1.Inhibitory effects of sulforaphane on hypoxia-inducible factor-1α in human prostate cancer cells

Objective To investigate the effects of sulforaphane (SFN) on the proliferation of prostate cancer DU145 cells and hypoxia-inducible factor-1α(HIF-lα) expression,and to explore the possibility of SFN in the prevention of prostate cancers.Methods The DU145 cells were treated with different doses and different time of SFN.The cell proliferation viability were measured by 3-(4,5)-dimethylthiahiazo (-zy1)-3,3,5-di-phenytetrazo`liumromide (MTT) proliferation and cytotoxicity assay kit.The hypoxia tumor microenvironment was stimulated by pretreating cells with 1% O2,HIF-lα expression and vascular endothelial growth factor (VEGF) activitity were detected by Western-blot and ELISA,respectively.Results SFN had an inhibitory effect on the proliferation of human prostate cancer DU145 cells in a dose-and time-dependent manner.The cell proliferation inhibition rate exceeded 50% in more than 20 μ mol/L SFN-treated groups after 48 hours.SFN could inhibit the expression of HIF-1α induced by 1% O2 and VEGF activity.10 μmol/L SFN showed inhibitory effect on the expression of HIF-1α after 24 hours.20 μ mol/L SFN showed a significant inhibitory effect on the expression of HIF-1α after 12 hours.And peroxidase had synergistic inhibition on HIF-1α.Conclusions SFN can inhibit the proliferation of DU145 prostate cancer cells and HIF-lα expression.SFN can be used as a potential chemotherapeutic agent against human prostate cancer.
2.BML-111, the analogue of lipoxin, inhibits Hela cell proliferation

Objective To investigate the effect of BML-111 (the analogue of lipoxin) on uterine Hela cell (cervix cancer cell line) proliferation and the underlying mechanism. Methods Hela cells were stimulated by 50, 100, 200 and 400 μg/L BML-111, respectively, and cell viability was determined by MTT assay. Hela cells were divided into three groups:the control group (no treatment), the BML-111(200μg/L) group and the BML-111(200μg/L)plus Boc-2 (10μmol/L)group. Expression and location of P53 protein were detected by immunofluorescence. Expressions of NF-κB p65,P53 and CyclinD1 protein were detected by Western blotting. Results BML-111 (100, 200 and 400 μg/L) could effectively inhibit Hela cell viability compared with the control group (P < 0.05). P53 expression was shown decreased in both the nucleus and the cytoplasm without any change of P53 location , however, Boc-2 could reverse this effect. BML-111 could effectively inhibit P53 and CyclinD1 expression via NF-κB pathway and the effects could also be inhibited by Boc-2. Conclusions BML-111 can effectively inhibit Hela cell proliferation via FPR2 and NF-κB pathway.
3.Protection effects of Xuezhikang on heart in hypertension patients.

Objective To investigate the effect of xuezhikang on heart in patients with hypertension. Meth-ods 60 systemic hypertension (HT) patients with normal cholesterol were randomized into placebo group and Xu-ezhikang group (1200 mg/d) for 72 weeks. Extended-released nifedipine were administrated by HT patients. Plasma was obtained at baseline, after 24 weeks and 72 weeks of Xuezhikang therapy. Em/Am ratio at atrioventricular ring , E/A ratio at mitral orifice, IVSTd, LVIDd, LVPWTd were measured by echocardiography. Type Ⅰ collagen car-boxypropeptide (PIP) was determined. Results After 72 weeks treatment, the differences of LVWT, IVSTd, LVP-WTd and LVIDd between Xuezhikang group and the control group were not significant; E/A,Em/Am ratios at atrio-ventricoiar ring were significantly increased except that Em/Am at right atrioventricular ring with right ventricular lateral wall(P <0.01 ,P <0.05). After 24 weeks treatment the levels of plasma PIP were significantly decreased in Xuezhikang group compared with the placebo group (P < 0.01, P < 0.05). Conclusion Xuezhikang exerts cooper-ativity in improving those parameters related to myocardial fibrosis so to protect heart function of hypertension pa-tients.
4.Protective effect of N-acetyl-L-cysteine on the cytotoxicity induced by iohexol in HK-2 cells

Objective To explore the protective effect and mechanism of antioxidant N-acetyl-L-cysteine (NAC)on the cytotoxicity induced by iohexol in HK-2 cells. Methods The incubated HK-2 cells were divided into four groups:control group,iohexol group,NAC group,and NAC+iohexol group(pre-incubated with NAC and then co-incubated with iohexol).The cell viability was tested by CCK-8 assay;cell apoptosis was determined by Hoechst 33342 fluorescence staining and flow cytometry with Annexin V-FITC/PI double staining.Intracelluar ROS waft detected by flow cytometry with DCFH-DA fluorescence staining.The signaling transduction pathways were investigated by Western blotting and immunofluorescence staining. Results Iohexol decreased cell viability,and increased apoptosis in a dose-and time-dependent manner.In iohexol(100 gl/L,6 h)group,ROS was increased by 1.30-fold of control(P<0.05).In NAC(5,10,15 mmol/L)+iohexol groups,the cell viability was increased by 104%,118%,130%respectively,and iohexol group was 63% (P<0.05, respectively); apoptosis rate was decreased by 13.51%, 13.46%, 12.23% respectively, and iohexol group was 24.41% (P<0.05, respectively); ROS was decreased by 1.05-fold, 0.93-fold, 0.86-fold respectively, and iohexol group was 1.3-fold (P<0.05, respectively).Iohexol induced the increase of p53 phosphorylatian and activity, then up-regulation of Bax and down-regulation of Bcl-2 protein expression. Iohexol induced the release of cytochrome C from mitochondria to cytoplasm, all of which caused final activation of caspase-3. The expression levels of p53, Bax and caspase-3 were decreased, while Bcl-2 protein expression level was increased by NAC. Conclusions Iohexol induces the increase of apeptosis rate and ROS generation in HK-2 cells. NAC attenuates this iohexol-induced cytotoxicity by decreasing intracelluar ROS, which is mairdy through the intrinsic pathway.
5.Prenatal sonographic diagnosis of fetal intracranial hemorrhage

Objective To investigate the diagnostic value of prenatal ultrasonography in the fetal intracranial hemorrhage.MethodsIn a retrospective analysis,the ultrasonographic findings of five fetuses with intracranial hemorrhage diagnosed in our hospital were reviewed and compared with other imagemodalities.ResultsIn the five fetuses with intracranial hemorrhage,the ultrasonographic features mainly includeddilateduni-orbilateralventriclesandintraventricularechogenicfociorperiventricular echodensities.The diagnosis of all cases were confirmed by prenatal magnetic resonance.Four of these cases chose termination of pregnancy,and the other fetus had a normal neurological follow-up after birth.Conclusions Fetal intracranial hemorrhage can be diagnosed accurately by prenatal ultrasonography,especially in the second and third trimester.It is rarely associated with other anomalies.Prenatal sonographic examination may detect the lesion and help to evaluate the prognosis.
6.Effects of phosphatidylinositol-3 kinase/serine threonine kinase pathway on expression of beta-site amyloid precursor protein cleaving enzyme-1 in the hippocampus neurons

Objective To investigate the effect of phosphatidylinesitol-3 kinase/serine threonine kinase (PI3K/Akt) signaling pathway on expression of beta-site amyloid precursor protein cleaving enzyme-1 (BACE1) in the hippocampus neurons of rat brain. Methods Forty SD rats were randomly divided into 4 groups: blank control group, sham-operated group, insulin group and wortmannin group. Insulin or the specific inhibitor of PI3K, wortmannin was injected into hippocampus neurons to activate or inhibit the signaling pathway in insulin group or wortmannin group, respectively. Immunoprecipitation and Western blot were used to analyze the proteins levels of PI3K/Akt and BACE1. Results In insulin treatment group,among the proteins downstream of signaling pathway, expression of Akt increased (0. 952±0.060 vs 0.835±0.029,t=4.9150, P=0.0001), phospho-Akt set473 increased (0.800±0.075 vs 0.657± 0.025,t=4.5598, P=0.0002), phospho-GSK-3α decreased (0.604±0.062 vs 0.726±0.041, t= 3.5871, P=0.0018 ), and the expression of mature BACE1 and β-CTF significantly decreased. In wortmannin group, the expression of Akt and phospho-Akt ser473 were inhibited; phospho-GSK-3α increased ; mature BACEI (1.004±0.096) and β-CTF (1.031±0.048) increased (t=11.5980, P= 0.0000 and t =4.2194, P =0.0004, respectively). Conclusions PI3K/Akt signaling pathway might effect the expression of BACE1, in which impaired signaling pathway may cause the amyloid precursor protein to be easily processed by BACE1, and thus involves the pathology of Alzheimer' s disease.
7.Comparative study of gross tumor volume shown on MRI and FDG PET/CT of nasopharyngeal carcinoma

Objective To compare gross tumor volume (GTV) of nasopharyngeal carcinoma (NPC) according to MRI and FDG PET/CT and to investigated four fixed threshold methods to delineate the GTV using FDG PET/CT.Methods Fifty patients with primary biopsy-proven NPC were prospectively were enrolled into the study.FDG PET/CT scans and MRI were carried out within one week prior to pretreatment,respectively.The GTV was named GTV-MRI (GTV were delineated according to MRI),GTV-PETvis,GTV-PET30,GTV-PET40,GTV-PET50 (GTV was delineated according to the PET-based GTVs obtained by visual interpretationor,by percentage of the SUVmax (30%,40%,50%) thresholds,respectively).The differences were compared among the GTV-MRI,GTV-PETvis,GTV-PET30,GTV-PET40 and GTV-PET50 in different by Wilcoxon test.Results Of 50 patients,the median of volume descending order were: GTV-MRI 27.8 cm3,GTV-PETvis 22.2 cm3,GTV-PET30 22.7 cm3,GTV-PET40 14.4 cm3 and GTV-PET50 9.0 cm3.However,there was no significant difference between GTV-PETvis and GTV-PET30 (Z=-0.05,P=0.958),as well as GTV-MRI and GTV-PETvis or GTV-PET30 in 25 patients who were T1-2 stage (Z =-0.93,-0.93,P=0.353,O.353),the other GTVs were all different in 50 patients' (Z=-5.74-2.09,P =0.000-0.037).Conclusions All the GTVs delineated by the different methods of using FDG PET/CT were less than the GTV delineated by MRI.The potential advantages with the GTV-PETvis or GTV-PET30 delineated by FDG PET/CT are reduction of biological metabolic tumor volume in GTV delineation and reduction of the size of the GTV in NPC patients.
8.Clinical observation of gefitinib in treatment of female patients with adenocarcinoma of lung WU

Objective To evaluate the efficacy and adverse effects of gefitinib in the treatment of fe- male patients with advanced adenocarcinoma of lung who had failed to previous chemotherapy.Methods These patients received 250mg of gefitinib orally,once daily until disease progression or development of intol- erable toxic reaction.They were evaluated one month after treatment and every other month thereafter.Results Among the 27 evaluable patients,there were 1 CR(3.7%),11 PR(40.8%),10 SD(37.0%)and 5 PD(18.5%). The overall response rate was 44.5%(95% CI 29%~68%);and 22 patients(81.5%)gained profit(CR+PR+ SD)from the clinical therapy(95% CI 62%~94%);the mean TTP was 7.2 months.Symptomatic improvement rate was 80.0%.The main adverse effects were mild rash and diarrhea.Conclusion gefitinib has significant efficacy in the treatment of female patients with advanced tung cancer who had failed to previous chemother- apy.Adverse effects are mild.gefitinib is a suitable therapy for these patients.
9.Comparison of swing shoulder and internal fixation for the treatment of proximal humeral fractures in elderly.

OBJECTIVETo compare clinical outcomes of swing shoulder and internal fixation in treating proximal humeral fractures.
METHODSFrom June 2007 to June 2012, totally 89 elderly patients with humeral proximal fractures were treated by swing of shoulder or internal fixation, and 81 patients were followed up. In swing shoulder group, there were 38 patients including 13 males and 25 females aged from 62 to 84 with an average of (67.11±6.18) years old; 27 cases were 2-part fractures and 11 cases were 3-part fractures according to Neer classfication. In internal fixation group, there were 43 patients including 16 males and 27 females aged from 60 to 80 with an average of (66.47±5.48) years old; and 29 cases were 2-part fractures and 14 cases were 3-part fractures according to Neer classfication. VAS score and complications were compared between two groups after treatment, and Constant-Murley functional scoring was used to evaluate shoulder function of patients.
RESULTSEighty-one patients were followed up from 13 to 26 months with an average of 18.3 months. There was no significant difference in preoperative VAS score between two groups. After treatment, VAS score in swing shoulder group was (3.11±0.95), and (3.88±1.14) in internal fixation group, and had significant difference between two groups (t=-3.313,P<0.05). There was no significant difference in Constant-Murley scores between swing shoulder group (79.53±3.73) and internal fixation group (77.98±4.11) (t=1.768,P>0.05). Postoperative complications in swing shoulder group was 18.4%(7/38), 39.5%(17/43) in internal fixation group, and had significant differences between two groups (χ2=4.313,P<0.05).
CONCLUSIONSwing shoulder for the treatment of proximal humeral fractures in elderly has advantages of low cost, less complications and good recovery of joint function; while internal fixation has a good therapeutic effect but increased complications.

10.Observation on ear point taping and pressing therapy for treatment of primary dysmenorrhea.

OBJECTIVETo observe therapeutic effect and prognosis of ear point taping and pressing therapy for primary dysmenorrhea.
METHODSOne hundred and fourteen cases were randomly divided into an ear taping and pressing group of 60 cases and a western medication group of 54 cases. The ear taping and pressing group were treated by ear point taping and pressing at Shenmen, Zigong (uterus), Neifenmi (endorine), Pizhixia (subcortex), Jiaogan (sympathesis) and Shen (kidney), which were taped and pressed by a adhesive plaster with Vaccaria seeds. The western medication group were treated with oral administration of Indomeixin enteric tablets. The therapeutic effects were assessed after treatment of 3 menstrual cycles.
RESULTSThe total effective rate was 91.7% in the ear taping and pressing group and 77.8% in the western medication group with a significant difference between the two group (P < 0.05); and the long-term therapeutic effect in the ear taping and pressing group was better than that in the western medication group.
CONCLUSIONEar point taping and pressing therapy has significant therapeutic effect on primary dysmenorrhea.
