1.Research progress on antitumor effects and mechanisms of phellinus.

Phellinus is a kind of rare medicinal fungus that has a variety of physiological activities include anti-cancer, anti-liver fibrosisa, antioxidant and so on. Phellinus contains polysaccharides, steroids, terpenoids, flavonoids, pyrone, furan, alkaloids and other substances. Polysaccharide extracts of phellinus showed obvious antitumor effect and has been a hot research field in recent years. It was also found other extracts of phellinus such as ethyl acetate extract exhibited anticancer activity. Thus, the antitumor effect of different extract, especially the anti-cancer mechanism were discussed in this review.

2.Construction of real-time polymerase chain reaction detection for infection-related cytokines of tree shrew.

Tree shrew is a novel and high-quality experimental animal model. In this study, the real-time polymerase chain reaction methods were established to detect infection-related cytokines interleukin-6 (IL-6), IL-8, IL-10, IL-17A, interferon-γ (IFN-γ) and housekeeping gene glyceraldehyde-phosphate dehydrogenase ( ) of tree shrew. The results indicated that the establised methods had good specificity. The high point of the linear range of these reagents reached 1 × 10 copies, and the low points ranged from 10 copies (IL-6, IL-17A), 100 copies (IL-10, ) to 1 000 copies (IL-8, IFN-γ). In this interval, the linear correlation coefficient of each reagent was greater than 0.99. The lowest detectable values of IL-6, IL-8, IL-10, IL-17A, IFN-γ and were 8, 8, 4, 8, 128 and 4 copies, respectively. The results showed that the established detection methods had good specificity, sensitivity and wide linear range. The methods were suitable for detection of multiple concentration range samples, and could be used for the subsequent studies of tree shrew cytokines.

3.Research on biological and genetic characteristics of human placenta mesenchymal stem cells cultured in vitro.

OBJECTIVETo investigate the biological characteristics and genetic features of human placenta mesenchymal stem cells (hPA-MSCs) cultured in vitro in order to assess its safety for clinical use.
METHODSThe shapes of the 1st, 3rd, 5th, 7th, 10th, 13th, 17th and 20th generation hPA-MSCs cultured in vitro using serum-free culture medium were observed. Their cell cycle, cell surface markers, and karyotype were analyzed, and relevant genes and cytokines were measured.
RESULTSThe shape of hPA-MSCs has remained as fusiform or short fusiform, and there was no significant change. About 93% of hPA-MSCs cells were in G0/G1 phase and remained stable. No obvious chromosomal translocation, loss or inversion was noted by karyotyping analysis. Cytokines expression level remained stable. Related gene expression level as a whole was on the decline, but the gene expression level of the first five generations showed very slight variations, with genetic characteristics remaining stable.
CONCLUSIONThe hPA-MSCs cultured in vitro with serum-free medium has retained stable in the first five generations.

4.Sperm Motility in Chronic Nonbacterial Prostatitis: Related to Antisperm Antibody and Seminal Cytokine and Seminal Plasma Composition.

Seminal inflammatory white blood cells in patients with chronic nonbacterial prostatitis may be related to the decrease of semen quality affecting fertility. So we evaluated the change of sperm motility, the levels of antisperm antibodies, the seminal levels of various cytokines and the change of seminal biochemical compositions that may affect sperm motility in the control group. We also examined chronic nonbacterial prostatitis with pyospermia to establish the role of seminal inflammatory white blood cells on alteration of sperm motility. The relation between various affecting factors and sperm motility were also evaluated. Titers of antisperm antibodies were low in patients with chronic nonbacterial prostatitis. Interleukin-1a and interleukin-8 of seminal inflammatory cytokines were present in increased quantities in patients with chronic nonbacterial prostatitis and interleukin-8 of these increases were correlated to sperm motility. Among the seminal composition, the level of zinc was decreased significantly which was not correlated to sperm motility. In conclusion, sperm motility was decreased and alterations in interleukin-8 of seminal cytokines were correlated with sperm motility in chronic nonbacterial prostatitis.

We examined changes in measures of motility after administration of cytokines to sperm in vitro. MATERIALS AND METHODS: Semen from 30 healthy men were obtained. With the swim-up procedure, actively motile sperm were isolated and incubated in Ham's F10 culture medium. Interleukin(IL)-1 alpha(10ng/mL) was added, and measures of sperm motility were examined ater 5 and 24 hours using the Hamilton-Thorne computer semen analysis system. These measures were expressed as a percentage of the values before incubation. RESULTS: The decreases of the average path velocity (VAP) in the IL-1 alpha-treated and IL-8treated specimens after 24 hours are statistically significant. None of the decrease in values for straight-line velocity (VSL) is statistically significant. There were statistically significant decreases in the curvilinear velocity (VCL)in IL-6-treated and TNF-1 alpha-treated specimens after 24 hours. None of the changes in the lateral head displacement (ALH)or the beat cross frequency (BCF)was statistically significant. CONCLUSION: The administration of cytokines decreases sperm motility in vitro. It is suggested that seminal cytokines may affect sperm motility in vivo.

Osteoarthritis (Osteoarthritis, OA) is a common clinical degenerative joint disease with increased incidence rate in recent years. Animal experiment is one of the important ways to explore pathogenesis and treatment of OA, while induced animal model is the most important part in animal experiment. Intra-articular injection of drugs is a classical method for establishing animal model of OA. Choose of animal should follows the principle of correlation, appropriateness and practicability, injections should perform in accordance with experimental purposes and subject, detections means and evaluation methods also should corresponding to experimental reality. The gold standard of OA animal model and intra-articular injections has not build, need further study.

8.Research advances in immunological pathogenesis of immunoglobulin A vasculitis.

Immunoglobulin A (IgA) vasculitis is the most common leukocytoclastic small-vessel vasculitis in children and mainly involves the small vessels in the skin, joints, digestive tract, and kidneys. Its pathogenesis is still unclear. Currently, it is believed that environmental factors can cause autoimmune dysfunction and lead to the deposition of IgA-containing immune complexes on the wall of arterioles on the basis of genetic factors. This article reviews the research advances in the role of immune factors in the pathogenesis of IgA vasculitis.

9.Effects of low-power wireless power transfer system on memory behavior and part of physiological properties of mice.

Wireless power transfer (WPT) is a new power transmission way, which can be widely used in electric vehicles and other fields. Its electromagnetic environment must be analyzed to ensure safe application. A low-power wireless power transfer system experimental platform was built, with 25 W receiving power and 47 kHz resonant frequency, which was used to carry out animal experiments. Treatment mice were exposed to environment of wireless power transfer system for 5 h a day and 6 days as one cycle. At the end of every cycle, learning memory behavior of mice were detected in T-shaped maze. The exposure experiment lasted for 12 weeks. Finally, immune parameters, sex hormones and part of organ physiological structure were detected. The results are as follows: as exposure time increased, memory behavior of mice did not change obviously with no statistical difference in sex hormone either ( > 0.05), the concentration of immune factors including tumor necrosis factor-α (TNF-α), interleukin 6 (IL-6) and interleukin-1 beta (IL-1β) significantly increased ( < 0.05), and the structure of some organs showed some changes. The experimental results show that the environment of the wireless power transfer system has no effect on the memory behavior of mice, and has some effect on physiological properties of mice.

10.Regulatory T cells and asthma.

Asthma is a chronic disease of airway inflammation due to excessive T helper cell type 2 (Th2) response. Present treatment based on inhalation of synthetic glucocorticoids can only control Th2-driven chronic eosinophilic inflammation, but cannot change the immune tolerance of the body to external allergens. Regulatory T cells (Tregs) are the main negative regulatory cells of the immune response. Tregs play a great role in regulating allergic, autoimmune, graft-versus-host responses, and other immune responses. In this review, we will discuss the classification and biological characteristics, the established immunomodulatory mechanisms, and the characteristics of induced differentiation of Tregs. We will also discuss the progress of Tregs in the field of asthma. We believe that further studies on the regulatory mechanisms of Tregs will provide better treatments and control strategies for asthma.
