1.Chinese Sanqi industry status and development countermeasures.

Sanqi (Panax notoginseng) is a valuable unique herb, and is also one of the very fast developed varieties of traditional Chinese medicines in recent years with increasing role in traditional Chinese medicine industry. This paper summarized the main experience, industry development and present situation, pointed out the main problems existing in the industry development. On this basis, we put forward the targets and measures for the development of the Sanqi industry in to provide decision-making reference for the sustainable development of the Sanqi industry in China.

2.Three-D gait patterns before and after total hip arthroplasty.

OBJECTIVETo determine how selected gait parameters may change as a result of total hip arthroplasty (THA).
METHODSFrom February 2006 to February 2007 the study was performed on 53 osteonecrosis of femoral head subjects prior to and 6 weeks, 3, 6 and 12 months after total hip arthroplasty surgery. Gait analysis was performed using the three-dimensional Infrared-based system (Dalian Dong Fang Xin Rui Company, China). Kinematic data were recorded for the lower limb. The results obtained from the osteonecrosis of femoral head subjects were compared with those of 40 individuals without osteonecrosis. Using a three-dimensional motion analyzer, the following were measured: (1) gait speed, stride length, cadence; (2) step length, stride width; (3) standing phase time, walking phase time; (4) gait cycle time, Joint movement angular.
RESULTSCompared with persons before total hip arthroplasty, THA patients showed significantly increased gait speed, gait frequency, step length and affected limb stand phase (P < 0.01). Angle timetable: preoperative hip-maximum flexion was 15 degrees, post-extension was 8 degrees, knee-maximum flexion was 70 degrees, ankle-maximum flexion was 15 degrees. Postop hip-maximum flexion was 27, post-extension was 17 degrees, knee-maximum flexion was 50 degrees, ankle-maximum flexion was 14 degrees; parameters of hip, knee between preoperative and postoperative was significance (P < 0.01).
CONCLUSIONThese results suggest that 3-D gait analysis before and after total hip arthroplasty can evaluate precisely hip vitodynamics variation.

3.Study on dynamic change of middle and micro element in Panax notoginseng plant soils with different interval year.

OBJECTIVETo study effects of different interval year on Panax notoginseng plant soils middle and micro element content.
METHODThe dynamic change of Ca, Mg, Mn, Cu, Zn, B from Mabai, Matang, Gumu and Panlong were determined under different planting patterns (new soil, interval 5 years soil and continuous cropping soil).
RESULTAll the micro elements (except Ca, Mg) of interval 5 years soil and continuous cropping soil were significantly higher than those of new soil. All the middle and micro elements (except B) of interval 5 years soil were significantly higher than those of the continuous cropping soil. Planting patterns had remarkable influence on the content of Mn, Cu, B, but not Zn Ca, Mg. Cu, Ca under the 3 planting patterns, and Zn under the continuous cropping pattern did not show significant quarter changes. B content increased with the elongation of implantation time. Zn in new soil and interval 5 years also increased with prolonging of planted time. Mg, Mn and Cu content reached to peak value on April next year, and reached to minimum on the end of this experiment. Compared with new soil, the proportion of Mn, Cu in total elements increased by 29%, 114%, Mg, B decreased by 18%, 38%, Zn and Ca changed slightly of interval 5 years soils; In continuous cropping soil, Mn, Cu and B increased by 50%, 120%, 22%, respectively, but Zn, Ca, Mg had no significant change.
CONCLUSIONContinuous cropping pattern could not induce the deficient of soil middle and micro elements, and thereafter might not result in continuous cropping obstacles. But the imbalance proportional of soil middle and micro elements in P. notoginseng plant soils may be one of the main reasons for continuous cropping obstacles.

4.Study on dynamic change law of N, P and K in Panax notoginseng plant soils with different interval year.

OBJECTIVETo study effects of different interval year on Panax notoginseng plant soils macro element content.
METHODThe dynamic change of total N, P, K and available N, P, K in soil from Mabai, Matang, Gumu and Panlong was determined under different planting patterns (new soil, interval 5 years soil and continuous cropping soil).
RESULTContents order of soil total N, P and available N, P were interval 5 years soil > continuous cropping soil > new soil. No significant quarter change on soil total N was found, but the other three showed inverted "v" curve, and the peak value appeared on April 2010. Content of soil total K did not change significantly, but the available K content order was new soil > continuous cropping soil > interval 5 years soil, the quarter change was similar as soil available N or P. The soil total N, P, K and available N, P, K were different of the 4 monitoring sites under the 3 interval planting modes. There was a significant correlation between soil total P and available P under all these 3 interval planting modes, but N and K. The propitiation of N-P-K of new soil, interval 5 years soil and continuous cropping soil were 1: 0.4: 2.4, 1: 0.4:1.4, 1:0. 4:2.0, respectively.
CONCLUSIONContinuous cropping pattern induce the accumulation of P, but deficient of K. The imbalance proportion of N, P and K was one of the incentives of continuous cropping induced obstacles. Strengthen the research of optimum proportion of soil N, P and K, and then eliminate continuous cropping obstacles by means of formulated fertilization is the future research direction.

5.Study on genetic diversity of herba Cistanches by RAPD.

OBJECTIVETo determine the genetic diversity of Cistanche species.
METHODTwo populations of Cistanche deserticola and four populations of C. tubulosa were analyzed by random amplified polymorphic DNA (RAPD) markers.
RESULTA total of 76 and 87 loci were amplified using 10 random primers each other. The average percentage of polymorphic loci of C. deserticola was 47.37%. The PPL were 39.47% and 35.53% for two populations. Average Nei's gene diversity was 0.1358, Shannon' s genetic diversity was 0.2072, and Gst was 0.2546. The average PPL of C. tubulosa was 27.59%. It was 19.54% to 25.29% in different populations and Andi'er population had the highest. Average Nei's gene diversity was 0.0823, and Shannon' s genetic diversity was 0.125 8, Cst was 0.175 5.
CONCLUSIONThe diversity of Cistanche deserticola is higher than that of C. tubulosa, but both has differentiation among populations, C. deserticola has already separated itself into two different ecotypes.

6.Study on the micropermeability of resin-dentin bonding interfaces with ethanol-wet bonding technique.

OBJECTIVETo evaluate the micropermeability on bonding hydrophobic adhesive to dentin with ethanol-wet bonding under simulated pulp pressure.
METHODSTwenty-four intact human third molars were used in the study. After the enamel of occlusal surfaces was removed, the molars were randomly divided into six groups. Adper Scotchbond Multi-Purpose was used in the control group; in the experimental groups, the dentin surfaces were saturated with ethanol for 20 s (group 1), 1 min (group 2), 2 min (group 3), 3 min (group 4) or with a series of increasing ethanol concentrations before application of hydrophobic adhesive (group 5). All the bonding procedures were done under simulated pulp pressure. After 24 hours, micro-tensile bond strength test were performed on the specimens. Bonding interfaces were observed under laser scanning confocal microscope (LSCM) after the pulp chamber were filled with a water-soluble fluoroprobe rhodamine B for 3 hours.
RESULTSCompared with the control group [(38.14 ± 4.97) MPa], bond strengths in group 1 [(21.02 ± 7.23) MPa] and group 2 [(29.64 ± 3.81) MPa] were statistically lower (P > 0.05), while bond strength in group 3 [(38.40 ± 5.03) MPa], group 4 [(37.26 ± 4.68) MPa] and group 5 [(40.12 ± 5.95) MPa] were similar to the control group (P < 0.05). The images taken by LSCM showed that with extension of ethanol-wet time, the deposition of fluorescent dye in hybrid layer and along the dentinal tubules decreased gradually. Especially in group 5, only spare fluorescent dye deposition could be detected in the hybrid layer.
CONCLUSIONSDentin saturated with ethanol for more than 2 min before bonding hydrophobic adhesive to dentin could provide favorable bond strength and decreased the micropermeability of bonding interfaces under simulated pulp pressure.

7.Immunohistochemical study of RET protein in Hirschsprung's disease and allied Hirschsprung's disorder.

OBJECTIVEHirschsprung's Disease (HD) and allied Hirschsprung's disorder (HAD) have very similar clinical manifestations, but there are many different theories about the two diseases. The present study was designed to understand the expression of Ret protein in HD and HAD, and to explore the role of Ret protein in the pathogenesis of HD and HAD.
METHODSColon specimens from patients with confirmed HD and HAD, including 15 cases of HD (male 12, female 3) and 11 cases of HAD (male 8, female 3) were collected for this study. At the same time normal colon specimens from 10 individuals with other diseases were used as control. The expression of Ret protein in the intestinal tissue was detected by using immunohistochemical SABC technique with mouse anti-Ret monoclonal antibody.
RESULTSIn the colon specimens from normal controls and the dilated segments of colon from HD and HAD patients, moderate to large number of Ret-positive cells were observed among the ganglion cells of myenteric plexuses and submucosal plexuses (P > 0.05). On the contrary, Ret-positive cells were not seen in the stenotic segment of colon from HD patients. But there was positive staining in the stenotic segment of the colon from HAD. Moreover, giant ganglion cells showing strongly positive staining could be seen. There were also displastic cells, small cells, and cells with irregular shape. Statistical analysis showed significant differences in Ret cells positivity between the stenotic segment of colon of HD and the normal control (P < 0.001) as well as between HD and HAD (P < 0.001).
CONCLUSIONRet protein may play an important role in the pathogenesis of HD and could not have definite relationship with HAD.

8.Free iliac crest grafts with periosteum for treatment of old acetabular defects.

OBJECTIVETo inquire into the therapeutic effectiveness of free iliac crest grafts with periosteum on old acetabular defects.
METHODSFrom February 1996 to June 2005, 9 patients were treated with free iliac crest grafts with periosteum to reconstruct old acetabular defects. There were 7 males and 2 females and the average age was 41.3 years. The acetabular defects were caused by traffic accidents in 6 cases and fall injury in 3 cases. The time from injury to treatment was 4-13 months and averaged 8 months. Intraoperatively we firstly removed the acetabular fracture fragments of the posterior wall. The femoral head was then reduced. Bone graft was harvested from the iliac crest with periosteum, which was sculpted with a rongeur to conform to the defect. The concave (iliac fossa) side of the graft was placed toward the femoral head. The graft was securely fixed by two to three leg screws.
RESULTSPostoperative syndrome was not found in any of the cases. Harris' score system showed that the score raised from 32.3 points preoperatively to 81 points postoperatively. The hip function was evaluated as excellent in 3 cases, good in 4 cases and fair in 2 cases.
CONCLUSIONSAlthough this procedure could not exactly reproduce the anatomy of the hip joint, it enables to restore the posterior stability, provide bone-stock for the hip joints and prevent dislocation of the femoral head.

9.Inhibitory effect of survivin antisense oligodeoxynucleotides on HepG2 cells by using polyamidoamine dendrimer as gene delivery system

Objective:To use polyamidoamine(PAMAM)dendrimer as gene delivery system for survivin gene anti- sense oligodeoxynucleotide(asODN)transfection for inhibition of HepG2 cancer cell growth.Methods:The first to the fifth generation of PAMAM and asODN were used to prepare a complex:PAMAM-asODN.The morphology of PAMAM- asODN was observed using agrose electrophoresis and atomic force microscope(AFM).PAMAM-asODN was then used to transfect HepG2 cells and cells transfected with asODN served as control.The transfection efficacy of PAMAM-asODN into HepG2 cells was observed under confocol microscope,the surviving mRNA expression was analyzed by RT-PCR,and the inhibition of HepG2 cell growth was determined by MTT assay.Results:Agrose electrophoresis showed strong complexing action between PAMAM and asODN and they formed a complex with a diameter of 25 nm.Confocol microscope showed the transfection efficacy of PAMAM-asODN was higher than that of asODN.RT-PCR showed a decreased expression of sur- vivin mRNA in PAMAM-asODN transfected cells.MTF results demonstrated that the growth of HepG2 cell was obviously inhibited after transfection of PAMAM-asODN and the inhibition rate increased with culture time,concentration of com- plex,the generation of PAMAM.PAMAM-asODN at 6.0?mol/L G4.0 resulted in a 55% inhibition of HepG2 cells 96 h after culture.Conclusion:PAMAM dendrimers can efficiently mediate the entry of survivin asODN into HepG2 cells,re- sulting in inhibition of HepG2 cells.PAMAM might be a promising gene carrier for potential molecular therapy of cancer.
10.Three-year follow-up outcomes of hepatocellular carcinoma patients undergoing liver resection versus liver transplantation

ObjectiveTo investigate the three-year follow-up outcomes of hepatocellular carcinoma patients undergoing liver resection (LR) versus liver transplantation (LT). MethodsA retrospective analysis was performed for 171 patients with hepatocellular carcinoma who underwent surgical treatment in Beijing YouAn Hospital, Capital Medical University, from March 2009 to March 2014, and according to the treatment method, they were divided into LR group(n=83) and LT group(n=88). Related clinical data were compared between the two groups. The chi-square test was used for comparison of categorical data between two groups; the Kaplan-Meier survival curve and the log-rank test were used for comparison of disease-free survival and overall survival between two groups, and the Cox proportional hazards model was used for the univariate and multivariate analyses of disease-free survival and overall survival. ResultsCompared with the LR group, the LT group had a significantly higher proportion of patients with single tumor [45.78% (38/83) vs 85.23% (75/88), χ2=29649,P<0.001], tumor size <3 cm [15.66% (13/83) vs 6705% (59/88), χ2=46.383,P<0001], or high Child-Pugh class [ 964% (8/83) vs 26.14% (23/88),χ2=7833,P=0.005] and a significantly lower recurrence rate of tumor [48.19%(40/83) vs 3295%(29/88) ,χ2=4121,P=0.042]. There was a significant difference in disease-free survival rate between the LR group and the LT group (46.02% vs 80.71%, P=0.006); the LT group had a higher overall survival rate than the LR group (86.99% vs 76.44%, P=0.219). Both univariate and multivariate analyses showed that treatment method was an independent risk factor for disease-free survival (risk ratio[RR]=3383,95% confidence interval[CI]: 1334-8579;RR=0239,95%CI:0093-0.612,both P<005), but the prediction of overall survival by treatment method did not reach statistical significance(P=0232). ConclusionLT is recommended for patients with early-stage hepatocellular carcinoma and can achieve a satisfactory three-year disease-free survival rate.
