1.Method of Isolating Deiters Cell from Guinea Pig Cochlea and Criterion of Cell Judgement

Objective To probe the method of dissociating isolated Deiters cells of cochlear and its judging criterion for living Deiters cells.Methods Deiters cells were dissociated from guinea pig cochlea separately by collagenase,papain,trysin digestion followed by trituation.After that ,the Deiters cells were observed and counted.Results Isolated Deiters cell looked like a comma, which was divided into cell body, stem and Deiters phalanx. About 20 to 32 isolated Deiters cells were obtained from every two cochleae.The membrane was smooth and the birefringence existed; the position of the nucleus was normal; full phalanx and no particles with Brownian movement.Conclusion Substantial sum of Deiters cell with sound vitality can be obtained by all three enzymes digestion followed trituation;The criterion used is reliable.

Colonic neoplasms is the third most commonly diagnosed cancer in men and second most commonlv diagnosed cancer in women worldwide.The morbidity of Colonic neoplasms is increasing year by year.Despite the fact that surgery is still the main treatment for patients with colon cancer,surgery alone hasn't improved the treatment effectiveness.The current management of colon cancer has come to multidisciplinary team (MDT) modality.MDT modality could offer the optimal treatment option and the internal communication.Under the pattern of MDT which integrates the surgery,chemotherapy,radiotherapy,interventional therapy,targeted therapy and immune therapy,there has been a big progress in the diagnosis and treatment of colon cancer.Patients with metastatic colon cancer should undergo a discussion by a multidisciplinary team before the initial treatment.
3.Cytotoxicity and antibacterial effect of silicone-based soft liner varnish containing Ag/TiO2: an in vitro study.

OBJECTIVEWe aim to evaluate the cytotoxicity and antibacterial effects of Silagum-Comfort (SLC) silicone-based soft liner varnish containing Ag/TiO2 in vitro.
METHODSSilicone-based liner circular specimens, with a diameter of 10 mm and a thickness of 2 mm, were created and divided into six groups randomly. Ag/TiO2 was incorporated into the SLC varnish based on the agent/material weight rate of 0, 0.5%, 1.0%, 1.5%, 2.0%, and 2.5%. The mixed varnish was used to polish the SLC specimens. Antibacterial specimens (ATSLC) were created. Methyl thiazolyl diphenyl-tetrazolium bromide assay method was used to evaluate the 3T3 cell cytotoxicity of ATSLC in vitro. Surface roughness (Ra) measurements of ATSLC were obtained. The antibacterial effect of ATSLC on Candida albicans was examined using the pelliclesticking method.
RESULTSThe cytotoxicity of ATSLC was graded from 0 to 2. The Ra of ATSLC increased when Ag/TiO2 agent concentration increased. However, the differences were not statistically significant compared with the 0 group. With increasing Ag/TiO2 concentration, the antibacterial agent concentration and the antibacterial rate of ATSLC also increased. When the Ag/TiO2 antibacterial agent concentration reached 2.0% and 2.5%, the antibacterial rate increased to 97.5% and 96.5, respectively.
CONCLUSIONVarnish containing Ag/TiO2 has excellent antibacterial effect and can significantly improve the antibacterial effect of silicone-based soft liner.

5.Therapeutic effects of ZK_(14), a novel nitric oxide donating biphenyldicarboxylate derivative, on hepatic fibrosis in rats

Aim: To study the therapeutic effects of ZK_(14), a novel nitric oxide donor, on hepatic fibrosis in rats. Methods: Rats were injected intraperitoneally with CCl_4 and induced to hepatic fibrosis, followed by intra-gasrtic administration with high dose of 20 mg/kg and low dose of 10 mg/kg of ZK_(14) for 4 weeks. The effects were evaluated in the content of nitric oxide of serum and nitric oxide synthases of liver, in serum levels of the hepatic functions indices and hepatic fibrotic index and in liver histopathology. Results: The biochemical indices and pathology grade were effectively improved in the group of ZK_(14) 20 mg/kg, and liver function was also effectively protected in this group. Conclusion: ZK_(14), a novel nitric oxide donating biphenyldicarboxylate derivative, showed preferable therapeutic effects on hepatic fibrosis of rats induced with CC14, and it may be of potential value in the treatment of hepatic fibrosis.
6.Clinical effect of small dose of glucocorticoid in the treatment of AECOPD patients with severe disease associated with low levels of cortisol

Objective To explore clinical effect of small dose of glucocorticoid in the treatment of AECOPD patients with severe disease associated with low levels of cortisol.Methods72 patients with AECOPD in our hospital were randomly divided into control group and treatment group.The control group was treated with conventional AECOPD method,the treatment group was treated on the base of the treatment group with a small dose of corticosteroids,CRP, IL-6, TNF-α, FEV1, PCT and other indicators, as well as patients with ventilation, shock and mortality were compared after the end of treatment.ResultsThe time of non mechanical ventilation, the time of non-mechanical ventilation, and the time of non-shock were increased, the shock and the mortality was lower in the treatment group,(P<0.05);The CRP level of patients in the treatment group was lower(P<0.05);IL-6, TNF-α, PCT levels of patients in the treatment group was lower (P<0.05);FEV1, FVC and FEV1/FVC level in the treatment group were higher(P<0.05).ConclusionSmall dose glucocorticoid can improve the pulmonary function of patients with AECOPD complicated by severe disease, and alleviate the disease, significantly reduce the levels of inflammatory factors, promote the improvement of the lung function.
7.Diagnosis and treatment of penile verrucous carcinoma

Objective To explore the diagnosis and treatment of penile verrucous carcinoma,a distinctive variant of squamous cell carcinoma. Methods The clinical data of 8 patients (mean age,46 years) with penile verrucous carcinoma were analyzed retrospectively.All the tumors showed exophytic,papillary lesions,ranging from 2 to 6 cm in the greatest dimension.The tumors were located at glans in 5 patients and invaded to shafts in the other 3.The diagnosis was established by biopsy of the lesion.Three patients with tumor invading to shafts and 1 patient with large tumor at glans underwent partial penectomy, while the other 4 with the tumors localized at glans were treated by local excision. Results Histopathologic examination of the specimens confirmed tumors-free surgical margins in all the cases.The tumor cells were mostly well-differentiated.Fourteen months postoperatively,local recurrence occurred in 1 patient with local excision and partial penectomy was performed; postoperative follow-up of 9 years showed no recurrence and metastasis. During a follow-up of 4 to 13 years,the other 7 patients were disease-free.The patients with local excision of the tumors reported satisfaction of their sexual lives. Conclusions Verrucous carcinoma of penis is characterized by slow growth and a locally aggressive nature.It seldom develops metastasis to regional lymph nodes or distant areas.The prognosis is favorable after appropriate treatment.
8.Obervations on TDP Therapy in Treatment of 110 Cases of Infantile Diarrhoea

110 cases of infantile diarrhoea were admitted to this institute in a period from Aug. 30 to Dec. 22 of 1982.The patients were randomly divided into two groups, the TDP group and the control group. The general condition and the age distribution of the patients of both groups were similar. The patients of the TDP group received only TDP radiation instead of antibiotics and those of the control group received antibiotics therapy but no TDP. Other treatments such as fluid replacement, dietary regulation, etc, were the same in two groups. Stool samples were sent for routine examination and bacterial culture and blood samples for the determinations of the electrolyte levels, CO2CP, and immunity function for all the patients right after admission as well as just before discharge. The cure rate and course of the disease were similar in two groups. However the pathogenic organisms could still be revealed in the stool of the patients of the TDP group after recovery. But the rate of lymphocyte transformation was significantly higher in the patients of TDP group.It is concluded that TDP radiation is a simple, safe and effective treatment for infantile diarrhoea but its therapeutic mechanism remains obscure.
9.Bacteriostatic effect of chitosan lactate with different relative molecular masses in vitro

Objective To observe the bacteriostatic effect of chitosan lactate with different relative molecular masses in vitro. Methods The MICs of the chitosans lactate with different relative molecular masses against the usual pathogens were detected,and the MBCS against two standard strains were also determined. Levofloxacin hydrochloride was used as a control drug. Results The MIC90 of chitosan lactate with 10,30,50kDa against 50 strains of gram-positive cocci and 50 strains of gram-negative bacilli were 0.5,1~4,0.5~4mg?mL-1 and 1~4,0.5~1 ,0.5~4mg?mL-1,respectively. The MIC90 of levofloxacin hydrochloride against the 50 strains of gram-positive cocci and 50 strains of gram-negative bacilli were 4~8 and 8~16?g?mL-1,respectively. ATCC 25923 strain of S.aureus and ATCC 25922 strain of E.coli could be killed by 10kDa chitosan lactate in the concentration of 4mg?mL-1. The MBCS of levofloxacin hydrochloride against ATCC25923 strain of S.aureus and ATCC 25922 strain of E.coli were 2 and 4?g?mL-1,respectively. Conclusion The 4 species of the experimental bacteria could be inhibited by the chitosan lactate with 10,30 and 50kDa Mr in vitro,but the inhibitory effect of the chitosan lactate with 10kDa Mr was stronger among them. ATCC 25923 strain of S.aureus and ATCC25923 strain of E.coli could be killed only by 10kDa chitosan lactate in vitro,not by others.
10.Effect of Jiuqiang Naoliqing on the TXA_2 and PGI_2 level in spontaneous hypertension rat plasma

ObjectiveTo investigate the effect of Jiuqiang Naoliqing(JNQ) on the TXA2 and PGI2 level in spontaneous hypertension rat (SHR) plasma.MethodsThe plasma was separated after the SHR and Wistar rats were treated with JNQ at the dose of 0.133g/kg,0.265g/kg,0.530g/kg and 1% carboxymethyl cellulose respectively for 5 weeks. The level of TXB2 and 6 keto PGF1α ,stable metabolin of TXA2 and PGI 2,in SHR plasma was tested by radioimmunoassay.ResultsThe level of TXB2 and the ratio of TXB2/6 keto PGF1α (T/P) in SHR plasma increased significantly(P<0.01),and there was no significant difference in the concentration of 6 keto PGF1α between Wistar rats and SHR plasma(P>0.05). JNQ could increase the generation of 6 keto PGF1α and decrease the level of TXB2 and T/P in SHR plasma after treated with different dosages for 5 weeks.ConclusionJNQ may improve the balance between TXA2 and PGI2 in SHR plasma.
