cases had arteriovenous malformations, 1 case had posterior communicating artery aneurysm,21 cases had cerebrovascular stenosis or occlusions. Shortly after intervention therapy using urokinase,muscle strength increased by one level in 13 cases,two levels in 3 cases,3 levels in 2 cases and no effect in 4 cases. The total effective rate reached 86%. Seven patients with ischemic stroke achieved 100% recanalization. Conclusion DSA is very important in diagnosis of cerebrovascular stenosis and occlusion.

4.Experimental study of gut absorptive capacity after trauma detected by D-xylose content test with micromethod

Aim To establish a rapid and sensitive diagnostic markerfor adjusting gut absorptive capacity after trauma. Methods A mi-cromethod for D-xylose test with phloroglucinal was established according toEberts with modification. The standard curves were repeated for fivetimes. Then the blood contents of fasting and orally taking 0. 5 g/kgD-xylose at 2nd and 4th hour were measured and the blood D-xylose con-tents of seven refilling rats with ischemia of intestine were mea-sured. Results The maximal absorption spectra was read at 554 nm byscanming of DU-7 Beckman. Good linearity of the D-xylose standard curvewas found with the range of 0 to 4 mmol/L(r=0. 9979 ±0. 0017) . Theblood content of rats at 2nd hour after orally taking D-xylose was ( 154 ± 6)mg/L amd that at 4th hour was decreased to (87 ± 11) mg/L. And theblood content at 2nd and 4th day after ischemic refilling was (162 ± 5)mg/L and (80 ± 8) mg/L respectively. The variation coefficients within andamong batches were 1.98% (n=6) and 3.10% (n=6), respective-ly. And D-xylose recovery rates were from 97.2% to 104. 3%. ConclusionMicromethod for D-xylose test appears to be simple, rapid and sensitive,and is an available index for estimating intestinal absorption after severetrauma.
5.Comparison of Scopoletin Contents in Cortex Mori Processed by Different Methods and Picking Periods

Objective: To study the best gathering period and processing method of Cortex Mori.Methods: A HPLC method was used to determine the content of scopoletin. Results: The scopoletin contents in Cortex Mori picked in Jan., Feb. Apr., Jul. and Aug. were higher than that picked in the other months. The scopoletin content in Cortex mori of which rough barks were unremoved was higher than that of which rough barks were removed.Conclusion: Spring and Summer are the best gathering periods, but the processing method shouldn't be stricthy limited.
6.Study on in vitro percutaneous adsorption of Huayu Babu Sticking Plaster

Object To research the effect of laurocapram upon the percutaneous absorption of Huayu Babu Sticking Plaster (HBSP) in vitro. Methods The improved Franz diffusing cells were chosen to study percutaneous absorption of piperine from HBSP. The content of permeated piperine was determined by HPLC; while the content of augenol was determined by GC to inspect the stability of HBSP. Results The result showed that the process of penetrating of piperine in HBSP through skin could be in accordance with zero-grade releasing equation and HBSP was stable during the course of experiment. Conclusion Laurocapram could promote percutaneous absorption of piperine.

9.Relationship between homocysteine metabolism abnormality and pre-clampsia

Objective To study the serum levels of homocysteine(Hcy) and the polymorphism of C677T mutation of methylenetetrahydrofolate reductase (MTHFR) gene in pregnant women to explore their role in the pathogenesis of pre-clampsia. Methods Fifty pregnant women with preclampsia (study group) including 14 mild pre-eclampsia and 36 severe pre-eclampsia and 40 healthy ones (control group) were selected. Hey concentration was determined by fluorescence polarization immunoassay. The mutations of the 677C→T and 1298A→C of MTHFR were investigated by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Results The mean level of plasma Hey in the study group was higher than that of control 1(12. 00±4. 59) μmol/L vs (7. 85:1:1.51) μmol/L, P<0.05], and that of the severe pre-eclampsia group was higher E(13. 30±2. 06) μmol/L, P<0. 01]. However, no significant difference was found between the control and the mild pre-clampsia group[(8. 63±3. 94) μmol/L, P>0.05]. There was significant difference in the genotype frequencies of MTHFR 677CT, TT genotype(C/T:42.0% ;T/T:14. 0%)and the total frequency of mutant T (T: 35.0%) allele between the study and control groups (P < 0. 05). Conclusions Hyperhomocysteinemia plays an important role in the pathogenesis of pre-clampsia. The polymorphism of MTHFR C677T might be associated with the development of pre-clampsia as it affects the metabolism of Hey.
10.Observations on the Efficacy of Interactive Thumbtack Needle Embedding plus Rehabilitation Instruction in Treating Knee Osteoarthritis

ObjectiveTo investigate the efficacy of interactive thumbtack needle embedding plus rehabilitation instruction in treating knee osteoarthritis.Method Seventy outpatients with knee osteoarthritis were randomly allocated to acupuncture-rehabilitation and control groups, 35 cases each. The control group received rehabilitation instruction alone and the treatment group, interactive thumbtack needle embedding in addition. The function of the affected knee was assessed using the Western Ontario and McMaster Universities Osteoarthritis Index (WOMAC) score and manual muscle test (MMT) in the two groups of patients before and after two courses of treatment. The therapeutic effect was evaluated using Nimodipine method accordingto 2002Guidelines on Clinical Study of New Chinese Herbal Medicine(trial).ResultAfter two courses of treatment, there were a statistically significant pre-/post-treatment differences in the WOMAC total score, pain score, stiffness score and function score in both groups (P<0.01). There were also statistically significant post-treatment differences between the two groups (P<0.05). There were statistically significant pre-/post-treatment differences in quadriceps femoris and hamstring muscles MMT in both groups of patients (P<0.01) but there were no statistically significant differences in the same items between the two groups (P>0.05). There was a statistically significant difference in therapeutic effect between the two groups (P<0.05).ConclusionInteractive thumbtack needle embedding plus rehabilitation instruction has a definite therapeutic effect on knee osteoarthritis.
