1.Detection and clinical implication of minimal residual cancer cells in patients with solid tumors

After surgical removal of a primary tumor the minimal residual cancer cells (MRCC) and metastases derived thereof are the actual targets for all theraputic approaches. Due to the great sensitivity and PCR-based detection systems, the molecular characterization of MRCC can provide information about their metastatic potential, availability of drug targets, drug sensitivity and development of therapy resistance, which will close the analytical gap between primary disease and the detection of metastases by conventional methods such as imaging procedures, and this will help therapy selection, monitoring of the treatment effects and predicting of the prognosis. Patients will benefit from a individualized therapy in the end.
2.Bilateral thyroid carcinoma: a case report.

We describe a case of bilateral thyroid carcinoma in a 48-year-old woman. She was admitted to our hospital as a case of cervical nodular goiter. Multifocality of the thyroid nodules were evaluated by preoperative ultrasonography. The patient received bilateral thyroid lobe total resection and bilateral IV lymph node dissection. Medullary thyroid cancer was confirmed by intraoperative frozen pathology in the right lobe of thyroid gland and papillary thyroid microcarcinoma in the left lobe of thyroid gland. No tumor recurrence and metastasis were found after 3-months follow-up.

3.The influence of acupuncture on blastocyst nidation when Cx43 expression at the acupoints is silenced

Objective To investigate the influence of acupuncture on promoting rat blastocyst nidation with Cx43 expression at the acupoints silenced, and to relate Cx43 with acupuncture's effects. Methods Ma-ted female rats were randomly divided into five groups: normal control, model, acupuncture, interference-control and interference. Mifepristone was used to induce a model of nidation disorder, and RNA interference technology was used to silence the expression of Cx43 at acupoints. P-Cx43-shRNA (1) was injected locally into the acu-points of rats in the interference group, and P-con-shRNA was injected in the interference-control group. Acu-puncture was administered to the acupuncture, interference-control and interference groups for 7 days. On the 8th day of pregnancy, the quality of blastocysts, pregnancy rates, and the average amount of blastocysts in each group were observed. Results ①The mRNA and protein level of Cx43 at the zusanli (ST36) acupoints in the interference group were significantly lower than in the acupuncture group ( P<0.05, P<0.01). There was no significant difference between the interference-control group and the acupuncture group. ② Compared with the normal control group, the quality of blastocysts in the model and interference groups was bad, and the amount was less. Their size was comparatively small, they were inhomogeneous, poorly distributed and had poor blood supply. The results improved in the acupuncture group and the interference-control group compared with the model group. ③The pregnancy rates in the normal control, model, acupuncture, interference-control and inter-ference groups were 100% , 50% , 75% , 75% , and 50% respectively, but these differences were not statisti-cally significant. ④The average amount of blastocysts in the model and interference groups was significantly less than in the normal group (P≤0.01). Compared with the acupuncture group, the average amount of blastocysts in the interference group was lower (P≤0.05). Conclusion Acupuncture can improve the quality and quan-tity of blastocysts in a rat model of nidation disorder. Local injection of Cx43 shRNA expression vector can si-lence the expression of Cx43 in acupoints and markedly influence acupuncture's effect. This demonstrated that Cx43 was involved in acupuncture's effects.
4.Making suggestions of nursing postgraduates' training mode reform through the state of their core competence

Objective To make suggestions of reform of nursing postgraduates' training mode according to the state of their core competence. Methods 297 nursing postgraduates were investigated using the assessment form of nursing postgraduate's core competence, the questionnaire of training mode and general conditions. Results Nursing postgraduate's core competence were not good, the four lowest scored ability respectively were English ability, scientific research ability, teaching ability, grasping ability of professional knowledge and skill. Conclusions In order to promote the teaching quality, we should reform the training mode from the following aspects: to make clear training target; to make clear professional direction, formulate and carry out training program effectively; to increase the courses related with profession; to reinforce the study of public English and professional English ; to improve the ability of scientific research; to strengthen and perfect the teaching practice; to consolidate the basic knowledge,the basic theory and the basic skill of nursing; to build a unified and normal evaluation system.
5.Quality Standard for Infantile Bronchitis Traditional Chinese Medicine Lotion

Objective:To establish the quality standard for infantile bronchitis traditional Chinese medicine lotion. Methods:TLC was used for the qualitative identification of Ephedra herba, Paeonia lactiflora Pall. , Poria cocos, Fructus aurantii and Epimedium brev-icornu Maxim. . HPLC was used for the content determination of ephedrine hydrochloride, pseudoephedrine hydrochloride and paeoni-florin. The chromatography conditions for ephedrine hydrochloride and pseudoephedrine hydrochloride were as follows: an AElichrom Pdar-Phenyl Polyphenyl ether-bonded phenyl-bonded silica column (250 mm × 4. 6 mm, 5μm) was used, the mobile phase was aceto-nitrile-0. 2% phosphoric acid solution (1 :99), the flow rate was 1. 0ml·min-1, the detection wavelength was 210 nm, the column temperature was 30℃ and the injection volume was 10 μl. The chromatography conditions for paeoniflorin were as follows:an Inertsil ODS-3 C18 column (250 mm × 4. 6 mm, 5 μm) was used, the mobile phase was acetonitrile-0. 1% phosphoric acid solution (15:85), the flow rate was 1. 0 ml·min-1, the detection wavelength was 230 nm, the column temperature was 30℃ and the injection vol-ume was 10 μl. Results:The TLC results of Ephedra herba, Paeonia lactiflora Pall. , Poria cocos, Fructus aurantii and Epimedium brevicornu Maxim. showed clear spots with good resolution. Ephedrine hydrochloride, pseudoephedrine hydrochloride and paeoniflorin had a good linear relationship within the range of 10.11-101.10 μg·ml-1(r=0.9996), 10.08-100.80 μg·ml-1(r=0.9991) and 20.50-102.5 μg·ml-1(r =0.9996), respectively. The average recovery was 98.80% (RSD =1.87%, n =6), 98.77%(RSD=1. 72%, n=6) and 99. 57% (RSD=1. 56%, n=6), respectively. Conclusion: The established quality standard can be used for the quality control of infantile bronchitis traditional Chinese medicine lotion.
6.Expression of phosphorylated cell cycle related proteins in developing and aging rat hippocampus

AIM:To observe the expression of phosphorylated cell cycle related proteins cyclin-dependent kinase(CDK) and the retinoblastoma protein(Rb) in the developing and aging rat hippocampus.METHODS:The immunofluorescent staining for anti-phospho-CDK2,anti-phospho-CDC2,anti-phospho-Rb and anti-NeuN antibodies was used to determine their expressions and distributions in Wistar rat hippocampus at different ages.The expressions of phosphorylated cell cycle related proteins were also assessed by Western blotting analysis.RESULTS:(1) The number of neurons of CA1 area in hippocampus decreased during the period from 1 day to 15 months.(2) The positive expressions of phospho-CDK2 and phospho-Rb increased with aging.(3) No apparent change in the expression of phospho-CDC2 in the hippocampus of all the five groups was observed.CONCLUSION:The expressions of phospho-CDK2 and phospho-Rb suggest that differentiated neuron can re-enter the cell cycle and more neurons enter the cell cycle in the aged rat brain.Re-entering the cell cycle of differential neurons probably results in apoptosis.
7.Effects of Lead on Vitality, Differentiation, Special Markers and Morphological Changes of Osteoblasts in Newborn Rats

Objective To investigate the effects of lead on the vitality, differentiation, special functional markers and morphological changes of the osteoblasts in newborn rats, which helps to clarify the toxicity mechanism of lead on the skeleton development in children. Methods The osteoblasts were isolated from the calvariae in newborn Wistar rats and cultured in DMEM medium. Lead acetate at different concentration (0, 0.1, 0.5, 1, 5, 10, 50, 100 ?mol/L) was added for 24 h, 48 h, 72 h. The vitality of the osteoblasts was measured by MTT methods; The activity of alkaline phosphatase(ALP) was measured by pNPP method. The protein content of the osteoblasts was determined by Coomassie brilliant blue method. The levels of osteocalcin in both secreted (in medium) and cellular of osteoblasts were determined by radiate immunochemistry methods. The optical structure was determined by LM. Results The vitality decreased significantly when the dose was at more than 50 ?mol/L, especially after 72 h. The activity of ALP decreased significantly when more than 0.5 ?mol/L, whereas the levels of secreted osteocalcin decreased significantly when at 0.1 ?mol/L. It appeared lead had some toxicity on the shape and number on the osteoblasts. Conclusion Lead acetate has adverse effect, at higher doses, on the vitality, protein content, ALP, osteocalcin and optical structure, maybe it is one of the mechanisms of lead influencing the development of the osteoblasts even the skeleton.
8.Study on the Mechanism of Butylphthalide Delaying Renal Fibrosis of Mice Model with Unilateral Ureteral Obstruction

Objective To study on the possible mechanism of butylphthalide delaying renal fibrosis of mice with obstructive nephropathy. Methods Totally 72 male CD-1 mice of clean grade were selected and randomly assigned into 4 groups: sham operation group (Sham), model group (UUO), control group (ACEI) and treatment group (NBP). The mice in the control group (ACEI) and the treatment group (NBP) were given benazepril and butylphthalide by gavage, and the mice in sham operation group (Sham) and model group (UUO) were given normal saline by gavage. Six mice were sacrificed at the third, 7th, 14th day, respectively. The obstructive renal tissue was selected for immunohistochemical staining and western blot. Results (1)With the longer time of ureteral obstruction, the expression of Nrf-2 was gradually strengthened in time-dependent manner;(2)Compared with the model group, the levels of Nrf-2 and γ-GCS in butylphthalide group were significantly increased, and the expression of type Ⅰ collagen was decreased significantly (P < 0.05). The expression of Nrf-2 and γ-GCS in each time points was stronger than that in the benazepril group (P < 0.05). Correlation analysis showed that: there was a positive correlation between Nrf-2 and γ-GCS (r = 0.930) and a negative correlation between Nrf-2 and the expression level of type Ⅰ collagen(r = -0.859). Conclusion Butylphthalide can relieve renal interstitial injury caused by oxidative stress and delay the progress of renal interstitial fibrosis by activation of Nrf-2 pathway and up-regulated expression ofγ-GCS.
9.Comparison of accuracy of Marsh model versus Schnider model for propofol target-controlled infu-sion system

Objective To compare the accuracy of Marsh model and Schnider model for propofol target?controlled infusion ( TCI) system. Methods Eighty patients, aged 20-60 yr, of American Society of Anesthesiologists physical status ⅠorⅡ, with body mass index of 17?5-28?0 kg∕m2 , scheduled for e?lective gynecological operation under general anesthesia, were equally and randomly divided into either Marsh model group ( group M) or Schnider model group ( group S) using a random number table. The target plasma concentration was set at 3 μg∕ml in both groups. During TCI and at different time points after the end of TCI, the blood samples were collected for determination of blood propofol concentrations by high per?formance liquid chromatography with fluorescence detector. The difference between measured and predicted concentrations (△C) at each time point was calculated. The median performance error ( MDPE) , median absolute performance error ( MDAPE) , and wobble of propofol TCI system were calculated in each group. Results In M and S groups, the MDPE was 9. 90% and 14?00%, respectively; the MDAPE was 11?43% and 14?49%, respectively;the wobble was 7?77% and 7?79%, respectively. There was no sig?nificant difference in △C at each time point during TCI between group M and group S (P>0?05). After TCI was stopped, △C at each time point was significantly lower in group M than in group S ( P<0?05) . Conclusion Marsh model provides higher accuracy than Schnider model for propofol TCI system in the pa?tients undergoing gynecological operation.
10.The effects of different doses of butylphthalide on Nrf-2 expression in mouse model with unilateral ureteral obstruction

Objective To explore the impact of different doses of butylphthalide on renal interstitial fibrosis in mice with obstructive nephropathy, and to discuss the correlation between Nrf-2 expression level and renal interstitial fibrosis. Methods Totally 72 male CD-1 mice of clean grade were selected and randomly assigned into 4 groups:sham operation group (Sham), model group (UUO), low dose of butylphthalide group (NBPL) and high dose of butylphthalide group (NBPH). In addition to the control group, UUO model was established in other three groups. NBPL group was given NBP 150 mg/(kg· d) by gavage since postoperative day 1,NBPH group was given NBP 220 mg/(kg·d) by gavage. Sham group and UUO group were given equal volume of saline by gavage. Six mice were sacrificed at the third, 7th, 14th day, respectively. The obstructive renal tissue was selected for immunohistochemical staining and Western blot assay. The expression levels of Nrf- 2 and type Ⅰ collagen were detected by Western blot assay. Results The IOD value of type Ⅰ collagen was increased with time in UUO group, and which showed a gradual decreasing trend in Sham group, NBPL group and NBPH group (P<0.05). The IOD values at different time points were significantly higher in UUO group, NBPL group and NBPH group compared to those of Sham group. The IOD values were significantly decreased in NBPL and NBPH groups than that of UUO group, and IOD value was lower in NBPH group than that of NBPL group (P<0.05). Western blot assay showed that IOD values of Nrf-2 and type Ⅰ collagen at 7 d and 14 d were increased in UUO group, NBPL group and NBPH group compared with those of Sham group. The IOD values of Nrf-2 protein were increased, and IOD values of type Ⅰcollagen were decreased, in NBPL group and NBPH group than those of UUO group. The IOD value of Nrf-2 protein was decreased in NBPL group than that of NBPH group, but the IOD value of type Ⅰcollagen was increased in NBPL group than that of NBPH group (P<0.05). Conclusion NBP can improve renal interstitial fibrosis in UUO mice, which may be related with the increased expression of Nrf-2 and the down-regulated expression of type I collagen.
