1.Study on stability and degradation kinetics of ketoprofen-paeonol conjugate.

OBJECTIVETo study the stability and degradation kinetics of Ketoprofen-Paeonol conjugate (Ket-Pae).
METHODRP-HPLC method was used to determine the solubility and partition coefficient of Ket-Pae. Stability test was carried out to investigate the factors affecting Ket-Pae. The kinetic studies of Ket-Pae degradation were conducted in different pH buffer solutions and 80% rat plasma at 37 degrees C.
RESULTKet-Pae showed significant degradation phenomenon at high temperature. The solubility of Ket-Pae was decreased about 200 to 300 times compared with parent drugs in water while the lnP increased about 4 times. The degradation curve displayed a V-shape, and kept maximum stability at week acidic (pH 5.0, t(1/2) = 11.4 d). Ket-Pae degraded quickly with very short half life of 1.3 min in plasma, therefore easily released ketoprofen and paeonol.
CONCLUSIONThe lipophilicity of Ket-Pae is increased, its stability is affected by temperature and pH value.

2.Expressions of aquaporins 1 and 3 in the mouse prostate and their significance.

OBJECTIVETo investigate the expressions of aquaporins (AQPs) in the mouse prostatic tissue and their significance, and to provide some evidence for a deeper insight into the physiological function and regulation of AQP expressions in normal and diseased prostatic tissues.
METHODSThe mRNA expressions of AQP0 - 4 in the mouse prostatic tissue were determined by RT-PCR, and the expressions and localizations of AQP1 and AQP3 proteins were characterized by Western blot and immunohistochemistry.
RESULTSRT-PCR exhibited the mRNA expressions of AQP1 and AQP3, but not those of AQP0, AQP2 and AQP4 in the prostate tissue, while Western blot showed the expression of the AQP1 protein with the relative molecular mass (RMM) of 28 000 and those of the glycosylated and non-glycosylated AQP3 proteins with the RMM of 35 000 and 27 000, respectively. Immunohistochemistry indicated the strong expression of AQP1 in the cyst and plasma membrane of the secretary cells and that of AQP3 in the stroma cells of the prostate.
CONCLUSIONThe AQP1 and AQP3 genes were expressed in the secretary epithelia of the mouse prostate tissue, which suggests that AQP1 and AQP3 may play an important role in the secretion of prostatic fluid.

3.Therapeutic effect of Qiliqiangxin capsule combined Xinhuosu on refractory heart failure and its influ-ence on miR-132 and miR-155

Objective:To study therapeutic effect of Qiliqiangxin capsule combined Xinhuosu on refractory heart fail-ure(RHF)and its influence on miR-132 and miR-155.Methods:A total of 100 RHF patients treated in our hos-pital were divided into Xinhuosu group(received Xinhuosu based on routine treatment)and combined treatment group(received Qiliqiangxin capsule based on Xinhuosu group).Both groups were treated for one month.Serum levels of N-terminal pro-brain natriuretic peptide(NT-proBNP),transforming growth factor(TGF)-β1,ma-trix metalloproteinase(MMP)-9,interleukin(IL)-6,tumor necrosis factor(TNF)-αand soluble intercellu-lar adhesion molecule-1(sICAM)-1,expression levels of miR-132 and miR-155 before and after treatment and total effective rate were compared between two groups.Results:Compared with Xinhuosu group,after treat-ment,there were significant reductions in serum levels of NT-proBNP[(2838.76±380.26)pg/ml vs.(2450.53±289.83)pg/ml],TGF-β1[(39.74±5.40)μg/ml vs.(35.34±4.72)μg/ml],MMP-9[(276.62±30.61)ng/L vs.(230.37±27.86)ng/L],IL-6[(7.09±1.54)ng/L vs.(5.10±1.25)ng/L],TNF-α[(90.11±16.38)ng/L vs.(63.82±12.25)ng/L]and sICAM-1[(239.30±45.30)ng/ml vs.(192.33±34.41)ng/ml],and ex-pression level of miR-155[(1.37±0.22)vs.(1.24±0.22)],and significant rise in expression level of miR-132[(1.47±0.24)vs.(1.62±0.33)]in combined treatment group(P<0.05 or<0.01).Total effective rate of combined treatment group was significantly higher than that of Xinhuosu group(94.00%vs.76.00%,P=0.012).Conclusion:Qiliqiangxin capsule combined Xinhuosu can significantly improve clinical therapeutic effect,relieve my-ocardial injury,improve myocardial fibrosis and ventricular remodeling,alleviate inflammatory response and regu-late expression of miR-132 and miR-155 in patients with refractory heart failure.
4.Meta-integration of qualitative research on breastfeeding experience in professional women

Objective:To systematically evaluate the real breastfeeding experience in professional women.Methods:Qualitative studies on breastfeeding experience and needs in professional women which were published up to August 20, 2020 were retrieved from English databases such as PubMed, Medline, The Cochrane Library, Embase, Web of Science, Ovid, CINAHL, Joanna Briggs Institute Library, Chinese databases such as CNKI, Wanfang, VIP, SinoMed, and grey literature databases like ProQuest. After literature screening and evaluation, a collection integration method was used to integrate the results.Results:A total of 12 studies were included, and 57 complete research results were extracted, which were summarized into 8 new categories integrated into 3 new results: working mothers benefited from breastfeeding but also had negative experience due to returning to work; their breastfeeding was affected by multiple promotion and obstacles after returning to work; and they dealt with breastfeeding challenges after returning to work through self-management.Conclusions:We should actively pay attention to the breastfeeding experience of professional women, improve the support for breastfeeding of working mothers, and help them improve their self-management ability, in order to promote their breastfeeding behavior after returning to work.
5.Construction of evaluation index system of occupational protection competency of nurses in Operating Room

Objective:To construct the evaluation index system of occupational protection competence of Operating Room nurses based on the competency model.Methods:A research team was established in October 2022 to preliminarily construct an evaluation index system for occupational protection competency of Operating Room nurses through literature retrieval and expert interviews. From November to December 2022, 2 rounds of expert letter consultation were conducted with 23 experts, and the evaluation index system of occupational protection competence of Operating Room nurses was finally established. The enthusiasm of experts was expressed through the effective response rate of the inquiry questionnaire, the degree of expert authority was expressed using the expert authority coefficient, and the degree of coordination of expert opinions was expressed using the Kendall's W coefficient. Results:The effective recovery rates of the questionnaire in 2 rounds of expert consultation were respectively 100.00% (23/23) and 91.30% (21/23), the expert authority coefficients were respectively 0.878 and 0.920, and Kendall's W of the overall index in 2 rounds of letter consultation were respectively 0.239 and 0.312 ( P<0.01). Finally, an evaluation index system of occupational protection competency of Operating Room nurses was formed, including 4 first-level indexes, 12 second-level indexes and 53 third-level indexes. Conclusions:The competency evaluation index system of occupational protection for Operating Room nurses based on competency model is scientific and reasonable, which can provide training ideas for future clinical training practitioners and college educators.
6.Summary of best evidence for non-pharmacological management of postoperative delirium in adult liver transplant recipients

Objective:To summarize the best evidence for non-pharmacological management of postoperative delirium in adult liver transplant recipients, providing reference for postoperative delirium management in liver transplant recipients.Methods:Evidence on non-pharmacological management of postoperative delirium in adult liver transplant recipients was retrieved through computer in databases such as UpToDate, British Medical Journal (BMJ) Best Practice, Embase, PubMed, Cochrane Library, Joanna Briggs Institute Evidence-Based Health Care Center Database, Guidelines International Network, China Biology Medicine disc, China National Knowledge Infrastructure, WanFang Data, and VIP. The search period was from database establishment to January 31, 2023. The types of literature included guidelines, clinical decision-making, best practices, systematic review, summary of evidence, and expert consensus. Two researchers conducted quality evaluation and evidence extraction on the included literature.Results:A total of 10 articles were included, involving 1 clinical decision-making, 4 guidelines, 3 systematic reviews, and 2 expert consensuses. A total of 25 best pieces of evidence for non-pharmacological management of postoperative delirium in adult liver transplant recipients were summarized from four aspects, including screening, evaluation, non-pharmacological prevention, and non-pharmacological treatment.Conclusions:The best evidence on non-pharmacological management of postoperative delirium in adult liver transplant recipients summarized provides a reference for postoperative delirium management in liver transplant recipients.
7.Mediating effect of individual resilience between work stress and work engagement among nurses in Emergency Department

Objective:To explore the relationship between individual resilience, work stress, and work engagement among nurses in Emergency Department.Methods:A questionnaire survey was conducted on 350 nurses from six Class Ⅲ Grade A hospitals in Shandong Province from April to September 2022, using the General Information Questionnaire, the Chinese version of the Stressor Scale for Emergency Nurse, the Chinese version of the Utrecht Work Engagement Scale, and the Medical Staff Composition Resilience Scale. Pearson correlation was used to analyze the correlation between nurse work stress, work engagement, and individual resilience. The structural equation model was constructed using a mediating effect testing program to analyze the relationships between variables.Results:A total of 350 questionnaires were distributed, and 330 valid questionnaires were collected, with an effective response rate of 94.28%. The scores of nurse work engagement, work stress, and individual resilience were (54.26±10.08), (77.36±25.48), and (67.76±12.33), respectively. Work stress was negatively correlated with work engagement and individual resilience (all P<0.01), while individual resilience was positively correlated with work engagement ( P<0.01). Individual resilience was a mediating variable between work stress and work engagement among nurses in Emergency Department, with a mediating effect accounting for 31.38% of the total effect. Conclusions:Most nurses of Emergency Department have moderate levels of work stress, work engagement, and individual resilience. Individual resilience partially mediates the relationship between work stress and work engagement among nurses in Emergency Department. Nursing managers should attach importance to the organizational culture atmosphere and develop corresponding measures to enhance the individual resilience of nurses and encourage them to be energetic and enthusiastic in their work.

The skeletal muscle atrophy could be induced by the injury of nerve. According to the source of denervated skeletal mus-cle atrophy, it could be divided into exogenous muscle atrophy and endogenous muscle atrophy. In recent years, the ex-ogenous muscle atrophy models are mainly established by operating, physically injuring or chemically injuring, while the endogenous muscle atrophy models are mainly established by the transgenic animals of amyotrophic lateral sclero-sis. The selection and optimazation of animal models are crucial for the basic studies of denervated skeletal muscle atro-phy.
9.Effects of Electroacupuncture on Autophagy-related Gene Expression in Skeletal Muscle in Denervated Rats

Objective To explore the effects and mechanism of electroacupuncture(EA)on denervation-induced atrophy. Methods A total of 21 male Sprague-Dawley rats were divided into sham group(n=7),model group(n=7)and EA group (n=7).The latter two groups were cut off their right sciatic nerve.Since one day after modeling,EA group accept-ed electroacupuncture at right Zusanli(ST36)and Huantiao(GB30)for eight weeks.Then,the gastrocnemius of all the rats were obtained,and measured the wet mass ratio.Cross-sectional area(CSA)and fiber diameter were measured after HE staining. The expression of autophagy-related gene ULK1, Atg13, Beclin1, Atg14, Atg7, Atg12,Atg5 and Atg16L1 were tested with reverse transcription real-time quantitative polymerase chain reaction. Results Compared with the sham group,the wet mass ratio,CSA and fiber diameter of gastrocnemius were lower signifi-cantly in the model group and EA group(P<0.001),and they were more in EA group than in the model group(P<0.05).Compared with the sham group,the mRNA expression of ULK1,Atg13,Beclin1,Atg14,Atg7,Atg12,Atg5 and Atg16L1 was more significantly in the model group (P<0.001), and they decreased in EA group compared with those of the model group(P<0.05). Conclusion Electroacupuncture can inhibit the overexpression of autophagy-related gene in denervated rats,which may steady skeletal muscle cells to delay atrophy.
10.Study on the acupoints belonging to the three yin meridians of foot reflecting the variation pattern of uterine qi and blood in women with moderate constitution

Objective: To observe the blood perfusion volume variation pattern in the body surface microcirculation at the Yuan-Primary and the Xi-Cleft points during the menstrual cycle in female college students with moderate constitution under normal physiological state of the uterus; to explore the specific laws of the body surface microcirculation at the Yuan-Primary and Xi-Cleft points in response to the uterine qi and blood changes under normal physiological conditions, and to provide the experimental basis for the specificity of acupoints reflecting the uterine function. Methods: Forty-three healthy and moderate constitution female college students with regular menstrual cycles, without dysmenorrhea and not yet giving birth were recruited. Bilateral Yuan-Primary points [Taichong (LR 3), Taibai (SP 3) and Taixi (KI 3)] and Xi-Cleft points [Zhongdu (LR 6), Diji (SP 8) and Shuiquan (KI 5)], belonging to the three yin meridians of foot and adjacent to the spinal cord segment of the uterus, were selected as the detection acupoints; the crossing point of the three yin meridians of foot [Sanyinjiao (SP 6)], the uterus-related meridian acupoint [Xuehai (SP 10)], the uterus-non-related meridian acupoint [Xuanzhong (GB 39)], and the non-meridian non-acupoint point were selected as the control points. The laser speckle blood flow imaging technique was used to monitor the blood perfusion volume in skin microcirculation at the above points at the menstrual, follicular, ovulatory, and luteal phases of the subjects. Results: The blood perfusion volume in the body surface microcirculation at the right Zhongdu (LR 6) at the ovulatory phase was higher than that at the menstrual, follicular and luteal phases (all P<0.05); there was no significant difference in the microcirculation blood perfusion volume at the other points among different phases (all P>0.05). Conclusion: The blood perfusion volume in the body surface microcirculation at Zhongdu (LR 6), the Xi-Cleft point of the Liver Meridian, shows a specific response to qi and blood changes in the uterus of women with moderate constitution.
