More than 100 countries have added hepatitis B vaccination to their routine immunization programmes.Hepatitis B vaccines progressed from blood vaccines to recombinant vaccines,then to peptide vaccines and DNA vaccines,to their functions extended from prophylactic to therapeutic.This article analyzes the relationship between hepatitis B vaccine R&D and the immunization programmes in different countries,to summarizes the R&D of both prophylactic and therapeutic vaccines,and points out the direction for futare vaccine development.

3.Differential expression of long chain noncoding RNA in PBMCs in health people and patients with rheumatoid arthritis and its pathogenesis

Objective To explore the differentia expression of long chain noncoding RNA (IncRNA) in peripheral blood mononuclear cells (PBMCs) of the patients with rheumatoid arthritis (RA) and normal human and to analyze to further clarify the pathogenesis of RA.Methods Twenty-two cases of RA in our hospital from January to April 2016 were selected as the observation group and 22 healthy persons served as the control group.The differential expression of IncRNA and mRNA in PBMCs of 5 RA cases and 5 healthy persons were determined by using Agilent human IncRNA chip;GO and Pathway were used to analyze the functional distribution of differentially expressed IncRNA,the co-expression pressionsnetwork was constructed and the Trans-and Cis -were adopted to predict the RA onset related IncRNA.Results There were 1623 differential expressions of IncRNA in PBMCs of RA patients and healthy persons and 851 differential expressions of mRNA.GO and Pathway analysis showed that the main function of differentially expressed mRNA was to participate in the binding and transcriptional regulation of protein kinase,nucleotide and metal ions,and also participate in the regulation of B cell receptor signaling pathway and TNF signaling pathway.After Cis-and Trans-analysis prediction and real-time PCR validation,the 3 IneRNA could be related with RA onset,which were NONHSAT 120696,uc.80+ and NONHSAT 031501.Conclusion IncRNA has differential expression in PBMCs of RA patients and healthy persons,three 031501 IncRNA of NONHSAT 120696,uc.80+,NONHSAT may be related to the pathogenesis of RA.
4.ERCC1 expression and prognosis in non-small cell lung cancer

Objective To explore the expression of Excision repair cross -completion 1 in non-small cell lung cancer and to analyze its clinical significance .Methods ERCC1 protein expression was detected by IHC method in 160 cases of non-small cell lung cancer tissue .We analyzed relationship between the ERCC 1 ex-pression and clinicopathological parameters of non -small cell lung cancer ,as well as its correlation between ER-CC1 expression and the prognosis .At the same time ,160 cases of intraoperative specimens were primarily cul-tured.The relationship between ERCC1 expression and the drug sensitivity of DDP was analyzed .Results ER-CC1 protein was mainly expressed in the nucleus .ERCC1 protein expression in lung cancer tissue showed no rela-tionship with age,gender,tumor size and pathological types .ERCC1 protein expression was associated with TNM stages and lymph node metastases .High expression of ERCC1was significantly correlated with poor prognosis in pa-tients,indicating that ERCC1 is the independent factor of NSCLC patients .MTT results showed that the positive ex-pression rate of ERCC1 in DDP sensitive groups was higher than in DDP resistant groups .Conclusion ERCC1 is an independent factor influencing the prognosis of non -small cell lung cancer ,and associated with DDP resistance .
5.Characterization and Sequence Analysis of 28S rDNA of Cryptococcus neoformans

Objective To investigate genetic relationships among five serotypes of two variants of Cryptococcus neoformans. Methods PCR mediated DGGE (denaturing gradient gel electrophoresis) and sequence analysis of 28S rDNA of C. neoformans were performed in ten reference strains, C. neoformans capsular-deficient strain CAP10, and nineteen clinical isolates from non-HIV patients. Results The results of DGGE and analysis of nucleotide sequences of 28S rDNA showed identical patterns and nucleotide sequences in the serotype A and D of C. neoformans var. neoformans, which were distinct from the serotye B and C of C. neoformans var. gattii. The patterns and sequences of serotype AD coincided with those of C. neoformans var. gattii. The patterns and nucleotide sequences of C. neoformans capsular-deficient strain CAP10 (serotype D) and serotype A and D were identical. Of the nineteen clinical isolates, seventeen had patterns of serotype A and D, and the others had patterns of serotype B and C. Conclusions PCR mediated DGGE integrated with sequence analysis of 28S rDNA is a valuable tool for the classification of C. neoformans. The clinical isolate of C. neoformans var. neoformans is predominant in Chinese non-HIV patients. Serotype AD is genetically close to C. neoformans var. gattii rather than C. neoformans var. neoformans. The data seem not to be in favor of previous study that serotype A, C. neoformans var. grubii, is a new variant of C. neoformans.
6.CT-guided biopsy of malignant lymphoma

Objective To evaluate the value of CT-guided biopsy of deep-located lesion in the diagnosis of lymphoma. Methods CT-guided percutaneous biopsy was performed in 58 patients with 16-20 gauge core-needle biopsy. The locations of lesion involved mediastinun, lung, retroperitonurn, spleen, kidney, adrenalal and musculoskeletal system. Pathology examination included hematoxylin and eosin staining and immunohistochemical assays. Results In 56 out of 58 cases,the biopsy findings could be confirmed by histologic examination including correctly diagnosed 47 malignant lymphomas, corresponding to a sensitivity of 81%;with fuithecmace subclassification 42 of the 47(89.4%) could be as diagnosed malignant lymphomas on the basis of CT-guided biopsy. Conclusions Biopsy of deeply located lymphoma mass under CT guidance has high diagnostic accuracy and low complication rate with convenience for subclassification of malignant lymphomas.
7.CT-guided percutaneous mediastinum biopsy

Objective To study the technique and clinical significance of percutaneous mediastinum biopsy guided by CT. Methods 35 cases with mediastinal masses were undergone this procedure. Results The successful rate of the procedure was 100%. The successful punctural rates reached 100% for malignant neoplasms(21/21)85.71% for benign lesions(11/14), and the diagnostic accuracy was 94.21%(33/35). The complication of pneumothorax occurred in 3 cases, and mediastinal bleeding presented in 1 case. Conclusions CT-guided percutaneous mediastinum biopsy possesses high diagnostic accuracy, simultaneously with great help for the management.

0.05).The transfected HepG2 cells by infection of SL3261 containing the vector were shown with GFP and the 930 bp target expression by RT-PCR.Conclusion:Attenuated Salmonella typhimurium SL3261 containing recombined eukaryotic expressing plasmid pIRES2-EGFP-4-1BBL is successfully constructed which can deliver recombinant plasmid into HepG2 cells.
9.LZTS2 tumor suppressor gene and advancements in tumor re-search

Leucine zipper tumor suppressor 2 (LZTS2) is a novel tumor suppressor gene that has been increasingly recognized in recent years. Currently, many studies illustrate that LZTS2 gene, the important candidate tumor suppressor gene, is already involved in the inhibition of tumorigenesis and aberrant proliferation of tumor cells, and other functions of tumor cells. Information from these stud-ies can contribute to the formulation of new strategies for the treatment of tumors.
10.Evaluation of eye injury degree of laser irradiation frequency and single duration

Objective:To explore the effects of laser irradiation parameters (irradiation frequency and single duration) on tear secretion, lens and retina.Methods:Thirty-six healthy guinea pigs were randomly divided into 6 groups with random number table method according to different frequency and single exposure duration of laser to the eye, namely, high frequency short time (HFST) group, high frequency long time (HFLT) group, medium frequency short time (MFST) group, medium frequency long time (MFLT) group, low frequency short time (LFST) group and low frequency long time (LFLT) group, 6 for each group.The right eyes were irradiated with 500 lx laser as experimental eyes, and the left eyes of the guinea pigs served as the control eyes.The high, medium and low irradiation frequencies were defined as 15 times, 10 times and 5 times, respectively, and the short and long period was defined as 30 seconds and 60 seconds each time, respectively.The right eyes were irradiated based on the grouping at a 10-minute interval.The tear secretion was detected by SchirmerⅠtest; lens opacity was assessed under the slit-lamp microscope; fundus photography was performed to evaluate the general morphology of retina; retinal function was evaluated by electroretinogram (ERG) record and the thickness of retinal outer nuclear layer was measured by histopathology examination.This study protocol was approved by the Medical Ethics Committee of Air Force Military Medical University (No.20181203), and the use and care of the experimental animals complied with the ARVO statement.Results:The tear secretion was 8.00(7.37, 9.00), 8.75(8.25, 9.00), 8.50(7.75, 9.50), 9.00(8.50, 9.50), 8.00(7.37, 8.75) and 8.25(7.75, 8.75) mm/5 min in the HFST group, HFLT group, MFST group, MFLT group, LFST group and LFLT group, respectively, without significant difference among the groups(χ 2=5.502, P=0.240); after laser irradiation, there were no statistically significant differences in tear secretion between the control eyes and laser-irradiated eyes in all the groups (all at P>0.05). The lenses were clear and the fundus was normal through the experimental duration in all the groups.The amplitude of ERG a-wave was significantly reduced in the HFST group in comparison with the LFST group (P<0.05), and there was no significant difference in the b-wave amplitude among the six groups (F=1.358, P=0.268). The ERG a-, b-wave amplitudes were not significantly different between the control eyes and laser-irradiated eyes in various groups (both at P>0.05). There was no significant difference in the thickness of the outer nuclear layer of retina among the HFST group, HFLT group, MFST group, MFLT group, LFST group and LFLT group (F=0.952, P=0.463). Conclusions:The 500 lx laser irradiation is safe to ocular surface and lens, but there are some injuries to retinal function, and the injury degree is related to laser irradiation frequency.
