1.Expression of Nek2, Plk1 and Cdk1 in gastric carcinoma and their clinicopathological signifi-cance

Purpose To detect the expression of Nek2, Plk1 and Cdk1 in gastric carcinoma and to explore their correlation with clini-copathological factors. Methods We used immunohistochemistry to detect the protein expression of Nek2, Plk1 and Cdk1 in 80 cases of gastric carcinoma. Results The positive rates of Nek2, Plk1 and Cdk1 expressions in 80 cases of gastric carcinoma were 64%, 79% and 85% respectively. While the positive rates of Nek2, Plk1 and Cdk1 expressions in 20 cases normal gastric tissue were 15%, 10% and 20% respectively,with statistical significance (P<0. 01). There were significant associations between Nek2 expression and differentiation of gastric cancer, invasion depth, lymph node metastasis and TNM stage (P<0. 05). There were no significant associa-tions between Nek2 expression and age, gender and tumor size (P>0. 05). The expression of Plk1 and Cdk1 had a correlation with tumor size, differentiation of gastric cancer, invasion depth, lymph node metastasis and TNM stage (P<0. 05). There were no signifi-cant associations between Plk1 and Cdk1 expressions and age and tumor size (P>0. 05). There were significant associations between Nek2 and Plk1, Plk1 and Cdk1, Nek2 and Cdk1 expression (P <0. 01). Conclusions Abnormal expression of Nek2, Plk1 and Cdk1 might be one of the mechanisms of tumorigenesis, especially of abnormal tumour proliferation. They may represent new potential targets for therapeutic intervention.
2.Anticancer effect of crocin

Crocin is a chemical extraction from saffron and it is the most important kind of pigment of saffron.Recently,the research on cervical cancer,bladder cancer,leukemia,tongue squamous cell carcinoma,colon cancer,pancreatic cancer shows that crocin has a role in inducing apoptosis of the tumor cells by activating caspase-3,regulating the ratio of bcl-2/bax and repressing the expression of survivin.Crocin has a strong inhibitory effect on tumor proliferation by inducing cell cycle to arrest through down-regulating the expression of cyclin D1 and up-regulating the expression of p27kip1.Crocin can also exert antitumor effects by liposome encapsulation to enhance cytotoxic effect.Crocin is a kind of effective chemical for tumor treatment and prevention with no significant toxico-and by-effect in vivo,and is likely to be used as an anticancer drug clinically in the future.
3.Morphogenesis and histochemistry of roots in Codonopsis pilosula

Objective To study the process of the root development of Codonopsis pilosula as well as theaccumulatinglocationsofthemainmedicinalcompositionandstoragenutritionintheroot. MethodsThe development process of the roots was studied by paraffin section methods. Storage nutrition was loca-lized by PAS reaction and Sudan Ⅲ reaction. The medicinal composition of C. pilosula, terpene and steroid were stained with mix reagent of 5% vanillin-acetic acid and perchloric acid. Results The development process of the root is shown as four stages: promeristem, primary meristem, primary structure, and secondary growth. Abundant laticifers exist in mature root and the prime laticifers could be found in phloem as early as the first vessel of primary xylem appears. Then, the laticifers exist in phloem at all time. Starch granule accumulated in mature root as reserved substance. Terpene and steroid accumulated in parenchyma and laticifer. Conclusion Parenchyma and laticifer in the roots of C. pilosula are the main parts of the roots to store medicinal composition and storage nutrition.
4.Observation on compressive strength of veneers on mottled teeth after pulse Nd:YAG laser irradiation

Objective To analyze experimentally the compressive strength of veneers on mottled teeth after Dul8e Nd:YAG laser irradiation.Methods Sixty motded teeth were extracted,including thirty maxdlary central incisors and thirty maxillary lateral incisors and each were divided equally into two groups randomly(experimental groups and control groups).The former were irradiated by pulse Nd:YAG lafer and then etched by 37%phosphorus acid.The latter only etched by 37%phosphorus acid.All the teeth were restored by composite resin veneers and tested with Instron 4505 electronic testing machine.The average collapse loads of the veneers of incisors were tested. The maximal compressive loads of the samples were obtained,then the experimental results were analyzed.Results The average collapse loads of central incisors of test groups and control groups were(0.342±0.053),(0.289±0.041) kN respectively,with a signiticant difference(t=3.16,P<0.05).The average collapse loads of lateral incisors of test groups and control groups were(0.321±0.041),(0.208 ±0.032)kN respectively.There was a significant difference when they were compared(t=7.66,P<0.05).Conclusions The compressive strength of the veneers incneases significandv when the mottled teeth are irradiated by pulse Nd:YAG laser.This method is worthy of recommending.
6.Effect of Restraint and Watered Stress of Different Durations on Gastric Empting in Rats

Objective To study the effect of restraint and watered stress of different durations on the gastric empting in rats. Methods 48 male SD rats (weight ±150 g) were divided randomly into 6 groups with 8 in each. The experimental rats were restrained and watered in 23℃ water box for 1 h/d, while the controls were free. The body weight, general conditions and behavioral indexes of the rats were observed and recorded. The gastric empting rate was measured in rats with semi-solid diet after 3 d,7 d and 28 d during the statistically significant differences among different groups of different stress groups,and between the 3 d test group and 3 d control group. The gastric empting rate was higer in the 7-day test than in 7 d controlled group, lower in the 28 d test group than 28 d control group. Conclusion Different chronic stress can cause changes of gastric empting rate. With the prolonging of stress time, the empting rate is rising and then decending.
7.Expression and clinical significance of CAK complex in estrogen receptor-positive breast cancer

Objective To evaluate the expression and clinical significance of cyclin dependent kinase (CDK)-activating kinase (CAK) complex including CDK7, cyclin H and accessory protein menage a trios 1 (MAT1) in estrogen receptor-positive breast cancer.Methods A total of 40 patients with estrogen receptor-positive breast cancer from Department of Galactophore, Baoji Maternal and Child Care Service Centre of Shaanxi Province were investigated in this study.Breast cancer tissues and adjacent normal tissues were obtained from patients undergoing surgery.The mRNA expressions of CDK7, cyclin H and MAT1 in two types of tissues were measured by real-time fluorescent quantitative (qRT)-PCR, and their correlations with clinicopathologic features of patients were analyzed.Results The expressions of CDK7, cyclin H and MAT1 in cancer tissues were 2.54±0.78, 2.21±0.56 and 2.46±0.58, while those in adjacent normal tissues were 1.26±0.30, 1.16±0.42 and 1.17±0.39, and there were significantly differences between different types of tissues (t=9.654, P<0.001;t=9.433, P<0.001;t=11.741, P<0.001).The higher expressions of cyclin H and MAT1 in patients′ cancer tissues had the lower clinical stage, with significant correlations (U=3.17, P=0.01;U=2.53, P=0.01).In addition, the tumors were smaller in patients with higher expression levels of MAT1 (χ2=14.16, P=0.01).Conclusion The expressions of CAK complex CDK7, cyclin H and MAT1 are elevated in estrogen receptor-positive breast cancer patients.Cyclin H and MAT1 are closely associated with clinical grade, and MAT1 is also significantly associated with tumor size.

Objective To evaluate retroperitoneal laparoscopic Anderson-Hynes pyeloplasty. Methods 6 patients underwent laparoscopic and 10 patients underwent open Anderson-Hynes pyeloplasty.The clinic outcome including subjective and objective findings was compared between the two groups. Results All the procedures were successfully completed.The mean operating time was 279 minutes for laparoscopic pyeloplasty being much longer than the open procedure 121 minutes(P0.05).No severe postoperative complications were found.3 months after the procedure,the pain-free rate was about the same between the two groups.However the open surgery group had much pain and prolonged convalescence due to the flank incision. Conclusions Laparoscopic pyeloplasty is a minimally invasive technique that provides similar clinical and radiographic results as open pyeloplasty.
