1.Construction of the Antibody Library and Selection of the Antibodies Specifically Binding to the Prostate Carcinoma Cells

Objective: To obtain phage-displayed ScFv library directly against prostate carcinoma cells, and select antibodies binding to prostate carcinoma cells specifically, so as to lay a foundation for developing diagnostic agents and clinical therapies of prostate carcinoma. Methods: Balb/c mice were immunized i. p . with purified membrane protein mixture of prostate carcinoma cells PC3, DU145. mRNA was isolated from the spleens of immunized mice, heavy and light chain genes ( VH and VL) of antibody were amplified separately by RT-PCR and assembled into ScFv gene with a specially constructed linker DNA. , the ScFv gene was ligated into the phagemid vector pCANTAB 5E and the ligated sample was transformed into competent E. coli TG1. The transformed cells were infected with M13K07 helper phage to yield recombinant phage. After five rounds of panning with PC3 cells, the positive clones were selected with the ELISA from the enriched phages. Results: A ScFv library of 3. 5 ? 106 was obtained and one phage-ScFv which can bind specifically PC3 cells was found. Conclusions: A prostate carcinoma specific antibody was identified , which paves a way for study of prostate carcinoma.
2.The relationship between the peripheral lung cancer and the bronchi,pulmonary artery and vein:a muitislice helical CT observation

0bjective To investigate the relationships between the peripheral lung cancer and pulmonary vessels or bronchi by 16-row muhislice computed tomography(MSCT)and analyze the related factors.Methods Fifty-four patients with peripheral lung cancer confirmed pathologically underwent contrast-erdaanced MSC TI Multiplanar reformation(MPR)and maximum intensity projection(MIP)in all patients were used to demonstrate the relationships between the peripheral lung cancer and pulmonary vessels,bronchi.The relationships were categorized five types:Type 1,erupted at the edge of nodule. Type 2,erupted at the center of nodule.Type 3,penetrated through the nodule.Type 4,contacting the nodule but stretched or encased.Type 5,contacting the nodule but smoothly compressed.The pathology type,stage,size,density and location of the peripheral lung cancer were recorded and the relationships with five types were evaluated by using Chi-square test and correlation analysis.Results (1)Tumor-bronchi relationship:type 1(33,61.1%)was more often seen in≥2.0 cm and solid lesions with stage Ⅱ-Ⅳ.while Type 2(14,25.9%)was often seen in<2.0 am and part-solid or non-solid lesions with stage Ⅰ.(2)Tumor-PA relationship:Type 1 was more often seen in≥2.0 am and solid lesions with stage Ⅱ-Ⅳ.while Type 2 was often seen in part-solid or non-solid lesions with stage Ⅰ.(3)Tumor-PV relationship:type 4 was the most common type(29,53.7%).Type 2(13,24.1%)was more often seen in part-solid or non-solid lesions.(4)Tumor-bronchi relationship and tumor-PA relationship had a positive correlation(r=0.5265,P<0.01).Conclusions MSCT can demonstrate the relations between the peripheral lung cancer and bronchi.PA and PV.It is useful for the differential diagnosis and prognosis evaluation of the lung csncer.
3.Correlation between serum thymosin α1 and cardiac function in patients with acute anterior wall ST-segment elevation myocardial infarction

Objective To explore the correlation between serum thymosin α1 level and left ventricular ejection fraction(LVEF)in patients with acute anterior wall ST-segment elevation myocardial infarction(STEMI)after receiving percutaneous coronary intervention(PCI).Methods A total of 74 patients with acute anterior wall STEMI(acute anterior wall STEMI group)and 38 patients having no myocardial infarction(control group),who were admitted to the Shanghai Sixth People's Hospital of China from December 2019 to February 2022,were enrolled in this study.According to the LVEF value after the recanalization of anterior descending coronary artery with PCI during hospitalization period,the patients of acute anterior wall STEMI group were divided into LVEF<50%subgroup(n=33)and LVEF≥50%subgroup(n=41).Serum thymosin α1 level was determined by enzyme linked immunosorbent assay(ELISA),the results were compared between the groups.Logistic regression analysis was used to analyze the correlation between thymosin α1 level and LVEF.The receiver operating characteristic(ROC)curve of serum thymosin α1 level for predicting cardiac function in patients with acute anterior wall STEMI after receiving PCI was drawn.Results The serum thymosin α1 level in LVEF≥50%subgroup was significantly higher than that in the LVEF<50%subgroup(P=0.032).During the post-PCI hospitalization period,the serum thymosin α1 level was positively correlated with LVEF.Logistic regression analysis revealed that serum thymosin α1 level was an independent predictor for LVEF<50%in patients with acute anterior wall STEMI after receiving PCI.The area under ROC of serum thymosin α1 level for predicting LVEF≥50%in patients with acute anterior wall STEMI during hospitalization was 0.644(P=0.034).The area under ROC of serum thymosin α1 level combined with peak troponin I level and with peak NT-proBNP level for predicting LVEF<50%in patients with acute anterior wall STEMI during hospitalization was 0.780(P<0.01)and 0.702(P=0.003)respectively.When taking the median serum thymosin α1 level as the cut-off value,the proportion of LVEF≥50%patients was higher among the patients having the post-PCI serum thymosin α1 level>2,890 ng/L.Conclusion In patients with acute anterior wall STEMI,the serum thymosin α1 level is closely related to the LVEF value during the post-PCI hospitalization period,it is an independent predictor for cardiac function improvement after PCI.It is expected that the serum thymosin α1 level may become a new indicator for predicting the improvement of cardiac function in patients with STEMI after recanalization of anterior descending coronary artery with PCI.
