5.Characterization of the nover late T cell activation proteins, 519 and 520.

The late T cell activation gene, 519, is expressed in antigen specific, growth factor dependent T cell lines and clones but not in T or B cell tumors, other hematopoietic cells, tonsil, muscle, lung, or liver. Resting peripheral blood lymphocytes (PBLs) express little or no 519mRNA, but levels increase dramatically 5~7 days after activation with alloantigen or mitogen. Four alternatively spliced transcripts of the gene exist. 520 cDNA, the predominate gene transcript, encodes a protein of 144 amino acids and has a potentially cleavable hydrophobic leader of 15 amino acids at the amino terminus. The 519 transcript encodes a 129 amino acid product. An polyclonal antiserum was produced by immunizing a rabbit with a synthetic peptide corresponding to the second hydrophobic region common to both 519 and 520. 14.3 and 15.5kD protein bands were immunoprecipitated (IP) from radiolabled in-vitro translated products, 519 and 520 respectively. A protein of 14.3kD was detected by IP of in-vivo 35S cysteine and methionine radiolabled cytotoxic T lymphocytes (CTL) cells. Western blotting of lysates from a CTI line and a B cell tumor line revealed a distinct band at 14.3kD only in CTL lanes. Both CTL and peripheral blood lymphocytes expressed the greatest amount of 519/520 protein in cells 5~7 days after stimulation. Western blotting of differential centrifugation samples localized 519/520 protein to the granule layer. Comparison of 519/520 proteins with other known protin sequences identified homology with surfactant associated and sphyngolipid activating proteins. A homologous gene(NKC5) is present in natural killer cells (NK). The increase in the amount of the 519/520 protein product in CTL and PBL parallel increases in mRNA expression. 519/520 and related gene products are present in CTL, PBL and NK cells, have structural similarity with lipid associating proteins.

7.Alteration of Serum Insulin-like Growth Factor(IGF)-I and IGF-Binding Protein-2 in Healthy Population with Aging.

PURPOSE:Insulin-like growth factors, IGF- I and IGF-II, are proteins that promote cellular growth and differentiation of the various organs including the kidney. These peptides circulate in serum bound to specific carrier proteins, called IGF binding proteins(IGFBPs). The IGFs are produced in most organs but liver is believed to be the principal source of circulating IGF-I. We studied the correlation of serum IGF-I and IGFBP-2 pattern with aging. METHODS:Sera were collected from 320 healthy population who were grouped according to age. IGF-I was seperated from IGFBPs by Sephadex G-50 acid chromatography. We measured serum IGF-I and IGFBP-2 by using radioimmuno-assay (RIA) and immunoradiometric assay (IRMA) respectively. RESULTS:Serum IGF-I levels were quite low in early childhood, rising slowly and reaching a peak during puberty and a significant decline(P<00.01) during adulthood. The age-dependent pattern of serum IGFBP-2 levels shows a pattern opposite to that of IGF-I which are high at birth, decline by late puberty and increase again with aging. CONCLUSION: Our results demonstrate the alteration of serum IGF-I and IGFBP- 2 pattern with aging. These data suggests that when these tests are performed in the clinic, their interpretation should be based upon age specific criteria.

8.A Case of Rapidly Progressive Glomerulonephritis in Henoch-Schonlein Purpura.

A case of primary non-clear-cell adenocarcinoma of the vagina is reported occurring in a 65-year-old woman without exposure to diethylstilbestrol (DES) in utero. The adenocarcinoma did not appear to be associated with vaginal adenosis. It lacked clear cell component and interestingly composed of columnar epithelial cells of endocervical-type. Cytologically round to oval nuclei revealed one or more small nucleoli and fine granular chromatin pattern. Cytoplasm was plump, faintly basophilic and homogeneously stained. Histologically well differentiated columnar epithelial cells were arranged in trabecular pattern mainly, and also occasional glandular lumina and small solid sheets were found. Mitoses were hardly found.

9.Clinical Observation of Congenital Urinary Tract Anomalies.

We present the cytologic features of a case of solid and papillary neoplasm of the pancreas. Cytologically, the tumor was composed of a monotonous population of polygonal cells containing eccentrically located round nuclei with one or two distinct small nucleoli and a finely stippled chromatin pattern. The tumor cells were similar to those of the islet cell tumor and showed isolated loosely aggregated and solid sheets or large cell clumps. The large cell clumps revealed a branching papillary structure containing fibrovascular central core, which is characteristic histologic feature of solid and papillary neoplasm of the pancreas. This case was confirmed by tissue examination including histochemical, immunohistochemical and electron microscopical studies. Ultrastructurally, the tumor cells contained a few membrane-bound electron dense granules.

10.A Case of Neonatal Diabetes Mellitus: Transient or Permanent?.

A case of neonatal diabetes mellitus is described. The child presented with low birth weight but was normal in appearance. She was acidotic and ketonuria was observed. The HLA typing was DR1 and 3, and insulin autoantibodies were negative. Genetic analysis with polymorphic DNA markers for chromosome 6 indicated biparental inheritance. She required insulin therapy for the control of hyperglycemia, and insulin dependence continues after 8 months of age.
