1.Over-expression and purification of recombinant spike protein fragment of SARS coronavirus

Objective:To obtain the spike glycoprotein(S protein) of SARS coronavirus (SARS-Cov) for detection of the corresponding antibody and preparation of diagnostic kits.Methods:Genes encoding S protein were picked out to form a new gene and divided into two parts according to the prediction of antigenicty.They were obtained by nucleotide synthesis and were cloned into expression vector pET32a(+) respectively.Proteins then were induced by IPTG and purified through Ni-NTA.Results:Target proteins were successfully expressed and purified.Conclusion:The recombinant S proteins of SARS-CoV could be of great help in diagnosing the disease and investigating the virus further.
2.Clinical use of mearing antithyroid peroxidase antibody in autoimmune thyroid disease

ObjectiveTo investigate the clinical application of TPO-Ab by comparing the relationship between TPO-Ab and TMAb in ATD. MethodsIn this study with the method of RIA, we measured the serum level of TPO-Ab and TMAb in 200 ATD patients, 50 patients with non-ATD and 30 normal controls, and analyzed the relationship of TPO-Ab and TMAb, compared the differ ence between TPO-Ab and TMAb for diagnosis ATD. ResultsTPO-Ab titer increased in ATD es pecially in Hashimoto's thyroiditis(P<0.01), at the same time it could influence the state of the throid function. A significant positive correlation was found between TPO-Ab and TMAb(γ= 0.83,P<0.01 ), but TPO-Ab appeared to be more sensitive and specific for diagnosising Hashimoto's thy roiditis than TMAb. ConclusionTPO-Ab is a sensitive and specific stantard for diagnosising ATD.
3.Evaluation of Curative Effects of Different Chronic Laryngitis by Voice Acoustic Analysis

Objective To investigate the application of the voice acoustic analysis in the evaluation of curative effects of different chronic laryngitis.Methods The adult patients with chronic laryngitis (80 cases)were divided into 3 different syndrome types what were as the Yin deficiency group (28 cases),the Qi deficiency group (25 ca-ses),and the TanNing blood stasis group (27 cases).According to the different compositions of golden voice series medicines,the medicines were administered,combined in two to three.A total of 30 normal people were set to the control group.The German ATMOS voice test and analysis system was used to detect continuous vowel signals and the results were compared before and after treatment.Results Before the treatment,the chronic laryngitis group, compared with control group,had higher jitters,shimmers and NHR.The MPT was significarry shorter and DSI smaller compared with the control group(P <0.05).After 1 month of medication,the Yin deficiency group was the best and there were no significant differences in almost all parameters compared with control group (P >0.05)ex-cept for the parameter-jitter.The jitter,shimmer,and NHR decreased significantly after 1 month treatment while the MPT and DSI increased significantly in the Yin deficiency group and the Qi deficiency group compared with be-fore treatment (P <0.05).There was a statistically significant difference on jitter,shimmer,NHR and DSI and no difference on MPT compared with before treatment in TanNing blood stasis group.Conclusion The voice acoustic a-nalysis could be objective and quantitative to analysis the chronic laryngitis patients'voice quality with different syn-drome types and evaluate the curative effects.
4.Family of multiple peptide fragments derived from angiotensin and their interaction

It is well established that renin-angiotensin system (RAS) is the major regulatory networkthat maintains blood pressure, fluid and electrolyte balance and the homeostasis of cardiovascular system.Most studies in the last decades centered on the pivotal members of RAS, that is, angiotensinⅡ (AngⅡ) and angiotensin converting enzyme (ACE). Recent identification of ACE-2, the homology of ACE,and the identification of the multiple products degraded from angiotensinⅠ, including AngⅢ, AngⅣ,Ang1 -9, Ang1 -7, Des-Asp-angiotensinⅠ(DAAⅠ), etc, have proved that AngⅡ is not the only bio-logical active compound of RAS. Peptides derived from angiotensinogen,that is, family of angiotensins,display independent and pleiotropic biological activitiesin vivo, and interact with each other both in me-tabolism pathway and the biologic effects. The imbalance of the network of angiotensin metabolites exhib-its significant pathophysiological role in cardiovascular disease.

Treating autoimmune diseases(AID) is a challenge for physicians.Traditionally,we use cortisone or other immune inhibitors to treat AID although we know that these kinds of drugs can cause some severe toxic action on patients if used longer.For over ten years,the biological therapy for AID,which specially targets to the pathogenesis of AID,has long been engaged in the study.These new drugs can protect the normal immune system while stopping undesirable immune reaction to the target organ.

7.Coracolavicar Ligament Reconstructed by Artificial Tendon Applied to Treat Complete Acromioclavicular Joint Dislocation

Objective To explore curative effects of coracolavicar ligament reconstructed with artificical tendon on treatment of complete acromioclavicular joint dislocation.Methods 32 patients with complete acromioclavicular joint dislocation received artificial tendon graft, 19 cases of whose implanted flexible carbonfiber tendon,and the others were treated with human-hair keratin artificial tendon (HHKAT). All patients were given bu internal fixation with Kirschner pins for 6 weeks after the operation.Results Fellowed-up for 4 months to 30 months showed the acromiclavicular join reduction and recovery of the anatomical relation of acromioclavicular joint checked by X-rayrecovered. According to karlsson scores, 21 cases graded excellent, 10 cases good and only one case bad, the excellent and good rate of recovery was 96 9%.Conclutions With the internal fixation with Kirschner pins in short term,and plus artificial tendon reconstructing coracoclavicalar ligament for patients with complete acromioclavicular joint dislocation, which had a little wound and a good curative effect, was easy to be performed and have worth in clinical application.
8.Effect of human umbilical cord blood stem cells on flash visual evoked potential in traumatic optic neuropathy in rats

Objective To investigate the effect of human umbilical cord blood stem cells on flash visual evoked potentials (F-VEP) of the traumatic optic neuropathy rats.Methods Forty-eight Sprague-Dawley rats were randomly divided into an injury group (Group A) and 3 treatment groups (Groups B,C,and D).A traumatic optic neuropathy model was built in Group A,and the rats in Groups B,C,and D were injected with the neurotrophic factor,human umbilical cord blood stem cells,and the mixture of the neurotrophic factor and human umbilical cord blood stem cells,respectively.F-VEP was recorded in both eyes of rats at the 1st h,1st week,2nd week,3rd week,and 4th week after the optic nerve injury.Results At all time points,there were significant difference in the wave latency and amplitude between Group A and normal control eyes (P<0.01).The differences of the wave latency and amplitude between Group A and Groups B,C,and D were statistically significant at various time points after the injury except for the wave latency at the 1st h post-operation (P>0.05).The amplitude in Group D was higher while the latency was shorter than those of Group B at all time points since the 1st week (P<0.05).The comparisons at the same point in the remaining treatment groups were not significantly different (P>0.05).Conclusion The mixture of human umbilical cord blood stem cells and neurotrophic factor has a promotion effect for the recovery of F-VEP of optic nerve in traumatic optic neuropathy in rats to some degrees.
9.Neuroprotective mechanisms of human umbilical cord blood stem cells to retinal ganglion cells in a partial optic nerve crush rat model

BackgroundOptic nerve injury lead to apoptosis of retinal ganglion cells ( RGCs ), and its mechanism of apoptosis is endoplasmic reticulum stress (ERS). So, decreasing of ERS may protect the injury of RGCs. ObjectiveThe present study was to investigate the mechanisms of endoplasmic reticulum stress(ERS) and the protective effects of human umbilical cord blood stem cells on partial optic nerve crush injury. MethodsThe optical nerves were crushed with a 40 g clip by holding for 60 seconds to establish the partial optical nerve injury model in the left eyes of 102 SPF SD rats,and 10 μl of mRNA and 10 μl of nerve growth factor were injected into the vitreous immediately after the establishment of the model. The morphological changes of retinal ganglion cells(RGCs) were examined under the light microscope after 3,7,14,21 and 28 days and the RGCs number was calculated. The apoptosis rates of RGCs were detected by the TUNEL technique after 3, 12,24,45,72 hours and 1 week. The expression levels of GRP78 mRNA and CHOP mRNA were detected by reverse transcription polymerase chain reation (RT-PCR). This procedure followed the Regulations for the Administration of Affair Concerning Experimental Animals by Hunan Provincial Science and Technology Committee.Results The number of RGCs was significantly decreased with the prolongation of time of optical nerve injury in the model injury group,whereas the number of RGCs in the human cord blood cells group was reduced at a slower rate( Ftime =20. 100,P =0. 007 ). At various time points after the injection of human cord blood cells, the survival of RGCs was evidently increased in comparison with the model group(P<0. 01 ). The apoptosis rate of RGCs was considerably elevated with injury time prolongation both in the model group and human cord blood cells group,but no apoptosis was seen from 3-24 hours after operation,and only a small amount of apoptotic cells were found in the human cord blood cells group from 48 hours through 1 week than in the model group(P<0. 01 ). In the human cord blood cells group,GRP78 mRNA level was significantly higher and the CHOP mRNA level was significantly lower than those in the injury group at identical time points(P<0. 01 ). ConclusionsIn the rat optic nerve partial crush model,ERS induces the apoptosis of RGCs. Human umbilical cord blood stem cells can protect RGCs from ERS injury by inhibiting apoptosis.
