Objective To investigate the characteristic and clinical significance of airway inflammation in children with acute asth-ma. Methods Underwent sputum induction and sputum induction in children (n=34) with acute asthma was repented in recovered children ( n = 24).Induced sputum were also taken from 15 healthy children as controls.Total and differential cell counts were per-formed. Interleukin(IL)-8、IL-6 were measured,The relationship between inflammatory cells and IL-8、 IL-6、peak expiratory (PEF) were analyzed.Results The inflammatory cell infiltrate was mixed including eosmophilic granulocytes, neutrophils, and macrophages. They decreased significantly, but eosinophilic granulocyte remained a higher percentage compared with healthy subjects. There was low-er percentage of lymphocytes at acute exacerhation.Eosinophilic granulocytes were correlated with the degree of airflow obstruction. Levels of IL-8、IL-6 were elevated during the acute exacerbation and decreased at resolution.IL-8 was correlated significantly with neutrophils at acute exacerhation and resolution.IL-6 was correlated signifficantly with eosinophilic granulocytes at acute exacerbation. Conclusions Airway inflammation is chataeterized by infiltration of eosinophils, neutrophils and macrophages.IL-8、IL-6 possibly is the important cytokines of airway inflammation in children with acute asthma. Increased eosinophils in induced sputum correlates with asthma severity. Therapy to the cytokines may have potential values.J Appl Clin pediatr,2004,19(12): 1023-1025

Objective To determine the changes of interleukin-12(IL-12) and IL-12 mRNA in gastric mucosa of children with helicobacer pylori (Hp) infection,and to study the effects of Hp infection on the expression levels of IL-12 and IL-12 mRNA,and to evaluate its possible roles in the pathogenesis of gastric mucosal inflammation in Hp related gastroduodenal diseases.Methods Biopsy specimens were taken from the antral mucosa on endoscopy in patients with or without Hp infection, which were diagnosed by urease test and Giemsa staining. The expression levels of IL-12 and IL-12 mRNA in gastric mucosa were determined by ELISA and RT-PCR.Results Inflammation of gastric antral mucosa was more severe in Hp-positive mucosa .The expression levels of IL-12 and IL-12 mRNA in Hp-positive mucosa were (66.42?15.15) ng/g and (59.21?15.03)%,which were more than those in (Hp-negative )(22.22?8.79) ng/g and (17.94?7.39)%(P

Objective To explore the effects of smoking and alcohol drinking on arsenic metabolism of people exposed to different concentrations of arsenic in drinking water.Methods Residents in Shanxi exposed to different concentrations of arsenic in drinking water and age ≥ 18 years old adults were chosen as the subjects for this study in 2008,the subjects were divided into three groups according to the concentrations of arsenic in drinking water: high-arsenic exposure group (more than 0.05 mg/L),low-arsenic exposure group (between 0.01 and 0.05 mg/L) and control group(less than 0.01 mg/L),excluded recently had eaten seafood and had poisoning symptoms of chronic arsenic in drinking water in the crowd.Smoking and alcohol drinking habits were investigated by questionnaire.Arsenic species in the urine samples were detected with hydride generation atomic absorption spectroscopy.Total arsenic(tAs) was the sum of iAs%,MMA% and DMA%.iAs%,MMA% and DMA% were calculated as iAs/tAs,MMA/tAs and DMA/tAs,respectively.The first methylation ratio(FMR) and the secondary methylation ratio(SMR) were calculated as (MMA + DMA)/tAs and DMA/(MMA + DMA),respectively.Results Three hundred and ninety-five adults were chosen in this study.In the high exposure group the alcohol drinking and smoking subjects had higher MMA%(16.24%) but lower SMR(82.19% ) than the non-drinking and non-smoking subjects (12.16% and 86.13%,respectively).The differences of both MMA% and SMR were significant(P < 0.05 ).No significant difference was observed between the non-smoking/non-drinking subjects and the smoking or the drinking subjects(all P > 0.05 ).In the low exposure group there were higher MMA%( 13.86%,13.99%) lower DMA%(72.87%,77.76%)and lower SMR (83.48%,83.90% ) in those with smoking or drinking/smoking compared with the non-drinking and non-smoking subjects (11.83%,80.35% and 86.54%,respectively,all P <0.05 ).No significant difference was observed between drinkers and non-drinking/non-smoking subjects(P > 0.05).In the control group there were a higher MMA%( 17.27%,17.06%) lower DMA% (73.89%,72.29%) and lower SMR (81.48%,82.58% ) in those with smoking or drinking/smoking compared with the non-drinking and nonsmoking subjects( 11.52%,79.68% and 87.19%,respectively,all P < 0.05).No significant difference was observed between drinkers and the non-drinking/non-smoking subjects (all P > 0.05).ConclusionThe arsenic methylation capacity of people with drinking and smoking is poorer than that of non-drinking and non-smoking subjects after arsenic exposure.

9.Effects of acupuncture at different acupoints on behaviors in depression model rats.

OBJECTIVETo observe therapeutic effects of acupuncture at different acupoints on depression.
METHODSSeventy adult male rats with similar Open-Field score were randomly divided into a normal control group, a model group, a blank control group, a drug treatment group, an acupuncture group I ["Baihui" (GV 20), "Shenting" (GV 24)], II ]"Neiguan" (PC 6), "Sanyinjiao" (SP 6)], and III ("Baihui", "Shenting", "Neiguan" and 'Sanyinjiao"), 10 rats in each group. Separation feeding, long term unpredictability and medium stimulation stress techniques were applied to develop depression model rats. Changes of behaviors were investigated with Open-Field method and sucrose consumption trial.
RESULTSCompared with the normal control group, the score for the behavior and the consumption of sucrose reduced in the depression model group; compared with the blank control group, the score for the behavior and the consumption of sucrose increased in all the treatment groups during treatment. There was no significant difference among the acupuncture groups in the increase of behavior scores 21 days after treatment (P > 0.05), and there were significant differences in the increase of consumption of sucrose in the acupuncture group I and the increase of score for vertical movement in the acupuncture group III 14 days after treatment (P < 0.05, P < 0.01).
CONCLUSIONAcupuncture can change the behavior abnormality induced by separation feeding, long term unpredictability and medium degree stimulation depression model rats and different acupoint selection is a factor in acuounture treatment.

10.Liquid chromatography frontal analysis of the protein binding of glimepiride.

AIMTo study the protein binding of glimepiride.
METHODSAn HPLC-FA method is performed by using Pinkerton GFF II-S5-80 internal-surface reversed-phase silica support (150 mm x 4.6 mm ID, 5 microm) at pH 7.4 in a 67 mmol x L(-1) isotonic sodium phosphate buffer at 37 degree C. Other conditions included flow rate of 0.2 mL x min(-1), UV detection at wavelength 230 nm and injection volume 900 microL.
RESULTSNonlinear regression parameter estimation was used for the association constant measurement of glimepiride to both primary and secondary sites, which were 5.1 (micromol x L(-1)-1 and 1 for K1 and n1, and 0.017 (micromol x L(-1))-1 and 7 for K2 and n2, respectively.
CONCLUSIONThe method is shown to be suitable for investigation of protein binding of glimepiride.
