1. Rapid analysis of polar components in ophiocordyceps sinensis by conventional liquid chromatography system

Objective: To develop a rapid high-performance liquid chromatography coupled with diode array detection (HPLC-DAD) method for the simultaneous determination of six polar compounds in Ophiocordyceps sinensis. Methods: A poroshell SB Aq column (50 mm × 4.6 mm, 2.7 μm) and gradient elution were used; The detection wavelength of compounds was set at 260 nm. The chromatographic peaks of the six investigated compounds in sample were identified by comparing their retention times with reference compounds. Results: All calibration curves showed good linearity (r > 0.999) within the tested ranges. The intra- and inter-day precisions of the six analytes were less than 0.8% and 2.1%, respectively, and the recoveries of the six analytes were between 95% and 103%. The validated method was successfully applied to the determination of six polar compounds in O. sinensis samples. Conclusion: The poroshell SB Aq column is suitable for the rapid analysis of polar components in Chinese materia medica on conventional HPLC system and the developed HPLC method is also helpful to the quality control of O. sinensis. © 2014 Tianjin Press of Chinese Herbal Medicines.
2.Experience in Managing 73 Patients with Positive Margins after Cervical Conization

ObjectiveTo study the proper treatment of the patient with positive margins after cervical conization. MethodsA total of 220 cervical conizations were performed in the department of obstetrics and gynecology in Conception Hospital in Marseille in France from January 1, 1996 to June 30, 1998. Among them, we studied 73 patients, who had positive margins, by using cervical cytology examination and colposcopy in 3,6,12,18 and 24 months after cervical conization. Results7 cases of cancer invasive were treated with radiotherapy; 17 underwent a second conization or hysterectomy, among whom 8 had residual disease of positive margins near internal orifice of uterus (residual rate, 47. 06 % ); 40 remained follow- up; but 9 were lost.ConclusionCytology and colposcopy are effec- tive in the surveillance of patients with positive margins after cervical conization, and can give the indica- tion of the second conization.
3. Influence of nitric oxide on growth of microorganisms: A double-edged sword

Recently increasing functions of nitric oxide (NO) has been discovered. Researchers around the world have reported that NO plays an important role in the anti-microbial infection; however, more recent studies have found that NO also protect the growth of microorganisms, for example, it can facilitate drug resistance of bacteria and it can protect microorganisms from oxidative stress and pressure. This paper reviews both positive and negative roles of NO on microbial growth and the related mechanisms.
5.Content Determination of 17 Kinds of Amino Acids in Sargassum and Its Adulterants

OBJECTIVE:To establish the method for content determination of 17 kinds of amino acids in Sargassum and its adulterants,and to carry out cluster analysis ,so as to provide reference for quality control of Sargassum. METHODS :Totally of 18 batches of sample (S1-S6 as certified product ,S7-S18 as adulterants )were collected. After acid hydrolysis ,amino acids contents were detected by using automatic amino acid analyzer. The separation was performed on LCAK 06/Na sulfonic acid cation exchange resin column with mobile phase consisted of buffer-regeneration system (gradient elution )at the flow rate of 0.45 mL/min (elution pump )and 0.25 mL/min(derivative pump ). The detection wavelengths were set at 440 nm(proline)and 570 nm(other amino acids ),and the sample size volume was 50 μL. PASW Statistics 18.0 software was used ,and cluster analysis was conducted by using group connection method of cluster analysis with “square Euclidean distance ”as the measurement standard. RESULTS :17 kinds of amino acids were well separated without interference from blank sample. The linear relationship between mass concentration and peak area was good (all r were over 0.998),and the upper and lower limits of the linear range were 48.06 μg/L (cystine)and 1.501 μg/L(glycine),respectively;RSDs of precision ,reproducibility and stability tests were lower than 2%. The average recoveries were between 90.60%-101.56%(RSDs were 0.88%-2.15%,n=6). 17 kinds of amino acids were detected in Sargassum and its adulterants ,among which the contents of glutamic acid ,aspartic acid ,leucine,alanine,glycine and valine were relatively high . Results of cluster analysis showed that 18 batches of sample were clustered into 4 categories,i.e. S 1-S6 into one category;S7-S9 into one category ;S10-S12,S16-S18 into one category ;S13-S15 into one category ;which was consistent with the identification result of Sargassum and its adulterants . CONCLUSIONS :The method is simple , rapid, accurate and reproducible,and can be used for the quantitative analysis and identification of amino acids in Sargassum and adulterants.
6. Effects of hydroxyl morpholine resistance on K562 cells of chronic myelocytic leukemia

OBJECTIVE: To investigate the inhibitory activity, induced differentiation-inducing activity and apoptosis-inducingactivity of hydroxyl morpholine (QDML-01) on chronic myelocytic leukemia cells line K562. METHODS: The cell growth curve was drawn based on cell counting method. The IC50 value of QDML-01 and positive control medicine to K562 cells were evaluated by methyl thiazolyl tetrazolium (MTT) assay method. Double soft agar assay method was carried out to study the ability of cell proliferation to determine efficacy of phamacognosy. The pathomorphism was analyed by the Wright-Giemsa staining method. The mechanism of cell apoptosis from morphology and gene level were investigated, by AO-EB double-staining method and DNA breakage test. The effect of QDML-01 on K562 cells from the protein level was determined by Western-blot. RESULTS: The growth curves showed the K562 cells had strong cell vitality. They came into logarithmic phase on the third generation. The MTT assay results showed that the IC50 values of QDML-01 and imatinib to K562 cells were 5. 81 and 596.88 nmol ·L-1. Double soft agar colony formation test showed that clone formed at 21 d and the inhibitory rate of QDML-01 was 81.7%. It indicated that K562 cells were sensitive to QDML-01. Morphology test result showed that QDML-01 induced K562 cells to normal cells. The results of AO-EB double-staining method showed that QDML-01 induced the apoptosis of K562 cells. The study of DNA breakage test indicated that QDML-01 can induce the apoptosis of K562 cells to produce DNA banding with step-like. Western-blot analysis result suggested that QDML-01 can downregulated the expression of P210bcr/abl protein. CONCLUSION: QDML-01 has the inhibitory activity on chronic myelocytic leukemia cells line K562 by promoting the apoptosis of K562 cells and inducing differentiation to normal cells.
7.Software development of blood drug concentration monitoring system and its application in data analysis

OBJECTIVE:To provide a blood drug concentration monitoring system(BDCMS program) which could automatic process data analysis with multi-function.It was very easy to query and input data for clinical pharmacy operator.METHODS:BDCMS program was written with visual basic 5.0 and run in windows 95 system.The blood drug concentration data,which was determinated with flurescence polarization immunoassay(FPIA) among the 26 months at our hospital,was analyzed with DBCMS program.RESULTS:2026 samples were measured,of which 63.8% was within the effective blood concentration range,24.2% was under the effective blood concentration range,and 12.1% was above the effective blood concentration range.The proportion of normal value in each age group(0~14a,14~40a,40~65a,65~100a) was separately of 49.6%,71.8%,60.0%,52.0%.The times of normal value had significant difference by X2-test(P<0.05).CONCLUSION:DBCMS program was a very practical tool to carry out blood drug concentration monitoring.It was very useful to monitor blood drug concentration especially for children,old people and kidney transplant patient who had been operated in three months.
8.Xanthogranulonatous pyelonephritis: report of 5 cases.

Xanthogranulomatous pyelonephritis (XGP) is an unusual form of chronic pyelonephritis in which the renal parenchyma is destroyed and replaced by lipid-laden foamy macrophages. It usually affects middle-aged women with a history of recurrent urinary tract infection, diabetes, or kidney stones. The inflammatory process is usually diffuse and can extend beyond the kidney. The rare focal forms may simulate primary renal tumours. The preoperative imaging diagnosis may be difficult. We reported five cases of XGP, The findings of ours were recorded including kidney size, shape, contour, the echogenecity of the renal parenchyma, the internal echoes of the dilate collecting system, the presence of perinephric fluid accumulation and obstruction. One of the 5 cases was a male patient, and the other four were female, with a mean age of 53 years. He affected kidneys of the 5 cases swelled in different degrees, and one of them was found with line-like anechoic fluid. Among the 5 cases, one kidney appeared as diffusely reducing of the parenchyma echogenicity, multiple hypoechoic areas, disappearance of corticomedullary differentiation and multiple hyperecho with shadow. A round cystic anechoic lesion was found in one kidney, with internal punctate echo and peripheral fluid. Ultrasonographic finding of 1 case was extremely hypoechoic lesion on the left kidney, protruding from the outline of the kidney, with the partial renal capsule discontinuous, the less clear boundary, and a little blood flow in it. Ultrasonographic demonstration of 2 cases was mild dilatation of the collecting system with irregular wall thickening and internal hypoechogenicity, and 1 case was solid lesion with less clear boundary to the pelvic wall and a small amount of blood flow signal, the another 1 case was showed floccule without internal blood flow. Three cases were caused by chronic obstruction verified by operation, of which one was staghorn calculi, one was poorly differentiated squamous cell carcinoma in the middle part of the ureter, and one was inflammatory stricture of upper ureteral. Through analysis of the above five cases and review of related literature, we explored diagnoses and management of the patients with XGP. Xanthogranulomatous pyelonephritis (XGP) is a rare chronic variant of pyelonephritis characterized by destruction of the renal parenchyma. Combining ultrasonographic features of XGP with clinical recurrent urinary infection and chronic obstruction, XGP can be included in the differentiation. The diagnosis of XGP suspected by ultrasound can be clarified by CT, MRI, contrast-enhanced ultrasound.

9.Study on electroacupuncture treatment of depression by magnetic resonance imaging.

OBJECTIVETo explore the changes of metabolites in brain after treatment by analysis on 1H-MRS examination of the hippocampus and prefrontal lobe in the healthy volunteers and the depression patients.
METHODSSeventy-five cases of mild and moderate depression were randomly divided into groups A, B and C, 25 cases in each group. The group A was treated with oral administration of Prozac capsule, 20 mg/d; the group B by electroacupuncture for 30-40 min and the needle was retained for 1 h, once each day, with main points Baihui (GV 20), Yin-tang (GV 29) and adjuvant acupoints selected; the group C by combination of the treatment methods in the groups A and B. They were treated for 6 weeks. Use PROBE-J sequence at the MRI system on Single Voxel of ROI of each lateral of hippocampus and frontal lobe in the depression patients of the 3 groups. Compare the differences of N-acetylasp artate/creatine (NAA/Cr) and choline/creatine (Cho/Cr) between the healthy volunteers and the patients before and after treatment.
RESULTSBefore treatment, NAA/Cr in the bilateral hippocampus decreased in the 3 groups as compared with the control group, and after treatment, NAA/Cr in the bilateral hippocampus of the group B and in the right hippocampus of the group C increased compared with that before treatment (P < 0.05), and NAA/Cr in the left hippocampus of the group C significantly increased as compared with that before treatment (P < 0.01). Before treatment, Cho/Cr in the bilateral prefrontal lobe in the 3 groups increased as compared with that in the control group (P < 0.05), after treatment, Cho/Cr in the bilateral prefrontal lobes of the groups A, B and C significantly decreased as compared with that before treatment (P < 0.05, P < 0.01).
CONCLUSIONThere are differences in contents of metabolites in corresponding parts in bilateral frontal lobes and hippocampus between the depression patient and healthy person.

10.MR imaging of knee osteoarthritis and correlation of findings with reported patient pain.

To evaluate lesion detection of MRI in knee joint osteoarthritis in patients with symptoms of pain, the correlation between MRI findings and varying degrees of reported pain was assessed. Twenty-eight patients (31 knees) with osteoarthritis were recruited for this study. The degree of knee pain was assessed by VRS scores. The knees were evaluated by plain film radiograph utilizing Kellgren-Lawrence scores. Multiple MR sequences were performed on a 1.5T MR-system, including sagittal and coronal dual fast spin echo (TR/TE 3660/11/120 ms, slice thickness 5 mm), coronal spin echo T1-weighted (TR/TE 360/9 ms, slice thickness 5 mm), sagittal fat saturated 3D-spoiled gradient-recalled echo (TR/TE 50/6 ms; slice thickness 1.5 mm; flip angle 40 degrees ), and 3D steady-state free precession (TR/TE 6/2.2 ms; slice thickness 1.6 mm; flip angle 30 degrees ) pulse sequences for the purpose of detecting abnormities of cartilage, menisci, the anterior cruciate ligaments, bone marrow edema-like lesions, osteophytes, synovitis, and joint effusions. MR findings were compared with the degree of pain using Fisher exact test with P values less than 0.05 indicating a statistically significant difference. The results showed that, of the 31 knees evaluated, mild pain was reported in 11 and severe pain in the remainder. Kellgren-Lawrence scores of all 31 evaluated OA knees were as follows: grade 1 lesions (n=6), grade 2 lesions (n=14), grade 3 lesions (n=8), and grade 4 lesions (n=3). Articular cartilaginous defects were found in 37.1% of knees. Abnormalities of the menisci and anterior cruciate ligaments, bone marrow edema-like lesions, osteophytes, synovitis, and joint effusions were detected in 32.3%, 38.7%, 45.2%, 100%, 15.1% and 67.7% of knees, respectively. Of these variables, only the differences in prevalence of joint effusions were significantly different in the mild and severe pain groups (P=0.004). It is concluded that MRI evaluates the entire joint structure of the osteoarthritic knee, demonstrating abnormalities of the cartilage, menisci, and anterior cruciate ligaments as well as bone marrow edema-like lesions, osteophytes, synovitis, and joint effusions. The difference in pain grading between OA patients reporting mild and severe degrees of pain is related to the presence of joint effusion.
