3.Comparison of rabbit retinal fixation techniques: chemical fixation and microwave irradiation

Background Conventional chemical method to fix the entire eyeball results in the edema of retinal nerve fiber layer and therefore influent the evaluation of tissue structure.Research showed that microwave irradiation can avoid this phenomenon. Objective This study was to investigate the appropriate energy and time duration of microwave irradiation for fixation of rabbit eyes specimen and compare the influence of different fixation methods on retinal structure. Methods Twenty-two New Zealand white rabbits were sacrificed by using venous air embolism.The eyeballs were enucleated from the rabbits and immersed in chemical reagents with glacial acetic acid,methyl aldehyde and chloroform for 2 days as the control group.The eyeballs were immersed in 400 ml physiological balance solution immediately,and then were fixed by microwave irradiation for 80,160,240 and 320 seconds under the lower power as the microwave irradiation groups.In the microwave irradiation+chemical fixation group,the specimens were immersed in chemical reagents for 1-2 hours after the microwave radiation.The morphology of retinal structure was examined and area of retinal detachment was calculated by hemotoxylin and eosin staining under the light microscope. Results A better fixation effect was obtained in the retinal section of microwave radiation for 240 seconds in comparison with that of the traditional chemical reagent fixation method for 2 days.The retina detachment ratio was 16.3%±11.5% for simple microwave method,50.0%±24.5% for chemical method,and 6.7%±7.8% for microwave+chemical method.showing a significant difference among 3 groups(F=32.43,P=0.000).Retinal staining was clear and retinal structure was almost normal in the specimens of microwave+chemical fixation group with 200-240 seeonds microwave irradiation and 1-2 hours chemical fixation. Conclusion Microwave irradiation method is a more ideal way for the fixation of retina because of taking short duration,lower toxicity and better staining.The combination of microwave radiation and chemical reagents can acquire an excellent quality of retinal section.

5. Preparation of sustained-release tablets of tilianin nanosuspension lyophilized powder

Objective: To prepare sustained-release tablets of tilianin nanosuspension lyophilized powder. The factors that might influence drug release and release mechanism were studied in present study. Methods: High pressure homogenization method was used to prepare tilianin nanosuspension. Lactose and mannitol (3:1) were employed as freeze-drying protective agent to prepare lyophilized powder. HPMC was used as framework material to prepare sustained-release tablets of tilianin nanosuspension lyophilized powder. Based on single factor test, the effects of proportion and amounts of HPMC K4M and HPMC K15, amounts of PEG 4000 and magnesium stearate on in vitro drug release of sustained-release tablets were investigated. Orthogonal test was designed to gain the optimum prescription. Results: The particle size and zeta potential of tilianin nanosuspension were (164.41 ± 9.72) nm and (-37.21 ± 2.38) mV, respectively. The particle size and zeta potential of re-dispersed freeze-drying products were (211.83 ± 11.26) nm and (-31.66 ± 2.92) mV, respectively. The optimum prescription was as follow: the proportion and amounts of HPMC K4M and HPMC K15 were 2:1 and 40 mg, amounts of PEG 4000 was 20 mg, and amounts of magnesium stearate were 0.5%. Sustained release tablets of tilianin nanosuspension were well accorded with Higuchi kinetics model. The equation was Mt/M∞ = 0.286 8 t1/2-0.073 8, r2 = 0.981 4. And the cumulative release could achieve 92.36% in 12 h. The drug release from the tablets was controlled by diffusion and degradation of the matrix. 6.Mechanical proprieties of four cements at different mixing ratios

To investigate the influence of mixing ratios on the hardness,wear resistance and compressive strength of the cements clinically used,Zinc Oxide Eugenol cement,Glass ionomer cements,Zinc Phosphate Cement,and Dual-curing composite cement were mixed at different ratios separately,then made into test pieces.Vickers hardness,wear resistance and compressive strength of these test pieces were measured by related machines.The results showed that vickers hardness,wear resistance and compressive strength of Zinc Oxide Eugenol cement,Glass ionomer cements and Zinc Phosphate Cement reduced with the decrease of powder/liquid ratios.As to Dual-curing composite cement,Vickers hardness,wear resistance and compressive strength decreased with the difference between A/B ratio and 1∶1 increase.Conclusively,mixing ratios could influence the mechanical proprieties of the cements clinical used,so the use of cements should be under the instructions of the product.

Objective To investigate the effects of hypoxia and sodium cyanide(NaCN)on oxidative stress in rabbit arterial blood.Methods An artificial hypobaric hypoxia chamber was used to simulate 4 000-meter high altitude.Twenty rabbits were randomly divided into 4 groups:hypoxia group with high-or low-dose,non-hypoxia with high-or low-dose.The animals in the non-hypoxia groups were operated under normal circumstances while those in hypoxia groups were subjected to chamber in low pressure for 72 h before receiving the hypoxia experiments.Femoral arterial cannulation was performed on all animals under anesthetization with pentobarbital sodium(30 mg/kg,iv)and NaCN(ip)at the doses of 1.5 mg/kg and 2 mg/kg.Blood samples were collected at 10 min before intoxication,and 5,10,15,20,30,60,120 and 180 min after intoxication and blood seperation was conducted.The activity of superoxide dismutase(SOD),contents of reduced glutathione(GSH)and malondialdehyde(MDA)were determined.Results The MDA content was significantly increased(P
8.Influence of sensation of uncertainty in illness on curative effect of two-week rehabilitation program in patients with acute myocardial infarction

Objective: To observe influence of sensation of uncertainty in illness on curative effect of two-week rehabilitation program in patients with acute myocardial infarction (AMI). Methods: A total of 85 AMI patients recently treated in our hospital were selected continuously. They received two-week rehabilitation program treatment and were assessed by Missals uncertainty in illness scale (MUIS). According to MUIS scores, AMI patients were divided into middle-high score group (74.8~117.4 scores, n=51) and low score group (32~74.7 scores, n=34), and they were compared with 43 patients with coronary heart disease (CHD) angina pectoris (CHD control group) received treatment in the same period. Results: Compared with CHD control group, there were significant increase in each dimension score and total score of MUIS [total score (60.61±12.42) scores vs. (78.34±15.20) scores]in AMI patients (P<0.05~0.01). The exercise peak heart rate of MUIS middle-high score group was significantly higher than that of low score group [(137.80±26.49) times/min vs. (126.12±20.51) times/min, P<0.01]. Compared with low score group, there were significant decrease in total scores of activity of daily living (ADL) scale [(84.15±16.38) scores vs. (73.92±14.21) scores] and the 36-item short-form general heath survey [SF-36, (45.22±6.86) scores vs. (37.95±6.43) scores], and significant increase in total score of symptom checklist (SCL)-90 [(138.35±36.47) scores vs. (151.87±42.61) scores], mean days in CCU [(2.53±0.26)d vs. (2.77±0.29)d], mean days on bed [(4.46±0.25)d vs. (5.38±1.22)d], mean hospital day [(20.48±3.16)d vs. (25.37±3.82)d] and mean inpatient fee [(39.1±8.2) thousand RMB vs. (45.7±9.3) thousand RMB] in middle-high score group, P<0.05~0.01. Conclusion: There is significant sensation of uncertainty in illness in patients with acute myocardial infarction, and it makes curative effect of two-week rehabilitation program significantly decrease.

The stability of cell genetic material is influenced by a variety of factors, both internal and external, which can cause various types of DNA damage, such as DNA alkylation, oxidation, mismatching, loop structure, atypical DNA structure, single-strand break, and double-strand break.These DNA damages disrupt cellular homeostasis and dynamic equilibrium, which cause gene mutations, chromosomal abnormalities, and even degradation, aging, and death at different biological levels.By searching and identifying DNA damage sites, the cell activates a series of biochemical pathways, coordinates the progress of DNA replication and transcription, and then repairs the DNA damage.In this way, the cell maintains its independence and stability.While radiotherapy plays a role in eliminating tumors by DNA damages, it also initiates DNA damage responses.Among the responses, base excision repair, nucleotide excision repair, mismatch repair, double-strand break repair, and post-translesion synthesis repair play a key role in repairing the damages.The dysfunction of these repair pathways will cause differences in tumor radiation sensitivity.This paper summarizes recent research results in DNA damage repair, and focuses on the types of DNA damage and their repair mechanisms, so as to promote the understanding of the great significance of this field and to provide a theoretical basis for exploring the application of DNA damage repair pathways in tumor therapy.
