1.Experience of diagnosis and treatment of iatrogenic injury by ureteroscopic surgery

Objective To summarize the experience of diagnosis and treatment of iatrogenic injury by ureteroscopic surgery.Methods Retrospective analysis of 13 cases with iatrogenic injury by ureteroscopy from December 2008 to December 2011,including 8 men and 3 women,aged 15 to 75 years.Among the 13 cases (Holmium laser lithotripsy under ureteroscope),there were 5 cases of ureterostoma severe disruption,4 cases of submucosa injury,2 cases of perforation,1 case of disruption,and 1 case of sleeve exfoliation of mucosa.Results Among these 13 cases with iatrogenic injury by ureteroscopy,10 cases underwent double J drainage (drainage duration:60 days),and 3 cases underwent open surgery immediately.There was no hydronephrosis when examined by IVU after six months to two years follow-up.Conclusions The skills and techniques of surgical operation should be improved when performing ureteroscopic operation,and it is essential to be familiar with ureteric dissection and alignment,which can avoid ureteric injury.Indwelling D-J tube is very important in dealing with mild ureteral injury secondary to ureteroscopes.Surgical intervention should be given to severe cases of ureteric injury in time.
2.Comparison of different operation methods for the treatment of localized and high risk prostate cancer

Objective To compare clinical efficacy and complications of three operation methods for treating the localized and high risk prostate cancer.Methods From July 2006 to July 2010,102 cases with localized and high risk prostate cancer were included in the study.Among them,51 cases received open radical prostatectomy (Group A).Their characters included aged (63.7±7.9),PSA (31.8±6.6) μg/L,Gleason scores (8.5±0.5).In this group,T2c stage was diagnosed in 38 cases and T3a stage in 13 cases.32 cases received laparoscopic radical prostatectomy(Group B).Their characters included aged (64.4± 8.3),PSA (29.9±5.2) μg/L,Glcason scores (8.7±0.4).In this group,T2c stage was diagnosed in 21 cases and T3a stage in 11 cases.19 cases received 125I implantation (Group C).Their characters included aged (61.4± 7.4),PSA (30.6±.5.7)μg/L,Gleason scores (8.6±0.6).T2v stage was found in 8 cases and T3a stage in 11 cases.Operation time,amount of bleeding,hospitalization time,drainage time,gastrointestinal function recovery time,medical expenses and survival and recurrence rate in 1 year,3 years,5 years were compared within those groups.Results Operation time in three group were (232.6±38.4) min,(186.3±31.4) min,(35.4±14.6) min,respectively.Amount of bleeding in three group were (413.6±132.4) ml,(273.9± 77.4) ml,(19.4±4.4) ml,respectively.Hospitalization time in three group were (20.9±3.7) d,(15.6± 2.2) d,(6.4±2.6) d respectively.Drainage time in three group were (8.3±1.8) d,(7.5±0.9) d,(3.2± 0.8) d,respectively.Gastrointestinal function recovery time in three group were (4.1 ±0.6) d,(3.2± 0.4) d,(0.4±0.1) d,respectively.Medical expenses in three group were (23±4) thousand yuan,(32±3) thousand yuan,(45t3) thousand yuan respectively.All those items exhibited the significantly statistical difference (P<0.05).The survival and recurrence rates had no significant difference during the follow-up in three groups.Conciusions Compared to the open radical prostectomy and laparoscopic radical prostectomy,particle implantation for prostate cancer had advantages in minimally invasive,less bleeding,short operation time,fast recovery and protection for Intestinal function.
3.Using standardized patients in training of assistant general practitioners

Thirty assistant general practitioners (GPs) of Jiading District Shanghai were trained for clinical skills and communication skills by using standardized patients (SP) in outpatient department from January to April 2014.The scores of clinical skills and communication skills were evaluated and compared before and two months after training.The results showed that the score of clinical skills [(72 ± 5) vs (56 ± 7), t=7.871, P<0.05] and communication skills [(71 ±4) vs (45 ±5), t =9.573, P<0.05] were significantly improved after training.After training, the scores of medical history taking [(36 ±5) vs (28 ± 6), t =4.352], clinical thinking [(17 ± 2) vs (14 ± 3), t =3.974], health education [(3 ± 1) vs (2 ± 1), t =3.090] and medical record v,riting [(13 ± 1) vs (8 ±2), t =5.317] were significantly higher than those before training (all P < 0.05).But there was no difference in reception time management [(4 ± 0) vs (4 ± 0), t =1.025] before and after training (P > 0.05).After training, the scores and the proportions of the trainees qualified for building physician-patient relationship [(20 ± 3) vs (15 ± 4), t =4.056],collecting information and providing feedback [(22 ±4) vs (13 ±3), t =4.682], understanding complaints from patients' perspective [(5 ± 2) vs (3 ± 2), t =3.107], sharing information and obtaining informed consent [(19 ± 4) vs (11 ± 5), t =4.322] and closing consultation [(6 ± 2) vs (3 ± 1), t =2.987] were higher than before training (all P < 0.05).The results indicate that training by using SP can effectively improve the clinical skills and communication skills of assistant GPs.
4. Genetic diversity of Lonicera macranthoides geographical populations revealed by SRAP markers

Objective: To research genetic diversity of different Lonicera macranthoides geographical populations. Methods: Seventeen materials were estimated by the approach of sequence-related amplified polymorphism (SRAP). The data of amplified bands were analyzed by Popgene 1.31 and Treeconw software. The system diagram of genetic relationship was built by UPGMA. Results: The 24 pairs of SRAP primers combination employed produced a total of 239 discernable and reproducible amplified fragments. Among them there were 210 polymorphic bands. The percentage of polymorphic bands within different populations was 87.8%; Genetic diversity analysis showed that Nei's gene diversity (He) was 0.239 9 and Shannon's genetic diversity index (I) was 0.372 4. Basis on the DNA molecular level, the results indicated that there was abundant genetic diversity among the tested materials. Genetic similarity coefficient ranges changed from 0.442 3 to 0.940 2. The dendrogram including all samples was obtained by UPGMA. In the dendrogram, there were two cluster groups. Conclusion: The genetic diversity of L. macranthoides geopraphical population in China is plentiful. SRAP markers can be effectively applied to genetic analysis in L. macranthoides populations.
5.Quality classification standard of Scrophularia ningpoensis seedlings.

The morphological indexes of the Scuophularia ningpoensis seedlings including the longth, diameter and weight were measured, clustering analysis was used to set up the standard quality grading of seedlings of S. Ningpoensis by SPSS. Field experiment was carried out to measure the indicators of plants growth and development, the yield and the quality. The results showed that the growth and yield of class I seedlings were better than those of class II and III. The content of main active ingredients was affected barely by seedlings classification. To ensure the quality, class II seedlings or above should be used for plantation. The established quality classification standard of S. ningpoensis seedlings was scientific and feasible, and provides the basis for the standardized cultivation of S. ningpoensis.

6. Analysis on genetic relationship of medicinal species of Dipsacus marked by ISSR, SCoT, and SRAP

Objective To detect the genetic relationship among nine medicinal species of Dipsacus in China. Methods The polymorphic bands were detected by three labeling methods (ISSR, SCoT, and SRAP). The genetic distances were calculated by TREECONW software and the systematic diagram of genetic relationship was clustered by UPGMA method. Results The percentage of polymorphism by ISSR, SCoT and SRAP markers showed little difference with value at 90.4%, 88.5%, and 88.2%, respectively, which indicated that the genetic polymorphism of the tested materials was very rich. The genetic distance between Dipsacus chinensis and D. japonicus was the largest, which indicated their genetic relationship was the most distant. Nine medicinal species of Dipsacus were all divided into three groups by three markers, that D. chinensi and D. lijiangensis, D. asperoides and D. japonicas were respectively clustered into one group, D. asperoides var. omiensis was alone clustered, the other four species were clustered into one group. The clustering results labeled by ISSR and SRAP were basically the same with a slight difference between D. daliensis and D. asperoides. Conclusion The clustering results by there markers was between well consistent with the classical classification, which confirmed that molecular markers can be used as one of the effective methods to reveal the genetic relationship among medicinal species of Dipsacus.
7.The enantioselective pharmacokinetic study of desvenlafaxine sustained release tablet in Chinese healthy male volunteers after oral administration.

A chiral LC-MS/MS method for the simultaneous analysis of desvenlafaxine (DVS) enantiomers in human plasma was developed and applied to a pharmacokinetic study on 12 Chinese healthy volunteers. d6-Desvenlafaxine was used as internal standard (IS). Chromatographic separation was performed on the Astec Chirobiotic V chiral column (150 mm x 4.6 mm, 5 μm). The assay was linear over the concentration range of 0.500-150 ng x mL(-1) for both enantiomers (r2 > 0.99). The method was successfully applied to a stereoselective pharmacokinetic study of 100 mg desvenlafaxine sustained release tablets on 12 Chinese healthy volunteers under fasting conditions. The results showed that the pharmacokinetic parameters were similar to both enantiomers in Chinese healthy volunteers. The AUC(0-t), and C(max) of the two enantiomers were about 1.5 times higher than those of blacks and whites reported in the literature.

8.Growth and developmental rhythm of Scrophularia ningpoensisin southwest middle mountain area of China.

Plant samples were collected and investigated periodically. According to the growth of different parts and the characteristics of dry substance accumulation of Scrophularia ningpoensis, the development of S. ningpoensis could be divided into four stages: seeding stage, stem and leaf growth stage, expanding period of root tubers, and dry substance accumulation stage of root tuber. Leaf numbers of S. ningpoensis grew gradually from one at first to 370 at the final stage, main stem leaf were 50 pieces. Leaf size increasesed with the fastest growth at the stem and leaf growth stage, average daily increase amount was 225 cm2. By the middle of August, leaf size reached to 16,270 cm2. Leaf area indexrose sharply in the seeding stage, and remained above 8 among stem and leaf growth stage and expanding period of root tubers, and rapidly reduced to zero in the stage of dry substance accumulation of root tuber. Leaf area ratio has a tendency of obvious dropping. The net assimilation rate had a small change ranges, two small peak were seeding stage and dry substance accumulation of root tuber. The value of specific leaf area was higher in seeding stage, and in the earlier stage of dry substance accumulation of root tuber. Relative growth rate changed with large ranges, higher in seeding stage, rapid decrease in stem and leaf growth stage, rose in expanding period of root tubers, and declined again in the stage of dry substance accumulation of root tuber. Crop growth rate was higher in the first and last stages, and smaller in interim stage. The growth parameters of S. ningpoensis such as relative growth rate, net assimilation rate, leaf area index, leaf area ratio, specific leaf area, crop growth rate changed along with the growth. The rule of dry matter accumulation was as follows: the dry matter increased slowly during the seeding stage and speeded up in the middle and late stages, and in dry substance accumulation of root tuber increased slower, the growth of dry matter all appeared an "S" curve, and accorded with logistic equation. Cultivation technologies of S. ningpoensis and the relevant management methods could be established according to the growth of different parts of S. ningpoensis and the characteristics of dry substance accumulation in different stage.

9.Analysis on genetic diversity among wild Dipsacus asperoides by SCoT.

To investigate the genetic diversity among wild Dipsacus asperoides in China, 66 germplasmic resources of D. asperoides were analyzed by Start Codon Targeted Polymorphism (SCoT) molecular markers. Genetic distance was calculated by TREECONW software and the systematic diagram of genetic relationship was clustered by UPGMA method. The results showed that the totals of 181 bands were detected using 20 primers , among which 109 were polymorphic bands. The average percentage of polymorphic bands was 60.13%. Genetic distance changed from 0.030 6 to 0.181 4. The clustering results showed that there was no significant correlation between the classification of the wild D. asperoides and their geographical origin. The relatively high genetic diversity of D. asperoides provides the basis for breeding new varieties.

10.Genetic structure and genetic diversity of Artemisia annua varieties (strains) populations based on SCoT markers.

To reveal the genetic diversity and genetic structure in Artemisia annua varieties (strains) populations, we detected the genetic polymorphism within and among eight varieties (strains) populations (192 individuals) by the approach of Start Codon Targeted Polymorphism (SCoT). The associated genetic parameters were calculated by POPGENE1.31 and the relationship was constructed based on UPGMA method. The results showed that, using 20 screened primers, a total of 145 bands were produced, of which 122 were polymorphic loci. At species level, there was a high level of genetic diversity among eight varieties (strains) populations (PPB = 84.1% ,H = 0.217 3 and H(sp) = 0.341 9). However, at the variety (strains) population level, genetic diversity was lower, the average of genetic parameters was PPB = 41.9%, H = 0.121 5, H(pop) = 0.186 8. The Nei's genetic differentiation coefficient was 0.441 0, indicate that most of the genetic variation in this species existed within the variety populations. The gene flow (N(m) = 0.633 9) was less among populations, indicating that the degree of genetic differentiation was higher. Genetic similarity coefficient were changed from 0.755 1 to 0.985 7. By clustering analysis, eight varieties (strains) were clustered into two major categories and it was also showed the same or similar genetic background varieties (strains) have a tendency to gather in the same group. Results suggest that, in variety breeding, breeders should strengthen the exchange of bred germplasm and increase mutual penetration of excellent genes, which would broaden the genetic base of A. annua.
