Objective To observe the expression of matrix metalloproteinase-9(MMP-9)/tissue inhibitor of metalloproteinase-1(TIMP-1) in tubulointerstitial fibrosis(TIF) rats and explore the effects of MMP-9 and TIMP-1 in TIF.Methods Forty-eight male Sprague-Dawley(SD) rats were randomly divided into sham-operated group(n=24) and model group(n=24).Results Tubulointerstitial fibrosis model was established via unilateral ureteral obstruction(UUO). AIM: To evaluate the clinical effects of compound trabeculectomy for primary angle-closure glaucoma with persistent high intraocular pressure.
●METHODS:Thirty-three cases ( 34 eyes ) with primary angle - closure glaucoma were executed compound trabeculectomy. All patients were divided into two groups: group A: 18 cases ( 18 eyes ) were executed compound trabeculectomy under high intraocular pressure higher than 35mmHg after using lOP-lowering medicine for 48-72h; group B: 15 cases (16 eyes) were executed compound trabeculectomy, the intraocular pressure was normal after using lOP-lowering medicine.
●RESULTS:Thirty-four eyes were successfully completed surgery with follow-up of 6 to 18mo and there were no serious complications, including suprachoroidal hemorrhage. The visual acuity before operation in group A and group B were 0. 02 ± 0. 01, 0. 04 ± 0. 02, respectively. And the postoperative visual acuity in group A and group B increased 0.2±0. 06, 0. 3±0. 07, respectively (P<0. 01). All of them, intraocular pressure with 30 eyes were controlled in 9-23 mmHg, 3 eye was controlled by normal through adding in several kinds different lOP-lowering medicine. One eye was failure. Compared to preoperation, postoperative intraocular pressure of two groups were decreased by normal, which were 17. 9 ± 9. 1 mmHg and 15. 4±8. 4mmHg, respectively (P<0. 01). There was no significant difference between two groups for postoperative intraocular pressure.
●CONCLUSION: The primary angle-closure glaucoma with persistent high intraocular pressure should be executed compound trabeculectomy decisively, even under high intraocular pressure, which prevent further damage and loss of visual function. As long as to fully consideration the preoperation, intraoperative careful operation, careful nursing, it is safety and effective to have compound trabeculectomy under the continuous high intraocular pressure for primary angle-closure glaucoma.

Objective To build a more perfect serum protein ifngerprint models for early diagnosis of ganglioneuroblas-toma (GNB) in children. Methods Thirty children with GNB and 30 normal control children were recruited. Serum samples were collected. Nonspeciifc serum protein was detected and studied by MB-WCX processing, SELDI-TOF-MS mass spectrom-etry system and MALDI-TOF/TOF platform. Results Through the SELDL-TOX-MS processing, a peak at 5920 m/z protein markers, and the expression of the markers was high in GNB children (6180.6±2328), compared with normal control children (419.1±493.3), the difference was statistically signiifcant (P<0.05);MALDI-TOF/TOF platform showed that the protein with a peak at 5920 m/z is identiifed as ApoC-Ⅲ. Conclusions m/z peak of 5920 protein is suggested as speciifc biomarker of GNB in children, can provide signiifcant reference for early diagnosis of ganglioneuroblastoma, and prognostic monitoring.

Objective To study the expression of Rho kinase and its functional activation in lung tissue from hypoxic pulmonary hypertension(HPH) rat model,and the effects of fasudil on HPH.Methods Seventy-two male Spraque-Dawley rats were randomly divided into the control group,hypoxic model group,and fasudil-intervention group[group with hypoxia and fasudil for 15 mg/(kg?d)],respectively.Mean pulmonary arterial pressure(mPAP) and right ventricle hypertrophy index(RVHI) were measured.Expression of Rho kinase mRNA and protein were examined by reverse transcriptase-polymerase chain reaction(RT-PCR) and Western blot,respectively.The phosphorylation of binding subunit of myosin phosphatase(MBS)-a substrate of Rho kinase was detected by Western blot and defined,as the mark of functional activation of the kinase.Results The expression of Rho kinase mRNA in hypoxic model group was markedly upregulated even before the onset of the third day after the experiment(HPH),and it was much lower in rats of fasudil group than that of hypoxic model group.The phosphorylation of MBS was significantly higher in hypoxic model group than that in control group,and it was positively correlated with the mPAP and RVHI(all P

Objective The objectives of this study were to investigate the relationship between DKK1 protein expression and lymph node invasion,and prognosis in patients with intrahepatic cholangiocarcinoma.Methods From January 2005 to December 2012,78 patients with intrahepatic cholangiocarcinoma were treated with endoscopic resection.Tissue microarray of intrahepatic cholangiocarcinoma and adjacent tissues were done and biochemical indexes were measured before operation.The patients were recruited every 3 months after operation.The Kaplan-Meier method was used to analyze the recurrence-free survival curve and the overall survival curve.The Cox proportional hazards regression model was used to analyze the multivariate analysis and to determine the factors that affect the disease-free survival(RFS)and overall survival(OS).Results There were significant differences in the expression of GGT,hepatic portal lymph node invasion,Child-Pugh grade,MMP9 and DKK1 protein in the negative and positive expression of intrahepatic cholangiocarcinoma(P ＜ 0.05).The positive expression rate of DKK1 protein in intrahepatic cholangiocarcinoma was 35.90％ (28/78),and the positive expression rate of DKK1 protein in hilar cholangiocarcinoma was 14.10％ (11/78).RFS and OS in 78 cases of intrahepatic cholangiocarcinoma were 51.28％ (40/78) and 50.00％ (38/78),41.03 ％ (32/78) and 38.46％ (30/ 78),25.64％ (20/78) and 23.08％ (18/78) after surgery for 1,3 and 5 years,respectively.The results from univariate and multivariate analysis showed that GGT,CA19-9,CEA,tumor size,DKK1 and hilar lymph node invasion were the prognostic factors of OS in patients with intrahepatic cholangiocarcinoma.CEA,tumor size,DKK1 and hilar lymph node involvement were prognostic factors for RFS in patients with intrahepatic cholangiocarcinoma.DKK1 protein expression positive and negative OS curve or RFS curve showed that 5 years after intrahepatic cholangiocarcinoma patients DKK1 protein expression positive and negative OS were 28.20％ and 20.51％;RFS were 24.36％ and 21.79％.Conclusion The expression of DKK1 protein is closely related to lymph node invasion in patients with intrahepatic cholangiocarcinoma.The expression of DKK1 protein is the prognostic factor of OS and RFS in patients with intrahepatic cholangiocarcinoma.

Objective To investigate the adjunctive therapeutic effects and safety of intravenous immunoglobin (IVIG) for treating pneumonia following kidney transplantation.Methods Sixteen cases of pulmonary infection after kidney transplantation were divided into two groups.Twenty-eight cases were subjected to IVIG therapy (0.2 g·kg-1 ·day-1) for 7-10 days besides the standard specific anti-bacterial,anti-fungal,and anti-virus treatment and regular immunosuppressive regimen with dose adjustment (IVIG group),and the control group was only treated with standard specific anti-pathogen therapy.The incidence and mortality ofsevere pulmonary infection,levels of serum IgG,T lymphocyte subsets,and creatinine in the two groups were observed.Results The effective power of IVIG group and control group was 100 ％ and 93.75 ％ (P＜0.05).The incidence of severe pneumonia in IVIG and control groups was 0 and 12.5％,respectively (P＜0.05),with the mortality being 0 and 6.25％,respectively (P＜ 0.05).The levels of serum IgG were significantly increased in IVIG group as compared with that before treatment and in control group.There were no significant adverse reactions associated with IVIG infusion.Conclusion As an adjunctive therapy,IVIG treatment for pulmonary infection can reduce the incidence of severe pulmonary infection and mortality after kidney transplantation,further increase the survival rate of patients after kidney transplantation.

ObjectiveTo prepare a novel etimicin-encapsuled chitosan/hydroxyapatite nano-scaffolds and offer assistances for bone defect or osteomylitis.MethodsDrug-carried chitosan nanoparticles which was prepared by ionotropic gelation were combined with nano-hydroxyapatite.The mixture were shaped in molds and then prepared into porous scaffolds by freeze-dry.The surface of one scaffold was scanned.The grinded,particles of the scaffold were detected by field emission scanning electron microscope; X-ray diffraction was used to analyze components of the scaffold and total porosity.Staphylococcus aureus was choosed as the experimental bacteria,we studied lasting antibacterial property of drug-carried bone scaffold by antibacterial experiments,long-term drug releasing experiments and accumulation drug releasing experiments.Bone mesenchymal stem cells were used to detect the histocompatibility and inductivity of etimicin-carried scaffold.ResultsFreeze-dried porous scaffold has a surface with proper pore distribution (total porosity 70.68％) and the grinded scaffold has a globular and coliformed microstructure known after scanned by electron microscope.The drug-carried scaffold has a typical wave of hydroxyapatite under X-ray diffraction.The lasting antibacterial property study indicated that the drug-carried bone scaffold had maintained an inhibition zone for more than 7 days.The long-term drug releasing experiments and accumulation drug releasing experiments show that the fictional drug-carried bone scaffold released above the bacteriostasis concentration after one week and the accumulative amount within the safety scale.The scaffold had not an inhibitory effect on bone mesenchymal stem cells.ConclusionThe etimicin-encapsuled chitosar/ hydroxyapatite nano-scaffolds has similar microstructure and components of bone tissue.It is promising in bone tissue engineering applications because of its slow-release,antibacterial properties and satisfactory histocompatibility.

Objective To perform pathogenic surveillance of scarlet fever in Xicheng District of Beijing in 2014 ,and provide sci‐entific data for developing reasonable prevention policy for the disease .Methods The throat swab specimens from 212 patients who were diagnosed with streptococcal infection ,tonsillitis and angina in surveillance hospital were collected ,and from which the patho‐gens were isolated and identified .Results In the 212 samples ,the positive rate of A group hemolytic streptococcus were 21 .2%(45/212) .The patients were mainly 4-15 years old ,especially 6- <8 years old .The positive rate was highest in 6- <7 years old children .The peak of disease incidence was observed in May and June .Conclusion Preschools and schools were the key sites for scarlet fever prevention and control ,therefore ,the surveillance and prevention should be further strengthened to prevent the out‐break .
