2.Dynamic analysis of alkaloids and flavonoids in Genus Morus L. in China during different harvesting time.

To investigate the optimum harvesting time and utilization of mulberry leaves during different growth periods based on the content of alkaloids and flavonoids, 88 samples of 11 species of mulberry leaves were collected and analyzed. UPLC-TQ/MS method was applied and the results showed that the ingredients of alkaloids and flavonoids in mulberry leaves are quite different in different growth periods and different species. There was a sharp decline of the average content of alkaloids in all samples from October, while the content of flavonoids dropped either from October but with less volatile. The content of flavonoids in M. atropurpurea was much higher than alkaloids, while M. australis was opposite completely. There was a sharp decline of alkaloids in M. cathayana and M. mongolica from Tuly to August, however, the content of alkaloids and flavonoids in M. alba is neither too high nor too low. In summary, it is more suitable to harvest tender mulberry leaves harvested from the end of September to beginning of October that provide a scientific evidence for rational harvest and comprehensive utilization of mulberry leaves.

4.Comparing the effect of An's Shaobei Injection (symbols; see text) with Xiaozhiling Injection (symbols; see text) in patients with internal hemorrhoids of grade I-III: a prospective cohort study.

OBJECTIVETo compare the effect of An's Shaobei Injection ([symbols; see text]) with Xiaozhiling Injection ([symbols; see text]) in patients with internal hemorrhoids of grade I-III.
METHODSThis cohort study included 1,520 internal hemorrhoids patients with grade I-III who were scheduled for liquid injection treatment from July 2003 to July 2009. The cohort included patients who underwent either An's Shaobei Injection treatment (the treatment group, 760 cases) or Xiaozhiling Injection treatment (the control group, 760 cases). All patients were followed up regularly for 3 years; the observing indices included anal function recovery and clinical response after operation.
RESULTSAmong the 1,520 patients, 1,508 (99.2%) completed the 3-year follow-up. The efficacy rate was 97.5% in the treatment group, significantly higher than the control group (91.8%, P<0.01). The recurrence rate in the treatment group was 0.5%, significantly lower than that of the control group (1.3%, P<0.01). In addition, perianal callosity occurred in 8 cases (1.1%) and anorectal stricture in 26 cases (3.5%) after operation in the control group. There was no perianal callosity and anorectal stricture in the treatment group.
CONCLUSIONThe treatment with An's Shaobei Injection demonstrated superior clinical effect to Xiaozhiling Injection with fewer adverse effects.

5.Cigarette smoking affects cyclogeny of spermatogenic cells in rats.

OBJECTIVETo determine the effects of cigarette smoking on the cyclogeny of spermatogenic cells in rats.
METHODSRat models of passive smoking were established using a self-made smoking device, and then allocated randomly into two passive smoking groups (A and B, n = 10) and two corresponding control groups (C and D, n = 10). Groups A and B were exposed to cigarette smoke for 8 weeks, followed by the sacrifice of the rats in Groups A and C. And the animals in Groups B and D were killed 48 days after the cessation of passive smoking. The spermatogenesis cycle of each group of rats was detected by flow cytometry, the levels of testosterone (T) and luteinizing hormone (LH) measured by radio-immunity method, and the testis histopathology analyzed by HE staining and transmission electron microscopy.
RESULTSCompared with Group C, Group A showed a significant decrease in the number of spermatids, spermatozoa ([18.76 +/- 3.58]%) and primary spermatocytes ([5.71 +/- 1.18]%) (P < 0.01), but an obvious increase in the spermatogonias ([55.98 +/- 5.35]%, P < 0.01), with a markedly decreased proliferation index ( P < 0.01). The rats of Group A also exhibited pycnosis of spermatocytes, nucleus aberration of Leydig cells, expansion and degranulation of the endoplasmic reticulum, decreased Golgi apparatus, increased lysosomes and fat drops of Sertoli cells, as well as a reduction in the thickness of the wall and the layers of seminiferous tubules and the number of spermatogonia. The T and LH levels were significantly lower in Group A than in C (P < 0.01). After the cessation of passive smoking, a remarkable increase was observed in the percentage of spermatozoa and primary spermatocytes and the levels of serum T and LH in Group B, although the latter were still lower than those of Group D.
CONCLUSIONSmoking damages spermatogenic epithelia, Leydig cells and Sertoli cells, reduces the T and LH levels, and block the proliferation of spermatogenetic cells. These changes can be partially reversed after cessation of smoking.

6.Comparative analysis of three diarrhea-household-surveys in Guangxi

Objective To study the epidemiological characteristics and the effects of control measurements in Guangxi by comparing the results from diarrhea-household-surveys conducted in the three different periods of time and to develop control strategies. Methods Data on the incidence, health care seeking and treatment of diarrhea from three household surveys conducted in 1988, 1995 and 2007 was analyzed using SPSS (version 13.0). Results The incidence rates of diarrhea over the three periods of time were 0.562, 0.456 and 0.221 per person-year, respectively (P<0.001). No significant difference was found in the incidence between males and females. The disease mainly attacked young age groups and those with lower educational levels. In recent years, longer durations of disease but less severe were observed in patients with diarrhea. The patients mainly visited first line health services with a health care seeking rate of 28.3%. Antibiotics were used by most of the patients (49.8%-90.2%), while the rate of using oral rehydrathon salts(ORS) was only 1.4%-11.5% but the use of traditional Chinese medicines has increased. Intake of untreated water, contaminated foods and contact with patients were important risk factors on diarrhea. Conclusion The prevalence of diarrhea in Guangxi had declined and the health seeking rate was low in the past two decades. It is necessary to further regulate the treatment, in order to strengthen the health education programs to the general population, in order to improve the accessibility of health services and to increase both the health care seeking rate and effective diagnosis rate.

Objective To characterize the biological characteristics of natural vectors of Yesinia (Y.) pestis in China.Methods Species and genera of natural vectors of Y.pestis in China and their faunal distribution were characterized with modem insect taxonomic techniques.The ecological roles of natural vectors of Y.pestis in natural plague foci were determined according to insect ecological experiments.Results There were 63 species of natural vectors of Y.pestis including 28 major reservoirs and 35 secondary ones.Conclusion The biology characteristics of major vectors on Y.pestis and their roles in natural plague foci were defined.
8.Analysis of relevant factors affecting results of Ritleng lacrimal duct intubation surgery and nursing intervention measures

Objective To find the relevant factors that can influence the results of Ritleng lacrimal duct intubafion surgery, apply effective nursing intervention measures, and cooperate with the success conduct of operation and perioperation. Methods 102 patients who underwent Ritleng lacrimal duct intubation were analyzed in our study. The relevant factors were found out, then various corresponding nursing intervention measures before and after the surgery were used in 293 patients that underwent the same operation from May 2007 to February 2008. Results After applying the effective nursing intervention measures, the cure rate of these 293 patients was 95.9%. Silicone tube was never broken in the operation. Conclusions Applying various corresponding nursing intervention measures before and after Ritleng lacrimal duct intubation can improve the success rate of surgery and reduce the complications.
9.Sensitivity of Oridonin on temozolomide through enhances the U251 and via down-regulating nuclear factor-κB pathway

Objective To study the timorazolamide sensitivity for U251 after the pretreatment by Oridonin.Methods All experiments were divided into 5 groups including control group (not drugs) and extreme low,low,medium,and high concentrations (Oridonin:10,20,40,80 μmol · L-1) groups.U251 cells in all groups were cultured in 24 h,48 h and 72 h.At different time points,U251 cells' proliferation and nuclear factor-κB (NF-κB) protein expression in all groups were detected by Western blot.After culturing U251 cells in medium group and high group for 72 h,cells in the two groups continued to be incubated in extreme low,low,medium,and high concentrations (Temozolomide:50,100,200,400 μmol · L-1) for 24 h and 48 h.Apoptosis ratio in the two groups were detected by flow cytometer.Results After culturing 48 h and 72 h,relative expression of NF-κB in medium group and high group were 0.67±0.11,0.55 ±0.21;0.45 ±0.12,0.51 ±0.17 respectively,compared with the control group,the difference had statisticalsignificantly (all P < 0.05).After culturing 72 h in medium group and high group,U251 cells continued to be incubated in 4 different concentrations temozolomide groups for 48 h,and apoptosis ratio were(50.13 ±5.44) %,(55.34 ±5.93) %,(55.58 ± 4.92)%,(57.51 ± 4.89)%;(55.67 ± 3.96)%,(60.34 ± 6.89)%,(59.77 ± 4.71)%,(63.45-± 5.67) %,compared with the control group,the difference had statistical significantly (all P < 0.05).Conclusion The NF-κB protein expression of U251 cell can inhibited by Oridonin,and the drug sensitivity of U251 cells can be enhanced by pretreatment of temozolomide.
10.The study of multiple RT-PCR-based reverse dot blot hybridization technique for detecting influenza viruses

Objective To establish a multiplex RT-PCR-based reverse dot blot hybridization technique to detect influenza viruses. Methods Obtain the HA nucleotide sequences of seasonal influenza H1 N1, seasonal influenza H3N2, influenza H1 N1 and human avian influenza H5 N1 from GenBank. Design primers in conservative district and probes t in high variable region respectively, after analyzing the HA nucleotide sequences of influenza virus through the Vector NTI 9.0. Establish and optimize multiple RT-PCR system by comparing amplification efficiency and specificity at different primer concentrations. Establish the reverse dot hybridization system after optimizing the concentration of probes. To compare the sensitivity and specificity of this technique and the general RT-PCR Method through extracting the viral RNA of the mentioned influenza virus which are to be the reference substance. Results Successfully establish a multiplex RT-PCR-based reverse dot blot hybridization technique for detecting influenza viruses. This technique is 100-1000 times more sensitive than gel electrophoresis method, and it has a good specificity.Conclusion Successfully established multiplex RT-PCR-based reverse dot blot hybridization technique for detecting influenza viruses.
