1.Rapid identification of Arg778Leu gene mutation in Wilson's disease by 4-primer ARMS-PCR

Objective The aim of this study was to establish a new method for genotyping ATP7B Arg778Leu gene mutation that does not require RFLP PCR or sequencing.Method 4-primer amplification refractory mutation system (ARMS)-PCR was performed to screen the Arg778Leu mutation in 47 unrelated Wilson's disease (WD) patients and 30 unrelated healthy controls.Direct sequencing was used to confirm the specific amplification products.Results PCR products were visualized on agarose gel electrophoresis.Among the 47 WD patients,4 were homozygous and 14 were heterozygous for this mutation.The total mutation rate was 38.3% (18/47).The results of direct sequencing completely consisted with the results of 4-primer ARMS-PCR.Conclusions The ATP7B Arg778Leu gene mutation is a hot spot for the research of Chinese WD patients.4-primer ARMS-PCR is a fast convenient and accurate method for typing mutation in high throughput population screening.This approach can be used to detect other point mutations.
2.The protective effect of prenatal hypoxia adaptation on the brain of newborn rat

Objective To examine whether the prenatal hypoxia adaptation has any protective effect on the brain of the newborn rat Methods 12 22 day pregnant Wistar rats were randomly divided into 2 groups: the control group and the treated group In the treated group the pregnant rats were placed in a tightly closed hypoxia adaptation chamber When the O 2% in the chamber dropped to 15%, the rat was taken out to breathe fresh air for 5 min then put back in the chamber This process was repeated until its natural delivery In control the chamber was not tightly closed (O 2%=21%) 40 newborn rats weighing 6 8g were selected and subjected to brain ischemia and hypoxia, Left common carotid artrey was ligated under ether anesthesia 2h after recovery from surgery the newborn rats were placed in hypoxia chamber (T=36℃?1℃,O 2%=9%)for 1 5h 24h later they were sacrificed and brain was removed for microscopic examination (optical and electron) and flow cytometry measurement Results In the treated group most newborns were normal There were a few apoptosis cells in early stage The rate of apoptosis was 2 9%, necrosis cells could hardly be seen In the control group, although most neurons were also normal but there were apoptosis cells in early, middle and late stage and even necrosis cells The rate of apoptosis was 9 51%, which was significantly different from that in the treated group Conclusions Prenatal hypoxia adaptation has protective effect on the brain of newborn rat

6.Mechanisms underlying the promotion of woundhealing by bletilla carrying exogenousrecombinant human epidermal growth factor

BACKGROUND:How to promote wound healing is always the research focus of the surgical physicians in the clinic. Recombinant human epidermal growth factor (rhEGF) can effectively promote wound healing. However, as a biological agent, it is easy to be decomposed under normal temperature.
OBJECTIVE:Toexplorethe mechanisms underlying the promotion of wound healing in the back of rabbits by bletila carrying exogenous rhEGF.
METHODS:Model rabbitswith ful-thickness skin defects in the back were treated with bletila carrying exogenous rhEGF (combined treatment group), bletila, rhEGF, or saline (control group).
RESULTS AND CONCLUSION:The time of wound healing was the shortest (P< 0.05) and the wound healing rate was the highest (P< 0.05) in the combined treatmentgroup. On postoperative days 3 and 10, newly formed granulation tissue, capilaries, and colagenous fibersshowed by hematoxylin-eosin staining staining and Masson staining and the strongest immunoreactivity of vascular endothelial growth factor determined by immunohistochemical staining were found in the combined treatment group (P< 0.05). These findings confirm that bletila carrying exogenous rhEGF promotes wound healing by accelerating the forming of granulation tissue, new-born capilaries, and colagenous fibers, and the effects are superior to either of them alone.
7.Influence of Epstein-Barr virus infection on T cell subset in children with infectious mononucleosis

Objective To observe the influence of Epstein-Barr virus(EBV) infection on T cell subsets in children with infectious mononucleosis(IM).Methods Eighty children patients with IM in the Hanzhong Municipal People's Hospital from January 2013 to January 2016 were chosen as the study subjects and received the anti-virus therapy.The negative conversion rate of EBV-DNA after treatment was observed.The distributions of T cell subsets werecompared between the children patients with EBV-DNA positive and patients with EBV-DNA negative.The related factors of EBV-DNA non-negative conversion were analyzed.Results After treatment,67 children cases (83.75%) were EBV-DNA negative conversion.The proportions of CD3+ and CD8+ after treatment in the EBV-DNA negative group were lower than those in the EBV-DNA positive group,and the proportion of CD4+ and CD4+/CD8+ were higher than those in the EBV DNA positive group,the difference had statistical significance (P<0.05).In the analysis on the related factors in the children patients with EBV DNA non-negative conversion,the gender,WBC count and lymphocyte count had no significant correlation with EBV-DNA negative conversion(P> 0.05),while interferon use,age and initial Ct demonstrated significant correlation with EBV-DNA non-negative conversion(P<0.05).Conclusion EBV infection can lead to disturbance of T cell subsets in children with IM,so clinic should adopt an active anti-virus treatment.
8.Advances in research on liver-specific biologically active peptides

The liver is an important organ of the body,with an extraordinary capacity for responding to physical or chemical injuries by regenerating. The mechanisms behind liver regeneration are very complicated as hundreds of substances are involved.However,most of them are not liver-specific,such as HGF,EGF etc. We recently isolated a pure protein with hepatic stimulatory activity from the extract of a weanling calf liver and named its hepatopoietin Cn (HPPCn).This paper,with reference to our own work, mainly reviews the development and bioactivity about hepatic stimulator substance(HSS),augmenter of liver regeneration(ALR),hepassocin(HPS) and HPPCn,which are liver-specific biologically active peptides.

Organophosphate chemicals are widely used as agricultural pesticides and war reagents, their biodegradation is emphasized on the theoretical and practical aspects. Organophosphate hydrolases play important roles in the biodegradation of organophosphate chemicals. Great advancement was achieved recently in the determination of crystal structure and catalytic mechanisms of the hydrolase. This paper reviewed the research progresses in the enzymology, protein structure, catalytic mechanisms and application of the organophosphate hydrolase, and predicted the future research in this field.
10.Analysis the level of hepatocyte growth factor and related factors in patients with hypertension

Objective To observe the level of serum hepatocyte growth factor(HGF)in patients with hypertension(HT).Methods HGF,fasting plasma glucose(FBG),postprandial plasma glucose(2hPG),glycosylated hemoglobin A1c(HbA1c),total cholesterol(TC),triglyceride(TG),systolic blood pressure(SBP),diastolic blood pressure(DBP),body mass index(BMI)and waistline(w)were detected in 30 patients with hypertension,30 patients with hypertension complicated by type 2 diabetes mellitus(T2DM),and 30 normal controls such indexes as.Results The level of serum HGF in HT group[(413.87±90.87)ng/L],and HT+T2DM group[(413.72±98.72)ng/L]were higher than that in control group[(120.45±25.11)ng/L](P<0.05),but there was no significance between HT group and HT+T2DM group(P>0.05).There existed a positive correlation between HGF and FBG,SBP(r=2.273,r=5.353,P<0.05).Conclusion Higher levels of HGF is found in patients with hypertension and hypertensin complicated with T2DM,which indicates the severity of dysfunction of vascular endothelium.
