1.The mechanism of repressive effects of transthyretitin on the growth of human retinal microvascular endothelial cells under high glucose and hypoxia environment

Objective To explore repressive effects of transthyretitin (TTR) on the growth of human retinal endothelial cells (hREC) under high glucose and hypoxia environment. Methods hRECs were divided into 8 groups, including normal glucose group (5.5 mmol/L glucose), hypoxia group, high glucose group (25.0 mmol/L glucose), high glucose and hypoxia group, normal glucose group+TTR, normal glucose and hypoxia group+TTR, high glucose group+TTR, high glucose and hypoxia group+TTR. Flow cytometry was used to analyze cellular apoptosis. The expression level of Akt, p-Akt, eNOS, Bcl-2 and Bax protein were measured by Western blot. Results Hypoxia could induce apoptosis as the apoptosis rate of normal and hypoxia group was higher than normal group (χ2=25.360, P<0.05), high glucose and hypoxia group was higher that high glucose group (χ2=17.400, P<0.05). The cell apoptosis rate of high glucose and hypoxia group+TTR were increased significantly as compared with high glucose and hypoxia group (χ2=9.900, P<0.05). There was no statistically significant difference on the cell apoptosis rate between normal group and high glucose group, normal group+TTR and normal group, high glucose group+TTR and high glucose group, normal and hypoxia group+TTR and normal and hypoxia group (P>0.05). Western blot showed that the expression of Akt did not change significantly in all eight groups(F=2.450, P>0.05). Compared to normal group, the expression of p-Akt, eNOS, Bcl-2 in normal and hypoxia group were decreased (t=9.406, 5.306, 4.819), and the expression of Bax (t=-4.503) was increased (P<0.05). Compared to high glucose group, same trend was found in high glucose and hypoxia group (t=8.877, 7.723, 6.500, -14.646; P<0.05). The expression of p-Akt in normal and hypoxia group+TTR was higher than normal and hypoxia group (t=-5.024, P<0.05) ,but there was no difference on the expression of eNOS, Bcl-2, Bax between these two groups (t=-2.235, -2.656, -0.272;P>0.05). Compared to high glucose and hypoxia group, the expression of p-Akt and Bcl-2 in high glucose and hypoxia group+TTR were decreased (t=4.355, 4.308; P<0.05), the expression of Bax was increased (t=-4.311, P<0.05), and there was no difference on the expression of eNOS between these two groups (t=-1.590, P>0.05). There was no statistically significant difference in the expression of p-Akt, eNOS, Bcl-2, Bax between high glucose group and normal group (t=-3.407, -4.228, -4.302, -2.076; P>0.05), normal group+TTR and normal group (t=-4.245, -4.298, -2.816, -1.326; P>0.05), high glucose group+TTR and high glucose group (t=4.016, -0.784, 0.707, -0.328; P>0.05). Conclusion Under high glucose and hypoxia, transthyretitin suppress the growth of hREC through Akt/Bcl-2/Bax, but not Akt/eNOS signaling pathway.
2.Effect of absorption combined with continuous venovenous hemofiltration on eliminating serum inflammatory medium and its nursing

Objective To discuss the effect of absorption (AP) combined with continuous venovenous hemofiltration (CVVH) on eliminating serum inflammatory medium for patients with systemic inflammatory response syndrome (SIRS) and its nursing essence.Methods Totals of 63 patients with SIRS were randomly divided into the treatment group (AP combined with CWH,n =31) and the control group (CWH,n =32).Data of serum inflammatory medium,such as TNF,CRP,IL-1,IL-6 and IL-10 were recorded and compared within and between two groups before and after the treatment.Results There was no statistically significant difference in TNF,CRP,IL-1,IL-6 and IL-10 between two groups before treatment (P > 0.05).After the treatment,levels of TNF,CRP,IL-1,IL-6 and IL-10 in 2,26 and 50 hours showed descending tendency in the treatment group,and the differences were statistically significant compared with before (P < 0.05).Levels of TNF,CRP,IL-1,IL-6 and IL-102,26 and 50 hours after the treatment in the control group also showed descending tendency,and the differences were statistically significant compared with before (P < 0.05) except data in 2 hours (P > 0.05).Levels of TNF,CRP,IL-1,IL-6 and IL-10 also had statistically significant differences between two groups at the same time (P < 0.05).For example,score of TNF was respectively (226.4±27.6),(165.3 ±24.5),(120.6 ± 19.2) ng/L 2,26 and 50 hours after treatment in the treatment group and (315.7 ± 38.3),(262.7 ± 29.4),(219.3 ± 25.6) ng/L in the control group,with statistically significant differences (t =-3.31,-7.19,-8.79,respectively; P < 0.05).Condusions AP combined with CVVH can effectively eliminate the serous inflammatory medium for SIRS patients,and close surveillance and nursing are key to smooth treatment and expectations achievement.
3.Establishment of HPLC Fingerprint of Pholidota chinensis and Its Cluster Analysis

OBJECTIVE: To establish HPLC fingerprint of Pholidota chinensis and cluster analysis the samples, and to provide reference for quality evaluation of P. chinensis. METHODS: HPLC fingerprints of 16 batches of P. chinensis from Guangxi, Guangdong and Fujian were established according to 2012 edition of Similarity Evaluation System for Chromatographic Fingerprint of TCM [chromatographic column was Agilent SB-aq column with mobile phase consisted of acetonitrile-0.05% phosphoric acid solution (gradient elution), at the flow rate of 1.0 mL/min. The detection wavelength of 220 nm, the column temperature was 30 ℃ and the sample volume was 5 μL].The cluster analysis was carried out by using SPSS 22.0 statistical software. RESULTS: Accoding to the established HPLC fingerprint, 11 common peaks were indentified, and the common peak 1 was identified as gastrodin; the similarity of 16 batch of samples was greater than 0.9. 16 batches of medicinal materials could be divided into 2 groups. CONCLUSIONS: The established HPLC fingerprint and cluster analysis can provide reference for the quality control of P. chinensis.
4.Analysis of prognostic factors and treatment of pulmonary metastasis from colorectal cancer.

OBJECTIVETo investigate effect of the treatments and prognostic factors of patients with pulmonary metastasis from colorectal cancer.
METHODSClinical data of 79 patients who suffered from lung metastatic diseases from colorectal cancer in 1990 - 2010 were retrospectively analyzed. The number of patients who had received lung operation was 22, and non-operated group contained 57 patients. Compared the prognosis of operated group and non-operated group and analyzed the prognostic factors.
RESULTSThe median survival time after the pulmonary resections was 34.5 months; the overall survival of 1-, 3- and 5-year survival rates were 90.9%, 45.4% and 4.5%, and the overall of 1-, 3-, and 5-year survival rate in non-operated group were 59.6%, 14.0% and 0. The surgery (RR = 4.805, 95% CI: 1.864 - 12.384, P = 0.001) and the number of metastasis (RR = 2.177, 95% CI: 1.431 - 3.314, P = 0.010) were the factors that could influence the patients prognosis.
CONCLUSIONThe surgery for pulmonary metastases from colorectal cancer is effective.
