The validity of fasting plasma glucose(FPG)combined with HbA_1c in diagnosing diabetes was assessed in patients with coronary heart disease.The results showed that the paired determination of FPG and HbA_1c helped to identify potentially diabetic subjects in patients with coronary heart disease.

Objective To detect the epidemiologic features of pediatric patients with fall from height injury in an effort to provide a reference for prevention and scientific treatment.Methods Data of pediatric patients with fall from height injury treated from January 2011 to December 2012 were collected in the form of questionnaires.Epidemiologic features and relationship of associated factors were measured.Results Pediatric fall injury occurred very often in warmer season,namely on April and May,and in a day,it was more likely to happen when parents slacked at noon and night.Stairs and balconies were the most common sites of falls and head was most vulnerable to the injury.Falling height was the primary factor to the severity of injury.Conclusion To strengthen the protection reform in key areas,the supervision in peak time for falls and the care to children of the migrant workers can reduce fall injury in a certain extent.

Objective To evaluate the effect of up-converting phosphor technology(UPT) in detection of plague antigen-antibody by receiver operating characteristic curve (ROC) method,and to provide a scientific basis for field application of UPT rapid detection technology in plague prevention and control.Methods Two hundred and twenty four serum samples were collected from Marmots and ground squirrels in the plague foci,Yersinia pestis antibody was detected by UPT,ELISA,Colloidal-gold Strips and IHA,respectively; 108 organs and bone marrow samples were collected,and Yersinia pestis antigens were detected by UPT,ELISA,PCR and RIHA,respectively.IHA was used as the gold standard for antibody test results,RIHA,PCR + Colloidal-gold Strips,PCR + ELISA were used as the gold standard for antigen test results.The results were evaluated using ROC method.Results Antibodies detection:the AUCs of UPT,ELISA and Colloidal-gold Strips were greater than 0.5.The difference between UPT and other methods was not statistically significant (z =1.204,P > 0.05).Antigen detection:the AUCs of UPT,ELISA,Colloidal-gold Strips and PCR were greater than 0.5.There was no statistical difference between UPT and other methods(z =0.866,P > 0.05).Conclusions UPT as a new technology works well in the detection of plague antigen-antibody.The technology is simple,fast,accurate,and suitable for on-site monitoring of plague,emergency treatment of sudden plague,and suitable for promotion.

Haizao Yuhu decoction (HYD) is a formula that has been used for approximately 500 years and famous for its efficiency in treating thyroid-related diseases in traditional Chinese medicine (TCM). HYD was first presented by Chen Shi-gong in a famous surgical monograph named Waike Zhengzong during the Ming Dynasty. We conducted the research to investigate the possible pharmacokinetic profile of different prescriptions of HYD in rats, in order to reveal the interactions of Haizao and Gancao drug pair with other herbs in HYD. Liquiritin, naringin, besperidin, peimine, peiminine liquiritigenin, glycyrrhizic acid, hergapten, nobiletin, osthole, glycyrrhetinic acid in blood samples were determined by UPLC-MS/MS. The result revealed tbat Haizao could enhance the peak concentration of glycyrrhizic acid. The other herbs in HYD may promote'the absorption of flavonoids in Gancao in normal rats, but inhibit the absorption of saponins and accelerate their metabolism. Gancao and Haizao drug pair could enhance the bioavailability of hesperidin, peimine, bergapten, nobiletin and osthole and prolong the elimination of peimine and naringin.

7.The effect of HPV16E7 DNA vaccine transdermal delivery with microneedle array.

OBJECTIVETo study the effects of DNA vaccine transdermal delivery with microneedle array.
METHODSThe pcDNA3.1-HPV16E7 recombinant vector acting as gene vaccine was established. The infiltration quantity of pcDNA3.1-HPV16E7 getting across the microchannels generated by microneedle arrays in vitro was observed. 30 BALB/c mice were divided into 3 groups (experimental group, in vain plasmid group, negative control). Each group had 10 mice. Then immunized BALB/c mice with a dose of 200 microg with microneedle array every two weeks. The control groups did the same as that as the study groups. Two weeks after the third immunization, the serum and lymphocytes were separated to detect the functions of humoral immunity with indirect immunofluorescence test, while, the functions of cellular immunity with lymphocyte transformation test was also detected.
RESULTSThe DNA vaccine could easily get across the microchannels generated by microneedle arrays in vitro. Moreover, the course was permanent and the whole infiltration quantity was comparatively high, reaching 0.73819 mg/cm2 at the 30th hour. And among immunized BALB/c mouse, DNA vaccine transdermal delivery with microneedle array could induce specific antibodies. Lymphocyte transformation test showed that there was significant difference for the lymphocyte transformation rate between experiment (the average of lymphocyte transformation rate was 47.25%) and control group (the average of lymphocyte transformation rate was 30.00%) (chi2 = 12.903, P < 0.001). Also, the difference was found between in vain plasmid group (the average of lymphocyte transformation rate was 43.00%) and negative control(chi2 = 7.292, P = 0.007). While, no difference was observed in the experimental group and in vain plasmid group (chi2 = 0.817, P = 0.366).
CONCLUSIONThe DNA vaccine combined administering with microneedle array might get across the microchannels generated by microneedle arrays in vitro and induce humoral and cellular immune response in vivo.

8.Prevalence of abnormal glucose regulation in patients with chronic ischemic heart failure.

OBJECTIVETo investigate the prevalence of diabetes mellitus (DM) and glucose abnormalities in patients with ischemic chronic heart failure (CHF).
METHODSA total of 1004 hospitalized eligible patients from 52 hospitals in 7 Chinese cities were included in this study.
RESULTSIn this survey, 420 out of 1004 patients had DM history (41.8%), 175 patients were newly diagnosed as DM (17.4%), 208 patients (20.7%) had impaired glucose tolerance (IGT). NYHA grade increases in proportion to severity of abnormal glucose metabolism [(r(s)) = 0.17, P = 0.001]. After adjustment of age and other factors, logistic regression analyses showed risk of suffering severe CHF symptoms (NYHA III/IV) increases with the severity of abnormal glucose metabolism: OR, 1.2, 95% CI: 0.7 - 1.7 in patients with IGT; 1.4, 95% CI: 0.9 - 2.1 in the newly diagnosed DM patients and 1.7, 95% CI: 1.2 - 2.4 in the DM history group, respectively.
CONCLUSIONSHigh prevalence of abnormal glucose metabolism was observed in patients with chronic ischemic hear failure and the severity of abnormal glucose metabolism was closely related to NYHA symptom grade.

Objective To observe the effects of agmatine on the expression of glial fibrillary acidic protein(GFAP)and the levels of IFN-γ and IL-10 in experimental allergic encephalomyelitis(EAE)in rats.Meth-ods EAE was induced in Wistar rats by immunization of spinal cords homogenate of guinea pigs and complete Freund's adjuvant.Rats gived with agmatine was intrapefitoneally injected with agmtine at dosage of 25 ,50,100 mg·kg~(-1),respectively,once daily for fourteen days after im-munization.The ethologic change is observed.and the number of GFAP in central nervous system(CNS)was examined with the method of immu-nohistochemistry and the levels of IFN-γ and IL-10 of the CNS in EAE were observed with the method of sandwich-enzyme-linked inmunosorbent assay.Resuts Compared with EAE model rats,agmatine treatment sig-nificanfly caused the decrease in morbidity and the improvement clinical manifestation of rats with EAE.The number of GFAP in and around the inflamatory demylinatve lession of CNS in EAE increased obviously(P<0.05),and the levels of IL-10 had significantly increased and the levels of IFN-γ had significantly decreased,compared with model group(P<0.05 or<0.0 1).Conclusion Agmatine could promote the expression of GFAP of central nervous system in EAE.regulate the ratio of Th1/Th2 in EAE,increased the levels of IL-10 and the levels of IFN-γ in the CNS of EAE.
10.Acetylated Modification of Polysaccharides from Stems and Leaves of Abelmoschus Manihot and Its Immunoregulatory Activity

OBJECTIVE Acetylated modification was applied to modify the structure of polysaccharide from stems and leav-es of Abelmoschus manihot,and the in vitro immunoregulatory activity of polysaccharides were evaluated,which was expected to provide the scientific evidence for the resource utilization of A.manihot disused parts.METHODS The crude polysaccha-ride(SLAMP)was obtained by water extraction and alcohol precipitation,and neutral polysaccharide(SLAMP-a)was further prepared by DEAE-52 ion exchange column.Acetic anhydride was used as acetylating agent to synthesize three acetylated de-rivatives (SLAMP-a1 ,SLAMP-a2 and SLAMP-a3).Their structures were preliminarily identified by chemical composition a-nalysis,pre-column derivatization HPLC method and IR spectrum.In addition,in vitro immunoregulatory activity of SLAMP-a and three acetylated derivatives was evaluated by splenocyte proliferation together with its effects on NO production of RAW264.7.RESULTS SLAMP-a showed little immunomodulatory activity and the total sugar content was 99.76%;Among the three derivatives,only SLAMP-a1 could significantly stimulate the proliferation of spleen cells and activate RAW264.7 to product NO.SLAMP-a1 mainly contained glucose with a few of mannose,galactose and arabinose,and the total sugar and DS was 82.50% and 0.62 respectively.CONCLUSION In vitro immunomodulatory activity of polysaccharide SLAMP-a from stems and leaves of A.manihot is significantly enhanced by acetylation and SLAMP-a1 possesses the potential to develop as immunoregulatory agents.The acetylation modification is an effective way for resource utilization of polysaccharide from the disused stems and leaves of A.manihot.
