BACKGROUND:Non-heart beating donors have become the most promising source of donor in recent years. Tofurther improve donor preservation solution, on the basis of conventional storage solution hypertonic citrate adenine solution, artificial cordyceps was added to prepare artificial cordyceps compound solution. It is hoped to reduce donor injury and to elevate donor quality by antioxidation. OBJECTIVE: To investigate the preservation effect of artificial cordyceps compound solution on isolated renal from non-heart-beating rats. METHODS: Medulas of 39 Sprague-Dawley rats were injured. Withoutin vitro respiratory and circulatory supports, non-heart-beating models were established and randomly divided into three groups: artificial cordyceps compound group (n=15), hypertonic citrate adenine solution group (n=15) and physiological saline group (n=9). The kidneys of rats in different groups were stored in artificial cordyceps compound solution, hypertonic citrate adenine solution and physiological saline at 4℃. RESULTS AND CONCLUSION: Compared with the hypertonic citrate adenine solution group, after 6 and 12 hours of preservation, morphological injury was mild, apoptotic index, malondialdehyde, Bax and Caspase-3 expression levels decreased, superoxide dismutase activity and Bcl-2 expression increased; after 24 hours, the degree of injury was similar, malondialdehyde content decreased, and superoxide dismutase activity increased in the artificial cordyceps compound group. These findings indicated that the preservation effect of artificial cordyceps compound on antioxidation and inhibition of apoptosis was better than that of hypertonic citrate adenine solution in non-hearting beating rats.

OBJECTIVETo explore the effect of curcumin on TGF-β2 regulated peroxisome proliferater activated receptor y (PPAR-γ)/platelet derived growth factor β (PDGF-β) signaling pathway in lung fibroblasts of mice.

METHODSC57BL/6 mouse lung fibroblasts were in vitro cultured with TGF-β2, curcumin, or TGF-β2 plus curcumin. The cell proliferation was detected by cell growth counting in the blank control group, low, middle, and high dose curcumin groups (5, 25, 50 μmol/L), the TGF-β2 (10 ng/mL) group, TGF-β2 (10 ng/mL) plus curcumin (5, 25, 50 μmol/L) groups. mRNA expressions of PPAR-γ, platelet-derived growth factor receptor β (PDGFR-β), fibroblast growth factor R1 (FGFR1) were detected using reverse transcription PCR. Protein levels of PPAR-γ and collagen-1 were detected using Western blot and ELISA in the blank control group, the TGF-β2 group, the TGF-β2 (10 ng/mL) plus curcumin 50 μmol/L group.

RESULTSCompared with the blank control group, curcumin 50 μmol/L showed the most significant inhibition on cell proliferation at 48 h and 72 h. Compared with the TGF-β2 group, TGF-β2 (10 ng/mL) plus curcumin 50 mol/L also showed the most significant inhibition on cell proliferation at 48 h and 72 h. Compared with the blank control group, mRNA expressions of PPAR-γ and PDGF-β, as well as protein expression of PPAR-γ increased, the collagen-1 expression also increased in the TGF-β2 group (P < 0.05). Compared with the TGF-β2 group, mRNA expressions of PPAR-γ obviously increased in the TGF-β2 (10 ng/mL) plus curcumin 25 μmol/L group and the TGF-β2 (10 ng/mL) plus curcumin 50 μmol/L group, higher than that in the TGF-β2 (10 ng/mL) plus curcumin 5 [μmol/L group (P < 0.05). mRNA expressions of PPAR-γ was higher in the TGF-β2 (10 ng/mL) plus curcumin 50 μmol/L group than in the TGF-β2 (10 ng/mL) plus curcumin 25 μmol/L group (P < 0.05). mRNA expressions of PDGF-β was lower in TGF-β2 (10 ng/mL) plus curcumin groups than in the TGF-β2 group (P < 0.05). Besides, PDGF-β mRNA expressions were lower in the TGF-β2 (10 ng/mL) plus curcumin 50 μmol/L group than in the TGF-β2 (10 ng/mL) plus curcumin 5 μmol/L group and the TGF-β2 (10 ng/mL) plus curcumin 25 μmol/L group (P < 0.05). There was no statistical difference in FGFR1 mRNA expressions between the TGF-β2 group and 3 TGF-β2 plus curcumin groups (P > 0.05). Compared with the TGF-β2 group, PPAR-γ protein expressions increased and collagen-1 protein expressions decreased in the TGF-β2 (10 ng/mL) plus curcumin 50 μLmol/L group (P < 0.05, P < 0.01).

CONCLUSIONSCurcumin not only could inhibit TGF-β2 induced proliferation of lung fibroblasts, but also could inhibit the synthesis of collagens. These might be associated with up-regulating PPAR-γ expressions and down-regulating PDGF-β expressions. Therefore, curcumin might inhibit the occurrence and developing of lung fibrosis through blocking PPAR-γ/PDGF-β signaling pathway.

OBJECTIVETo evaluate the expression and clinical significance of macrophage migration inhibitory factor (MIF) in patients with bladder urothelial cell carcinoma.

METHODSImmunohistochemical staining for MIF was performed on tissue sections of 110 patients with bladder urothelial cell carcinoma and 10 normal controls, and the correlations between MIF and clinicopathological characteristics and prognosis were also analyzed.

RESULTSNormal bladder urothelium from control subjects showed negative or weak staining of MIF. Of the cancer specimens, 72/110 (65.5%) showed a moderate to strong staining of MIF. The expression of MIF protein was found predominantly in the tumor cell cytoplasm and inversely correlated with tumor stage. 27 cases also showed a positive intranuclear staining of MIF, which was inversely correlated with tumor grade, stage and tumor size. Kaplan-Meier analysis showed that the expression of MIF in the cell nuclei was associated with disease-free survival for the cancer patients, but multivariate analysis showed that MIF was not an independent prognostic factors.

CONCLUSIONSThe expression of MIF in non-muscle invasive bladder cancer tissues was more frequently than that in muscle-invasive disease, the positive staining of MIF in cell nuclei might be a favorable biomarker for patients with bladder urothelial cell carcinoma.

OBJECTIVETo study the clinical and pathological features, diagnosis, therapy and prognosis of primary small intestine malignant tumor.

METHODSA retrospective analysis was performed on the clinical data from the 120 cases of primary small intestine malignant tumor.

RESULTSAbdominal pain, gastrointestinal bleeding, anemia, abdominal mass and jaundice were the main clinical features. The pathology was confirmed by abdominal X-ray, gastrointestinal barium, CT, MRI, endoscopy and surgical exploration. Most tumors originated in the duodenum (54.1%), and adenocarcinoma (55.8%) was the main pathological type. The median survival time of the patients was 19.2 months and the 1-year survival rate was 55.4%. Chemotherapy did not seem to significantly improve the 1-year survival rate of the patients (P=0.842).

CONCLUSIONPrimary small intestine malignant tumors lack specific clinical manifestations and surgical resection should be performed as early as possible.

Objective To evaluate the clinical significance of serum Klotho protein levels in the early diagnosis and prognosis of acute kidney injury (AKI) among adult patients in the intensive care units (ICU).Methods The study was prospective and observational.Blood samples and clinical data of AKI patients admitted to the ICU of the First Affiliated Hospital of Xinjiang Medical University between July 1 and August 31,2016 were collected.ELISA was used for the detection of Klotho and NGAL.Receiver operating characteristic curve (ROC) and the area under the curve (AUC) were used to compare the predictive performance among Klotho,NGAL and serum creatinine,evaluating the sensitivity and specificity of Klotho on the diagnosis of AKI.The correlation between Klotho and prognosis of AKI was investigated by comparing serum Klotho levels and early AKI predictors.Results The patients were divided into AKI group of 52 cases and non-AKI group of 98 cases.The baseline serum Klotho level in AKI group was significantly lower than that in non-AKI group (P ＜ 0.001).The AUC of Klotho predicting for AKI was 0.945(95％ CI:0.892-0.997) and the best cut off value was 1.76 μg/L(sensitivity 92％,specificity 94％).The predictive ability of Klotho was significantly higher than serum creatinine (Scr),and the sensitivity is higher than NGAL (sensitivity 87％,specificity 96％).Serum Klotho combined with Scr predicted better AKI (AUC=0.958,95％ CI:0.915-1.000,sensitivity 96％,sensitivity 92％).The level of Klotho in patients with AKI was significantly different between the renal function recovery group and non-recovery group (P=0.047),while there was no significant difference between the two groups in the level of NGAL and Scr (P ＞ 0.05).There was no significant correlation between the Klotho level at diagnosis of AKI and peak Scr,peak eGFR,Scr at discharge and eGFR at discharge (r=0.026,P=0.853;r=-0.127,P=0.368;r=0.243,P=0.082;r=-0.187,P=0.184).Conclusion Serum Klotho may be a potential biomarker for early diagnosis of AKI,but the association between serum klotho and the prognosis of AKI requires further study.

Abstract?AlM: To observe the effects of Shuangdanmingmu capsule on VEGF expression and retinal vascular morphology in rats with diabetic retinopathy ( DR) .?METHODS: DR rats were fed with Shuangdanmingmu capsule. By comparing with the normal group, the model control group, and positive control group, the effect of Shuangdanmingmu capsule on retinal tissue of DR rats was observed under electron microscopy. After HE staining, retinal structure was observed under the light microscope. lmmunohitochemical staining was used to detect the VEGF expression in retina.?RESULTS:Two months after treatment, the layers tissue of retina presented mild edema, capillary pericytes performed edema, mitochondria showed mild swelling and less clear structure, some endothelial cells showed slight proliferation in Shuangdanmingmu group. Compared with the normal group, the expression level of VEGF in retina increased in the other groups, especially in model control group. A significant differential in expression of VEGF was found between Shuangdanmingmu group, positive control group and model control group (P<0. 01).? CONCLUSlON: Shuangdanmingmu capsule can effectively improve the retinal microvascular, reduce edema and necrosis of each layer of retina, improve the ultrastructure of retina's tissue and inhibit VEGF expression in DR rats.

BACKGROUNDThe main risk factor for chronic obstructive pulmonary disease (COPD) is cigarette smoking. However, only 10% - 20% of chronic heavy smokers develop systematic COPD. We hypothesized that the inheritance of gene polymorphisms could influence the development of COPD, which was investigated by studying two single nucleotide polymorphisms (SNP) in exon 1 of the transforming growth factor-beta1 (TGF-beta1) gene.

METHODSWe enrolled 219 patients with COPD as the research group and 148 healthy people as the control group, all of whom were Chinese Han people. The polymorphisms of the TGF-beta1 gene, 869T/C and 915G/C, were analyzed using the method of amplification refractory mutation system-polymerase chain reaction (ARMS-PCR).

RESULTSThe occurrence of the TGF-beta1 gene 869T/C polymorphism in patients with COPD was significantly different from the control group (P < 0.05), in which the relative risk of this disease increased in cases who had the C allele (OR: 1.131, 95%CI: 1.101 - 1.539). There was no increased frequency of TGF-beta1 915G/C gene in COPD patients compared with control subjects (P > 0.05).

CONCLUSIONSThe polymorphism 869T/C in TGF-beta1 gene has a significant association with disease occurrence in COPD patients and the C allele might be a risk factor. The homozygous wild-type CC of 869T/C on TGFbeta1 could be a predisposing factor in COPD and those who carry the C allele might have particularly susceptibility to developing COPD.
