1.Prevalence of Cardiovascular and Cerebrovascular Diseases in Army Old Male Hypertensive Patients in Beijing

Objective To investigate the prevalence of cardiovascular and cerebrovascular diseases in the army old male hypertensive patients in Beijing. Methods From October 2011 to April 2014, old male patients in the geriatric department of the General Hospital of Chi-nese People's Liberation Army were enrolled and categorized into 3 groups: normotensive, well-controlled hypertensive and poorly-con-trolled hypertensive groups. The prevalence of cardiovascular and cerebrovascular diseases was recorded and binary logistic regression was used to analyze the differences among the groups. Results 719 old male patients were enrolled, 228 in normotensive group, 389 in well-con-trolled hypertensive group and 102 in poorly-controlled hypertensive group. Compared to normotensive group, there was more incidence of angina (OR=2.547, P<0.001), myocardial infarction (OR=2.493, P=0.003), cerebral thrombosis (OR=3.438, P=0.003) and peripheral artery atherosclerotic disease (OR=1.766, P=0.026) in the well-controlled hypertensive group, and it was more of angina (OR=4.123, P<0.001), la-cunar infarction (OR=2.810, P=0.020), cerebral thrombosis (OR=3.484, P=0.001) and peripheral artery atherosclerotic disease (OR=3.773, P<0.001) in the poorly-controlled hypertensive group. There was not significant difference of prevalence of cerebral hemorrhage (0.9%, 1%and 1%, respectively). Conclusion In the army old male hypertensive patients in Beijing, control rate of blood pressure was higher than com-mon population, but the prevalence of ischemic diseases was still higher than in the normotensive patients. Anti-hypertension and control of other risk factors could reduce the mobidity of cerebral haemorrahge.

3.Prevalence of cardiovascular disease in subjects hospitalized due to chronic obstructive pulmonary disease in Beijing from 2000 to 2010

Objectives To investigate the overall prevalence of cardiovascular disease (CVD) in subjects hospitalized for chronic obstructive pulmonary disease (COPD),and explore the prevalence of the major CVD complications and trends in patients with COPD over a 10-year period.Methods Medical records in the PLA General Hospital,Beijing Union Medical College Hospital,and Beijing Hospital from 2000/01/01 to 2010/03/03 were retrospectively reviewed.A total of 4960 patients with COPD were reviewed in the study (3570 males,mean age,72.2 ± 10.5 years; 1390 females,mean age,72.0 ± 10.4 years).Results The prevalence of CVD in COPD patients was 51.7%.The three most prevalent CVDs were ischemic heart disease (28.9%),heart failure (19.6%),and arrhythmia (12.6%).During the 10-year study period,the prevalence of various CVDs in COPD patients showed a gradual increasing trend with increasing age.There was higher morbidity due to ischemic heart disease (P < 0.01) in male COPD patients than in the female counterparts.However,heart failure (P < 0.01)and hypertension (P < 0.01) occurred less frequently in male COPD patients than in female COPD patients.Furthermore,the prevalence of ischemic heart disease decreased year by year.In addition to heart failure,various types of CVD complications in COPD patients tended to occur in younger subjects.The prevalence of all major types of CVD in women tended to increase year by year.Conclusions The prevalence of CVD in patients hospitalized for COPD in Beijing was high.Age,sex and CVD trends,as well as life style changes,should be considered when prevention and control strategies are formulated.
5.Research of dietary intervention on self-efficacy and metabolic control of patients with type 2 diabetes

Objective To evaluate effects of dietary intervention on self-efficacy and metabolic control of patient with type 2 diabetes. Methods A self-control designed research was conducted to 70 patients with type 2 diabetes at clinic receiving diet intervention. Before intervention and 1 month after, body mass index,waist, total cholesterol, high density lipoproteins, low density lipoproteins and uric acid were tested to evaluate the metabolic control. After 3 month, HbA1c was monitored to evaluate glycemic control. Meanwhile,before intervention and 1,3 month after, self-efficacy was investigated. Results One month later, body mass index, waist, blood lipid were lower than before, HbA1c was reduced 0.78%, the total score of self-efficacy and the dimension of nutrition, monitoring and medical were improved. Conclusions The dietary intervention was beneficial for improving metobolic control and self-efficacy of patients.
6.Analysis of salt resistance on the poplar transferred with salt tolerance gene.

Poplar(Pupulus x Xiao zhannica, cv. "balizhuang-yang") transferred with mtl-D gene was used as an experimental material. Through tissue culture, aquaculture and pot culture transgenic poplars are tested with variant Nacl salt gradient. In it transgenic poplar raises its initial days for differentiation, differentiation rate, bud intensity, bud height and growth potential than control plant. At the same time, the transgenic poplar has higher rooting rate and more top roots, side roots and higher root length than control under the same salt stress. The result shows that the transgenic poplar has better tolerance ability than control plant in the salt 4@1000 contensity mediums.

7.Effect of Guanmaitong Tablet on ERK and p38 Protein of TLR2 Pathway Expression in Cerebral Ischemia/Reperfusion Rats: an Experimental Study.

OBJECTIVETo explore the inflammatory cascade mechanism through Toll like receptor 2 (TLR2) pathway after cerebral ischemia/reperfusion, and to study molecular mechanisms of Guanmaitong (GMT) Tablet for protecting brain damage.
METHODSWe used bolt-line method to block/release the middle cerebral artery, causing cerebral ischemia/reperfusion (I/R) injury model. GMT Tablet was given by gastrogavage. Rats were then divided into the high dose GMT group (1200 mg/kg), the middle dose GMT group (600 mg/kg), the low dose GMT group (300 mg/kg), the positive control group (Tanakan, 20 mg/kg). Their right brain tissues were fixed in 10% neutral formalin. TLR2 expressions were detected by immunofluorescence staining. The total protein was extracted from right brain tissues by ultrasonica- tion. Expression levels of extracellular regulated protein kinases (ERK), phospho-extracellular regulated protein kinases (p-ERK), p38-mitogen activated protein kinases (p-ERK), phospho-p38-mitogen activated protein kinases [p-p38-MAPKs(p-p38)] were assessed by Western blot. Abdominal aortic blood was withdrawn. IL-6 and IL-1β levels were detected by ELISA in brain tissues and serum.
RESULTSCompared with the sham-oepration group, expression levels of TLR2, ERK, p-ERK, p38, p-p38 protein were up-regulated (P < 0.05, P < 0.01), and contents of IL-6 and IL-1β in brain tissues and serum were increased in the model group (P < 0.01). Expression levels of TLR2, ERK, p-ERK, p38, p-p38 were down-regulated (P < 0.05, P < 0.01), and contents of IL-6 and IL-1β were reduced in brain tissues and serum in middle and high dose GMT groups (P < 0.05, P < 0.01).
CONCLUSIONSTLR2 pathway was involved in cerebral I/R injury. GMT protected neurons by down-regulating protein expressions of TLR2, ERK, p-ERK, p38, p-p38 and contents of IL-1β and IL-6.

8.Alterations of DPC4/SMAD4/MADH4 gene detected in paraffin-embedded tissues of human pancreatic carcinomas.

OBJECTIVETo demonstrate the alterations of DPC4/SMAD4/MADH4 gene in paraffin-embedded tissues of pancreatic carcinomas.
METHODSForty-six cases of resected specimens containing carcinomatous tissue and normal pancreatic tissue were analysed for possible DPC4 gene mutations by polymerase chain reaction (PCR)and single-strand conformation polymorphism (SSCP). The DNA sequencing technique was applied to determine the patterns of gene mutation in the PCR-SSCP positive cases. Fifty-six cases of pancreatic carcinoma along with the specimens corresponding normal pancreatic tissues were studied by in situ hybridization (ISH) and immunohistochemistry (IHC) techniques for gene expression in mRNA and protein level.
RESULTSThe homozygous deletion rate of exon 1, 2, 3, 4, 8, 11 of DPC4 gene in pancreatic carcinoma was 28.26%, while the mutation rate of DPC4 gene was 21.74%. In these tumors, there were 3 cases of nonsense mutation, 5 cases of missense mutation, 1 case of deletion and missense mutation, 1 case of insertion mutation. Positive rates of SMAD4 in carcinomatous tissues detected by the ISH and IHC were 53.57% and 58.93% respectively, whereas they were 91.07% and 89.29% in the matched normal tissue respectively. There were significant difference between cancer and normal tissue (P < 0.05). Thrity-two cases were positive of DPC4/SMAD4 with all methods mentioned above, the coincident rate was 87.50% (28/32). The coincidence between gene detection and ISH of SMAD4 was 87.50%, and it was 96.88% between gene detection and IHC of SMAD4. Of all 56 cases, the coincidence of the positive rates of SMAD4 detected by ISH and IHC was 91.07%. No significant difference among the positive rates of DPC4/SMAD4 as detected by the three different techniques (P > 0.05).
CONCLUSIONSThe main mechanisms of inactivation of DPC4 gene in pancreatic carcinoma are homozygous deletion and mutation. The product of DPC4 expression is significantly decreased in cancer group compared with the normal tissues. As a tumor suppressor gene, DPC4 alteration is an important molecular event in pancreatic carcinoma, and probably plays a crucial role in cancer development and progression.

10.Experimental research on spontaneous benign prostatic hyperplasia in old dogs.

OBJECTIVESTo investigate the pathoanatomize histological and biochemical characteristics of benign prostatic hyperplasia (BPH) by use of old dogs with spontaneous BPH as animal models.
METHODSOld dogs aged 6 to 13 years were recruited after anus check, B-ultrasonic examination by recta spy and measurement under surgical exploration. Ten dogs with notable prostatic hyperplasia were used as models, and 6 with non-hyperplasia prostate as control. Serum testosterone (T), estrogen (E2), ACP and prostatic specific antigen (PSA) were analyzed, and prostates were checked histologically.
RESULTSProstate volume of the BPH group was significantly bigger than those of the control group, (14.7 +/- 2.3) and (13.8 +/- 1.9) cm3 vs (8.4 +/- 1.0) and (8.4 +/- 1.9) cm3, P < 0.01. Serum T [(14.3 +/- 2.9) vs (16.4 +/- 4.0) nmol/L] and E2 [(137.6 +/- 70.8) vs (164.4 +/- 82.0) pmol/L] were not different between the two groups (P > 0.05). ACP of the BPH group was higher than that of the control group [(6.63 +/- 2.76) vs (4.92 +/- 2.19) U/L], but the difference was not statistically significant (P > 0.05). There was significant difference between the BPH group and the control group in PSA level [(5.6 +/- 0.78) vs (3.1 +/- 0.54) microgram/L, P < 0.01]. The tissue slides of the BPH prostates showed hyperplasia with raised height of epithelium, and many long and offsetting mammillae in the gland cavity due to epithelium hyperplasia.
CONCLUSIONOld dogs with spontaneous BPH are useful animal models for the etiological and pharmacological researches of human BPH.
