Objective To investigate metabolic characteristics and insulin resistance in newly diagnosed type 2 diabetes pa-tients with nonalcoholic fatty liver disease .Methods Two hundred patients with newly diagnosed type 2 diabetes mellitus including 140 with nonalcoholic fatty liver disease (NAFLD) (NAFLD group) and 60 without NAFLD (non-NAFLD group) were recruited. Metabolic characteristic were measured and recorded .homeostasis model assessment insulin resistance ( HOMA-IR) index and HOMA-C were used to assess insulin resistance .Results were compared between two groups .Results Compared with non-NAFLD group, body mass index (BMI), fasting blood glucose (FBG),alanine aminotransferase (ALT),gamma-glutamyl transferase (GGT), uric acid ( UA) , triglyceride ( TG) , low density lipoprotein cholesterol ( LDL-C) ,fasting insulin ( FINS) , fasting C peptide ( FCP) , gly-cated hemoglobin a1c (HbA1c), HOMA-IR, and HOMA-C were significantly higher in the NAFLD group ( P <0.05).Logistics re-gression analysis showed that BMI , TG, GGT, and UA were risk factots for NAFLD ( P <0.05, OR =1.82, 1.53, 1.37, and 1.09 ) .Conclusions Compared patients without NAFLD , newly diagnosed type 2 diabetes patients with NAFLD present more meta-bolic disturbance, and severely insulin resistance.Obesity and the increased level of postprandial UA , triglyceridemia, and GGT might increase the risk of NAFLD in the patients with newly diagnosed type 2 diabetes mellitus .

Objective To investigate the cytoprotection of curcumin against rotenone(Ro)-induced injury and the molecular mechanisms underlying in PC12 cells.Methods The insulted model of PC12 cells was established with Ro.Cell viability was determined using MTT reduction assay.The content of reactive oxygen species(ROS)was determined by the method of DCFH-DA staining.Chromatometry was used to measure the total activity of SOD,DCFH-DA staining to measure the level of intracellular ROS,and flow cytometry to assay the apoptosis rate.Results 0.5 and 1.0 μmol/L curcumin significantly decreased the inhibitory rate of Ro on the growth of PC12 cells for 24 h as compared with the Ro group(P＜0.01).0.5 and 1.0 μmol/L curcumin significantly ameliorated the changes in morphology of PC12 cells,increased the activities of intracellular SOD as compared with the Ro group(P＜0.05),decreased the production of intracellular ROS and inhibited the apoptosis of PC12 cells induced by Ro for 24 h as compared with Ro group(P＜0.01).Conclusion Curcumin can resist Ro-induced cytotoxicity probably by the mechanism of scavenging intracellular ROS and increasing the activity of antioxidase.

Methods Sixty aged patients with VBI were selected as experimental group for testing of visuo-oculomotor system reaction, spontaneous nystagmus, caloric test and positional nystagmus. Objective To investigate the inhibitory effects of exogenous WAF 1 S gene on human laryngeal cancer Hep 2 cell line, and to explore the potential use of WAF 1 S in gene therapy for laryngeal cancer. Methods A eukaryotic expression vector containing 2 1kb human full length WAF 1 S cDNA was transfected into human laryngeal cancer Hep 2 cell line by using lipofectamine. Expression of exogenous WAF 1 S gene was detected by dot blot hybridization. By using Western blot and confocal microscope, expression of p21 protein was quantitatively analyzed in situ . The growth state of transfected Hep 2 cell was determined by flow cytometry and MTT. Results It was found by dot blot hybridization that WAF 1 S gene could express in Hep 2 cell. The expression of the exogenous p21 gene in Hep 2 cells was markedly higher than that in the control group. It was confirmed with flow cytometry that WAF 1 S gene could induce apoptosis of laryngeal cancer Hep 2 cell line, and the progression of cell cycle was arrested at G 1 phase. Conclusion Laryngeal cancer cells could be arrested at G 1 /S phase and the growth of the cells could be significantly suppressed by exogenous WAF 1 S gene

Background and purpose: Peritoneal metastasis of gastric cancer is mainly discovered in the ad-vanced cancer. Nonetheless, the clinical applicability of each tumor biomarker in peritoneal metastasis of gastric cancer is still ambiguous. Therefore, this study investigated the diagnostic value and clinical significance of CEA, CA125 and CA72-4 in gastric carcinoma patients with peritoneal metastases. Methods: A total of 108 gastric carcinoma patients with peritoneal metastases from Jan. 2008 to Dec. 2013 were studied. All patients were diagnosed by imaging, operations and pathological examination, and also received intravenous or intraperitoneal chemotherapy. Serum tumor markers such as CEA, CA125 and CA72-4 were determined during diagnosis and before each chemotherapy. The diagnostic sensitivity of single marker and combined detection with 2 or 3 markers were analyzed. The correlations among the serum tumor markers and clinical pathological factors, chemotherapeutic effects and survival time were analyzed. Results: Positive rates of CEA, CA125 and CA72-4 were 20.4%, 46.3% and 45.4% in gastric cancer patients with peritoneal metastases, respectively. For these patients, the positive rates of CEA/CA125, CEA/CA72-4, CA125/CA72-4 and CEA/CA125/CA72-4 were 54.7%, 52.8%, 69.5% and 79.6%, respectively. The combined detection of 3 tumor markers was much better than single marker detection (P<0.05). Positive rates of CEA, CA125 and CA72-4 were correlated with the ECOG scale (P<0.05). Positive rate of CA125 was associated with ascites (P<0.001), while positive rate of CA72-4 was associated with ovarian metastasis (P<0.05). Median survival time of patients with positive rates of CEA, CA125 and CA72-4 was significantly lower than that of the patients with normal levels of these markers (P<0.05). Compared with pre-treatment, the levels of all three tumor markers significantly declined after three cycles of chemo-therapy (P<0.05). The decline in CA125 level after chemotherapy was significantly correlated with decreased amount of ascites (P<0.05). The tumor markers turned negative after 3 cycles chemotherapy in patients with positive markers upon initial diagnosis, their survival was significantly prolonged (P<0.001). Conclusion: Combined detection of serum CEA, CA125 and CA72-4 can significantly promote diagnostic rate of gastric cancer with peritoneal metastasis, and may be helpful in evaluating chemotherapeutic effects and predicting prognosis.

Background and purpose:The cinobufacini injection is a traditional antitumor drug.However,its mechanism iS still unclear.The purpose of this study was to observe the effect of cinobufacini injections in DNA TOPO Ⅰ of human hepatocellular carcinoma HcpG-2 cells.Methods:The cells that were proliferated were assessed by MTT assay.Cell cycles were shown through FCM.TOPO Ⅰ mRNA expression was analyzed through RT-PCR.The activity of TOPO Ⅰ was measured by TOPO Ⅰ mediated super coiled PHR322 relaxation.Supercoiled PBR322 was also used to determine the direct DNA breakages.Results:Cinobufacini injections significantly inhibited HepG-2 cells proliferation in ways that were dependent on dosages and time.Induced tumor cells arrest at the S-phase.TOPO ⅠmRNA expression decreased in a manner that was dependent on dosages which inhibited the TOPO Ⅰ mediated DNA relaxations.However,the cinobufacini injections could not directly induce DNA breakage at any concentration.Conclusion:Cinobufacini injections can inhibit human hepatocellular carcinoma HepG-2 cells proliferation.The regulation of topoisomerase Ⅰ activity and mRNA expression may be one of the mechanisms that causes the cinobufacini injection to contribute against tumor.
