1.Spatial autocorrelation analysis of brucellosis in Zibo of Shandong Province, 2013-2015

Objective To explore the spatial distribution and spatial clustering of brucellosis in Zibo,2013-2015.Methods Spatial autocorrelation analysis was used to analyze the surveillance data of brucellosis at town level.Township as a spatial analysis unit,spatial distribution of brucellosis in small scale in Zibo City was analyzed.Results The global Moran's I indexes of brucellosis were not significantly different in 2013-2015.According to local Moran's Ⅰ statistic,the high-high regions were Xindian Street and Jiangjunlu Street in 2013.The high-high regions were Tangfang Town,Jinshan Town,Luocun Town,Zhaili Town,Xihe Town and Longquan Town in 2014.The high-high region was Wangcun Town in 2015.Condusions Our study has showed that the spatial distribution of brucellosis is local clustered in Zibo.The detection of hotspots could provide guidance for formulating regional prevention and control strategies.

4.Clinical study on biocompatibility of four dialyzers with different membranes

Objective To elucidate the biocompatibility differences of 4 dialyzers with different membranes in maintenance hemodialysis (MHD)patients. Methods A total of 60 MHD patients were enrolled in the prospective,randomized,control,cohort study.In baseline,synthetic polysulfone(PS)membrane dialyzer was used in all the patients for at least 3 months.Then the patients were randomly divided into three groups:ployethersulfone(PES)membrane group,cellulose triacetate (CTA)membrane group,and synthetic polymethylmethacrylate(PMMA)membrane group.Study duration was 6 months.No dialyzer was reused.The biocompatibility markers were detected repeatedly at different time points and compared with each other in different dialyzer groups. Results The blood levels of high sensitive C reactive protein,interleukin-1β and interleukin-13 were not significantly different among different groups on every time point.However,the blood complements levels and WBC count were significantly different among four kinds of dialyzer.When the dialyzers changed from PS to PMMA membrane,C3a levels and WBC count changed significantly(P<0.05).Moreover,the change of C5a level was significantly different between PES group and PMMA group on month 3(P<0.05). Conclusion There are some differences of biocompatibiliy among different dialyzer membranes.
6.Systematic continuous sequence approach combined with three-dimensional ultrasonography in the diagnosis of fetal hand deformity

Objective To explore the value of systematic continuous sequence approach combined with two- and three-dimensional ultrasonography in screening of fetal hand deformity. Methods Systematic continuous sequence approach was performed with two- and three-dimensional u1trasonography in 28 541 cases to detect the fetal hand from January 2011 to December 2014 in the 105th Hospital of PLA. Prenatal ultrasonic diagnosis was compared with clinical delivery follow-up and pathology results of induced labour, then prenatal ultrasound features of various fetal hand deformities and the causes of missed diagnosis were analyzed. Results Thirty-four cases of fetal hand deformity were diagnosed out of 28 541 fetuses by prenatal ultrasonography (43 hands). In the 34 cases, there were 5 cases of cleft hand, 13 cases of ectrodactyly with fingers abnormal morphology, 3 cases of forearm and hand dysplasia, 7 cases of wrist or finger abnormal posture and 6 cases of hand absence of abnormal. Three missed cases included 1 case of polydacty, 1 case of middle phalanx and distal phalanx of the little thumb absence and 1 case of middle phalanx of little thumb absence. Hand deformity rate was 0.13%(37/28 541). The detection rate ofprenatal ultrasonography was 91.89%(34/37). The rate of hand deformity complicated deformity with one or more other organ was 52.94%(18/34). The rate of chromosome abnormalities was 13.51%(5/37). Cleft hand showed that fetal hand from the central longitudinal split into two halves. Ectrodactyly with fingers abnormal morphology showed that one or multiple fingers were absent combined with residual finger abnormal morphology. Forearm and hand dysplasia showed that the forearm was abnormally developed, the ulna and radius were short and the structure of the wrist disappeared. Wrist or finger abnormal posture showed that a hook-shaped wrist or half fist shaped hand, thumb adduction flexion, the index finger bending baroclinic on the dorsal of the middle finger and small finger bending baroclinic on the dorsal of the ring finger dorsal. Hand absence showed that no fetal hands. Conclusions Application of systematic continuous sequence approach combined with real time three-dimensional ultrasonography in the diagnosis of fetal hand deformity, such as ectrodactyly with fingers abnormal morphology and wrist or finger abnormal posture, can make up for the shortage of two-dimensional ultrasonography and obtain more diagnostic information.
7.Anti-tumor efficiency of cytotoxic T-lymphocyte induced by activated B lymphocyte after hepatocellular carcinoma alpha fetoprotein mRNA transfection

Objective To investigate the anti-tumor efficiency of cytotoxic T-lymphocyte (CTL) induced by activated B lymphocyte after hepatocellular carcinoma (HCC) alpha fetoprotein (AFP) mRNA transfection.Methods B lymphocytes were fractionated,purified and activated by recombinant human soluble sCD40L.PGEM4Z/AFP/A64-EGFP plasmid was established in vitro,mixed with polymerase T7RNA,and then transcribed into AFP mRNA with Poly (A) sequence.B lymphocytes electrotransfected with AFP mRNA were in the experimental group,B lymphocytes electrotransfected with GAPDH mRNA were in the negative control group,and untreated B lymphocytes were in the blank control group.The expressions of antigen-presenting cell (APC)markers (CD19,CD20,CD21,CD40,CD80,CD83) and major histocompatibility complex (MHC) of the 3 groups were detected.B lymphocytes of the 3 groups were cultured with T lymphocytes at ratios of 1∶40,1 ∶ 20,1∶10 and 1∶5 to induce and ampify CTL,and then the absorbance values were detected to evaluate the proliferation ability of T lymphocytes.The killing activity of CTL was investigated with HCC cell line SMMC7721 as the target cells.All data were analyzed using the paired t test,one-way analysis of variance or Tamhane's T2 test.Results The expressions of CD19,CD20,CD21,CD40,CD80 and CD83 of the experimental group were 74 ± 11,78 ±8,80 ± 10,90 ± 11,82 ± 6,56 ± 5,which were significantly higher than 51 ± 5,60 ± 7,53 ± 5,73 ± 8,50 ± 5 and 49 ± 6 of the negative control group,and 46 ± 3,54 ± 5,41 ± 3,56 ± 5,52 ± 6 and 21 ± 4 of the blank control group (t =5.302,4.812,7.627,5.932,9.142,7.813; 11.581,7.036,13.592,12.873,9.235,14.619,P < 0.01).The proliferation of CTL of the experimental group was significantly higher than that in the negative control group and blank control group (t =18.203,23.714,15.062,9.417 ; 16.833,19.392,13.871,6.592,P <0.01).When the T lymphocytes were mixed with the HCC cell line SMMC7721 at the ratios of 40∶ 1,20∶1and 10∶1,the killing rates of HCC cells by CTL of the exprimental group were 43% 4%,32% ± 4% and 22% ±3%,which were significantly higher than 15% ± 5%,7% ± 3% and 6% ±2% of the negative control group,and 7%±3%,8%±3% and 9%±4% of the blank control group (t =9.141,13.272,11.901; 14.372,12.835,9.507,P < 0.01).Conclusion Activated B lymphocytes after HCC AFP mRNA transfection may effectively induce CTL to kill HCC cells.
8.Epidemiological characteristics of natural focus and vector borne infectious diseases in Zibo City Shandong Province between 2004 and 2015

Objective To analyze the incidence trends and epidemiological characteristics of natural focus and vector borne infectious diseases in Zibo City between 2004 and 2015,and to provide evidence for control and prevention of the diseases.Method Surveillance data of natural focus and vector borne infectious diseases in Zibo City between 2004 and 2015 from the National Disease Reporting Information System were analyzed via descriptive epidemiological methods.Results A total of 10 natural foci and vector borne infectious diseases and 3 287 cases including 55 death cases were reported in Zibo City from 2004 to 2015.The average annual incidence was 6.24/100 000 and the case fatality rate was 1.67% (55/3 287).The incidence rate increased in recent years after the lowest rate in 2009 (2.83/100 000),and the rate in 2015 was 8.83/100 000.The top three diseases with higher incidence were hemorrhagic fever with renal syndrome (3.44/100 000),brucellosis (1.48/100 000) and tsutsugamushi fever (0.53/100 000).The top three high-prevalence areas with higher incidence were Yiyuan County (13.22/100 000),Zichuan District (9.73/100 000) and Boshan District (6.13/100 000).The cases mainly occurred from March to May and September,October,which accounting for 27.47% (903/3 287) and 24.64% (810/3 287),respectively of the total.The 40-59 year-oldage group was the highest,accounting for 48.13% (1 582/3 287)of the total.And 75.48% (2 481/3 287) of reported cases were farmers.Conclusions The overall incidence of the natural focus and vector borne infectious diseases in Zibo City has a annual increasing trend in recent years,while the incidence of brucellosis and tsutsugamushi fever have continued to rise since 2004.These diseases have showed a significant seasonal distribution and population distribution.It is necessary to conduct targeted prevention and control strategies.
9.Prevalence trend of neural tube defect in Zhengzhou city, 1996-2005.

OBJECTIVETo describe the prevalence trend and epidemiological characteristics of neural tube defects (NTDs) in perinatal in Zhengzhou city from 1996 to 2005.
METHODSData collected from hospital were used to depict the epidemiology of NTDs in Zhengzhou. All perinatal fetuses born in hospitals had an access within 7 days after delivery. The prevalence were calculated by perinatal'year, sex, birth area (urban versus rural) and maternal age. All monitored perinatal (162,074) accounted for 32.66% from totals (496,203).
RESULTSAll 238 cases were found NTDs, and the overall prevalence rate was 14.68/10,000. The annual prevalence rate presented a decreasing trend during that period. The rates in rural and urban area, in male and female birth were 29.28/10,000 and 9.63/10,000, 11.42/10,000 and 17.74/10,000 respectively. There were significant differences among maternal-age-specific prevalence rates (chi2 = 22.952, P = 0.000). The rates of <20 years group(53.76/10,000) and >35 years group(21.74/10,000) were higher than others.
CONCLUSIONThe prevalence rates of NTDs in rural area is higher than that in urban, female's is higher than male's in Zhengzhou. The annual prevalence rates of NTDs presents a decreasing trend in the past ten years.

10.Preliminary study on chemical constituents seperated from Cayratia japonica.

OBJECTIVETo study effective active constituents of Cayratia japonica,a genuine herbal medicine from Fujian.
METHODSuch chromatographic methods as Macroporous, Sephadex LH-20, ODS and normal phase silica gel column chromatography were adopted to separate the chemical components of C. japonica.
RESULTThirteen compounds were obtained, and their structures were identified by analyzing multiple spectral data as luteolin(1), apigenin(2), triethyl citrate-(3), 3-formylindole(4), esculetin(5), bis(2-ethylhexyl)-phthalate(6), calendin(7), ethyl-trans-3,4-dihydr-oxycinnamate(8), luteolin7-O-D-glucoside(9),5-hydroxy-3,4-dimethyl-5-pentyl-2(5H-furanone(10),ethyl-3,4-dihydroxybenzoate(11), eriodictyol(12) and daucosterol(13).
CONCLUSIONAmong them, compounds 3-8 and 10-12 were separated from the plant for the first time.
