AIM:To study the mechanisms of effects of the ?-opioid receptor(?-OR) on neonatal rat hypertrophic myocardial cells induced by isoproterenol(ISO).METHODS:Myocardial cells of neonatal rats were cultured in vitro. The total protein content was detected by the Lowry's assay. The volume of myocardial cell was measured by computer photogragh analysis system and the protein synthesis was assayed with[3H] leucine intake method. The degree of extracellular signal-regulated kinase(ERK) phosphorylation was determined by western-blotting.RESULTS:U0126(an inhibitor of ERK) decreased the total protein content, the cell size,the[3H]leucine incorporation and the ERK phosphorylation degree in hypertrophic myocardial cells induced by ISO.1 ?mol/L U50,488H(a ?-opioid receptor antagonist) could decrease the total protein content, the cell size,the[3H]leucine incorporation and the degree of ERK phosphorylation. But these effects could be diminished when incubated with U0126.CONCLUSION:?-opioid can abolish the hypertrophic response induced by ISO of cardiac myocytes, which was associate with ERK pathway. The inhibitor of ERK significantly inhibits the ISO-induced hypertrophied effect.

Background and purpose:Connective tissue growth factor(CTGF) is a member of CCN family,it has been reported that CTGF involve in many biological processes and various pathological conditions. In our study, the correlation of CTGF expression and biological effects on breast cancer cell lines were investigated. Methods:The eukaryotic expression vectors containing CTGF open reading frame(ORF) pcDNA3.0/CTGF were constructed and transfected into breast cancer cell lines. The relationship between CTGF expression and breast cancer cell growth ability and proliferation, cell cycle distribution, apoptosis, cell motility and invasive capacity in vitro were observed. Results:Upregulation of CTGF expression in MCF-7 cell line could inhibit its growth ability and proliferation, increased the proportion of G0G1 phase cells, enhanced apoptosis and inhibited its invasive ability in vitro.Downregulation of CTGF expression in MDA-MB-231 cell line increased its growth ability and proliferation, decreased apoptosis and promoted its invasive ability in vitro. There were no differences of cell motility among different groups in MCF-7 and MDA-MB-231 cell lines. Conclusions:CTGF can inhibit breast cancer cell growth by increasing cell apoptosis and/or the proportion of cells in G0G1 phase.CTGF can also inhibit breast cancer cell invasive ability.

Objective To study the role of CXC chemokine receptor 5 (CXCR5) in the pathogenesis of bullous pemphigoid (BP) by detecting the expression of CXCR5 on follicular helper T (Tfh) cells in skin lesions of BP.Methods An immunohistochemical procedure was used to detect CXCR5 expression in biopsy specimens from skin lesions of 16 patients with BP and normal skin of 10 healthy individuals. Results CXCR5 was mainly expressed in the cytoplasm and membrane of Tfh cells in the basal cell layer of normal skin as well as in both basal cell layer and prickle cell layer of BP lesions. The number of CXCR5-positive ceils per high power field (× 400) was 11.16 ± 4.47 in normal skin specimens, significantly lower than that in lesional skin specimens (35.70 ± 12.20,t =6.07,P < 0.01).Conclusion CXCR5 may be involved in the pathogenesis of BP.

Objective To study the current situation of human brucellosis infection in a population at high risk in Qian'an,and to provide a scientific basis for prevention and control of the disease. Methods Towns with centralized residents working in sheep breeding, transporting, slaughtering and processing in Jianchangying,Muchangkou and Xiaguanying of Qian'an were selected. In each selected town,2-3 villages with relatively centralized households working in sheep farming, transportation and slaughtering were chosen. All of the people who contacted the sheep or their excrement were chosen as monitoring objects, and serological antibody was tested with rose Bengal plate test(RBPT) and serum agglutination test(SAT).Regional, gender, age and occupational distribution of brucellosis were analyzed. Results A total of 367 blood samples were tested,46 of them were positive in both RBPT and SAT with a ratio of 12.53% (46/367).Male positive rate [13.51% (30/222)] was slightly higher than that of females [11.03%(16/145)].The rate in Jianchangying was higher than that of other two towns with a ratio of 13.38%(40/299).The veterinary population had the highest ratio of 33.33%(1/3).Conclusions It is necessary to carry out the surveillance on brucellosis and to further strengthen communication with the animal husbandry department, and strengthen protection on key population. At the same time,in order to control the spread of the disease, extensive health education and intervention measures should be carried out.

BACKGROUND: Fibrin glue has been demonstrated to function as a kind of biomaterial with high quality. It has been used in nerve tissue engineering and proved to be a kind of scaffold for some cells.OBJECTIVE: To explore the biocompatibility of fibrin glue and olfactory ensheathing cells (OECs).DESIGN, TIME AND SETTING: An in vitro control trial based on cytology was performed at the Institute of Neurobiology,Nantong University from August 2007 to February 2008.MATERIALS: Fibrin glue was made of fibrin and catalyst, and OECs derived from rats' olfactory bulb were normally primary-cultured.METHODS: OECs were divided into control (OECs clone spheres were cultured alone) and in fibrin glue (OECs clone spheres were cultured and combined with fibrin glue) groups. After 1 week of culture, the proliferation of OECs were observed by convert microscope and detected by S-100 immunofluorescence histochemical staining.MAIN OUTCOME M EASURES: OECs morphology, cell count, the area of the cell bodies and the perimeter of the cell were determined.RESULTS: OECs could survive, migrate in fibrin glue, and float in the fibrin glue in the lower layer. After 7 days of incubation, cell body exhibited fusiform or triangle, predominantly bipolar or bipolar. The number of the S-100 positive cells was more, and cell bodies were larger in fibrin glue group than control group (P < 0.05). However, there was no obvious difference between two groups in cell perimeter (P > 0.05).CONCLUSION: Fibrin glue has good biocompatibility with OECs, and OECs can survive and migrate in fibrin glue.

Objective To investigate the effect ofYindanxinnaotong (YDXNT) capsule on learning and memory abilities, super oxide dismutase (SOD) activity and malondialdehyde (MDA) content in the hippocampus of rat models of vascular dementia (VD). Methods Animals were randomly divided into sham-operated group,VD vehicle group (Group VD) and YDXNT treatment group (Group VD+YDXNT); the 4-vessel occlusion was employed to establish the VD rat models in the Group VD and Group VD+YDXNT,while rats in the sham-operated group were not performed occlusion of the bilateral common carotid artery.Rats of the Group VD+YDXNT were given 1 mL liquid (9 granules of YDXNT into 360 mL normal saline) through intragastric administration 3 times daily for a consecutive 4 weeks; and rats of the other 2 groups were given the same volume of saline. The changes of learning and memory abilities were observed by Morris water maze test; the activity of SOD and the content of MDA in the hippocampus were measured. Results As compared with those in rats of the Group VD, the learning and memory abilities in rats of the Group VD+YDXNT were significantly improved (P<0.05):time in finding the platform was obviously decreased and swimming distance in finding the platform was obviously shortened in rats of Group VD+YDXNT.As compared with those in rats of the Group VD,the activity of SOD in the hippocampus was significantly increased and the content of MDA was obviously decreased in rats of the Group VD+YDXNT (P<0.05). Conclusion YDXNT could increase oxygen radical metabolism to improve the learning and memory abilities of rats with VD.
