1. Double modality fusion between CT and MRI for human head based on surface anatomic characters

Objective To explore the accuracy of double modality fusion between CT and MRI for human head, based on pair anatomical points registration. Methods CT and MRI were approached in 4 patients, digital imaging and communications in medicine (DICOM) files were loaded on 3D Slicer (version 4. 8. 1), the surface and brain were reconstructed automatically with volume rendering as reconstructed models, 10 surface characters were tested with localization error and would be used as fiducial points, 8 of the points would be chosen as pair-points for registration according the localizing error after 5 localizations, 5 registration groups based on 4, 5, 6, 7, 8 fiducial points were divided. Singular value decomposition (SVD) was employed for registration as a basic algorithm, and fiducial registration error (FRE) was used to evaluate the registration error. Results FRE values of double modality fusion were 2. 68-6. 54 mm. FRE values of 4, 5, 6, 7, 8 fiducial points were (5. 14± 0.97) mm, (4. 71 ±0. 64) mm, (4. 45±0. 59) mm, (4. 13± 0. 55)mm, (3. 54±0. 72) mm respectively. There was no statistical difference between neighbor groups, while in 4 and 8 points groups, the FRE values were different statistically. Conclusion Surface anatomic characters can be used for double modality fusion between CT and MRI; FRE values after CT-MRI double modality fusion are reduced with the increase of points.

3.Comparative imaging study for soft tissue changes in osteomyelitis and malignant bone tumor

Objective To investigate the value of the imaging findings of soft tissue abnormality in the differential diagnosis between osteomyelitis and malignant bone tumor.Methods The CT and MRI findings of soft tissue changes in 57 cases of osteomyelitis and 70 cases of malignant bone tumor were retrospectively defined,observed,recorded and statistically analyzed.Results In 57 cases of osteomyelitis, 54 cases were examined with CT,and soft tissue swelling was presented in 52 cases (degree Ⅰin 19 cases, degreeⅡin 16 cases,degree Ⅲin 17 cases).Abscess-like cysts in soft tissue occurred in 6 cases,masses in 5,air in 1,fat-fluid level in 1 and sinus tract in 1.Among 14 cases examined with MR imaging,soft tissue swelling was presented in all cases (degreeⅠin 2 cases,degree Ⅱin 6 cases and degree Ⅲin 6 cases). Abscess-like cysts appeared in 3 cases and showed high signal in diffusion weighted imaging, mass in Ⅰand fat-fluid level in 1.Among 54 cases examined with CT in 70 cases of malignant bone tumor, soft tissue swelling was presented in 44 cases (degreeⅠin 29 cases,degreeⅡin 12 cases,degreeⅢin 3 cases).Soft tissue masses appeared in 49 cases,bone shell and shell-like calcification in 16 cases and neoplastic bone and neoplastic calcified cartilage within soft tissue mass in 25 cases.Among 49 cases examined with MR imaging,soft tissue swelling was presented in 46 cases (degree Ⅰin 21 cases,degreeⅡin 17 cases and degree Ⅲin 8 cases),and soft tissue masses appeared in 43 cases.The degree of soft tissue swelling and the occurrence of abscess-like cyst,mass,bone shell or shell-liked calcification in the rim of mass,neoplastic bone or neoplastic calcified cartilage in masses showed significant difference(P

Objective To explore the best time point for the ipsilateral hepatocellular apoptosis and the contralateraI hepatic hypertrophy after selective portal vein embolization(SPVE)in rabbit.Methods In a randomized study design,forty rabbits were divided into 5 groups with 8 rabbits per-group,including one as the control and the other 4 were treated with SPVE during open surgery.The rabbits were killed postoperatively,in 3,7,14,21 days respectively after the embolization.The hepatic lobes volume,the ipsilateral hepatocellular necrosis rates and apoptosis index,and liver functions were determined as well. Results In the treatment groups,the average amount of the right liver volumes decreased from 46.4 cm~3 preoperatively to 46.0,44.4,42.0,39.7 cm~3 in groups of 3,7,14,21 days postoperatively;meanwhile,the left liver volumes increased from 54.0 cm~3 preoperatively to 54.5,56.3,61.7,63.9 cm~3 respectively during 3, 7,14,21 days after the procedures.The rates of future remaining live volumes(FRLV)increased from 53.8% preoperatively to 54.2%,55.9%,59.0%,61.0% at 3,7,14,21 days postoperatively.The apoptosis indexes of hepatocells from group A to E were 8.1%,12.2%,19.4%,20.1%,14.2% respectively.Conclusions SPVE leads to atrophy of the ipsilateral hepatic lobe and hypertrophy of contralateral lobe,indicating that hepatocytes undergone apoptosis,rather than necrosis.The time point is 7 to 14 days.
5.Study on the tongue manifestations for the blood-stasis and toxin syndrome in the stable patients of coronary heart disease.

OBJECTIVETo explore the tongue manifestations for the blood-stasis and toxin syndrome in the stable patients of coronary heart disease (CHD) according to the acute cardiovascular events (ACEs) in one-year follow-up which based on the pathogenesis hypothesis of "blood-stasis and toxin causing catastrophe".
METHODSTotally 254 stable CHD cases were enrolled after diagnosed by coronary angiography, their tongue appearances were recorded by the digital camera of uniform type, 29 cases with ACEs during one-year follow-up were assigned in ACEs group. The non-ACEs were matched in proportion of 2:1 according to the gender, age (±2.5 years), diabetes mellitus history and previous acute coronary syndrome hospitalization history in the non-ACEs group, and 54 cases were eligibly included. The differences of tongue appearance between the ACEs and non-ACEs group were compared.
RESULTSFifteen cases manifested with bluish tongue (including bluish-grounding or bluish purple tongue), among which 11 cases (37.9%) in the ACEs group and four cases (7.4%) in the non-ACEs group, and there was significant difference (P<0.002). Twenty six cases showed yellow tongue coating in the non-ACEs group, which was significantly higher than that in the ACEs group (48.1 vs. 10.3%, P=0.001). The tongue of sticky greasy coating was more frequently occurred in the non-ACEs group than that in the ACEs group (66.7% vs. 41.4%, P=0.026). The proportion of purplish-red sublingual vessel was higher in the ACEs group than that in the non-ACEs group (41.4% vs. 20.4%, P=0.041). Odd ratio (OR) analysis showed that the patients with bluish tongue, purplish-red sublingual vessel, dry-greasy or dirty greasy coating were more likely to experience ACEs during one-year follow-up (OR: 11.67, 95%CI: 3.34 year 3.34-40.81, P<0.001; OR: 2.76, 95%CI: 1.02 1.02-7.44, P<0.05; OR: 3.12, 95%, CI: 0.89 0.89-10.92, P=0.066).
CONCLUSIONSThe bluish tongue (including bluish-grounding or bluish purple tongue) and purplish-red sublingual vessel were potential tongue manifestations of blood-stasis and toxin. The tongue coating changing from sticky greasy to dry greasy or dirty greasy was also probably a tongue manifestation of "transforming toxin", which need demonstration by further study.

6.Expression and significance of HER-2/neu mRNA and RECK mRNA in breast carcinoma tissue.

OBJECTIVETo investigate the expression of HER-2/neu mRNA and RECK mRNA in breast carcinoma tissue and relationship with progression of breast carcinoma.
METHODS92 cases of breast carcinoma tissue and homologous paraneoplastic breast tissue were selected, expressive level of HER-2/neu mRNA and RECK mRNA were detected with RT-PCR, expressive difference of HER-2/neu mRNA and RECK mRNA were contrasted between breast carcinoma tissue and homologous paraneoplastic breast tissue, along with different TNM stage breast carcinoma tissue and differentiate breast carcinoma.
RESULTSThe level of HER-2/neu mRNA was higher and RECK mRNA was lower in breast carcinoma tissue than homologous paraneoplastic breast tissue, the expressive level of HER-2/neu mRNA and RECK mRNA was remarkably different along with different TNM stage breast carcinoma tissue and differentiate breast carcinoma, there was a negative correlation between expressive level of HER-2/neu mRNA and RECK mRNA in breast carcinoma tissue.
CONCLUSIONSExpression of HER-2/neu mRNA and RECK mRNA was abnormal in breast carcinoma tissue, HER-2/neu mRNA and RECK mRNA regulate each other and mutual participate occurrence and progression of breast carcinoma.

AIM To investigate the equilibrium solubilities,oil-water partition coefficients and in vitro skin permeation features of brucine and strychnine in total alkaloids from Strychni Semen.METHODS Saturated dissolution method was applied to determining the equilibrium solubilities of two constituents in ethanol (10%,20%,30%,60%,90%,anhydrous ethanol),trichloromethane,n-octanol and surfactants (0.5% tween,0.5% sodium deoxycholate,0.5% oleic acid).Shake-flask method was adopted in detecting their oil-water partition coefficients in PBS (pH 2.5,4.0,5.0,5.8,6.8,7.0,7.4,9.0).Modified Franz diffusion cell method was used for evaluating their in vitro skin permeation features in PBS,20% ethanol and anhydrous ethanol.RESULTS Both brucine and strychnine showed the highest equilibrium solubilities in trichloromethane and the lowest equilibrium solubilities in surfactants.The equilibrium solubility of strychnine was higher than that of brucine in ethanol (> 20%) or PBS (pH < 8.0),which reached the highest in 60% ethanol and pH 2.5 PBS,respectively.The similar oil-water partition coefficients of two constituents,proportional to pH value,reached the highest at pH9.0.And they exhibited the highest accumulated transdermal absorptivities in anhydrous ethanol and pH 9.0 PBS,respectively.CONCLUSION Solvent type has obvious effects on the equilibrium solubilities,oil-water partition coefficients and in vitro skin permeation features of both brucine and strychnine.This study can provide a reference for the bioavailability improvement of transdermal drug delivery and development of related preparations.

Objective To study the clinical value of serum cytokeratin 19 fragment (CYFRA21-1) as a biomarker in evaluating its prognosis,monitoring and follow-up of the postoperative patients with non- small cell lung cancer (NSCLC).Methods Serum CYFRA21-1 was determined by radioimmunoassay for 207 patients with NSCLC before and after surgical operation at Tongji Hospital,Shanghai.Relationship between serum CYFRA21-1 and the prognosis,recurrence and metastasis of lung cancer was analyzed retrospectively.Results The patients were followed-up for 37 months in average.Preoperative serum CYFRA21-1 was positive in 42.0% (87/207) of all the cases,48.8% (60/123) of squamous cell earcinama and 34.6% (27/78) of adenocarcinoma,with statistical significance (X~2=3.901,P

Chronic myelogenous leukemia (CML) is a clonal myeloproliferative disease of transformed hematopoietic progenitor cells. In order to investigate the role of sphingosine kinase-1 (SphK-1)/sphingosine 1-phosphate (S1P) signal pathway in the expression of CML cells, and to explore whether P210(bcr/abl) involved is activating SphK-1/S1P signal pathwey, the expressions of SphK-1 and S1P receptor mRNA in bcr/abl positive K562 cells and bcr/abl positive primary CML cells were detected by RT-PCR, the imatinib mesylate, the specific inhibitor of P210(bcr/abl) was employed to inhibit the P210(bcr/abl) tyrosine kinases of K562 cells and CML primary cells, and then the intracellular SphK-1 activity was assayed. The results indicated that after being cultured with 2.5 micromol/L imatinib mesylate for 0.5, 2, 6, 24 and 48 hours, the intensions of inhibiting SphK-1 activity were 0.007%, 38.9%, 34.6%, 28.1% and 76.1% resepectively. SphK-1 activity in CML cells also was reduced by 2.5 micromol/L imatinib mesylate (16.8% - 41.9% decrease). It is concluded that the CML cells express SphK-1 and different S1P receptor, and P210(bcr/abl) fusion protein in CML cells can activate SphK-1.

10.Construction and application of conditionally replicative adenovirus for selective cytotoxicity in CEA positive colorectal cancer cells

Objective To construct a new conditionally replicative adenovirus (CRAds) targeting carcinoembryonic antigen (CEA) positive colorectal cancer cells. Methods The DNA fragment of the CEA gene promoter was amplified through PCR and cloned into the vector carrying fusion reporter gene EGFP-Luc to construct expression plasmid pCEA-EGFPLuc. The constructed plasmid pCEA-EGFPLuc was transfected into CEA positive and negative cells by liposome. The activity of CEA gene promoter was evaluated by detecting the expression of EGFP and luciferase activity. The conditionally replicative adenovirus Ad.CEA-E1A/CMV-TK carrying suicide gene HSVtk was constructed, in which the E1A gene was controlled under CEA promoter. CEA positive(Lovo and SW620)and negative tumor cells(HeLa) were infected with Ad.CEA-E1A/ CMV-TK. The selective cytotoxicity of Ad.CEA-E1A/CMV-TK and the synergistic effect of the virus with GCV in CEA positive tumor cells were evaluated by the expression of E1A, cytopathic effect and cell survival rate. Results CEA promoter possesses a good specificity as well as high activity. The expression of E1A only presented in CEA positive tumor cells. After infection with Ad. CEA-E1A/CMV-TK, the cell survival rates of Lovo and SW620 were (36.72±2.49)% and (39.82±4.76)%, respectively, significantly lower than that of Hela[(87.44±2.76)%1 (P<0.01). When combined with GCV, Ad.CEA-E1A/CMV-TK had better oncolytic effect on Lovo and SW620 cells, with cell survival rates of (17.26±3.65) % and (23.93±5.40) %, respectively, significantly lower than those without GCV[(36.72±2.49) % and (39.82±4.76) %, respectively] (P<0.01). Conclusion Ad. CEA-E1A/CMV-TK under the control of CEA promoter has selective cytotoxic effect on CEA positive colorectal cancer cells, and the effect can be enhanced when combined with GCV.
