1.The mechanism by which bone marrow mesenchymal stem cells participate in apoptosis of hepatic stellate cells

BACKGROUND:Control of hepatic stelate cel activation and proliferation is the focus of developing strategies against liver fibrosis. Human or murine bone marrow mesenchymal stem cels can induce apoptosis of hepatic stelate cels through paracrine of hepatocyte growth factors. OBJECTIVE:To explore the mechanism by which bone marrow mesenchymal stem cels participate in apoptosis of rat hepatic stelate cels. METHODS:Hepatic stelate cels and bone marrow mesenchymal stem cels were seeded and co-cultured in the upper and lower chambers in a co-culture system, serving as a co-culture group. In the blank control group, only hepatic stelate cels were involved. In the c-Met inhibitor group, hepatic stelate cels and bone marrow mesenchymal stem cels were treated with 3 mg/L C-Met inhibitor. In the RhoA inhibitor group, both kinds of cels were treated with 3 mg/L RhoA inhibitor. RESULTS AND CONCLUSION:The concentration of c-Met inhibitor was 3.0 mg/L. RhoA inhibitor at 30μmol/L exhibited a greater inhibitory effect than at other concentrations. RhoA mRNA and protein expression in the co-culture, c-Met inhibitor and in particular RhoA inhibitor groups was obviously greater than in the blank control group. Hepatocyte growth factor concentration in each group was gradualy decreased with time, hepatocyte growth factor activator concentration in each group was gradualy increased with time, and the changes were most obvious in the c-Met inhibitor group. Apoptosis rate of hepatic stelate cels in each group was gradualy increased with time, and highest apoptosis rate appeared in the RhoA inhibitor group, and lowest apoptosis rate in the c-Met inhibitor group. These findings suggest that bone marrow mesenchymal stem cels participate in and promote the apoptosis of hepatic stelate cels by activating hepatocyte growth factors and downregulating Rho activity.

3.The treatment of upper respiratory infection induced coughing with atomizated joint of compound liquorice oral solution and ipratropium bromide

Obiective To observe the clinical effect of treating upper respiratory infection induced coughing with atomizated ioint of compound liquorice oral solution and ipratropium bromide.Methods 69 cases of respiratory tract infection induced coughing were randomly recruited into A,B,and C group.Group A was treated with atomizated joint of compound liquorice oral solution and ipratropium bromide;Group C was treated with azithromycin and levofloxaein;Group B was treated with the combined therapy of Group A and Group C.Clinical effects and side effects were observed after the treatment.Results Symptom scores of cough in group A and group B were lower than those in group C.The difference was statistically significant(P<0.05).Dry mouth and throat complaints disappeared in group A and group A and group B after the treatment.Conclusion Tomizated ioint of compound liquorice oral solution and ipratropium bromide had good therapeutic effects in treating cough induced by upper respiratory infection.
4.Septum strut implantation for rhinoplasty simultaneous with cleft alveolar bone grafting in cleft lip and palate patients

Objective To pursue a more efficient and effective treatment for cleft lip and palate deformities. Methods Patients with unilateral complete cleft lip and palate at their age of 9 years after were chosen for simultaneous cleft alveolar repair and nasal deformity correction. Muco-periosteal pocket and iliacgranular bone was prepared, and bone grafting was performed conventionally. At the same time of iliac cancellous bone harvesting, a cortical plate was taken and sculpted into a strut of 18 mm in length, 6 mm in width, and 1.5 mm in thickness. A flying bird incision was made at the alar ram and across columella in a V-shape. Then the alar cartilage was detached from the overlying skin, a socket was made at the site of anterior nasal spine. The strut was inserted into the socket between the two medial crura of the alar cartilage. The medial crura was lift 3 mm above the superior edge of the strut, and mattress suture technique was used to secure the bilateral medial crura to the strut graft. Results 24 patients were treated by this technique. All the patients healed uneventfully. Depressed alar base, tilted columella and lower nasal tip were corrected satisfactorily. Conclusion There is no interference in simultaneous cleft alveolar bone grafting and rhinoplasty. Septum strut can provide favorable support for tilted nasal structure and satisfactorily correct nasal deformities. Simultaneous with alveolar grafting, it is much easier in harvesting, and the time of anesthesia and operation is also decreased.[
5.Analysis of IL-8 secretion in THP-1 cells stimulated by lipopolysaccharide and capsular polyssacharide extracted from Porphyromonas gingivalis

Objective·To isolate lipopolysaccharide (LPS) and capsular polysaccharide (CPS) from Porphyromonas gingivalis (Pg) and analyze their ability to induce THP-1 cells to secrete interleukin 8 (IL-8) in vitro. Methods·LPS and CPS were isolated from the polysaccharides by gel filtration chromatography, which had already been extracted from Pg ATCC33277 and SJD4. The isolated LPS and CPS from different strains were used to stimulate THP-1 cells in different doses and for different periods. The level of IL-8 in the cell culture suspension was measured by ELISA. Results·LPS and CPS were successfully separated from the polysaccharides in Pg ATCC33277 and SJD4. The relative molecular masses of LPS and CPS were 20000-50000 and 150000-500000, respectively. LPS and CPS could induce THP-1 cells to produce IL-8 in vitro in a time- and dose-dependent manner. The effect of LPS from ATCC33277 on secretion of IL-8 was greater than that of CPS with the dose of 0.1 μg/mL (P<0.05). However, the difference could only be found at 24 h in SJD4, and the effect of CPS was greater (P<0.05). Conclusion·As a component of polysaccharides extracted from Pg, CPS can induce THP-1 cells to secrete IL-8 in vitro in a time- and dose-dependent manner.

8.Study on diarylheptanoids from green peel of Juglans sigillata.

The chemical constituents from green peel of Juglans sigillata were isolated by column chomatographies over silica gel, Sephadex LH-20, and MCI. Four diarylheptanoids were isolated and their structures were characterized as dihydropterocarine(1), 3',4″-epoxy-1-(4'-hydroxy-phenyl)-7-(3″-methoxyl-phenyl)-heptan-3α-ol(2), pterocarine(3), and 1-(4'-hydroxy-phenyl)-7-(3″-methoxy-4″-hydroxyphenyl)-heptan-3α-ol(4). Compound 1 is a new compound, named as dihydropterocarine. Compounds 2-4 were isolated from the plant of J. sigillata for the first time.

Objective To investigate the clinical effect of highly aspherical lenslets(HAL)in myopia control in adolescents.Methods Totally 101 cases(202 eyes)of adolescent myopia patients treated in ophthalmology outpatient department of our hospital from January 2021 to February 2022 were selected as the study objects.Of these,43 cases(86 eyes)wore HAL lenses as an observation group and 58 cases(116 eyes)wore single-vision spectacle lenses(SVL)as a control group.After 12 months of follow-up,the equivalent spherical lens degree and axial length were recorded in 6 months and 12 months respectively.Results The change of equivalent spherical lens in the HAL group was(-0.18±0.22)D and that in the SVL group was(-0.47±0.21)D,and the change in the HAL group was less than that in the SVL group,the difference was significant(P<0.001).The equivalent spherical lens degree in the HAL group was(-0.33±0.32)D and the HAL group was(-0.82±0.20)D at the 12th month,and the change in the HAL group was less than that in the SVL group,the difference was significant(P<0.001).The axial length of the eye in the HAL group was(0.09±0.09)mm and the SVL group was(0.20±0.07)mm in June,and the change in the HAL group was less than that in the SVL group,and the difference was significant(P<0.001).The axial length of the eye in the HAL group was(0.14±0.13)mm and that in the SVL group was(0.37±0.08)mm,and the change in the HAL group was less than that in the SVL group,and the difference was significant(P<0.001).Conclusion Wearing HAL lens for 12 months can effectively slow down the growth of myopia degree and axial length in adolescent myopia patients.
