1.THE DISTRIBUTION OF OXYTOCIN IMMUNOREACTIVE NEURONS IN THE SUBEPENDYMAL PLEXUS OF THE THIRD VENTRICLE IN THE RAT

Immunohistochemical method was used to investigate the morphology and distribution of the oxytocin immunoreactive (OXT-ir) neurons and their dendrites in the subependymal plexus (SEP) around the third ventricle (OXT-SEP). The OXT-ir neurons were of magnocellular neurosecretory type which often intercalated between the ependymal cells, their dendrites were generally long and straight and mostly paralleled to or traveled within the ependyma,some of them could protrude into the ventricle, and most of them appeared heavily vacuous and have a strong tendency of grouping into fascicles. The plexus was composed of dorsal and ventral parts, the former corresponding to the extent of the magnocellular paraventricular nuclei and was extensive and dense, the dendrites were well organized; the latter was more sparse. The cell bodies and dendrites within and between different parts might mix or contact with each other. The functional significance of the plexus is discussed.
2. Application of Drosophila melanogaster in pharmacological research

As a model organism, Drosophila melanogaster has the characteristics of short life cycle, high fecundity and high gene conservation. At present, Drosophila melanogaster has been widely used in the studies of pharmacology and pathogenesis, including aging, neurodegenerative disease, insomnia, tumor and other fields, which show potential values in accelerating the speed of drug screening and target discovery. In this paper, the advantages of Drosophila as a model organism, the applications and limitations of Drosophila in pharmacological studies were reviewed.
3.Long-term efficacy of fluorouracil + oxaliplatin + gemcitabine combined with prophylactic hepatic arterial infusion chemotherapy after surgical treatment of primary hepatocellular carcinoma

Objective To analyze long-term efficacy after surgical treatment of primary liver cancer with fluorouracil plus oxaliplatin plus gemcitabine joint implementation to prevent the overall intervention effect of hepatic arterial infusion chemotherapy and radical resection of hepatocellular carcinoma following ascension.Methods60 cases of primary hepatocellular carcinoma were randomly divided into study group and control group according to the random number method in our hospital from February 2011 to November2013, 30 cases in each group.All patients underwent radical resection of liver cancer and liver (partial) resection.In the control group, the patients were treated with anti viral or immune enhancement after surgery, and the follow-up treatment was not carried out.In study group were given anti-virus or immune intensive treatment and at the end of surgery 3 and 7 weeks after the implementation of a prophylactic transcatheter arterial infusion chemotherapy and selecting drug 5-FU, oxaliplatin and gemcitabine.During 3 years of follow-up, the incidence of adverse reactions in the 3 groups was statistically analyzed, and the recurrence free survival rate, disease-free survival rate and overall survival rate of the two groups were statistically analyzed.ResultsAll patients completed the follow-up, there was no loss of follow-up cases.In adverse reactions, the study group patients were successfully tolerated hepatic artery infusion chemotherapy, no obvious adverse reactions or drug toxicity, more no interruption of chemotherapy cases.The recurrence free survival rate, disease-free survival rate and overall survival rate of the study group were 83.33%, 70.00%, 86.67%, respectively, which were significantly higher than those in the control group (60.00%, 43.33%, 63.33%) (P<0.05).ConclusionPrimary hepatocellular carcinoma should be treated with prophylactic hepatic arterial infusion chemotherapy in time after radical operation, which can greatly improve the survival rate of patients and ensure long-term curative effect.
4.Cloning of human interleukin-24 gene and its high efficiency expression in E. coli

Objective To construct a recombinant expression vector of human interleukin-24(hIL-24) gene and express it in E.coli M15,and to evaluate the bioactivity of IL-24 fusion protein.Methods The human IL-24 cDNA fragment was amplified from plasmid by polymerase chain reaction(PCR),and sequenced.PQE/hIL-24 was constructed by gene rearrangement,then it was transformed into E.coli M15.The expression of the target protein was induced with IPTG and purified by Ni2+-NTA agarose column.The expressed recombinant hIL-24(rhIL-24) was identified by SDS-PAGE and Western blotting.Normal peripheral blood mononuclear cells(PBMCs) were cultivated with the expression protein for 48 and 72 h,the levels of IL-6,IFN-? and TNF-? of PBMCs stimulated with rhIL-24 were detected by ELISA.Results The recombinant prokaryotic expression vector PQE/IL-24 was constructed successfully and expressed stably in E.coli M15.At about 18 400 of molecular weight,there was an induced protein band.The levels of IFN-?,IL-6 and TNF-? in the group of cultivated with the expression protein were obviously higher than those in the groups without the expresson protein(P
5.Application and assessment of the standard score in instructional supervision

The paper introduced the theory of the standard score and its application in instructional supervision and analyzed the difference between the standard score and original score in instructional supervision recording and revealed the reasons behind the difference.Besides,the paper pointed out the advantages which included scientificity and stability,providing information of the grade level,being suitable for longitudinal comparison and the crosswise comparison,and disadvantages such as complex calculation and depending on the distribution of original records.Finally,the paper put forward some recommendations on using standard score in instructional supervision:keeping the stability of the supervision expert team,insurance of supervision times,carrying out classified supervision and viewing the scores in its entirety.
6.A review of detection methods for human bocaviruses.

Human bocavirus (HBoV) 1-4 have been detected both in respiratory and stool samples since the first HBoV was discovered in 2005. HBoV-1 is mostly associated with respiratory infection, while HBoV 2-4 are usually associated with intestinal tract infection. A variety of signs and symptoms have been described in patients with HBoV infection, including cough, wheezing, pneumonia, and diarrhea, but the research on pathogenic mechanism of HBoV is limited because HBoV cannot be cultured in vitro due to the lack of appropriate host cells. Three-dimensional epithelial cell culture, reverse genetics, and viral metagenomics are identified as novel tools that may promote the research on pathogenic mechanism of HBoV and the discovery of new viruses. This review summaries currently available diagnostic approaches such as electron microscopy, cell culture, PCR, and immunoassay in order to provide a method reference for indepth research on HBoV.

7.Effects of bioactive extracts from Fructus Jujubae in attenuating the inflammation induced by Radix Kansui

Aim: To evaluate the effects of bioactive extracts from Fructus Jujubae in attenuating the inflammation induced by Radix Kansui. Methods: Exoteric splenic lymphocyte ( SPL) of mouse and peritoneal macrophage ( PMψ) of rat were used as cell model to evaluate the anti-inflammatory effects of fractions from Fructus Jujubae. MTT method was used to assay SPL proliferation, and Griess method was used in NO release of PMψ. Firstly, Radix Kansui fraction was applied to induce inflammatory effects of the cells, and then, the anti-inflammatory effects of five extracts of Fructus Jujubae were investigated to determine the active fractions in Fructus Jujubae, which probably contribute to the attenuation of Radix Kansui. The effects of Fructus Jujubae extract in interfering the gastrointestinal acute damages caused by the extract of Radix Kansui were observed according to the pathological sections of mice. Results: The extract RK-3 ( diterpenes) exhibited the most significant pro-inflammatory effect. The bioactive extracts B (micromolecule saccharides and amino acids), D(flavonoid glycosides), and E (triterpenes) of Fructus Jujubae could attenuate the action of RK-3. The effective extract of Fructus Jujubae protected stomach and duodenum of mouse from acute damages caused by the extract of Radix Kansui. Conclusion: Fraction RK-3 (diterpenes) of Radix Kansui had pro-inflammatory effects, and some extracts of Fructus Jujubae could attenuate this effect of Radix Kansui. The results helped to clarify that Fructus Jujubae attenuates the toxic effect of Radix Kansui in certain cases, and the bioactive extracts of Fructus Jujubae discovered in the experiments offer the basis for further research to the active components of Fructus Jujubae.
8.Application of water jet-assisted liposuction in augmentation mammoplasty with autologous fat graft

Objective To investigate the clinical value of breast augmentation with autologous fat graft using water jet-assisted liposuction (WAL) technique.Methods From October 2010 to December 2012,20 patients underwent Water Jet-Assisted Lipoplasty and breast augmentation with autologous fat tissue (average,210 ml volume injected per breast).Pretreatment and posttreatment TN,bilateral SN and MP were measured,and the outcomes were statistically analysed using paired t test,combined with photos of pretreatment and posttreatment.Follow-up ranged from 3 months to 2 years.Results The results were satisfactory.All the patients got good shape of breasts.There was no infection in all the patients.TN and SN were signifigantly increased at 3 month after operation [(88.7±2.9) cm vs.(87.2±4.2) cm,P<0.01],[left (20.1± 1.5) cm,right (20.1 ±1.5) cm,vs left (19.5±1.5) cm,right (19.5±1.6) cm,P<0.01].MP was also increased compared with measurement of pre-operation [(4.6±0.4) cm vs.(4.2±0.9) cm,P<0.05].Conclusions It is a good option to use water jet-assisted liposuction technique in the treatment of breast augmentation with autologous fat graft.

10.Association of polymorphisms in angiotensin Ⅱ receptor gene with primary aldosteronism

Objective To investigate the relationship between the polymorphisms of angiotensin Ⅱ receptor gene and the risk of primary aldosteronism (PA).Methods Polymerase chain reaction -restriction fragment length polymorphism (PCR -RFLP)was used to examine the 1 1 66A /C polymorphism of AT1 R gene and 1 675A /G poly-morphism of AT2R gene in 85 patients with PA and 1 00 healthy controls.Results There was no significant difference of AT1 R 1 1 66A /C genotypes (AA,AC,CC)and allele (A and C)frequency among patients and controls (χ2 =0.430,P =0.806).There was obvious difference of AT2R 1 675A /G genotypes (AA,AG,GG)and allele (A and G) frequency among two groups (χ2 =6.1 21 ,P =0.01 3).The G allele was higher than A allele in PA group (χ2 =6.767,P =0.009).Conclusion Homogenic mutation of 1 675A /G site in AT2R gene may be one of risk factors of PA.
