1.Current status and countermeasures of application of mercury thermometer

Objective To investigate the current ~atus of mercury thermometer applied in China and the replacement of it by electric thermometer.Methods A systematic review of the literatures was undertaken,and the mercury pollution caused by mercury thermometer was reviewed,the measures taken by various countries to reduce the use of mercury thermometer were introduced.The difficulty in promoting electric thermometer in China was analyzed,and some measures were proposed accordingly.Results Most of the population including medical staffs had few awareness of the hazard of mercury thermometer to the environment,while the promotion of electric thermometer was obstructed due to the deficiencies in propaganda andpolicies.Conclusion It's suggested emphases be placed on propaganda and measures to replace mercury thermometer by electric one to decrease mercury pollution.
2.HPLC Fingerprint Analysis of Ganzhixiao Decoction

Objective To establish a fingerprint analysis method for Ganzhixiao Decoction. Methods RP-HPLC was applied with Phenomenex Synergi 4?Hydro-RP 80A as the chromatic column,methanol-0.1 %H3PO4 solution (gradient elution) as the mobile phase,detection wavelength at 280 nm,flow rate being 1.0 mL/min,and column temperature at 30 ℃. Results Twenty common peaks were presented in 10 batches of Ganzhixiao Decoction and the positions of 20 characteristic peaks were identified,including 5-hydroxymethyl furfural,chlorogeni C acid,Salvianolic acid B. Conclusion The method is reliable and accurate,and can provide a reference for the research of Ganzhixiao Decoction.
3.HPLC fingerprint analysis on Shuangbai Powder

AIM:To establish the method of fingerprint analysis on Shuangbai Powder(Radix et Rhizoma rhei,Cacumen platycladi,Cortex phellodendri amurensis,Herba lycopi and Herba menthae) to distinguish the characteristic fingerprint.METHODS:HPLC with ZORBAX Eclipse XDB-C18 was used,acetonitrile-0.1% H3PO4 solution(gradient elution) as a mobile phase and detection wavelength at 254 nm,flow rate was 1 mL/min,and column temperature was 40 ℃.RESULTS:Thirty-seven common peaks were separated from 10 batches of Shuangbai Powder.The characteristic peaks were the summation,22 peaks were from Radix et Rhizoma Rhei,6 peaks were from Cortex phellodendri Amurensis,4 peaks were from Cacumen Platycladi,3 peaks were from Herba Lycopi,and 2 peaks were from Herba Menthae,there was one new characteristic peak.CONCLUSION:Ten peaks gathered from Shangbai Powder consist of rhein,emodin,chrysophanol,aloe-emodin,physcion,gallic acid,berberine hydrochloride,palmatine;quercitrosid,and linarin.

5.Detection and genotype determination of beta-human papilloma virus in actinic keratosis lesions

Objective To detect beta-human papilloma virus (HPV) and determine its genotype in actinic keratosis (AK) lesions.Methods Tissue specimens were collected from the lesions of 39 patients with AK and normal skin of 40 healthy controls.A nested PCR was performed to detect alpha-HPV and beta-HPV DNA in these specimens.The genotype of beta-HPV was determined in beta-HPV DNA-positive specimens by a common PCR using specific primers targeting 12 HPV genotypes,including HPV 5,8,15,17,19,20,21,23,36,38,49 and 80.Results The detection rate of beta-HPV DNA was 84.6% (30/39) in the patients with AK,and 30.0% (12/40) in the healthy controls (x2 =6.76,P < 0.05),while no significant difference was observed in the detection rate of alpha-HPV DNA between the two groups (12.8% vs.7.5%,x2 =0.91,P > 0.05).HPV 38 was the predominant genotype of beta-HPV in these patients with a detection rate of 36% (12/33),followed by HPV36.The prevalence of all the 12 genotypes of HPV was consistently low in the healthy controls.Mixed HPV infections were observed in 10 AK lesions,but in none of the healthy controls.No statistical difference was noted in the positivity rate of beta-HPV among patients at different ages,of different genders,with different occupations or clinical courses (x2 =0.53,0.94,0.81,0.73,respectively,all P > 0.05).Conclusions Compared with healthy controls,the patients with AK showed a higher beta-HPV infection rate,with HPV38 as the predominant genotype.

AIM: To establish a method of fingerprint analysis on Cacumen Platycladi. METHODS: HPLC was adopted with ZORBAX Eclipse XDB-C18,acetonitrile-0. 1% H3PO4 solution (gradient alution) as a mobile phase and detection wavelength at 254 nm,flow rate was 1 mL/min,and colum temperature was at 40 ℃. RESULTS: Eleven common peaks were separated from 14 batches of Cacumen Platycladi. As compared with standard sample,quecitroside and quecitin were checked out. CONCLUSION: The method is reliable,accurate and can be used as a quality control method for Cacumen Platycladi.
8.Effects of Yupingfeng Powder and Guomin Decoction Combined with Zhisou Powder on Lung Injury Rats Caused by Imbalance of Th17/Treg Induced by PM2.5

ObjectiveTo observe the lung injury of rats caused by PM2.5 induced imbalance of TH17/Treg immune system and the intervention effect of two different TCM treatments.Methods Wistar male rats were randomly divided into normal group, model group, TCM-treated group1 and TCM-treated group 2. PM2.5-induced lung injury model was established by airway instillation. Model group was given normal saline for gavage. TCM- treated group 1 and 2 were given Yupingfeng Powder and Guomin Decoction combined with Zhisou Powder for gavage. The pathological changes of bronchial and lung tissues, the contents of IL-8, IL-10, IL-17, NE, and MUC5AC in serum and BALF were compared, and the expressions of Foxp3 and IL-17 in lung tissue of each group were analyzed.Results Compared with normal group, the contents of IL-8, IL-17, NE and MUC5AC in serum and BALF of model group increased significantly, while IL-10 decreased significantly (P<0.05,P<0.01); the expression of IL-17 increased significantly and the expression of Foxp3 decreased significantly in lung tissue (P<0.01). Compared with model group, the contents of IL-8, IL-17, and NE decreased in TCM-treated group 1 and 2, while the content of BALF IL-10 increased significantly (P<0.05). The content of IL-10 in serum increased significantly in TCM-treated group 2 (P<0.05); the protein expression of IL-17 of lung issue decreased significantly, and the protein expression of Foxp3 increased significantly (P<0.01). The pathological changes were improved significantly.Conclusion PM2.5 can induce lung injury caused by the imbalance of TH17/Treg. Both two treatments can significantly improve the lung injury induced by PM2.5 and the imbalance of TH17/Tregs immune system.
9.Analysis of clinicopthologic features and prognosis of 4 cases primary gastric anaplastic large cell lymphomas

Objective To evaluate clinicopathologic features and prognosis of primary gastric anaplastic large cell lymphomas (ALCL).Methods Clinical data and parafiin blocks of 4 patients diagnosed with primary gastric ALCL were obtained. The diagnosis of all cases was based on the criteria of WHO classification of hematolymphoid neoplasm.Furthermore,chromosomal rearrangement involving ALK gene was detected by interphase fluorescence in situ hybridization (FISH) and Epstein-Barr virus (EBV) status was determined by in situ hybridization(ISH) for EBV-encoded small RNAs (EBERs).Results The patients (3 males and 1 female) were from 27 to 87 years old, with a median age of 58.5 years. All the four cases presented with a solitary ulcerative mass in stomach. Morphologically, the normal architecture of gastric wall was effaced by the diffuse infiltration of tumor cells in which the characteristic hallmark cells were easily identified.The tumor cells of all cases showed a consistently strong expression of LCA and CD30,and CD3e was expressed in 3 of the 4 cases.Both ALK expression and ALK gene rearrangements were negative in all cases.Two cases underwent total or partial gastrectomy followed by CHOP chemotherapy. Another one patient was treated with chemotherapy and autologous stem cell transplantation. None of these 3 patients developed a relapse or progression till the last follow-up on Nov 30,2011. While the rest one patient refused to take any treatment and died 20 months after diagnosis. Conclusions Primary gastric ALCL is very rare and usually ALK negative. Its pathologic features as well as the clinical outcome are quite similar to the ALK negative ALCL from other sites, except the more frequently positive CD3e Early diagnosis and proper therapy are of great significance to the prognosis.
10.The analysis of polymorphism on PINK1 gene of exon 5 in 50 patients with Parkinson disease

Objective To study polymorphism of phosphatase and tensinhomologue (PTEN)-induced kinase 1(PINK1) gene of exon 5 in Parkinson patients who located in Guangxi ,and its relationship with Parkinson disease (PD) .Methods PCR ,single strand conformation polymorphism (SSCP) and sequencing analyze were conducted to analyze the PINK 1 gene′s exon 5 polymorphism in 28 cases of early-onset PD patients ,22 cases of late-onset PD patients(early-onset PD patients + late-onset PD patients = PD group) and 55 of control group .Results The intronic intervening sequence 5-5G-A (IVS5-5G-A ) polymorphism and G12164A polymer-phism which were located on PINK1 gene of exon 5 were chain relation .The G/A ,A /A genotype frequency was significantly higher in PD group (42 .0% ) than that in control group (23 .6% ) (χ2 = 4 .034 ,P= 0 .045) .The frequency was also significantly higher in late-onset PD patients (45 .5% )than that of 38 cases who were older than 50 years old in control group(21 .1% )(χ2 = 3 .951 ,P=0 .047) .There were no significant differences in alleles .Conclusion This research suggests that chain relation polymerphism at IVS5-5G-A and G12164A in PINK1 gene may be a susceptible factor for PD patients in Guangxi .
