1.Biocompatibility of polycaprolactone with bone marrow stromal cells

BACKGROUND: Bone defect repair is a difficulty in orthopedic field all the time, researches on the tissue engineered bone tissue have provided completely new thoughts and methods for bone defect repair, and it is an important link to detect the biocompatibility of the biomaterials.OBJECTIVE: To investigate the biocompatibility of PCL with bone marrow stromal cells (BMSCs).DESIGN: A controlled observation.SETTING: Department of Orthopaedics, the First Affiliated Hospital of Xinjiang Medical University.MATERIALS: The experiments were carried out in the orthopedic laboratory of the First Affiliated Hospital of Xinjiang Medical University. Healthy 4 to 8-week-old New Zealand rabbits of about 2 kg were used.METHODS: ① Bone marrow was extracted from bilateral femurs of the rabbits, then mixed into 10 mL RPMI1640 complete medium, and then entered the passage culture. ② The cells were inoculated and then divided into PCL group and control group, the cells were only inoculated in the control group, and the BMSCs were co-cultured with PCL in vitro in the PCL group. The morphological observed, cell proliferation, protein content and enzymological determinations were conducted.MAIN OUTCOME MEASURES: The growth and the adhesion of BMSCs on PCL biomaterials were mainly observed.RESULTS: In the control group, most of the BMSCs changed to the shape of fusiform or multi-angles. In the PCL group, the BMSCs could adhere and proliferate on PCL, and the growth and function were not affected, PCL also played a certain role in promoting the cell proliferation.CONCLUSION: PCL possess satisfactory biocompatibility, and it is possible to be used as the carrier of BMSCs in tissue engineering.
2.Non-autophagic degradation roles of autophagy receptors.

A growing body of evidence has indicated the important role of autophagy receptors in directing ubiquitinated or non-ubiquitinated cargos towards autophagy. Autophagy receptors bind to LC3 (microtubule-associated protein 1 light chain 3) on phagophore and autophagosome membranes, and recognize signals on cargoes in the delivery system of autophagy. However, the diverse domains in the receptor structures determine that their roles would never be limited to autophagy. Up to date, increasing numbers of the receptor proteins have been demonstrated to serve as a molecular link or switch participating in autophagic degradation, apoptosis or cell survival signals. Here, we highlight the non-autophagic roles of these receptor proteins to draw attention to this growing research topic.

3.Proteomic analysis of lymph nodes affected by esophageal squamous cell carcinoma by two-dimensional gel electrophoresis

Objective To analyze the differential expression of proteome between metastatic lymph nodes and normal lymph nodes in esophageal squamous cell carcinoma.Methods Two dimensional polyacrylamide gel electrophoresis(2-DE)technique and computer-assisted image analysis were used to separate the protein spots and analyze the differential expression of proteome in the metastatic lymph nodes and normal lymph nodes.Results There were 23 protein spots significantly differentially expressed,of which 4 protein spots were newly found,1 protein spot disappeared,14 protein spots increased markedly,and 4 protein spots decreased significantly.Conclusion The proteomic expression in metastatic lymph nodes of esophageal squamous cell carcinoma is significantly different from that of the normal lymph nodes,which may be associated with the mechanism of metastasis of esophageal squamous cell carcinoma.
4.Effect of continuous adenosine infusion on mesentery microcirculation during cardiopulmonary resuscitation in rats with asphyxia

Objective To evaluate the effect of continuous adenosine infusion on mesentery microcirculation during cardiopulmonary resuscitation in rats with asphyxia. Methods Rat model of asphyxia was established in 22 healthy Wistar rats. The animals were then randomly divided into normal saline group (group A, n=10), epinephrine group (group B, n=6) and epinephrine plus adenosine group (group C, n=6). After a 3-min asphyxia without intervention, cardiopulmonary resuscitation (CPR) was started. The cardiac compression was carried out with an electric cardio-pulmonary resuscitation machine (200 times/min). Respiration was restored and maintained with a ventilator with tidal volume of 4ml, breathing rate 50times/min and FiO2 100%. After a 4-min CPR, rats in group A and group B were given normal saline and epinephrine (bolus of 90?g/kg) respectively, and in group C epinephrine (bolus of 90?g/kg) plus 70?g/kg adenosine were given. Electric defibrillation would be initiated if there was ventricular fibrillation. The reperfusion rate of mesentery arterioles and venules, diameter of blood vessels and relative blood velocity were observed. Human genne myo-inositol monophosphatase 2(IMPA2) was recently a novel and promising gen that was associated with disease,especially in mental and neuro diseases.Its locus is in chromosome 18p11 2,about 15 kb.It encodes IMPA2 enzyme.IMPA2 enzyme as a mainly and catalytic inositol plays an important role in cell signaling system.The mechanism of action of IMPA2 gene is still unclear.But in basic research on genetic structure and IMPA2 product,the biochemical functions and crystal structure have gradually been recognized;In the clinical application,the association with disease has been detected in manic depression,schizophrenia and febrile seizuers.IMPA2 even has been a susceptible gene to certain diseases.IMPA2 has increasingly been the hotspot in gene research.
7.Immunogenicity of Different Genetic Type Rotavirus NSP4 in Mice

NSP4, as the diarrhea-related protein of rotavirus, is becoming an attractive candidate for vaccine development. To compare the immunogenicity of NSP4 from different genetic groups, we constructed eukaryotic expression plasmids comprising the NSP4 genes from four different genetic types using the pCI vector. The recombinant vectors were designated as pCI-97B6, pCI-97S36, pCI-97S34 and pCI-97SZ8, respectively. Following the conformation of the transient expression of the constructs in 293 cells, the plasmids were respectively subjected to the 5 round i. m. inoculation of BALB/c mice. The specific antibodies against NSP4 as well as the IgG1/IgG2a subclasses of immunoglobulin in mice sera were examined with indirect ELJSA after each immunization. The results showed that the immunization of plasmids expression NSP4s could elicit not only humoral but also cellular immunity, but the humoral immune response is dominant. There is a difference of immunogenecity among the NSP4 of different genetic type. Further studies were needed to focus on the relationship between the immunogenicity and protection effect.

9.Comparative study of dosimetry between volumetric-modulated arc therapy and intensity-modulated radiation therapy for brain metastases

Objective To compare volumetric‐modulated arc therapy(VMAT) with intensity‐modulated radiation therapy (IMRT) for brain metastases with regard to the dosimetric character .Methods Sixty patients who were diagnosed with brain me‐tastases were included in this study .The target area received two dose levels using late addition amount technique ,WBRT (30 Gy/10 F) with following addition (20 Gy/10 F) to 59 Gy .For a fair comparison ,VMAT and IMRT treatment plans were respectively designed for every patient with the same dosimetric constraints .Dosimetric comparisons between VMAT and IMRT plans were ana‐lyzed to evaluate :target coverage and homogeneity ,conformity of PTV ;sparing of OARs ;monitor units (MUs) .Results Two treatment plans all reached the treatment need .When compared with IMRT ,there was no significant difference in Dmean of eyeball , len ,optic never ,visual chiasma ,parotid ,brain stem ,and external auditory canal of VMAT (P>0 .05) .The Dmax of eyeball ,len ,pa‐rotid ,and external auditory canal of VMAT were lower than that in IMRT group (P<0 .05) .The VMAT group has the less MUs (P=0 .017) and less treatment time .Conclusion VMAT can reach the big‐dose radiotherapy need on brain metastases clinically . There are no significant diffference between VMAT and IMRT on Dmax ,Dmean ,CI ,and HI .The Dmax of eyeball ,len ,parotid ,and external auditory canal of VMAT were lower than that in IMRT group .The VMAT can reduce the radiotherapy time .

Objective To study topiramate′s neuroprotection on primary cultured hippocampal neurons which were injuried by glutamate and its mechanism.Methods The primary cultured hippocampal neurons were made as the research object,and excitotoxicity model was exected with glutamate.Hippocampal neuron survival was assessed by MTT method and hippocampal neuron mitochondrial membrance potential(MMP) was evaluated by fluorescence microscope and flow cytometry.Results Hippocampal neuron survival of normal control was(98.4?0.8)%,and the survival of glutamate model group was(59.6?3.2)%,at the same time,two topiramate′s groups′ cell survivals was(74.1?0.5)% and(79.2?3.4)%,and topiramate with two levels all could obviously increase hippocampal neuron survival((P
