1.Evaluation of passage stability of H5N1(NIBRG-14) influenza virus vaccine strain in MDCK cells of serum-free suspension culture

Objective To evaluate the passage stability of H5N1(NIBRG-14) influenza virus vaccine strain in MDCK cells(sMDCK)of serum-free suspension culture.Methods H5N1(NIBRG-14) influenza virus working-bank vaccine strains were passed 15 consecutive times in sMDCK cells.The 8-segment nucleotide sequences(HA,NA,M,NP,NS,PA, PB1 and PB2 genes) of the main-bank,working-bank,virus of P1,P2,P3,P5,P10 and P15 generations were detected for genetic stability by second and first generation sequencing.The stability of amino acid sequences of hemagglutinin(HA)and neuraminidase(NA),the main antigens of the working-bank,P5 and P15 generation viruses,were evaluated by using peptide coverage as indicators;Influenza vaccine was prepared with working-bank,P5 and P15 generation viruses,with which the female BALB/c mice were immunized i.m.with 10 in each group,15 μg HA per mouse,and boosted 28 d later at the same dosage and route.At 28,42 and 56 d after the primary immunization,the mice were detected for the titer of neutralizing antibody in serum to evaluate the stability of immunogenicity.Results No segment insertion or deletion was detected in each generation of influenza virus,and the nucleotide sequence was completely consistent with the main-bank;Single nucleotide polymorphism(SNP) mutations did not occur in the main-bank,working-bank,P1,P2,P3,P5 and P10 generations of viruses,while the possibility of SNP mutation showed in many gene loci of P15 generation virus,with heterozygous SNP accounting for 91.62%.The coverage rate of HA and NA protein peptides of P5 and P15 generation viruses ranged from96.7% to 100%.There was no significant difference in serum neutralizing antibody titer of mice in the working-bank,P5 and P15 groups(H=2.253,2.029 and 1.408,P=0.324,0.363 and 0.495,respectively) at 28,42 and 56 d after the first immunization.Conclusion H5N1(NIBRG-14) influenza virus vaccine strain has good genetic stability in sMDCK cells,which is expected to be used in the production of sMDCK cell matrix pandemic influenza vaccine.

4.The expression and effect of elastin in full-term rats with hyperoxia induced by chronic lung diseases

Objective To investigate the expression and effect of elastin in full-term rats with hyperoxia induced by chronic lung diseases. Methods One hundred and forty-four full-term rats are randomly exposed to hyperoxia (hyperoxia group)and room air(room air group).Chronic lung disease(CLD)is induced by hyperoxia exposure. Gomori's stain for elastic fibers and in situ hybridization methods were used to detect the expressions of secondary crest and tropoelastin mRNA on the 1st,3 rd, 7th, 10th,14th and 21st days after exposure. Results The expressions of secondary crest decreased significantly in hyperoxia group, compared with room air group on the 3rd to 14th days(P<0.05).The expressions of tropoelastin mRNA decreased significantly in hypemxia group, compared with room air group on the 3rd to 10th days (P<0.05),otherwise increased significantly from the 14th to 21 st days(P<0.05). Hyperoxia exposure can delay the peak of tropoelastin mRNA. Conclusion Elastin is involved in the inhibition of alveolarization and lung fibrosis in the development of CLD.

This study was aimed to evaluate the clinical effect of 66 yin-deficiency acne cases treated with body constitution adjustment using acupuncture and moxibustion. In this research, 66 acne patients with the body con-stitution of yin-deficiency were selected . These patients were randomly divided into two groups ( 33 every group ) . The acupuncture treatment group was treated by acupuncture and local treatment. And moxibustion treatment group was treated by indirect moxibustion and local treatment. The treatment was given twice a week. And the improvement condition of acne skin lesions and yin-deficiency body constitution were evaluated after 12-week treatment. The results showed that certain curative effects were received in both groups. However, the moxibustion treatment group had higher curative effect compared with the acupuncture treatment group. There were significant statistical differences between two groups ( P < 0 . 05 ) , which meant moxibustion with local treatment had a higher curative effect than acupuncture with local treatment to acne. In the aspect of skin lesion improvement, according to the statistics, the moxibustion treatment group were statistically significant compared with acupuncture group (P< 0.05), which meant more obvious improvement of skin in the moxibustion treatment group. In the aspect of body constitution, the body constitution scores in both groups were significantly declined and the moxibustion treatment group was more obvious. There was statistical significance between two groups (P < 0.05), which meant patients in the moxibustion treatment group obtained a more obvious body constitution improvement. After the treatment, follow-up was given to all patients for 4 weeks. The results showed that both the acne skin lesion con-dition and yin-deficiency body constitution score were decreased and not achieve the recurrence diagnostic stan-dard. It showed that acupuncture and moxibustion are able to activate the self-regulation and disease resistibility of the body. It was concluded that acupuncture and moxibustion not only improve skin lesion of acne patients with the body constitution of yin-deficiency with inner heat , but also regulate yin-deficiency body constitution . Moxibustion had higher efficacy in the treatment of yin-deficiency with inner heat acne than acupuncture.
9.Experimental study on quick determination of viability of Oncomelania hu-pensis by staining

Objective To find out a quick,simple and convenient method of determining the viability of Oncomelania hupen-sis. Methods O. hupensis snails were stained for 30 minutes by 0.05%water soluble dye neutral red,0.5%methylene blue,red ink,methylene blue-eosin-borax(MEB)and 0.4%trypan blue,respectively. The soft tissue samples of the snails were observed by a stereoscopic microscope after crushing their shells. Results The vital snails were stained and the dead were unstained in the neutral red. The vital and dead snails were unstained in methylene blue. However,the vital and dead snails were stained in red ink. The partial vital and dead snails were stained in MEB. The vital snails were stained and the partial dead were stained in trypan blue. Conclusion The use of 0.05%water soluble dye neutral red is simple,rapid and accurate in determination of the viability of O. hupensis.
10.Structure and level of the stress of nursing students in clinic practice

Objective To describe the structure and the level of the stress of nursing students in clinic practice,and supply reference for reform of clinical teaching method.Methods Totally 100 undergraduate nursing students in clinic practice,who would graduate from nursing school and begin to work in 2013,were interviewed and investigated.Results The main stress sources were divided into five aspects:recognition,clinical nursing ability,career development,field response and exhaustion of body and mind.Conclusions The structure and level of the stress of nursing students changed a lot.Some new appropriate reform should be introduced into clinic education,in order to help nursing students to complete psychological change and role adaptation.
