1.Research on the Stress Resistance of Saccharomyces cerevisiae Industrial Strains for Molasses

The stress resistance of Saccharomyces cerevisiae industrial strains for molasses to high-concentration ethanol,high temperature,high osmotic pressure,furfural toxicity,phenol toxicity,acetic acid toxicity and G418 toxicity were analyzed by the spot dilution growth assays in this paper. The results showed that the stress resistances among these industrial strains were obviously different. The strains AS2.1189 and AS2.1190 are more resistant to the tested stress factors than any others .The strain 396 is the most resistant to the acetic acid toxicity and G418 toxicity,and the strain 2610 is the most resistant to the high temperature.
2.Microvascular free muscle flap reconstruction of soft-tissue defects with bone exposure after the distal tibial fracture

Objective To present a method to treat soft tissue defects with bone exposure after the distal tihial fracture.Methods Twenty-six patients who had suffered from the distal tibial fracture complications leading to exposure of the bone or fixation material sought medical help in our department.The internal fixation material was removed and radical revision of dead and infected tissue was carried out in all cases.Five of the 26 cases were treated using a free microvascular muscle flap and the mesh skin graft by one stage.Twenty-one of them were re- paired with the same method by two-stage.Results Follow-ups were 10 to 24 months.The microvascular re- construction was successful in 22 patients.Two of the 26 cases had partial necroses of the skin graft,and the other two cases had delayed wound healing.All the fractures united.Conclusion Microvascular free muscle flap re- construction of the leg is regarded as a reliable method to treat legs with massive soft-tissue defects or defects with bone exposure and chronic infection or fistulation at the distal leg.
3.Construction of eIF-4E Adenovirus and Its Effect on Metastasis Ability of Human Breast Cancer Cell Line MCF-7

Objective: To reconstruct adenovirus vector of breast eukaryotic initiation factor 4E and to observe its effect on the metastasis ability of breast cancer cell line MCF-7. Methods: eIF-4E gene was constructed into adenovirus vector pAD-X by gene recombination technique, which was transformed into 293 packaged cell for high titer adenovirus. Real-time PCR was applied to detect eIF-4E gene expression. eIF-4E siRNA was applied and then transwell cabin assay was used to observe changes of invasion and motor ability of MCF-7 cells transfected with reconstruction adenovirus. Result:The finding of digestion was coincided with expected. eIF-4E gene over-expression was detected in transfected MCF-7 cells with real-time PCR. 4. Preparation of baicalin nasal gel and its permeation through nasal mucosa

Objective: To prepare the nasal gel of baicalin and to evaluate its permeation in vitro. Methods: The orthogonal design was carried out using Cabopol-936 as gel matrix, glycerine as solubilizer, and triethanolamine as pH value and viscosity adjuster. The permeating rate of baicalin from nasal gel was determined through pig nasal mucosa in vitro by transdermal diffusion method. Results: The optimum nasal gel was made by 0.5% Carbomer-936, 30% glycerine, and 2% baicalin, with pH value 6.85-7.00, and the average viscosity was 1850-2350 Pa·s. The permeation of baicalin through nasal mucosa was fitted with first-order kinetics, the regression equation was Y=0.2806 X+0.5643, r=0.997, and the permeating rate was 0.2806 μg/(mm2·h). Conclusion: The preparation of baicalin nasal gel is simple. Baicalin is almost permeated through nasal mucosa by simple diffusion.

6.Influlance of different drying methods on quality of Schisandrae Chinensis Fructus.

OBJECTIVETo study the influence of different drying methods on the quality of Schisandrae Chinensis Fructus and thus provide useful reference for its proper drying methods.
METHODSchisandrae Chinensis Fructus was processed by eight drying methods including vacuum freeze drying, natural drying in the shade, drying in the sun, oven drying and vacuum drying under different temperature. The contents of the functional ingredients includes chisandrin, gomisin D, gomisin J, schisandrol B, angeloylgomisin H, angeloylgomisin Q, gomisin G, schisantherin A, deoxyschisandrin, schisandrin B, schisandrin C, 5-HMF, total aids and total sugars. The main components change after drying were analyzed by HPLC, ultraviolet spectrophotometry and potentiometric titration. Principal component analysis (PCA) was carried out by SPSS software to evaluate the quality of different processed products from Schisandrae Chinensis Fructus.
RESULTAll these results are in accordance with the requirements of Chinese Pharmacopoeia published in 2010, the contents of schisandrin and total eleven lignans were the highest using vacuum drying, and 5-HMF were the lower, oven drying made little difference but with lower schisandrin and higher 5-HMF as the heat increased.
CONCLUSIONDifferent drying methods have significant influence on the quality of Schisandrae Chinensis Fructus. Oven drying under 5°C should be adopted to substitute drying in the sun according to the China Pharmacopoeia published in 2010 for Schisandrae Chinensis Fructus by comprehensive analysis of the cost, content and practicality.

7.A clinical observation of combined application of magnetic attachments and extracoronal attachments.

OBJECTIVETo explore the clinical effect of combined application of magnetic attachments and extracoronal attachments in prosthodontics.
METHODSTwenty-two cases of dentition defect with isolated residual root or residual crown were selected. All the cases accepted the restorative treatment combined magnetic attachments with extracoronal attachments. The clinical effect of dentures and the condition of abutment teeth were evaluated by chief complaint of patients and clinical examination. The follow-up time ranged from 2 years to 4 years.
RESULTSSatisfactory functional, stable and esthetic results of dentures were achieved for all the cases. And there was no abutment loosening and no secondary caries. Alveolar bone loss around abutment occurred in 1 case, gingivitis of abutments appeared in 5 cases. Chewing pain occurred in 1 case and unstable state of dentures appeared in 5 cases after the use of denture for 2 years. The dentures of above-mentioned cases were used normally after symptomatic treatment.
CONCLUSIONThe combined application of magnetic attachments and extracoronal attachments in prosthodontics is an effective treatment option for cases of dentition defect with isolated residual root or residual crown.

8.The blood supply of third intestinal artery to the free jejunal transplantation:an applied anatomical study

Objective To assess the effective length of jejunal graft when the 3~(rd) intestinal artery is u- tilized as vascular pedicle and afford a reliable theoretic base for clinical esophageal reconstruction.Methods In 32 formalin preserved and 21 fresh cadaver specimens,the diameter of 1st to 5th intestinal arteries and diameter of arterial arches are measured with linear calibre.Measure the length of jejunum that can be harves- ted as graft when the arches are extended.In the 21 fresh specimens,the 1st,2nd,4th and 5th intestinal ar- teries are ligated,acetic ester stained with red dye were injected into the lumen of 3rd intestinal artery via catheter.Extent of distribution of the arteries to the jejunum was observed.And then red ABS solution was in- jected into the 3rd intestinal artery to make into cast specimen.The blood supply distribution of jejunum through 3rd intestinal artery-arterial arch and communicating system were observed again.Results The di- ameter of the 3rd intestinal artery was the largest among the 1st to 5th intestinal arteries.The length of jejunum vascularized by 3rd intestinal artery can be as long as (142.2?62.3) (69.0~206.60cm) in acetic ester in- filtrated specimens.While in ABS east specimen,the average available extent of donor jejunum was(30.8?7.3) (23.0~37.3cm).Conclusion As observed by this applied anatomy study,the jejunum graft vascu- larized by 3rd intestinal artery alone has sufficient length to meet the need of esophageal reeonstrution.
9.Significance of the examination of path between eosinophils in peripheral blood and marrow stem cells for evaluating the effect of hormone therapy in patients with allergic rhinitis

Objective By means of the detection of the numbers of CD34+ cells and eosinophils(EOS),and the level of IL-5 in peripheral blood from normal controls and patients with allergic rhinitis pre-or post-treatment.the role of EOS-stem cells paths for treatment effect in allergic rhinitis(AR)was studied so as to find the convenient and quick indicators which could be used to evaluate the clinical therapeutic effect and adiust the methods of the hormone therapy.Methods There were 2 groups.(1)experimental group:44 patients,including 24 males and 20 females with a age range of 7 to 68 years old.The patients received the treatment of flutieasone spurt for four weeks.(2)Control group:30 cases,from normal health examination.The EOS numbers,CD34+ cell numbers and the IL-5 level were examined in control group as well as before and after therapy in experimental group.Results The IL-5 level and CD34+ numbers before thearpy in experimental group were(88.25±33.47)ng/L、(9.24±2.15)/105,significantly higher than that after thearpy and experimental group which were(44.34±16.32)ng/L,(6.31±1.83)/105 and(31.24±8.43)ng/L,(3.47±1.32)/105 respectively.The IL-5 level was positively correlated with the CD34+ number pre-and post treatment in experimental group(r=0.64,P<0.01;(r=0.61,P<0.01).The EOS number was positively correlated with the level of IL-5(r=0.64,P<0.01).Conclusions IL-5 and CD34+ cells can be regarded as indicators to evaluate the therapeutic effect.The EOS、CD34+ cells and the level of IL-5 in the peripheral blood are correlated with the pathogenesis of AR,suggesting that there is a related path between the local nasal tissue of AR patients and the marrow.
10.Synthesis and drug resistant reversal activities of taxane-like multi-drug resistant reversal agents.

AIMTo design and synthesize a series of new taxoids with a 5-O-sidechain, and to test the multi-drug resistant reversal activity of these compound on KB/V200 cells which is 180 times more resistant to vincristine.
METHODSUsing Sinenxan A as a common synthetic starting material, three different types of 5-O-sidechain molecules were synthesized through different route. For type I compounds, 14-acetoxy of Sinenxan A was selectively removed by hydrolysis, xanthation and reduction with tributyltin; A C-10-oxo group was introduced by PCC oxidation; 5-O-acetyl group was selectively removed by potassium tert-butoxide and finally the side chain was introduced by acylating with the corresponding acid. For type II compounds, 5-O-sidechain was introduced to the 5-deacetyl Sinenxan A which was obtained by selective hydrolysis with tBuOK. For type III compounds, 9-acetoxy group was introduced, then 5-OH was left free by thorough hydrolysis and reacetylation. Acylation at 5-position, the final product was obtained. Structure of the compounds have been confirmed by FABMS and 2DNMR. The activity of the compounds in vitro was tested on KB/V200 resistant cell line using MTT method.
RESULTSNine compounds showed resistant reversal activity and enhancing the cytotoxicity of vicristine against KB/V200 cells. Compounds I2, I3, I4 restored the sensitivity of KB/V200 towards vicristine to a level of IC50 at 1 x 10(-8) mol.L-1 which is better than the positive control Verapamil.
CONCLUSIONThe drug resistant reversal activity of taxane derivatives can be affected by substitution at different positions and the length of side chains of Sinenxan A. It is worthy to be further studied.
