1.Imbalance of T cell subpopulations in immunologic pathogenesis of throm boangiitis obliterans

The T cell subpopulations of peripheral blood from 60 TAO patients and 40 normal subjects were detected with 3 kinds of monoclonal antibodies using indirect immunofluoresence technique. Meanwhile, the antiarterial antibodies (AAA) and circulating immune complex (CIC) from the sera of above patients and normal subjects were also determined. The results of the experiment suggests: (1) The functional disorders of immunoreguiating T cell (T4~+, T8~+),especially the severe decrease of T8~+ cell percentage play an important role in the pathogenesis of TAO;(2) The imbalance of T cell subpopulations have close relation to the production of antiarterial antibodies.
3.Effects of arteriovenous fistula on structure and function of heart in patients with hemodialysis

Objective To investigate the effects of artervenous fistula on structure and function of heart in patients with hemodialysis in the Second Clinical Hospital of Chinese Medical University from 2004 to 2005.Methods AVF、LAD、RVD、LVD、LVEDV、SV and EF were examined and analysed before and 8 weeks after AVF in 55 patients with hemodialysis.Results LAD、RVD、LVD、LVEDV after AVF increased significantly and EF reduced evidently after AVF.Conclusion AVF can cause extension of LAD、 RVD、 LVD and decrease of EF.

The development of body donation remains relatively slow in China.One of its main influencing factors restricted to the development of donation is the anatomy teachers' quality.Lots of volunteers to donate and their families are not satisfied with it,which eventually leads to the failure of the donation.Therefore,enhancing anatomy teachers' quality has become one of the key steps in the donated work.It needs to improve the idea awareness of teachers,to standardize the behavior of teachers,to enhance the role models of teachers,and to carry out a regular humanities education and training to truly realize "people-oriented" goals.
6.Pattern of motoneuron death following transection of the sciatic nerve in adult rats

Objective To investigate the pattern of motoneuron death induced by transection of the sciatic nerve in adult rats. Methods Sciatic nerve transected model was selected. The rats were sacrificed at 48 h, 7, 15, and 30 d after operation, respectively. The L 4-L 6 segments of the spinal cord were harvested after transcardiac perfusion with 4% paraformaldehyde, and were stained by TUNEL method and examined electron microscopically. Results A typical pattern of morphological changes of motoneuron apoptosis in the anterior horn of spinal cord was observed by both TUNEL staining and electron microscopy. Conclusion Motoneuron apoptosis in the spinal cord anterior horn induced by peripheral nerve transection demonstrates that neuron apoptosis may be the main pathological course of rat spinal cord neuron death following peripheral nerve injury in adult rats.
7.The practice of CAD custom femoral prosthesis and the computer simulated biomechanical experiment.

The present study was carried out to verify the accuracy and feasibility of the CAD program and to demonstrate whether the custom prosthesis is superior to the commercial ones. We modified the previously developed CAD (computer aided design) program, and buildt up a three-dimensional (3D) model of the custom femoral prosthesis by using the modified program and Canny algorithm; therefore, a basic foundation for the CAM (computer aided manufacture) was laid. Through the computer simulated biomechanical experiment on the prostheses, we came to the conclusion that the custom designed prosthesis has a better biomechanical characteristic. The future for this kind of prosthesis in THA(total hip arthroplasty) is very promising and it can be successfully applied in clinical practice through the CAM.

8.Comparative study of the image observation of earlypostburn brain edema in severely burned dogs.

OBJECTIVETo explore the diagnostic value of MRI and SPECT in early postburn brain edema in severely burned dogs.
METHODSTwenty-six mongrel dogs were randomized into control (n = 6) and burned groups in which every 5 dogs were allotted to each of following time points: 6, 12, 18 and 24 postburn hours (PBHs). The dogs in burn groups were inflicted with 50% TBSA of III degree skin burn and were infused with 5% glucose solution after 6 PBHs, so that severe early postburn brain edema was produced. MRI and SPECT were employed to observe dynamically the brain of dogs in all groups. The results were collected and compared with one another.
RESULTSThe results indicated that with MRI brain morphological change of early brain edema could be shown as early as within 12 PBH and diffuse brain edema became more obvious with elapse of time. The changes might be difficult to be found by MRI when T(1)WISIR decreased below 10%. T(2)WI SIR increased by 8.29% at 24 PBH with blurred demarcation between the brain gray and white matters. There was diffused and progressive nuclide ((99)TCm-ECD) concentration in the brain tissue as shown by SPECT at 6 PBH. The radio-nuclide taking ratio increased significantly after 12 PBH, especially at 24 PBH (P < 0.01) when compared with that before burn.
CONCLUSIONCombined application of MRI and SPECT could evidently increase sensitivity and specificity in the diagnosis of early postburn brain edema.

9.Antigen measurement and biomechanical characteristics of the inbred-line Banna mini-pig acellular bone matrix.

In this study, we prepared the acellular bone matrix of the inbred-line Banna mini-pig by using tissue engineering method and evaluated its possible application in bone tissue engineering. Histological analysis, xenoantigen expression and biomechanical measurement were performed on the matrix. HE staining and scanning electron microscopy showed the cellular components were almost removed. Immunohischemical result demonstrated that the xenoantigen, alpha-gal,was also eliminated. There was no statistically significant difference between the acellular bone matrix group and control group. The acellular bone matrix can provide appropriate space structure and strength for grafts. In conclusion, our data suggest that acellular bone matrix is a new kind of ideal bone scaffold material.

10.Preparation and application of monoclonal antibodies to recombinant human IFN alpha.

OBJECTIVETo prepare and apply monoclonal antibodies (McAb) against recombinant human interferon alpha (rHu IFN-alpha).
METHODSFive cell lines (2E9, 4G1, 2A7, 2C9, 4G10) secreting McAbs against rHu IFN-alpha were established by hybridoma technique.
RESULTSAll the cell lines secreted monoclonal antibodies stably. Functions of secreting antibodies of the five cell lines lasted for 6 months in BALB/c mice and 8 months in cell culture. The specificity of antibody was constant. The Ig subclasses of the McAbs were IgG1. Anti-IFN McAb affinity purification column was prepared by coupling the anti IFN-alpha McAb to Sepharose 4B. The combining rate reached was higher than 95%.
CONCLUSIONSThe highest purification efficiency was obtained by using 4G10 column.
